The biochemical reactions of zebra mussels Dreissena polymorpha (Pallas, 1771) exposed to different concentrations of cadmium (Cd) in the acute period (24 and 96 hours after treatment) were studied. Four variants of experimental treatment were examined: 0 μg/L Cd (control), 10 μg/L Cd, 20 μg/L Cd and 40 μg/L Cd. Changes in the enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione (GSH) and thiobarbituric acid reactive substances (TBARS) levels were determined using ELISA kits. In D. polymorpha exposed to the three experimental concentrations of cadmium, there was a statistically significant decrease (p<0.05) in SOD enzyme activity with increasing concentration or exposure times. No change was observed in CAT enzyme activity with increasing concentrations. The decrease in the activity depending on the exposure times was not statistically significant (p>0.05). As for GPx enzyme activity, a significant increase was detected after Cd application compared to the control (p<0.05). TBARS and GSH levels increased significantly in all Cd application groups compared to the control at 24 and 96 hours. The effects of different concentrations of Cd on D. polymorpha in the acute period were associated with changes in SOD, CAT, GPx enzyme activities. TBARS and GSH levels were effective parameters in determining the oxidative damage caused by Cd.

In this study, Gammarus pulex (L., 1758) individuals obtained from Tunceli Munzur Stream were exposed to different concentrations of two insecticides containing cyfluthrin and dimethoate active ingredients, and LC50 values were determined from acute toxicity tests. During the study, parameters such as water temperature, pH, and dissolved oxygen were constantly controlled to ensure that they did not change. In this study, experiments were carried out by placing 0.5 liters of water in 1-liter glass aquariums. 10 G. pulex individuals were used for each concentration group. To determine the LC50 value, the mobility of the living things was observed and recorded in 24-hour periods. G. pulex, which lost its motility, was removed from the aquarium and excluded from the study. The study was carried out in 3 replications and the LC50 value for dimethoate was determined as 170.51 ± 8.15 µg/l, while the LC50 value for cyfluthrin was determined as 0.800 ± 0.12 µg/l.

İncele - MakaleAçık Erişim2021Farklı Sıcaklık ve Farklı Konsantrasyonlarda Kadmiyuma Maruz Bırakılan Gammarus pulex'te Bazı Biyokimyasal Parametrelerin Değişikliklerinin Belirlenmesi
Serdar, Osman; Çalta, Metin; Aydin, Rahmi
Bu çalışmada Gammarus pulex üzerinde kadmiyum (Cd) toksisitesinin su sıcaklığına bağlı olarak oksidatif stres etkileri araştırılmıştır. Test organizması bireyleri, her sıcaklık (10, 14, 18 °C) için belirli oranlarda (C1; 1/10, C2; 1/20 ve C3; 1/40) LC50 değerlerinin 96 saat boyunca subletal konsantrasyonlara maruz bırakıldı. Cd'ye maruz kalan G. pulex'te belirlenen sıcaklıklarda malondialdehid (MDA) seviyesi, glutatyon peroksidaz (GPx) ve katalaz (CAT) enzim aktiviteleri araştırıldı.

In this study, some biochemical responses of Beta-Cyfluthrin (β-CF), which is a commercial insecticide in Dreissena polymorpha, were investigated. The 96 hour LC50 value of β-CF on D. polymorpha was calculated as 509.62 µg L-1. D. polymorpha was exposed to subletal concentrations (1/16, 1/8 and 1/4 of LC50 value: 32, 64 and 128 µg L-1) of β-CF for 24 and 96 hours. Malondialdehyde (MDA) and reduced glutathione (GSH) levels and Acetylcholinesterase (AChE) enzyme activities were determined in D. polymorpha individuals. In D. polymorpha exposed to β-CF, MDA levels increased compared to control and with increasing concentration. It was determined that GSH level decreased and AChE activity was inhibited compared to control and with increasing concentration. In conclusion, exposure to β-CF resulted in increased oxidative damage and has been found to cause neurotoxicity even at low concentrations.

