Korkmaz, Volkan; Yildirim, Numan; Erguven, Gokhan Onder; Durmus, Barbaros; Nuhoglu, Yasar

In this study, the bioremediation capacity of three isolates of Bacillus aryabhattai, Pseudomonas azotoformans and Sphingomonas pseudosanguinis and their consortia at glyphosate added soil medium investigated via the reduction of chemical oxygen demand (COD) and total organic carbon (TOC) parameters in filtrate waters obtained from a designed bioremediation setup. Additionally, mortality assays were performed on the filtrate waters with model organism Daphnia magna. According to the results of the experiments, at the end of the 11 days, the highest COD reduction rate seen in media with S. pseudosanguinis as 92.1% while TOC rate was 69.13% in consortia media. While the mortality rate was 10, 10 and 5% on B, P and S application groups at the end of the 72th respectively, these rates were 100% in control group at the end of the 48th. According to the results obtained from bioremediation and mortality assessment, these bacteria can reach the high bioremediation rate one by one and in consortia media.

Investigation on Microbial Biodeterioration of the Stone Monuments in Yildiz Technical University—Yildiz Campus—Istanbul—Turkey

Özdemir, Aysun; Erguven, Gokhan Onder; Adar, Elanur; Nuhoglu, Yasar

The stone samples of historical monuments around Yıldız Technical University Besiktas Campus were investigated using DNA extraction-PCR-DGGE methods, scanning electron microscopy (SEM), XRF, and other analytical methods to assess stone decay over the centuries. Microbial diversity was examined by classical cultivation and modern diagnostic methods besides modern analysis techniques. The number of the microorganisms in per gram of stone samples was calculated by microbial culture methods. SEM analysis showed that stone surfaces have too many pores, decaying pieces and microbial colony. It is put forth by XRF analysis that stone materials have some elements serving the growth of microorganisms. It was concluded that there is a close connection the stone structure and microbial growth, most likely mineralogical composition, hardness and porosity of stone. Cyanobacterial microorganisms lived on stone surfaces were also determined using denaturing gradient gel electrophoresis (DGGE) of PCR-amplified 16S rRNA gene fragments. It was revealed DNA-based molecular analysis of 16S rRNA that 23 bacterial/Cyanobacterial clones were inhabited to stone materials.

Bioremediation of fluazifop-p-butyl herbicide by some soil bacteria isolated from various regions of Turkey in an artificial agricultural field

    Erguven, Gokhan Onder; Nuhoglu, Yasar

The bioremediation rate of fluazifop-p-butyl (C19H20F3NO4) was monitored. Bacteria were isolated in agricultural soil samples. Fifteen sterilised glass jars were inoculated with 2, 5, 10, 20 cm3 of a homogenised bacterial mixture (109 CFU/cm3), then sterile agricultural soil and 60 µg of fluazifop-p-butyl (in liquid form) were added to each jar. Each week, filtrated water drained from bottles was analysed for fluazifop-p-butyl concentration, chemical oxygen demand (COD), biochemical oxygen demand (BOD5) and total organic carbon (TOC). Additionally, pH and dissolved oxygen concentration were monitored. The highest biodegradation rate was observed in the soil sample containing 20 cm3 of the culture media. In this media, fluazifop-p-butyl, COD, BOD5 and TOC removals were measured as 91, 83, 96 and 86%, respectively, at the end of the 2 months. The DO level was measured between 3 and 6 mg O2/dm3 in the first month for all cultures. An increase of pH was recorded during the first month and after this time a pH decrease was noted.
