The objective of the present study was to obtain data about cooking time and temperature of kiymali pide in the restaurants and to investigate thermal inactivation of E. coli O157:H7 during experimental kiymali pide making. A field study was conducted in randomly selected 23 of 87 pide restaurants. Processing parameters including oven temperature, cooking period and post-cooking temperature were determined. Kiymali pide samples were prepared using ground beef filling experimentally inoculated with E. coli O157:H7 (7.6 log(10) CFU/g). Pide samples were cooked at a conventional oven at 180 degrees C for 180, 240, 270, 300 and 330 s. Results of the current study suggest that cooking kiymali pide at 180 degrees C for at least 330 s (5.5 min) may provide sufficient food safety assurance (>= 6 log(10) CFU/g) for E. coli O157:H7. Effect of Potassium Sorbate on Some Microbiological Properties of Cokelek Stored at Different Temperatures

Öksüztepe, Gulsum; Ilhak, O. Irfan; Dikici, Abdullah; Çalicioglu, Mehmet; Patir, Bahri

In this study, the effect of potassium sorbate at 0.01%, 0.05% and 0.1% on the some microbiological (total aerobic mesophilic bacteria-IAMB, coliform bacteria, Lactobacillus-Leuconostoc-Pediococcus, Stapylococcus-Micrococcus. enterococcus and yeast-mold) and chemical (pH, acidity) properties of cokelek stored at 4 +/- 1 degrees C or 22 +/- 1 degrees C was investigated. It was seen that adding potassium sorbate at 0.01%, 0.05% and 0.1% to cokelek did not have an effect on numbers of TAMB, coliform bacteria, lactobacillus- leuconostoc- pediococcus, staphylococcus-micrococcus in cokelek stored at both 4 +/- 1 degrees C and 22 +/- 1 degrees C (P>0.05). However, adding potassium sorbate at 0.05% and 0.1% to cokelek had an effect on numbers of enterococci and yeast-mold in cokelek stored at 4 +/- 1 degrees C (P<0.05). It was also not observed significant difference in pH and acidity levels between control group and treatment groups containing potassium sorbate during storage period (P>0.05).

Presence of E. coli O157:H7 in Cakes Marketed in Elazig

Öksüztepe, Gulsum; Patir, Bahri; Çalicioglu, Mehmet; Ilhak, O. Irgan; Dikici, Abdullah

The present study was undertaken to investigate the presence of Escherichia coli O157:H7 in cakes (plain, with cacao, with fruit) marketed in Elazig. A total of 200 cake samples as 100 plain, 50 with cacao, and 50 with fruit were analysed. Following the enrichment, the samples were plated on to CT-SMAC agar. A total of 208 typical colonies (colourless) as 138 colonies from 65 plain cakes, 25 colonies from 8 cakes with cacao, and 45 colonies from 19 cakes with fruit were isolated. These colonies were subjected to further analysis given in the materials and methods section for identification of serotype O157:H7 of E. coli. However, no colonies representing E. coli O157 were detected in any of the samples. Therefore, analysis was not proceeded for determining H7 in any of the isolates. E. coli O157:H7 was not detected in any of the cake samples analyzed. Although this finding is a good news for public health, considering other pathogens, which may exist in cakes, it is concluded and reemphasized that strict hygienic practices should be applied at every stage of cake production.
