Background: In cancer, apoptosis relevant proteins-such as CaM kinase, Bcl-2 or P53, topoisomerase I, cell migration feature and DNA/BSA-macromolecules represent significant targets for current chemotherapeutics. Objective: We recently reported two coordination compounds-[Cu(C6H16N2O2)(2)][Ni(CN)(4)] (1) and [Cu(C6H16 N2O2) Pd(CN)(4)] (2)-together with their IR spectra, magnetic properties, thermal analyses and crystal structures. Herein, we describe the ability of these complexes to induce apoptosis in relevant proteins and stimulate topoisomerase I activity, cell migration velocity and DNA/BSA binding properties. Method: The in vitro antiproliferative effects and cell toxicity of both compounds were investigated through pharmacological measurement techniques, and interactions between both compounds and CT-DNA/BSA were studied with UV-Vis spectroscopy and fluorescence spectroscopy. Results & Conclusion: Studies on cells revealed that 2 (i) demonstrated a high antiproliferative effect, which was higher toward HeLa and C6 cancer cells than toward healthy Vero cells; (ii) impaired the migration of HeLa cells; (iii) altered the P53-Bcl-2 ratio in favor of apoptosis; (iv) strongly bound to DNA/BSA macromolecules; and (v) inhibited human topoisomerase I and KpnI or BamHI restriction endonucleases. In conclusion, this preliminary information demonstrates that 2 may represent a promising antiproliferative agent and a potential candidate for a therapeutic approach against HeLa.

