Mercury is an industrial toxin with detrimental effects on the reproductive system. This study explored the impact of naringenin (NAR) on testicular toxicity and apoptosis in rats chronically exposed to mercury chloride (MC). The research involved 41 adult male Wistar-Albino rats aged about 2.5 months, which were allocated into 5 groups: group 1 was a control group, group 2 received 100mg/kg of NAR, group 3 was exposed to MC, group 4 received both MC and 50mg/kg of NAR, and group 5 received both MC and 100mg/kg of NAR. For the treatments, the control group took 1mL/kg saline via intraperitoneal (ip) injection and 1mL/kg corn oil via gavage. MC was administered at 0.4mg/kg/day to the MC groups via ip, and NAR was delivered in corn oil at 50 and 100mg/kg/day to the respective NAR groups via gavage. All treatments were performed over a period of 20 days, with MC administration starting 1 hour after NAR administration. The irregularity and loss in spermatogenic cell organization, degeneration and vacuolization in tubules, edema, and congestion in interstitial areas of the testicular tissue of the MC group were alleviated by NAR administration. In addition, the increase in Caspase 3, iNOS, and eNOS immunoreactivities of the MC group were alleviated by NAR administration. In conclusion, it was found that NAR had protective and therapeutic effects against oxidative damage, histopathological changes, apoptosis, and altered spermatological parameters resulting from MC exposure in the testis, and 100mg/kg NAR provided positive effects on altered histopathological analysis, spermatological parameters, and apoptosis.

Esen, Engin; Parlak Ak, Tuba; Yaman, Mine
Neuropeptides are diverse neuronal signaling molecules that regulate cellular and physiological functions. Phoenixin (PNX) and Nucleobindin 2 (NUCB2)/nesfatin-1, which are known to co-localize and interact in some hypothalamic nuclei such as arcuate nucleus (ARC) and paraventricular nucleus (PVN), have important pleiotropic roles. The aim of this study was to investigate the immunohistochemical distribution of these neuropeptides in biological tissues of adult female and male rats. For this purpose, a total of 12 Spraque-Dawley rats, 6 female and 6 male, aged 3–4 months, were sacrificed and hypothalamus, pituitary, brain, cerebellum, adrenal gland, ovary and testis tissues were taken. Serum gonadotropin-releasing hormone (GnRH), plasma PNX-14, and NUCB2/nesfatin-1 concentrations showed no statistically differences between sexes. PNX-14 immunoreactivity was more prominent in the ARC of male rats, whereas NUCB2/nesfatin-1 immunoreactivity was markedly higher in the PVN of females. In the anterior pituitary, PNX-14 and NUCB2/nesfatin-1 staining was more intense in females relative to males. PNX-14 immunoreactivity in the brain was quite high in female and male rats, and this immunoreactivity in the cerebellum was low in both sexes. NUCB2/nesfatin-1 exhibited moderate immunoreactivity in these organs in both sexes. In the adrenal cortex of both sexes, PNX-14 immunoreactivity was quite intense, while NUCB2/nesfatin-1 was a moderate intensity. PNX-14 immunoreactivity was more intense in the ovary, whereas NUCB2/nesfatin-1 immunoreactivity was more prominent in the testis. In conclusion, we demonstrated the immunohistochemical localization of PNX-14 and NUCB2/nesfatin-1 neuropeptides, in conjunction with serum GnRH levels, in organs associated with the hypothalamic-pituitary-gonadal (HPG) and hypothalamic-pituitary-adrenal (HPA) axes of adult male and female rats.

Pollutants such as copper nanoparticles (CuNP), which form residues in the environment, especially in water environments, cause serious damage to the environment and the organisms living in the environment. In this study, Navicula cryptocephala var. veneta was selected as a model organism. The EC50 values of CuNP on Navicula cryptocephala var. veneta were determined as 4.94 mg\L and 3 different application concentrations were determined considering this ratio. The experiments were carried out in 3 replicates and samples were taken at 24 and 96 hours. At the end of 120 hours, samples were taken to determine the elimination values. CuNP accumulation amounts in the samples were measured by electrothermal atomic absorption spectrophotometer (ETAAS). For biomarker analysis, thiobarbituric acid (TBARS) and glutathione (GSH) levels and superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities were performed using CAYMAN ELISA kit. According to the study data, as CuNP concentration and application time increased, the amount of accumulation in Navicula cryptocephala also increased. Increasing-decreasing changes were observed in elimination. CuNP caused oxidative stress in N. cryptocephala and caused changes in TBARS and GSH levels. In addition, it caused increases in SOD activities and statistically significant decreases (P<0.05) in CAT and GPx activities.