İncele - MakaleYalnız künye2020Scopus SJRQ4Examining the biochemical effect of malathion pesticide on gammarus pulex (l., 1798)
Yukse, Fahrettin; Aydin, Rahmi; Serdar, Osman; Pala, Avsegul
This study determined the effect of a pesticide with malathion as active substance on Gammams pulex in terms of oxidative stress and antioxidant status. Firstly, 96-hours LC50 value of malathion on G. pulex was determined. Then, the organisms were exposed to determined sublethal concentrations (at the rate of 1/40, 1/20 and 1/10 LC50 value) of malathion for 24 and 96 hours. Thiobarbituric Acid Rcactive Substances (TBARS), rcduccd glutathione (GSH) levels, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) enzyme activities in the control group and samples that were exposed to malathion for 24 and 96 hours were determined. The results showed that TBARS level significantly increased in G. pulex that was exposed to malathion for 24 and 96 hours compared to the control group (p <0.05). However, GSH level, SOD, CAT and GPx activities dccrcascd (p <0.05). Malathion exposure caused oxidative stress in G. pulex. These results revealed that MDA, GSH levels, SOD, CAT and CiPx activities can be used as effective biomarkcrs in G. pulex.

- MakaleYalnız künye2020Scopus SJRQ2Temporal and seasonal variations in phytoplankton community structure in Uzuncayir Dam Lake (Tunceli, Turkey)
Kutlu, Banu; Aydın, Rahmi; Danabas, Durali; Serdar, Osman
Seasonal variations in species composition and growth dynamics of phytoplankton in Uzuncayir Dam Lake (Tunceli, Turkey) were studied in relation to ecological factors by collecting horizontal water and phytoplankton samples for over a year. A total of 46 phytoplankton taxa were recorded, where Bacillariophyta was the most abundant in species number representing 24 taxa. Diatoms were the most conspicuous phytoplankton in terms of abundance as well as occurrence frequency. Charophyta, Chlorophyta, Cyanobacteria, Ochrophyta, Miozoa, and Euglenozoa were the least abundant. However, some Diatoms (Gyrosigma attenuatum, Hippodonta hungarica, Meridion circulare, Nitzschia clausii, Nitzschia frustulum, Pantocsekiella ocellata, Rhoicosphenia abbreviata), Charophytes (Cosmarium granatum, Mougeotiopsis calospora), Cyanobacteria (Leiblenia epiphytica, Planktothrix agardhii); Dinoflagellates (Ceratium hirundinella, Parvodinium africanum, Parvodinium inconspicuum, Peridinium cinctum), and Ochrophyta (Dinobryon pediforme) were also noticeable, with their occurrence in all seasons. Canonical correspondence analysis (CCA) and principal component analysis (PCA) were employed to analyze the relationship between occurrence and abundance of phytoplankton species as well as the ecological factors. The results showed that pH and the concentrations of NH4, NO3, and PO4 influenced seasonal occurrence and abundance of phytoplankton species in the lake. Uzuncayir Dam Lake may be classified as an oligotrophic lake with the mean value of nitrate 2.62 mg L−1 and phosphate 0.23 mg L−1 as supported by the occurrence of various phytoplankton groups. Findings related to pollution indices yielded that Uzuncayir Dam Lake is on the edge of the moderate state of organic pollution.

İncele - MakaleAçık Erişim2019Scopus SJRQ3Possible indices for the assessment of ecological stream quality based on macroinvertebrates in Euphrates tributaries (Turkey)
Gültekin, Zuhal; Hellmann, Claudia; Aydin, Rahmi; Winkelmann, Carola
The aim of this study was to support the development of ecological stream quality assessment tools in order to provide a method for sustainable water management in Turkey. Therefore, we present two new or adapted indices based on benthic invertebrates. To develop and adapt the indices, 17 streams were studied and separated into three quality classes, which were supported by four community indices (EPT [%], EPTCBO [%], number of Individuals, evenness), and 23 taxa were identified as indicators for these three quality classes. As a first biological index, we adapted the Hindu Kush-Himalaya biotic score (HKHbios) to the Euphrates catchment by establishing a new and ecoregion-specific score list (Euph-Scores) by scoring 93 taxa depending on their distribution between the quality classes. Based on these scores, several average score per taxon values (ASPT value) were calculated. All ASPT values of the Euph-Scores separated the quality classes significantly. After a comparison of the different ASPT values we recommend to use the weighted ASPT, because the weighting enabled a sharper differentiation between the quality classes and named it Euphrates Biotic Score (EUPHbios). As a second biological index, we propose the proportion of habitat specialists. To calculate this index, a habitat score was developed by analysing the habitat preferences of several benthic invertebrates. Habitat score values were assigned to the 20 most common taxa from the streams in the best quality class (natural streams). The proportion of habitat specialists, identified using the new habitat score, differed significantly between the three quality classes, with higher values in natural streams than in polluted streams. In the light of the results, the presented methods appear to be suitable for developing a multi-metric index for assessment programs for the mountainous regions of the Middle East.