Zinc nanoparticles (ZnNPs) are among the least investigated NPs and thus their toxicological effects are not known. In this study, tilapia (Oreochromis niloticus) were exposed to 1 and 10 mg/L suspensions of small size (SS, 40-60 nm) and large size (LS, 80-100 nm) ZnNPs for 14 days under semi-static conditions. Total Zn levels in the intestine, liver, kidney, gill, muscle tissue, and brain were measured. Blood serum glucose (GLU), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and lactate dehydrogenase (LDH) were examined to elucidate the physiological disturbances induced by ZnNPs. Organ pathologies were examined for the gills, liver, and kidney to identify injuries associated with exposure. Significant accumulation was observed in the order of intestine, liver, kidney, and gills. Zn levels exhibited time- and concentration-dependent increase in the organs. Accumulation in kidney was also dependent on particle size; NPs SS-ZnNPs were trapped more effectively than LS-ZnNPs. No significant accumulation occurred in the brain (p>0.05) while Zn levels in muscle tissue increased only marginally (p0.05). Significant disturbances were noted in serum GOT and LDH (p<0.05). The GPT levels fluctuated and were not statistically different from those of controls (p>0.05). Histopathological tubular deformations and mononuclear cell infiltrations were observed in kidney sections. In addition, an increase in melano-macrophage aggregation intensity was identified on the 7th day in treatments exposed to LS-ZnNPs. Mononuclear cell infiltrations were identified in liver sections for all treatments. Both ZnNPs caused basal hyperplasia in gill sections. Fusions appeared in the gills after the 7th day in fish treated with 10 mg/L suspensions of SS-ZnNPs. In addition, separations in the secondary lamella epithelia were observed. The results indicated that exposure to ZnNPs could lead to disturbances in blood biochemistry and cause histopathological injuries in the tissues of O. niloticus. (c) 2016 Wiley Periodicals, Inc. - MakaleAçık Erişim2016Scopus SJRQ1Chronic exposure of tilapia (Oreochromis niloticus) to iron oxide nanoparticles: Effects of particle morphology on accumulation, elimination, hematology and immune responses
Ates, Mehmet; Demir, Veysel; Arslan, Zikri; Kaya, Hasan; Yilmaz, Sevdan; Camas, Mustafa
Effects of chronic exposure to alpha and gamma iron oxide nanoparticles (alpha-Fe2O3 and gamma-Fe2O3 NPs) were investigated through exposure of tilapia (Oreochromis niloticus) to 0.1, 0.5 and 1.0 mg/L (9.2 x 10(-4), 4.6 x 10(-3) and 9.2 x 10(-3) mM) aqueous suspensions for 60 days. Fish were then transferred to NP-free freshwater and allowed to eliminate ingested NPs for 30 days. The organs, including gills, liver, kidney, intestine, brain, spleen, and muscle tissue of the fish were analyzed to determine the accumulation, physiological distribution and elimination of the Fe2O3 NPs. Largest accumulation occurred in spleen followed by intestine, kidney, liver, gills, brain and muscle tissue. Fish exposed to gamma-Fe2O3 NPs possessed significantly higher Fe in all organs. Accumulation in spleen was fast and independent of NP concentration reaching to maximum levels by the end of the first sampling period (30th day). Dissolved Fe levels in water were very negligible ranging at 4-6 mu g/L for alpha-Fe2O3 and 17-21 mu g/L for gamma-Fe2O3 NPs (for 1 mg/L suspensions). Despite that, Fe levels in gills and brain reflect more dissolved Fe accumulation from metastable gamma-Fe2O3 polymorph. Ingested NPs cleared from the organs completely within 30-day elimination period, except the liver and spleen. Liver contained about 31% of alpha- and 46% of gamma-Fe2O3, while spleen retained about 62% of alpha- and 35% of the gamma-polymorph. No significant disturbances were observed in hematological parameters, including hemoglobin, hematocrit, red blood cell and white blood cell counts (p > 0.05). Serum glucose (GLU) levels decreased in treatments exposed to 1.0 mg/L of gamma-Fe2O3 NPs at day 30 (p < 0.05). In contrast, GLU levels increased during the elimination period for 1.0 mg/L alpha-Fe2O3 NPs treatments (p < 0.05). Transient increases occurred in glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and lactate dehydrogenase (LDH). Serum Fe levels did not change during exposure (p > 0.05), but increased significantly within elimination period due to mobilization of ingested NPs from liver and spleen to blood. Though respiratory burst activity was not affected (p > 0.05), lysozyme activity (LA) was suppressed suggesting an immunosuppressive effects from both Fe2O3 NPs (p < 0.05). In contrast, myeloperoxidase (MPO) levels increased significantly in treatments exposed to alpha-Fe2O3 NPs (p < 0.05), and the effect from gamma-polymorph was marginal (p > 0.05). The results indicate that morphological differences of Fe2O3 NPs could induce differential uptake, assimilation and immunotoxic effects on O. niloticus under chronic exposure. (C) 2016 Elsevier B.V. In this study, the effects of exposure to engineered nickel oxide (NiO 40-60 nm) and cobalt oxide (CoO < 100 nm) nanoparticles (NP) were investigated on Artemia salina. Aggregation and stability of the aqueous NP suspensions were characterized by DLS and TEM. Acute exposure was conducted on nauplii (larvae) in seawater in a concentration range from 0.2 to 50 mg/L NPs for 24 h (short term) and 96 h (long term). The hydrodynamic diameters of NiO and CoO NPs in exposure medium were larger than those estimated by TEM. Accumulation rate of NiO NPs were found to be four times higher than that of CoO NPs under the same experimental conditions. Examinations under phase contrast microscope showed that the nanoparticles accumulated in the intestine of Artemia, which increased with increasing exposure concentration. Differences were observed in the extent of dissolution of the NPs in the seawater. The CoO NPs dissolved significantly while NiO NPs were relatively more stable. Oxidative stress induced by the NP suspensions was measured by malondialdehyde assay. Suspensions of NiO NPs caused higher oxidative stress on nauplii than those of CoO NPs. The results imply that CoO and NiO NPs exhibit toxicity on Artemia (e. g., zooplankton) that is an important source of food in aquatic food chain.

Can, Safak Seyhaneyildiz; Demir, Veysel; Can, Erkan
Wastewater generated daily in urban localities could provide nourishment to algae for potential production of biofuel and biochemicals. This study presents an integrated approach to the use of Spirulina platensis cultures for the treatment of domestic wastewater and lipid extraction. In this experimental approach, wastewater subjected only to the primary settling process was used as nutrient media at different concentrations of 100%, 75%, 50%, and 25% wastewater diluted with distilled water, and Spirulina platensis was cultured in these mediums for 20 days. The inoculated medium was incubated at 30 degrees C in the presence of light (42 mu mol photon m(-2).s(-1)) by using light/dark cycle of 12 hours. S. platensis consumed nitrite, nitrate, phosphate and ammonium from wastewater, and it increased its biomass and its lipid amount. Although the highest biomass increase was observed at the wastewater medium without dilution (100% wastewater) (the control group was not included), the highest lipid production was found in 25% wastewater. On the other hand, the percent reduction in the level of nitrite, nitrate, phosphate and ammonium in 75% wastewater medium was 91%, 99%, 99% and 99%, respectively. Despite the low lipid content, since Spirulina platensis can live in harsh conditions and increase its biomass rapidly, they can be easily cultivated in wastewater. Therefore, their use in wastewater for biodiesel production was found appropriate. The resulting algae biomass after oil extraction can be also used in different industrial sectors.

