Tatar, Sule; Okuduci, Ebru
In this study, the accumulation of some heavy metals in water, sediment, and Typha latifolia plant species was investigated in a previously operated mining area located in the Hozat region of Tunceli. In the first phase of the study, water and soil samples were collected from sites where T. latifolia was dominant. The metal uptake capacities from soil to roots, stems, and leaves of the plants were examined for different metals as silver (Ag), barium (Ba), cadmium (Cd), copper (Cu), nickel (Ni), lead (Pb), strontium (Sr), zinc (Zn). Water, sediment, and plant samples collected from the field were analyzed for the above elements at ultra-low detection limits using ICP-MS at ACME Analytical Laboratories in Canada. Bioconcentration factors (BCF-R, BCF-S, BCF-L, respectively) for the root, stem, leaf, and translocation factor (TF) values were calculated. The highest TF value was found to be 0.85 for Sr. The highest bioconcentration factors in the root, stem, and leaves of the plant were also found to be 3.67, 1.55, and 1.79 for Sr, respectively. According to the results obtained, T. latifolia was found to be a suitable plant for the bioaccumulation of Cd, Pb, Cu, and Sr under the existing environmental conditions, but not a suitable bioaccumulator for Ag, Ba, Ni, and Zn. Based on the results, it was determined that some heavy metal pollutants can be remediated using certain plants, and that these plants can function as bioaccumulators for these metals.

Serdar, Osman; Yildirim, Nuran Cikcikoglu; Yildirim, Numan; Tatar, Sule

Tanyol, Mehtap; Cikcikoglu Yildirim, Nuran; Tatar, Sule; Serdar, Osman; Yildirim, Numan
The biochemical biomarkers (Catalase (CAT), glutathione s-transferase (GST), cytochrome P4501A1 (CYP1A1450) activities and glutathione (GSH), malondialdehyde (MDA) levels) of D. polymorpha exposed to the basic blue 41 (BB41) solutions for 24 and 96 h before and after the Fenton process (FTP) were evaluated to investigate the efficiency of the FTP. The optimum conditions for the removal of BB41 were set using central composite design (CCD), which is a response surface methodology (RSM) module in the Design-Expert® software. The optimal conditions were found to be 27.87 min, 3.49 mg/L, and 0.04 for reaction time, initial BB41 concentration, and Fe2+/H2O2 molar ratio, respectively, resulting in maximum (99%) removal efficiency. After FTP, the MDA and GSH levels decreased significantly during the 24 and 96 h. CAT activities were increased after treatment. No statistically significant changes were observed in the CYP1A1 levels after FTP. GST activities decreased during 24 h but increased during 96 h after FTP. CYP1A1450, catalase, GST activities and MDA, GSH levels of D. polymorpha are useful biomarkers to evaluate the efficiency of FTP. This study suggests that BB41 dye must be further treated after the FTP to minimise the ecotoxicological risk.

Onder Erguven, Gokhan; Tatar, Şule; Serdar, Osman; Yildirim, Nuran Cikcikoglu
This study reveals out detoxifying and antioxidant enzyme response of Gammarus pulex exposed/polluted to chlorpyrifos-ethyl insecticide before and after biodegradation/bioremediation by Methylobacterium radiotolerans and Microbacterium arthrosphaerae. Cytochrome P450 1A1, glutathione S-transferase, catalase, and superoxide dismutase activities in G. pulex exposed to chlorpyrifos-ethyl before and after bioremediation/biodegradation by these two bacteria during 24 and 96 h tested by using commercial ELISA kits. The activity of catalase enzyme was decreased depending on chlorpyrifos-ethyl before and after bioremediation/biodegradation the enzyme activity was increased repeatedly. Superoxide dismutase activity level increased after chlorpyrifos-ethyl exposure in 96 h (p > 0.05). Following bioremediation, superoxide dismutase enzyme activity decreased again during 24 h (p > 0.05) and increased during 96 h (p < 0.05). Statistical differences were not found in cytochrome P450 1A1 enzyme activity before and after the process (p > 0.05). No significant differences were determined during the activity of glutathione S-transferase in 24 h (p > 0.05). The activities of glutathione S-transferase were increased after exposure of chlorpyrifos-ethyl during 96 h. After bioremediation; the activity of glutathione S-transferase increased even more (p < 0.05). The results determined that activities of G. pulex at superoxide dismutase, catalase, and glutathione S-transferase are common biomarkers for revealing out the efficiency of bioremediation of chlorpyrifos-ethyl with these two types of soil bacteria. - MakaleYalnız künye2020Scopus SJRQ2Investigation of antioxidant responses in Gammarus pulex exposed to Bisphenol A
Tatar, Şule; Türkmenoğlu, Yener
One of the most important environmental problems in the world is micro-pollutants. The aim of this study was to investigate the antioxidant responses of Gammarus pulex to Bisphenol A (BPA), an endocrine-disrupting agent. For this purpose, sublethal concentrations of BPA were applied to G. pulex and biochemical responses were studied. Enzymatic antioxidants superoxide dismutase (SOD) and catalase (CAT) activities and nonenzymatic antioxidants glutathione (GSH) and thiobarbituric acid reagents (TBARS) levels in G. pulex were determined in four different groups during 24 and 96 h. Biochemical biomarkers were measured using commercial kits in a microplate reader. When we compared with control, SOD enzyme activity increased in all groups during both administration periods and CAT enzyme activity decreased in all groups. GSH and TBARS levels were increased after 24 and 96 h of application periods in all groups when compared with control. For changes in SOD and CAT activities and GSH, TBARS levels have been determined to be useful as biomarkers against BPA in G. pulex tissues. It has also been proven that G. pulex is an effective bioindicator that shows BPA pollution in water. The different results of biochemical biomarkers can be evaluated as a marker of possible metabolic processes, and the biochemical response of G. pulex can reveal to some extent the environmental consequences of BPA pollution resulting from industrial waters.

