Seasonal investigations were conducted on 640 Cyprinus carpio specimens obtained through commercial fishing at four different stations (Gerger, Sitilce, Hosirge, and Samsat) in the Atatürk Dam Lake. MDA (malondialdehyde) levels were measured in the liver and muscle tissues of male and female fish; the results revealed a significant increase in MDA levels at stations exposed to point-source discharge. This indicates that pollution-induced oxidative stress in fish varies significantly across seasonal and regional scales. Among the stations compared, the lowest MDA values were detected in fish tissues from the Gerger station, while the highest values were recorded at the Sitilce and Hosirge stations. Analysis of SOD, GSH-Px, and GSH values across all stations showed that these parameters fluctuated in correlation with MDA levels thereby maintaining the oxidant-antioxidant balance. This study investigated seasonal and spatial variations in oxidative stress biomarkers in common carp (Cyprinus carpio) collected from four stations in Atatürk Dam Lake. A total of 640 specimens were analyzed for malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and reduced glutathione (GSH) levels in liver and muscle tissues. Results showed that MDA levels were significantly higher in fish collected from Sitilce and Hosirge stations, indicating elevated lipid peroxidation associated with point-source pollution. In contrast, the lowest MDA levels were observed at the reference station, Gerger. Antioxidant enzyme activities (SOD, GSH-Px, and GSH) exhibited tissue-, season-, and site-dependent variations, suggesting adaptive responses to oxidative stress. Overall, the findings demonstrate that anthropogenic pressures in Atatürk Dam Lake significantly alter the oxidant–antioxidant balance in fish, highlighting the usefulness of these biomarkers for environmental monitoring.

Anti-oxidants are vital for protecting sperm and can mitigate the negative effects associated with cryopreservation. This study was conducted to evaluate the protective effects of adding anti-oxidants (vitamins and amino acids) to extender in sex-reversed rainbow trout (Oncorhynchus mykiss) sperm. The collected sperm was diluted at the ratio of 1: 5 by the extenders supplemented with different anti-oxidants including 1.00 mM of ascorbic acid and L-tryptophan, 2.00 mM of cysteine and α-tocopherol, and their combination. After dilution, the semen was aspirated into 0.50 mL straws, and the straws were placed on the tray, frozen for 10 min, and plunged into liquid nitrogen. Straws were thawed in a 30.00 ˚C water bath for 15 sec. The Sperm Class Analyzer System was used to evaluate sperm kinematics. The activity of anti-oxidant enzymes and lipid peroxidation were determined as oxidative stress indices. Our data indicated that the incorporation of anti-oxidants and amino acids increased sperm motility duration. The elevated activity of glutathione peroxidase, superoxide dismutase, and catalase in post-thaw samples indicates that the anti-oxidant system in sex-reversed rainbow trout sperm likely plays a crucial role in protecting membrane compounds from oxidation. In conclusion, the combination of 1.00 mM L-tryptophan and ascorbic acid to the extender media caused a prolonging effect in sperm motility after thawing and they have the potential to serve as effective agents for improving sperm cryosurvival.

As a potential pollution source, slaughterhouse wastewater (SWW) with development of the meat production and processing sector is a threat in the world. Therefore, the determination of economical, favourable, cost-effective and proper treatment method is crucial for sustainable environmental protection. This study focused on the performance and efficiency of a two-stage process combining biological treatment (BT) with freshwater snails and electrocoagulation (EC) of SWW. Viviparus contectus, Planorbis rubrum, Melanoides tuberculata were used in primary treatment while secondary treatment was realized with EC process. The EC of SWW was performed for reducing both turbidity and chemical oxygen demand (COD) concentration to levels compliant with direct-discharge regulations. In addition, the stirring speed, current density, initial pH and reaction time were assessed for evaluation the optimum EC condition. Oxidative stress biomarkers and some antioxidants were assessed in freshwater snails exposed to SWW after 24 and 96 h. The best removal efficiencies of COD (91.43%) and turbidity (92.42%) were obtained at pH 5 and 20 min of operational time by P. rubrum. Notably, MDA (Malondialdehyde) was highest level in M. tuberculata compared to V. contectus and P. rubrum. Overall, removing organic and inorganic pollutants from SWW could be provided with the hybrid process BT/EC as a combining approach.

Aroclor 1254 impairs sperm quality, fertilization ability, and embryo development of rainbow trout (Oncorhynchus mykiss)

Kocabaş, Mehmet; Kocabaş, Filiz Kutluyer; Aksu, Önder; Benzer, Fulya; Erişir, Mine; Sahilli, Yeliz Çakir

The polychlorinated biphenyls (PCBs) in aquatic environment adversely affect non-target organisms, including fish. Especially, the male reproduction and next generation can be damaged through high exposure to these pollutants. Hence, the sperm cells were exposed to sublethal concentrations of Aroclor 1254 (0, 1, 5, 10, or 25 mg/l) for 4 h. The sperm quality parameters were analyzed by SCA (Sperm Class Analyzer). The fertility, eyeing, and hatching rates were determined as gamete markers. Lipid peroxidation (malondialdehyde-MDA), glutathione (GSH), and antioxidant enzymes [superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT)] were measured for determination of oxidative stress. Our results showed that Aroclor 1254 negatively affected the motility rate and duration, fertilization rate, embryogenesis, and hatching and also triggered antioxidant defense mechanisms at the highest concentration (25 mg L−1). Furthermore, linear speed (VSL), linearity index (LIN), and amplitude lateral head (ALH) were significantly changed after exposure to 25 mg L−1, and the lowest concentrations (1 and 10 mg L−1) did not significantly affect the motility and fertilizing capacity. The embryogenesis and hatching were significantly affected by sperm exposure to 1, 10, and 25 mg L−1 of Aroclor 1254. Consequently, Aroclor 1254 causes potential hazards in male germ cells, and the exposure of sperm cells to pollutants can adversely affect next generation of wild populations.