Tanyol, Mehtap; Yonten, Vahap; Demir, Veysel
The study investigated phosphate adsorption from aqueous solutions using chemical- and thermal-modified bentonite in batch system. The adsorbent was characterized by SEM, BET, and FTIR spectroscopy. Contact time, beginning phosphate concentration, pH of the solution, and the effects of the temperature on phosphate adsorption capacity were determined by a series of experimental studies. In a wide pH range (3-10), high phosphate removal yields were obtained (between 94.23 and 92.26 %), and with the increase in temperature (from 25 to 45 degrees C), phosphate removal increased. Langmuir and Freundlich isotherms were used to determine the sorption equilibrium, and the results demonstrated that equilibriumdata displayed better adjustment to Langmuir isotherm than the Freundlich isotherm. Phosphate sorption capacity, calculated using Langmuir equation, is 20.37 mg g(-1) at 45 degrees C temperature and pH 3. Mass transfer and kinetic models were applied to empirical findings to determine the mechanism of adsorption and the potential steps that control the reaction rate. Both external mass transfer and intra-particle diffusion played a significant role on the adsorption mechanism of phosphate, and adsorption kinetics followed the pseudo-second-order-type kinetic. Furthermore, thermodynamic parameters (Delta H degrees, Delta G degrees, Delta S degrees) which reveal that phosphate adsorption occur spontaneously and in endothermic nature were determined. The results of this study support that bentonite, which is found abundant in nature and modified as an inexpensive and effective adsorbent, could be used for phosphate removal from aqueous solutions.

The effects of alpha-iron oxide (alpha-Fe2O3) and gamma-iron oxide (gamma-Fe2O3) nanoparticles (NPs) on marine microalgae species (Nannochloropsis sp. and Isochrysis sp.) were investigated in this study. Both Fe2O3 NPs covered the surface of algae with the agglomerates of the nanoparticles. This form of physical NP toxicity significantly decreased the sizes of phytoplankton. Both NPs were toxic to the tested algal species, while alpha-Fe2O3 showed less toxicity than gamma-Fe2O3 NPs for both algal species. A comparative analysis of growth data of the two algal species treated with alpha-Fe2O3 or gamma-Fe2O3 NPs revealed that Isochrysis sp. are more sensitive than Nannochloropsis sp. Toxicity of these widely used NPs to primary producers forming the base of the food chain in aquatic environments might result in widespread adverse effects on aquatic environmental health.

Ates, Mehmet; Arslan, Zikri; Demir, Veysel; Daniels, James; Farah, Ibrahim O.
Dietary and waterborne exposure to copper oxide (CuO) and zinc oxide (ZnO) nanoparticles (NPs) was conducted using a simplified model of an aquatic food chain consisting of zooplankton (Artemia salina) and goldfish (Carassius auratus) to determine bioaccumulation, toxic effects, and particle transport through trophic levels. Artemia contaminated with NPs were used as food in dietary exposure. Fish were exposed to suspensions of the NPs in waterborne exposure. ICP-MS analysis showed that accumulation primarily occurred in the intestine, followed by the gills and liver. Dietary uptake was lower, but was found to be a potential pathway for transport of NPs to higher organisms. Waterborne exposure resulted in about a 10-fold higher accumulation in the intestine. The heart, brain, and muscle tissue had no significant Cu or Zn. However, concentrations in muscle increased with NP concentration, which was ascribed to bioaccumulation of Cu and Zn released from NPs. Free Cu concentration in the medium was always higher than that of Zn, indicating CuO NPs dissolved more readily. ZnO NPs were relatively benign, even in waterborne exposure (p0.05). In contrast, CuO NPs were toxic. Malondialdehyde levels in the liver and gills increased substantially (p<0.05). Despite lower Cu accumulation, the liver exhibited significant oxidative stress, which could be from chronic exposure to Cu ions. (c) 2014 Wiley Periodicals, Inc. Environ Toxicol 30: 119-128, 2015.

