Onal, Ali; Demir, Hulya; Ince, Olcay Kaplan; Ince, Muharrem

Hydrogels because of their unique potentials, such as highwater content and hydrophilicity, are of interest for the controlled release of drug molecules. In the present study, a biofilm was produced using chitosan, a natural polymer. Characterization analyses of the synthesized biofilm were performed using Fourier-transform infrared spectroscopy (FT-IR), thermogravimetric analysis-differential thermal analysis, x-ray diffraction, and scanning electron microscopy-energy dispersive x-ray spectrometry. The analysis results of the film before loading were compared to the analysis made after adding vitamin B12 to the film. The release percentage of vitamin B12 was investigated by using ultraviolet-visible spectrophotometery. The FT-IR bands of vitamin B12 were in the range of 2,800-3,500 cm-1 and 1,000 cm-1-1,750 cm-1. When pH increased from 3 to 7, release of vitamin B12 from the biopolymer increased up to pH 5. While examining the release potential, parameters, such as pH, initial concentration of vitamin B12, release time, and solution volume, were optimized by using response surface methodology. It was determined that with the increase in release time and the initial concentration, pH contributed positively to the release of vitamin B12. The following conditions were determined for release of vitamin B12 from the biofilm: pH: 4.2, initial concentration of vitamin B12: 54.0 mg L-1, the release time: 117 min, and volume of solution: 2.1 mL. The analysis of variance results showed that the determination coefficients for the use of synthesized biofilm in vitamin B12 release were high and R2 =0.9704.

Kaplan Ince, Olcay; Ince, Muharrem; Onal, Ali

Risk assessment of cadmium (Cd) for human health play a key role because of toxic effect. Researchers are undergoing constant development for modelling approaches to make a risk assessment. It was carried out a central composite design combining with response surface modeling to optimize and modeling of Cd2+ ions in this study. Also, this study was performed to provide information about possible risk associated with alcoholic beverage consumption. Some independent process parameters including contact time and pH were chosen for optimization. Under optimal parameters, 142.9 mg g−1 was determined as Cd2+ ions removal effectiveness. Significance independent parameters and their interactions were checked using analysis of variance. These results were verified by correlation coefficients (R2 = 0.9967) of the statistical prediction. Estimated daily intake (EDI) and target hazard quotients (THQ) were calculated, to assess health risk. The Cd2+ ions EDI values (µg/day/person) were less than the recommended dietary allowance values and also, its THQ value was less than 1, also. When the obtained results compared with the levels set by regulatory authorities, the studied alcoholic beverages consumption do not pose a toxicological risk. Ince, Olcay Kaplan; Alp, Hevidar; Ince, Muharrem; Onal, Ali

In the present study, a novel biosorbent, namely Agaricus campestris, was used for the removal of Methylene Blue (MB) from an aqueous medium. The biosorbent was characterized using fourier-transform infrared spectrometer spectra, scanning electron microscopy-energy dispersive X-ray, and thermogravimetric analysis. The influence of important process parameters including pH (4–7), temperature (25°C–35°C), contact time (15–25 min), and biosorbent amount (40–80 mg) on the uptake of MB solution were also optimized, which significantly affected the removal efficiency of MB. A central composite design (CCD) combined with response surface modeling (RSM) was carried out to maximize MB removal by A. campestris for the adsorption process. A regression model was derived using RSM to predict the responses and analysis of variance (ANOVA), and to check model adequacy lack of fit test was used. The A. campestris adsorption capacity was determined as 24.2 mg g–1. The ANOVA showed a high coefficient of determination (R2 = 0.9987). This study clearly revealed that RSM was one of the most effective and appropriate method for the optimization of operating conditions. The possibility of reusing A. campestris was also investigated and, as a result, it was found quite possible and showed high performance in the recovery of MB.

