The current study focused on the ameliorating impact of TRP (5 mM) on Salmo abanticus spermatozoa exposed to sub‑lethal concentrations of GLY (2.5, 5, and 10 mg/L). Sperm quality parameters, oxidative stress indices, and DNA damage were analyzed in the post‑exposure sperm. Our data reveal that GLY has an adverse effect on motility rate and duration, sperm kinetics parameters, antioxidant status and DNA damage, and triggers antioxidant defense mechanisms at the highest dose (10 mg/L) and it has insufficient therapeutic effect. Overall, GLY poses potential hazards to male reproductive cells and may adversely affect subsequent generations of natural populations.

Spermatologic characteristics and sperm motility alterations caused by short-term copper exposure in Brook Trout Salvelinus fontinalis

Kutluyer, Filiz; Kocabaş, Mehmet; Başçınar, Nadir

Spermatozoa movement in many aquatic animals with external fertilization is directly related to the external aqueous environment. In particular, environmental toxic metals can cause harmful effects and impair sperm cell functions. The high concentrations of copper (Cu) may be harmful for aquatic animals, while low levels are required for metabolic interactions. Herein, we investigated the effect of different copper concentration [0 mg L−1 (Control),.5 mg L−1, 1 mg L–1, and 2 mg L−1] on spermatozoa of brook trout Salvelinus fontinalis. Motility and survival of sperm cells were assessed. Our results indicated that Cu exposure caused to an evident decrease in motility and longevity of spermatozoa with increasing concentrations (p <.05). Overall, we showed that Cu exposure can impair sperm motility of S. fontinalis. Our study suggested that sperm motility has the potential to be a sensitive bioindicator of freshwater environmental pollution.

Metal bioaccumulation in selected tissues of barb (Barbus sp.) and common carp (Cyprinus carpio, Linnaeus 1758) from the Keban Dam Lake, Turkey

The aquatic life has been negatively influenced by harmful effects of environmental toxic elements, including heavy metals. The elevated concentrations of metals may be harmful for aquatic animals and human health. Herein, the present study deals with assessment of the bioavailability of metals [cadmium (Cd), chrome (Cr), copper (Cu), zinc (Zn), iron (Fe), lead (Pb), manganese (Mn), mercury (Hg), nickel (Ni), and selenium (Se)] in organs such as muscle, gills, liver, kidney, and gonad of the commercially consumed Barbus sp. and Cyprinus carpio (Linnaeus, 1758) collected from Keban Dam Lake, eastern Turkey. The fish were captured from four different stations of Keban Dam Lake. Our results indicated that Fe and Zn concentrations were the highest in all tissues of two fish species while Cd concentrations were the lowest levels. In addition, detected metals accumulated in the muscle of common carp at higher levels compared to Barbus sp. The results from the present study also demonstrated significant variations and considerable differences in the concentrations of all 10 elements between the four stations and different fish tissues. Overall, the concentrations of detected metals in the muscle were many folds higher than the permissible level prescribed by some national and international legislation, which raises health risk concerns.

Effect of the organophosphate insecticide chlorpyrifos exposure on oxidative stress and quality of Salmo coruhensis spermatozoa

Kutluyer, Filiz; Kocabaş, Mehmet; Erişir, Mine; Benzer, Fulya

The use of pesticides has been increasing along with increasing farming activities and has caused deleterious environmental impacts. Non-target organisms in particular, including fish, are affected by pesticides. In this work, the impacts of Chlorpyrifos (CPF) on sperm oxidative stress markers and sperm motility were investigated in vitro. CPF concentrations were 0 μg/L (control), 5 μg/L, 10 μg/L and 15 μg/L. Lipid peroxidation [malondialdehyde (MDA)], nonenzymatic antioxidants [glutathione (GSH)] and enzymatic [superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT)] activities in sperm cells were examined for the determination of oxidative stress status. Our findings showed that motility and survival of sperm cells significantly decreased with exposure to Chlorpyrifos. Biochemical assays revealed that CAT activity and levels of MDA and, GSH increased in spermatozoa based on CPF concentration while activity of GSH-Px and SOD decreased. Consequently, spermatozoa were highly sensitive to CPF exposure. It can be deduced that CPF has the potential to disrupt sperm quality and to cause to oxidative stress in sperm cells of S. coruhensis.