In this study, Artemia salina (crustacean filter feeders) larvae were used as a test model to investigate the toxicity of aluminum oxide nanoparticles (Al2O3 NPs) on marine microorganisms. The uptake, toxicity, and elimination of -Al2O3 (50 nm and 3.5 m) and -Al2O3 (5 nm and 0.4 m) NPs were studied. Twenty-four and ninety-six hour exposures of different concentrations of Al2O3 NPs to Artemia larvae were conducted in a seawater medium. When suspended in water, Al2O3 NPs aggregated substantially with the sizes ranging from 6.3 nm to >0.3 mu m for spherical NPs and from 250 to 756 nm for rod-shaped NPs. The phase contrast microscope images showed that NPs deposited inside the guts as aggregates. Inductively coupled plasma mass spectrometry analysis showed that large particles (3.5 m -Al2O3) were not taken up by Artemia, whereas fine NPs (0.4 m -Al2O3) and ultra-fine NPs (5 nm -Al2O3 and 50 nm -Al2O3) accumulated substantially. Differences in toxicity were detected as changing with NP size and morphology. The malondialdehyde levels indicated that smaller -Al2O3 (5 nm) NPs were more toxic than larger -Al2O3 (0.4 mu m) particulates in 96 h. The highest mortality was measured as 34% in 96 h for -Al2O3 NPs (5 nm) at 100 mg/L (LC50 > 100 mg/L). -Al2O3 NPs were more toxic than -Al2O3 NPs at all conditions. (c) 2013 Wiley Periodicals, Inc. Crayfish (Astacus leptodactylus Eschscholtz, 1823) is the native crayfish species in Turkey. It was exported regularly to Western Europe. In this study, bioaccumulation and temporal trends of some trace elements (arsenic: As, cadmium: Cd, copper: Cu, mercury: Hg, lead: Pb, and zinc: Zn) in edible abdomenmuscle of crayfish from Keban Dam Lake (Elazig, Turkey) were investigated for the 2006-2012 period. Sequence of metal concentration levels was Zn > Cu > Hg > Pb > Cd > As in muscle tissues. The highest concentration of Zn (21.69mg kg(-1)) was detected in 2006, while the lowest (4.35mg kg(-1)) in 2009. In general, it was found that the concentrations of trace elements investigated were lower than the maximum permissible limits of the food regulations of the Ministry of Food, Agriculture, and Livestock (MFAL), the Turkish Food Codex and Commission Regulation (EC). If the crayfish selected for the study are recognized as bioindicators of environmental pollution, then it is possible to conclude that the changes in studied trace elements concentrations in the Keban Dam Lake are being steady.

Batch tests were conducted on several Texas native bioretention media to estimate the removal capacity of lead and copper from contaminated water and to understand the effect of the presence of phosphate (due to application of phosphate based fertilizers in the watershed) on metal removal. The results show that the 77% of lead and 58% of copper precipitated as the phosphate added to the contaminated water even before adding any media. Cedar mulch and hardwood mulch performed best and removed 99.2% of lead and 97.5% of copper from aqueous solution. When phosphate was added, removal increased to 97.8% for copper and 100% for lead (under detection limits). The removal capacity of metals was with the order of cedar mulch > hardwood mulch > pine mulch > sawdust > loam soil > silica sand. The leaching of metals from the media was much lower than the original solution and decreased when phosphate was added.