Ince, Muharrem; Kaplan Ince, Olcay; Onal, Ali

Present study was performed using a Box–Behnken Design (BBD) combined with a response surface methodology (RSM). The ultimate goal of this research was to apply BBD for modeling of copper (Cu2+) ions from alcoholic beverages and evaluate health risk assessment of Cu2+ via consumption of alcoholic beverages. The importance of independent paramaters and their interactions were examined using analysis of variance (ANOVA). Natural clay was investigated to remove Cu2+ ions from some alcoholic beverage samples including beer and wine. X-ray diffraction (XRD), fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM) coupled with energy dispersive X-ray (EDS) analysis were used for adsorbent characterization. Same analyzes were applied for Cu2+ loaded clay. In addition, experimental studies showed that Freundlich isotherm was better than Langmuir isotherm. Finally, optimized method was applied to alcoholic beverages. Besides target hazard quotients (THQ), estimated daily intake (EDI) values were calculated, to assess a health risk resulting from these alcoholic beverages regular consumption. The EDI values of Cu (µg/day/person) through consumption of investigated drinks were less than the recommended dietary allowance values. Also, the THQ values of Cu2+ were less than 1. According to estimated target hazard, it can be said no long life health concerns of Cu2+ is associated with the consumption of studied drinks. Obtained results were compared with the data from literature and levels set by regulatory authorities, it can be clearly stated that the consumption of mentioned alcoholic beverages do not pose a toxicological risk.

Biosorptive removal of eriochrome black-T using agaricus campestris: Parameters optimization with response surface methodology

Alp, Hevidar; Ince, Muharrem; Ince, Olcay Kaplan; Onal, Ali

In this study, Eriochrome Black T (EBT) removal from aqueous media by Agaricus campestris (AC) which is an eco-friendly biosorbent was investigated using a central composite design (CCD) combined with response surface methodology. The CCD was chosen as the statistical prediction method to reduce the number of experiments which, in turn, will directly save time and chemicals. To opti-mize the process, various experimental independent variables such as the pH of the solution, contact time, adsorbent dosage and temperature were selected. The optimal experimental conditions were obtained as 6.5, 30°C, 20 min and 20 mg, for solution pH, temperature, contact time and adsorbent dosage, respectively. Under these optimized conditions, maximum EBT (100 mg L–1 initial concentra-tion) removal efficiency was calculated as 64.2 mg g–1. The independent variables and their interaction significance were checked using analysis of variance and it was found that the EBT biosorption on AC was highly significant (p < 0.001). The results were justified by the relatively high correlation coefficients (R2 = 0.9967) of the statistical prediction. The EBT biosorption was confirmed by fourier transform infrared spectroscopy (FTIR) analysis monitoring the functional groups during EBT biosorption on AC. This was the first report on the parameter optimization of EBT biosorp-tion using AC in an experimental design. The used biosorbent was characterized by FTIR, scanning electron microscope coupled with energy dispersive spectrometer and thermogravimetric analysis. The proposed biosorption process efficiently removed EBT, present in various aqueous mediums and can be used in industrial processes.

Investigation of curcumin role on doxorubicin-induced tissue damage in terms of trace metal levels using etaas

Ince, Olcay Kaplan; Kandemir, Fatih Mehmet; Ince, Muharrem; Benzer, Fulya; Onal, Ali; Kucukler, Sefa

Doxorubicin (DXR) is one of the commonly used chemotherapeutic agents, which is highly toxic and causes gastrointestinal and cardiac toxicity. Curcumin is used as a spice and coloring agent in food, but also possesses anti-proliferating, potent antioxidant, antitumor-promoting and anti-carcinogenic properties in vitro and in vivo. In this study, the change in element concentrations in the tissues of healthy animals was examined by administering doxorubicin and curcumin separately, or both at the same time. For this purpose, 35 wistar albino male rats were selected and randomly divided into five groups of seven each. The objective of this study was to assess and compare the levels of copper, iron, zinc and selenium in the liver, kidney, heart, and testis of the control group and in the doxorubicin-induced tissues. According to the results, the Cu, Fe, Zn, Se amounts and the Cu/Fe, Cu/Zn, and Cu/Se ratios were present at different levels in each of the five groups. In terms of element concentration, the most affected tissue by doxorubicin application is the heart, and the least affected is the kidney. The ratios of Cu/Fe and Cu/Se in the liver and the heart were highest in Group II and Group III, respectively.

