Parlak Ak, Tuba; Seven, Pinar Tatli; Seven, Ismail; Mutlu, Seda Iflazoglu; Yaman, Mine; Sur, Aslihan; Gul, Burcu

The aim of this study was to evaluate the effect of chrysin on testicular damage induced by carbon tetrachloride (CCl4) in rats. Twenty-four male Spraque-Dawley rats were divided into four groups as the control group, Chrysin group, CCl4 group, and Chrysin+CCl4 group. Chrysin (100 mg/kg/body weight/day) and CCl4 (0.5 ml/kg/body weight/day in corn oil (1:1)) were administered by gavage for 14 days. Compared to the control group, it was determined that malondialdehyde (MDA) level increased, reduced glutathione (GSH) level, and superoxide dismutase (SOD) and catalase (KAT) activities decreased in the CCl4 group. It was also determined that many testicular lesions were induced, Johnsen’s Mean Testicular Biopsy Score (JMTBS) decreased, and caspase-3 immunoreactivity increased in this group. Compared to the CCl4 group, it was determined that GSH level and SOD and CAT activities increased, MDA level decreased in the Chrysin+CCl4 group. It was also observed that testicular lesions were alleviated, JMTBS increased, and caspase-3 immunoreactivity was decreased in this group. As a result, it was concluded that chrysin might have beneficial effects in the treatment of CCl4-induced testicular toxicity as a result of its antioxidant and anti-apoptotic properties.

Ozcelik, Mehtap; Erisir, Mine; Guler, Osman; Baykara, Murat; Bulmus, Funda Gulcu

The antioxidant and protective effect of resveratrol against irradiation induced oxidative damage were researched. Twenty-one rats were seperated into 3 groups; First group was the control group. Second group was the irradiation group; received physiological saline intraperitoneally (i.p.) for three days, then 9 Gray (Gy) irradiation was applied. Third group was irradiation+Resveratrol group; received 10 mg/kg resveratrol i.p. for three days and then the same dose of radiation was applied. As a result of irradiation, a significant increase was found in the level of malondialdehyde (MDA) in the liver, kidney and brain tissues of rats (P<0.001). Glutathione peroxidase (GSH-Px) activity in all the tissues and superoxide dismutase (SOD) activity, Glutathione (GSH) levels in liver decreased (P<0.01), GSH levels in kidney, spleen and ovarium tissues significantly increased (P<0.05). The giving of resveratrol significantly reduced the MDA levels increasing in liver, kidney, and brain as a result of irradiation (P<0.001) and the liver MDA levels reached to the control levels. By giving resveratrol, it was determined that all the antioxidants studied reduced significantly compared to the control group and/or the irradiation group (P<0.05). According to the data obtained as a result; It was concluded that resveratrol shows a corrective effect by changing the oxidant/antioxidant balance against oxidative stress caused by irradiation, therefore resveratrol is a powerful antioxidant.

The Effect of Naringenin on Oxidative Stress in Some Tissues (Heart, Lung, Brain, Spleen, Muscle) of Lead-treated Rats

Benzer, Fulya; Dağ, Üzeyir; Erişir, Mine; Özkaya, Ahmet

Naringenin, a flavonoid increases resistance against oxidative stress. The present study aims to investigate the effect of lead on lipid peroxidasyonu and some antioxidants in heart, lung, brain, spleen, muscle and the effect of naringenin on these parameters. Naringenin was administered by orogastric gavage (50 mg/kg, dissolved in corn oil) and lead acetate was given as daily 500 parts per million in drinking water for 4 weeks. In the rats exposed to lead, MDA levels in the brain, lungs, spleen and muscle tissues significantly increased (P<0.001), in the heart tended to increase but not significantly (P>0.05). Lead caused a statistically significant decrease in GSH-Px activity of the brain, spleen and muscle tissues, in CAT activity of the heart, lung and muscle tissues, in GSH concentration of the lungs and muscle tissues (P<0.05). The administration of alone naringenin caused a statistically significant decrease in MDA levels in the heart and muscle, but a significant increase in the lung (P<0.05). Although increased MDA levels in the heart, lungs, brain, muscle and spleen tissues were significantly decreased by use of the naringenin together with lead (P<0.05), increased MDA levels in the lungs and spleen tissues could not be recovered to the normal level. Generally the decreased GSH-Px and CAT activities in these tissues due to lead were significanly increased by naringenin supplementation (P<0.05). In addition, the reduced GSH levels in lung and muscle, the inceased GSH levels in heart with effect of lead were returned to the normal by naringenin. Lead particularly causes to an increase in lipid peroxidation by consuming antioxidants of cells. Naringenin was able to prevent as tissue-specific (brain, muscle) the oxidative stress caused by lead.