- MakaleAçık Erişim2025Scopus SJRQ1ADAMTS-1, a multifunctional proteinase, in the uterus of both estrous cycle rats and ovariectomized rats can be regulated via hormones
Parlak Ak, Tuba; Yaman, Mine; Bayrakdar, Ali; Bulmus, Ozgur
Remodeling of the extracellular matrix (ECM) throughout the estrous cycle is one of the most striking features of the uterus. A disintegrin and metalloprotease with thrombospondin type I motifs (ADAMTS-1) is a metalloproteinase responsible for the degradation of some proteoglycans, which are ECM components. In this study, ADAMTS-1 distribution was analyzed in the uterus of ovariectomized rats administered 17β-estradiol (E2) and progesterone (P4) and in the uterus at different estrous stages. Ovariectomized (OVX) rats were subjected to single and combined E2 (0.2 mg/kg) and P4 (10 mg/kg) hormone replacement therapies. E2 was administered for 3 consecutive days, followed by E2, P4, or E2 + P4 for 4 consecutive days. The serum level of E2 decreased from the proestrus phase to the diestrus phase, but that of P4 was the highest in the estrus phase. During the estrus phase, the serum level of luteinizing hormone (LH) was the lowest and that of follicle-stimulating hormone (FSH) was the highest. P4 level increased significantly in the OVX + P4 and OVX + E2 + P4 groups compared with the OVX group. The serum levels of LH and FSH decreased in the OVX + E2 and OVX + P4 groups compared with the OVX group, and were the lowest in the OVX + E2 + P4 group. ADAMTS-1 immunoreactivity in luminal, glandular, and stromal cells of the uterus decreased from proestrus to diestrus. When immunoreactivity in hormone replacement groups was compared, weak immunoreactivity was observed in the OVX group. ADAMTS-1 immunoreactivity gradually increased in OVX + P4 and OVX + E2 groups, and was particularly notable in luminal, glandular, and stromal cells in the OVX + E2 + P4 group. ADAMTS-1 distribution was affected by the estrous cycle process and hormone replacement therapy in the OVX procedure.

This study was conducted to determine the effects of addition of jujube (Ziziphus jujuba Mill.) fruit at different rates into the mixed feed on intestinal morphology, antioxidant and some blood parameters of broiler chickens. A total of 160 male broiler (initial body weights were determined as 209.25±2.17, 209.18±2.13, 209.18±1.74 and 209.20±2.58, respectively) chickens (Ross-308), including 40 in each group, were used in the study. The groups were divided into 4 sub-groups, each of which include 10 broiler chickens. The study groups were the control group (the group without jujube fruit), the H-0.5% group (the group with 0.5% jujube fruit), the H-1 group (the group with 1% jujube fruit), and the H-2 group (the group with 2% jujube fruit). At the end of the experiment (42nd day), the control group had the highest levels of blood glucose, triglycerides, total cholesterol, very low density lipoprotein cholesterol, alanine aminotransferase, aspartate aminotransferase, and uric acid, and the lowest level of creatine (P<0.05). While the lowest malondialdehyde level was detected in the H-2 group (P<0.001), the highest glutathione peroxidase (P<0.01) and catalase (P<0.05) levels were observed in the H-1 and H-2 groups. The control group also had the lowest values for villus height and crypt depth and the highest values for villus height/crypt depth (P<0.001). As a result, it was concluded that jujube fruit, which is mainly used for its antioxidant properties, can be used in poultry compound feeds especially at the level of 1% due to its positive effects on antioxidant and blood parameters and intestinal morphology.

Lipopolysaccharide (LPS), known as a stimulant of inflammation, causes acute liver injury by inducing the production of inflammatory mediators and oxidative stress. The purpose of this study is to determine whether of a nonsteroidal anti–inflammatory drug (NSAID) Flunixin meglumine (FM) and herbal an medicine Ginkgo biloba L. extract (GBE) show antioxidative, anti–inflammatory or antiapoptotic effects in liver tissue in LPS–induced hepatotoxicity. Animals were separated to 6 groups as control, sepsis (1 mg·kg-1, 7th day single dose, intraperitoneal (ip)), sepsis + FM (1 mg·kg-1, 7th day single dose, ip + 2.2 mg·kg-1 day, ip), sepsis + GBE (1 mg·kg-1, 7th day single dose, ip + 50 mg·kg-1 day, gavage), FM and GBE and the study continued for 7 days. Liver tissues taken from rats sacrificed were analyzed biochemically, histologically and immunohistochemically. Accordingly, LPS caused liver function markers alteration, inflammation, oxidative stress, and apoptosis, as well as histopathological changes in liver tissue. However, it was observed that LPS–induced changes were regulated by FM and GBE application. FM and GBE was demonstrated to have antioxidant, antiinflammatory and anti–apoptotic properties in LPS– induced hepatotoxicity.