The environmental and geological condition of a current solid waste disposal site in the city of Tunceli, Turkey was investigated. Improper geological structure and soil properties of this unsanitary solid waste storage site as well as leachate from this site caused significant pollution in air, soil, and surface water. Natural soil characteristics and geology of the site are presented in this paper. Petrographic properties of vulcanized rubber rocks composed of tuff and volcanic sandstone at the bottom of storage sites and the mineralogical composition of the base composed of these rocks were examined. Permeability of the natural soil and index properties were determined using soil mechanics experiments (moisture content, specific gravity, Atterberg limit tests, sieve analysis, hydrometer analysis, falling-head permeability test, and standard proctor test) conducted on disturbed samples collected from excavated sites and the relationship between mineralogical composition and soil hydraulic behavior was investigated. Natural soil samples of six different parts of the site indicated that, the soil class in four samples is silty sands, sand-silt mixtures, one is clayey sands, sand-clay mixtures and one is clayey silts with slight plasticity. The values obtained using permeability tests for subsoil samples vary from 10(-5) to 10(-8) cm/s. Low permeability (10(-7) and 10(-8) cm/s) at the south part of the site increases to 10(-5) cm/s at the natural soil of the North side. Extensively faulted and fissured volcanic sandstone levels at the site give potential to waste leachate to reach deep levels. The soil composition, hydraulic conductivity, topographic conditions, and other location criteria in the current solid waste disposal site showed that the current disposal site is not suitable for this purpose. Environmental monitoring of leachate quality from an open municipal solid waste dumping site in Tunceli, Turkey was studied in this research. The most commonly examined pollution parameters were determined on a seasonal basis. The annual average 5-day biological oxygen demand (BOD5) and chemical oxygen demand (COD) values of station points were measured as 70 and 425 mg/L, respectively, and also the average BOD5/COD ratio (a measure of biodegradability) was calculated as 0.20. The low ratio of biodegradability and slightly alkaline pH values in the leachate samples indicated that the site was characterized by methanogenic conditions. The mean ammonium-nitrogen (NH4 (+)-N) and corresponding phosphate (orthophosphate) values were assayed as 70 and 11 mg/L, respectively. The average solids content in the leachates was measured as 4,681 mg/L (total solids) and 144 mg/L (suspended solids). Very low concentrations of iron, manganese, copper, and zinc in the leachate samples were found and the concentration of cadmium was measured below detection limits. Excessive amount of nutrients and high organic and inorganic pollutant content in the leachates pose serious pollution potential to the environment. Since no drainage system or bio treatment exists in this open dumping site, high permeability of natural soil at the site and in the surrounding area and very fractured and crackled rocks under natural soil are indicators of high groundwater pollution potential in this site.

The increased use of nanosized materials is likely to result in the release of these particles into the environment. It is, however, unclear if these materials are harmful to aquatic animals. In this study, the sublethal effects of exposure of low and high concentrations of titanium dioxide nanoparticles (TiO2 NPs) on goldfish (Carassius auratus) were investigated. Accumulation of TiO2 NPs increased from 42.71 to 110.68 ppb in the intestine and from 4.10 to 9.86 ppb in the gills of the goldfish with increasing exposure dose from 10 to 100mg/L TiO2 NPs. No significant accumulation in the muscle and brain of the fish was detected. Malondialdehyde as a biomarker of lipid oxidation was detected in the liver of the goldfish. Moreover, TiO2 NPs exposure inhibited growth of the goldfish. Although there was an increase (8.1%) in the body weights of the goldfish for the control group, in the low and high exposure groups 1.8% increase and 19.7% decrease were measured, respectively. The results of this study contribute to the current understanding of the potential ecotoxicological effects of nanoparticles and highlight the importance of characterization of NPs in understanding their behavior, uptake, and effects in aquatic systems and in fish.

This study is aimed to determine the bioaccumulation of copper and zinc in hemolymph of freshwater crayfish (Astacus leptodactylus) fed with zebra mussel (Dreissena polymorpha) collected from Keban Dam Lake. This is the first study on bioaccumulation of Cu and Zn in hemolymph of males and females of A. leptodactylus. The average concentrations of Cu and Zn in the hemolymph samples were found out as 0.54 and 35.07 ppm respectively for male and 0.42 and 15.59 ppm respectively for female control freshwater crayfish. The levels of Zn in the hemolymph of both male and female ones among control and diet groups were statistically significant (P < 0.05). The Cu levels among control and diet groups in female ones were statistically significant (P < 0.05) but not in the male ones (P > 0.05). The results of current study revealed that feed treatment with boiling was not effective at the levels of metal bioaccumulation in hemolymph. It was observed that this metal bioaccumulation is more related with sex differences.