Effects of Different Plant Oils Having Different Oxygen Radical Absorbance Capacity (ORAC) on Hematological Parameters of Rainbow Trout (Oncorhynchus mykiss) at High Stocking Density

Aksu, Önder; Altınterim, Başar; Kutluyer Kocabaş, Filiz

In this study, macerated oils of different plant [sunflower (Helianthus annuus), onion (Allium cepa), garlic (Allium sativum), basil (Ocimum basilicum), opium poppy (Papaver somniferum), ginger (Zingiber officinale), turmeric (Curcuma longa)] were obtained. The study was conducted at a total of 7 groups, including the control group (sunflower oil), in 30 liter aerated tanks without water circulation during a week. Twenty rainbow trout per tank (14.6±1.0 g) were stocked at high density. Macerated oils were applied to the skin of the fish along the lateral line on the first day. For determination of effects of macerated oils on water and fish, oxidationreduction potential (ORP) of water and hematological parameters [granulocyte (GRAN), white blood cell (WBC), red blood cell (RBC), hemoglobin (HGB), mean corpuscular hemoglobin (MCH), red cell distribution width (RDW-SD), mean platelet volume (MPV), platelet distribution width (PDW), platelet large cell ratio (P-LCR)] were examined in fish. The results indicated that decreases were determined in ORP level of water. It was determined that the highest survival rate was in the application of basil oil. At the end of 7 days, the highest WBC, LYM, HGB, MCV, MCH, MPV values were determined in the application of ginger oil (ORAC: 28.811). It was determined that the highest RBC value was in the application of turmeric oil (ORAC: 159.277), the highest GRAN value was in the application of poppy oil (ORAK: 481), the highest HCT, PLT values was in the application of basil oil (ORAC: 67.553). At the end of the study, it was determined that the macerated oil application to lateral line induced blood parameters.

Effects of green tea (Camellia sinensis) Oil on Feed Utilization and Hametological Parameters of Rainbow Trout (Oncorhynchus mykiss)

Aksu, Önder; Öztürk, Ekrem; Kutluyer, Filiz; Altınterim, Başar

In the study, experiments were conducted to determine effects of green tea oil (Camellia sinensis) on the growth rate, feed utilization and hematological parameters of rainbow trout (Oncorhynchus mykiss). Fish (76.25±2.75 g) were fed diets supplemented with different concentrations of green tea oil (%0.25, %0.5, %1.0). Weight gain (WG), feed conversion ratio (BDI) and some blood parameters were determined in the rainbow trout. It was determined that the values of WBC, GRAN, HGB, HCT, MCH, MCHC, PLT, PDW and PCT were increased with respect to control group at the end of 42 days feeding with green tea oil. However, groups treated with green tea oil stimulated megakaryocytes (increased PLC-R levels), which also increased platelet production. Feed evaluation ratio varied between 1.83-4.53. At the end of the experiment, it was determined that the feed evaluation ratio decreased with the increase of the amount of green tea oil in the groups. It is thought that the bioactive substances in the lipophilic property of green tea oil provide thermogenic stimulation in fishes by synergistic effect, which causes weight loss. Besides, it was determined that antioxidants such as hematopoiesis and catechins in hematopoietic organs of green tea oil act on the granulocyte cell group.