Modelling cadmium bioaccumulation in Gammarus pulex by using experimental design approach

Serdar, Osman; Pala, Aysegul; Ince, Muharrem; Onal, Ali

Recently, although dynamic approaches have been used increasingly to describe metal bioaccumulation in aquatic organisms, the validation of such laboratory-derived modelling is rarely assessed under environmental conditions. The present study aims to evaluate and verify statistical and mathematical models for the bioaccumulation of cadmium (Cd) and to evaluate the suitability of Gammarus pulex as a biomonitor for Cd. To optimise Cd2+ ion bioaccumulation conditions a Box–Behnken design was used and whether G. pulex can be used as an estimated tool to assess aquatic quality was investigated. A Box–Behnken experimental design combined with response surface modelling (RSM) was employed for Cd2+ bioaccumulation in G. pulex. Independent variables including temperature, contact time and Cd2+ ion concentration significance and their interactions were tested using analysis of variance. The selected variables’ optimum values were obtained by solving the quadratic regression model along with by analysing the response surface contour plots. Based on Box–Behnken design, the applied model is significant because model F-value and R2 were obtained as 5121.49, 0.9998, respectively. Bioaccumulation of Cd amount was analysed using electrothermal atomic absorption spectrophotometer. The obtained data suggest that G. pulex can be evaluated as Cd biomonitor.

Modeling approach with Box-Behnken design for optimization of Pb bioaccumulation parameters in Gammarus pulex (L., 1758)

Pala, Aysegul; Serdar, Osman; Ince, Muharrem; Onal, Ali

The aim of this research was to evaluate the suitability of Gammarus pulex (L., 1758), an amphipod crustacean, as a bio-monitor for lead (Pb). A Box– Behnken experimental approach combined with response surface modeling (RSM) and quadratic programming was employed to increase the knowledge about Pb2+ bioaccumulation in Gammarus pulex (L., 1758). Independent variables, such as temperature ranging from 18 to 22 ºC, contact time from 8 to 48 hours, and Pb2+ ion solution concentrations from 50 to 100 µg L-1, were studied and tested using analysis of variance (ANOVA). The standardized effects of the independent variables and their interactions on the dependent variable were also investigated according to a Pareto chart. The selected variables' optimum values were obtained by solving the quadratic regression model along with analyzing the response surface contour plots. Optimum experimental conditions were obtained at 22.0 ºC, contact time of 8 hours, and Pb concentration at 69.5 µg L-1. Based on the Box-Behnken design, the applied model is significant because the model F-value and R2 were obtained at 77.80 and 0.9901, respectively. Bioaccumulation of Pb was determined using electrothermal atomic absorption spectrometry (ETAAS). The obtained data suggest that G. pulex can be used as a Pb biomonitor.

Response surface modeling for Pb(II) removal from alcoholic beverages using natural clay: Process optimization with box–behnken experimental design and determination by electrothermal AAS

Ince, Olcay Kaplan; Ince, Muharrem; Onal, Ali

A three-level four-factor Box–Behnken experimental design (BBD) combined with response surface modeling (RSM) was used for the optimization and modeling of Pb(II) adsorption from alcoholic beverages onto natural clay. The BBD method was chosen as the statistical prediction method to reduce the experimental numbers which saves time and chemicals and reduce analysis cost. Various independent process variables including solution pH (X1: 3.0– 7.0), contact time (X2: 2–60 min), adsorbent dosage (X3: 0.01-0.1 g), and agitation speed (X4: 50–150 rpm) were optimized. The optimal conditions used for Pb(II) removal were pH of 5.3, contact time 38.9 minutes, 0.071 g adsorbent dosage, and 98.7 rpm agitation speed, resulting in a maximum removal efficiency of 120 mg g-1. Pb was determined by electrothermal AAS. The significance of the independent variables and their interactions were tested by means of the analysis of variance (ANOVA) and are based on the ANOVA statistical value. The adsorption of Pb(II) onto clay was found to be highly significant with very low probability (p) values (<0.001). These results were justified by the relatively high correlation coefficients (R2 = 0.9965, R2 adj = 0.9930, and R2 Pred= 0.9819) of the statistical prediction. FTIR analysis confirmed major participation of amide, amines, ketones, including the primary alcohol groups, during Pb(II) biosorption. Clay was characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscope (SEM), coupled with energy dispersive X-Ray (EDX). The proposed adsorption process efficiently removes Pb(II) present in various aqueous media and can be used for industrial wastewater or drinks.