Gentamicin (GM), which causes nephrotoxicity, is an aminoglycoside antibiotic commonly prescribed to treat of gram–negative infections. Erythropoietin (EPO), which has several biological functions including neuroprotection, wound healing and nephroprotection, is a glycoprotein hormone that controls erythropoiesis. Hypericum perforatum (HP) is a medicinal herb with antibacterial and nephroprotective effects. The aim of this study is to demonstrate the efficacy of EPO and HP in GM nephrotoxicity using combined biochemical, histopathological and immunohistochemical evaluations together. A total of 36 male Spraque–Dawley rats were divided into as control, GM (100 mg·kg-1 day), GM+EPO, GM+HP, EPO (1000 IU·kg-1 three consecutive days apart) and HP (200 mg·kg-1 day) groups (n=6) and the experiment lasted for 9 days. GM–induced increased relative kidney weight and increased serum urea nitrogen (BUN), creatinine and urea levels were reduced by EPO and HP. EPO and HP reduced the level of malondialdehyde (MDA), which increased with GM application, and increased the activities of reduced glutathione (GSH), glutathione peroxidase (GSH–Px), and catalase (CAT). GM nephrotoxicity resulted in tubular degeneration, vacuolization and hyaline deposits, glomerular degeneration and interstitial mononuclear cell infiltration. EPO and HP attenuated these histopathological changes. Also, EPO and HP also reduced caspase–3 immunoreactivities, which increased with GM application. It was shown that EPO and HP have attenuating effects on GM–induced kidney injury, and especially the intense antioxidant content of HP has a regulatory effect on the negative consequences of oxidative stress.
- MakaleAçık Erişim2024Scopus SJRQ2Effects of naringenin on oxidative damage and apoptosis in liver and kidney in rats subjected to chronic mercury chloride
Kahramanoğullari, Merve; Erişir, Mine; Yaman, Mine; Parlak Ak, Tuba
Mercury chloride is a type of heavy metal that causes the formation of free radicals, causing hepatotoxicity, nephrotoxicity and apoptosis. In this study, the effects of naringenin on oxidative stress and apoptosis in the liver and kidney of rats exposed to mercury chloride were investigated. In the study, 41 2-month-old male Wistar-Albino rats were divided into five groups. Accordingly, group 1 was set as control group, group 2 as naringenin-100, group 3 as mercury chloride, group 4 as mercury chloride + naringenin-50, and group 5 as mercury chloride + naringenin-100. For the interventions, 1 mL/kg saline was administered to the control, 0.4 mg/kg/day mercury (II) chloride to the mercury chloride groups by i.p., and 50 and 100 mg/kg/day naringenin prepared in corn oil to the naringenin groups by gavage. All the interventions lasted for 20 days. Mercury chloride administration was initiated 1 h following the administration of naringenin. When mercury chloride and the control group were compared, a significant increase in plasma urea, liver and kidney malondialdehyde (MDA) levels, in kidney superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) activities (p <.001), and a significant decrease in liver and kidney glutathione (GSH) levels (p <.001), in liver catalase (CAT) activity (p <.01) were observed. In addition, histopathological changes and a significant increase in caspase-3 levels were detected (p <.05). When mercury chloride and treatment groups were compared, the administration of naringenin caused a decrease aspartate transaminase (AST), alanine transaminase (ALT), lactate dehydrogenase (LDH) (p <.01), urea, creatinine levels (p <.001) in plasma, MDA levels in liver and kidney, SOD, GSH-Px, GST activities in kidney (p <.001), and increased GSH levels in liver and kidney. The addition of naringenin-100 increased GSH levels above the control (p <.001). The administration of naringenin was also decreased histopathological changes and caspase-3 levels (p <.05). Accordingly, it was determined that naringenin is protective and therapeutic against mercury chloride-induced oxidative damage and apoptosis in the liver and kidney, and 100 mg/kg naringenin is more effective in preventing histopathological changes and apoptosis.
