Schiff bases and their metal complexes have antiviral, antimicrobial, antitumoral, radical scavenging and antioxidant activities. In this study, effects of Schiff base ligand and its metal complexes on the fatty acids and lyphophlilic vitamins of the liver of rats were investigated. The fatty acids of the liver were determined by GC and lyphophlilic vitamins of the liver determined by HPLC. The amounts of stearic acid (18:0), oleic acid (18:1) and monounsaturated fatty acids (MUFA) considerably decreased in the Mn, Ni and Zn complexes groups, whereas the amount of linolenic acid (18:3) and eicosatrienoic acid (20:3) considerably increased in the Ni an Zn complexes groups compared to the control group (P<0.01). However, there was no significant difference (P>0.05) in the amounts of arachidonic acid (20:4), docosahexaenoic acid (22:6), total saturated fatty acids (SFA) and polyunsaturated fatty acids (PUFA), total omega-3 and omega-6 fatty acids between groups. The amounts of vitamin K-1, vitamin D-2 considerably increased (P<0.05) in the Mn and Ni complexes groups as compared to the control. It was observed that the amount of a-tocopherol increased both in the Ni and Co complexes groups when compared to the control group but this increase was parallel to the amount in the same groups (P<0.05). As a result, it can be think that with the application of toxic metals such as manganese, cupper, nickel, cobalt, and zinc unsaturated fatty acids affected the activities of enzymes which are in the liver tissue and in charge in the fatty acid chain elongation.

THE EFFECTS OF HYDROXYUREA DERIVATIVE 1, 3, 4-THIADIAZOLES ON SERUM BIOCHEMICAL PARAMETERS AND ANTIOXIDANT STATUS IN LIVER OF RATS

Karagozoglu, Yusuf; Türkoglu, Semra; Parlak, Akif E.; Alayunt, Naci O.

In this study, it was aimed to investigate the antioxidant and antihepatotoxic effect of hydoxyurea derivative 1,3,4-thiadiazoles on serum biochemical parameters (AST, ALT, LDH, urea, creatinine and total bilirubin) and antioxidant parameters (SOD, CAT, GPX, MDA). In our study, a total of 35 adult male wistar rats were divided into 5 equal groups. DMSO diluted with only corn oil, was injected to the control group. 25 mg/kg ligand, 25 mg/kg thiadiazole-manganese, 25 mg/kg thiadiazole-cadmium and 25 mg/kg thiadiazole-chrome complexes was injected to the experimental groups rats subcutaneously for 15 days with three-day interval during the test. After blood samples were centrifuged AST, ALT, LDH enzyme activites hepatic injury; and then, the activites of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), and the level of malondialdehyde (MDA) in liver tissues were determined spectrophotometrically. The level of AST was found to be lower (p<0.05) in the Cr-L group than in other groups. The levels of ALT and total bilirubine were found to be lower (p<0.05) in the Cd-L group than in other groups. The level of SOD was found to be higher (p<0.05) in the Cd-L group than in other groups. The levels of GPX was found to be lower (p<0.05) in the Cr-L group than in other groups. As a result, it can be concluded that their pharmacological characteristics can be beneficial in many fields of application because hydroxyurea derivative 1,3,4 thiadiazole compounds show the antioxidant and antihepatotoxic activity.
