The present study focused on changes in the C-reactive protein (CRP) and adrenomedullin (ADM) levels in the liver and gill tissues of Capoeta umbla as biomarkers of environmental pollution and ecosystem health in inland waters of Tunceli (Turkey). The CRP levels in liver and gills were not significantly changed in March and September at every site (p>0.05) except for site 3 in gill tissues (p<0.05). The results of statistical analysis indicate that significant differences in CRP levels were found in liver and gills among sites (p<0.05), but no changes in the gills at March (p>0.05). In March, ADM levels in gills and liver were statistically changed among sites (p<0.05), while no statistical changes were determined in September (p>0.05). The results of this study indicated that CRP levels in the tissues of the C. umbla could be utilized as a significant bioindicator for disease pathogenesis in fish infections.

Immunological biomarkers reflect the effects of exposure to environmental contaminants. In this study, the suitability and sensitivity of cytokine responses, interleukin 1 beta (IL-1 beta), interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-alpha) in gill tissues of Capoeta umbla (Heckel, 1843), collected from different regions, as early warning indices of environmental pollution and ecosystem health was evaluated. Fish and water samples were taken from ten stations in March and September 2011 and 2012. Tumor necrosis factor-alpha, IL-1 beta and IL-6 levels were determined in samples of the gill tissues by using an ELISA kit. Significant variations of TNF-alpha, IL-1 beta and IL-6 levels observed between stations and seasons. The results of this study show that seasonal variations of cytokine responses in gills of Capoeta umbla are sensitive to the contaminants present in Uzuncayir Dam Lake (Tunceli, Turkey) water and are valuable biomarkers for environmental pollution and ecosystem health.

The aim of this work was to determine the seasonal changes in the activities of selected biomarkers in Capoeta umbla (Heckel, 1843) caught from Uzuncayir Dam Lake (Tunceli, Turkey) and to evaluate the effects of environmental factors on these activities. Fish were sampled on seasonal basis, and superoxide dismutase, catalase, glutathione peroxidase activities and levels of glutathione and malondialdehyde in gills were determined. Significant variations of oxidative stress biomarkers were observed between seasons and sites. The results of this study show that seasonal variations of oxidative stress responses and lipid peroxidation in gills of C. umbla are sensitive to the contaminants present in water of Uzuncayir Dam Lake and selected parameters are in valuable biomarkers for monitoring of water systems, since they give an early warning signal of effects of xenobiotics on aquatic organisms at molecular levels which help to prevent their effects at organismal level. The discharge of textile effluents into the environment without appropriate treatment poses a serious threat for the aquatic organisms. The present study was undertaken to investigate biochemical response of crayfish Astacus leptodactylus exposed to textile wastewater (TW) treated by indigenous white rot fungus Coriolus versicolor. Glutathione S-transferase (GST), cytochrome P450 1A1 (CYP1A1), and acetylcholinesterase (AchE) levels in hepatopancreas and abdomen tissues of crayfish exposed to untreated, treated, and diluted rates (1/10) in both TW during 24 and 96 h were tested. Physiochemical parameters (electrical conductivity (EC), chemical oxygen demand (COD), pH, and total dissolved solid (TDS)) of TW were determined before and after treatment. Physiochemical parameters of TW decreased after treatment. The GST activity and AchE were generally increased, but CYP1A1 activity was decreased in hepatopancreas tissue of crayfish exposed to different kinds of untreated TW. After treatment by indigenous white rot fungus (C. versicolor), GST and CYP1A1 activities were returned to control values, while AchE activities were increasing further. In this study, only GST and CYP1A1 activities of A. leptodactylus confirmed the efficiency of TW treatment with C. versicolor.

- MakaleYalnız künye2014Scopus SJRQ3Assessment of immunomodulator biomarkers (Tnf-α, Il-1β and Il-6) in liver of Capoeta umbla for biomonitoring of pollution in Uzuncayir Dam Lake (Tunceli, Turkey)
Yildirim, N. C.; Danabas, S.
This study was aimed to monitor water pullution of Uzuncayir Dam Lake using the changes of IL-6, IL1 beta and TNF-alpha levels in Capoeta umbla (Heckel, 1843) liver tissue at ten stations in March and September 2011. In this study C. umbla (Heckel, 1843) was used as the indicator organism. Tumor necrosis factor-alpha (TNF-alpha), interlekuin1 beta (IL-1 beta) and interleukin-6 (IL-6) levels were determined in samples of the liver tissue by ELISA kit. The lowest mean IL-1 beta levels were found at station 6. The mean IL-1 beta was reached its maximum level at station 2. The difference between the mean levels of IL-6 was found to be significant (p<0.05) amoung stations. The IL-6 levels were significantly increased in September at stations 1, 2, 7 and 8 (p<0.01) compared to the values in March. The mean levels of TNF-alpha were found to be significant (p<0.05) amoung stations. The TNF-alpha levels significantly decreased in September at stations 1 and 9 (p<0.01). TNF-alpha, IL-1 beta and IL-6 levels in C. umbla can be used as early diagnostic indicators against adverse environmental events and useful and reliable bioindicators in determining the pollution of the aquatic ecosystem.
