Investigation of Air Quality in Terms of Microbiological Density in Some Regions of the European Side of Istanbul Province

Erguven, Gokhan Onder; Aydin, Sumeyye; Bayhan, Hurrem; Ikizoglu, Bahar

Microbial fauna of the outdoor ambient air of Istanbul has been determined in European side of Istanbul province at four differentdistricts (Bakirkoy, Fatih, Basaksehir and Esenler) by active and passive sedimentation sampling methods. In addition tometeorological factors, the effect of urbanization and traffic density for the air microbial load is also determined. Outdoor airsampling was performed between April 2014 and November 2015. The sedimentation method was done by standart plate count agar(PCA) opening the cover of the petri dishes containing a prepared medium. Standart plate count agar (PCA) medium was used for thedetermination of total bacteria while malt extract agar (MEA) was used for fungi. The colonies formed at the end of this period werecounted and total number of bacteria and fungi per unit volume and area were determined. Air temperature, relative humidity, windspeed and direction were also recorded. As a result of the active sampling method, the most counted units of bacteria was determinedat 35°C in Basaksehir station as 222 CFU(Colony forming unit)/m³, maximum fungal was determined as 286 CFU/m³at 20°Cincubation temperature. The maximum amount of bacteria in the passive sampling method results at 35°C was in Bakirkoy station as9250 CFU/m³.

A research on coliform bacteria in the golden Horn Estuary (Sea of Marmara, Turkey)

Spatio-temporal variation of the total and fecal coliform groups as bacteriological indicators in the marine environment was investigated in surface water of the Golden Horn Estuary in monthly intervals between June 2012 and May 2013. Temperature, salinity, Secchi depth, pH, dissolved oxygen and oxygen saturation were measured at 10 stations simultaneously with bacteriological analyses. Highest total coliform counts in surface water was 940×103 CFU/100 ml at the Alibeykoy Creek in November and highest fecal coliform was 178×103 CFU/100 ml at the same area in October. The lowest coliform counts were observed during summer, while the highest counts were in autumn. During rainy days, some domestic wastewater overflowing from sewerages are discharged to the estuary, which those water adversely affect water quality. The total and fecal coliform counts in surface water increased gradually from the lower to upper section of the estuary. Although the Black Sea water pumping from the Strait of Istanbul to the Golden Horn by Kagithane Creek is an important factor in the decrease of pollution and better water quality, there are still some runoff materials including uncontrolled discharges particularly in the Alibeykoy Creek. The results of this study showed that that the bacteriological pollution increased due to municipal wastewater resulting from overflow after the precipitation and the surrounding waters of the Golden Horn basin mix with surface waters. Thus, the most important factor in order to decrease the bacteriological pollution is to provide a full control of the surface discharges.

Monitoring Aclonifen Remediation in Soil with a Laboratory-Scale Research

Erguven, G. Onder; Bayhan, Hurrem; Demir, Goksel; Ikizoglu, Bahar; Kanat, Gurdal

Aclonifen was used as the most preferable type of herbicide which is used in sunflower farming in the Thrace Region. Bacteria and fungi were isolated from the soil samples taken from the Thrace Region, and a mixed culture was prepared from equal volumes (10(8) CFU/mL) of liquid cultures of microorganisms. Including the witness sample, five different setups were prepared, to which Aclonifen, with a concentration of 1900 mu g/L, was added. Each unit was diluted with distilled water for six weeks. The water filtered at this stage was measured for chemical oxygen demand (COD), biochemical oxygen demand, total organic carbon (TOC), and Aclonifen. Additionally, pH and dissolved oxygen measurement was performed. According to the results of the study, the highest bioremediation was observed in the soil sample to which 10 mL of mixed culture of microorganisms was added and Aclonifen, COD, BOD5, and TOC remediation was observed as 93.2%, 97.8%, 98.8%, and 98.7%, respectively.

ROLE OF SOME ISOLATED SOIL CULTURES ON REDUCTION OF HERBICIDE TRIFLURALIN

    Erguven, Gokhan Onder; Bayhan, Hurrem

The study is about the removal of herbicide Trifluralin as COD parameter and monitoring the population dynamic of isolated cultures. These cultures were isolated from soil samples taken from these areas for their identifications before the use of herbicide. Removal of Trifluralin was determined by microbiological degradation with these produced bacteria and fungi in liquid media. With these growing bacteria and fungi, removal rate by bioremediation method studies were carried out. In addition to biodegradation mechanism, with the measurements of the turbidity; the colony number of microorganisms is monitored with isolated bacteria and fungi species. The removal efficiency for Bacillus simplex was 95% and Clostridium tetani species was 86%, by the fungi Penicillium simplicissimum, and Metacordcyceps chlamydosporia species, were 59% and 80% respectively in five days. Bioremediation study results have shown differences with respect to species in liquid media. Degradation time of these herbicides in liquid culture media with isolated cultures varied between 5 min. and 135 min.