- MakaleYalnız künye2020Scopus SJRQ2Can Toxicities Induced by Insecticide Methomyl be Remediated Via Soil Bacteria Ochrobactrum thiophenivorans and Sphingomonas melonis?
Tatar, Sule; Yildirim, Nuran Cikcikoglu; Serdar, Osman; Erguven, Gokhan Onder
The research study was about revealing the biochemical response of Gammarus pulex related to insecticide methomyl before and after bioremediation by two soil bacteria species, Ochrobactrum thiophenivorans and Sphingomonas melonis. Catalase (CAT), glutathione S-transferase.(GST), cytochrome. P4501A1 (CYP1A1) activities in G. Pulex related to methomyl solution were investigated in 24 h and 96 h. ELISA method was used for test studies. CAT enzyme was decreased in Gammarus pulex that was exposed to methomyl after all exposure period (P < 0.05). CAT activities were returned to control results after bioremediation assays. GST enzyme activity was decreased depending on methomyl exposure during 24 h but increased during 4 days (P < 0.05). After 8 days of bioremediation period, GST activity increased again during 24 h while decreased during 4 days (P < 0.05). CYP1A1 activity increased in Gammarus pulex that was exposed to methomyl after all exposure period (P > 0.05). After bioremediation, statistically significant changes were not revealed in CYP1A1 activities (P > 0.05). According to the results of our study, CYP1A1, CAT, and GST activities in G. pulex sanctioned the capability of Ochrobactrum thiophenivorans and Sphingomonas melonis in methomyl bioremediation. Isolated and enriched Ochrobactrum thiophenivorans and Sphingomonas melonis that were added to 2.5 ppb concentrations of methomyl for 8 days. Each day, chemical oxygen demand (COD) and biochemical oxygen demand (BOD5), pH and dissolved oxygen parameters were monitored. At the final phase of the bioremediation step, it was determined that these bacteria have efficient methomyl bioremediation properties in a mixed corsortia at a rate of 86%. These results show that these bacteria can be used for bioremediate the receiving environments that are polluted by these kinds of insecticides.

İncele - MakaleYalnız künye2019Antioxidant Enzyme Responses to Lead Exposure of Gammarus pulex
Serdar, Osman; Yildirim, Numan; Tatar, Sule; Cıkcıkoğlu Yıldırım, Nuran
It is aimed to investigate the activity response of the antioxidant enzymes superoxide dismutase, catalase and glutathione peroxidase of G. pulex exposed to lead. The G. pulex was exposed synthetic solutions containing 10, 30, 50 µg/L lead for 96 h. Superoxide dismutase, catalase, glutathione peroxidase activities were determined by using ELISA kit. The differences in superoxide dismutase activities were statistically insignificant (P>0.05). Catalase decreased in all treatment groups after lead exposure (P<0.05). The glutathione peroxidase activities were decreased depending on lead exposure (P>0.05). Our findings suggest that lead can induce oxidative stress by generating reactive oxygen species and inhibiting sulfhydryl dependent antioxidant enzymes activities. In conclusion, alterations in antioxidant enzymes may potentially be used as sensitive biomarkers for risk assessment of lead in the environment and may contribute to the establishment of discharge regulations.
