Küçükgül, Azime; Küçükgül, Altug; Korkmaz, Tulay Metin

Fish cells have been accepted as an alternative to in vivo assay for inflammatory effects of therapeutic experimental systems. To reveal the anti-inflammatory effects of Inula viscosa (IV) fish cell line RTG-2, derived from the gonadal tissue of rainbow trout, was infected with lipopolysaccharide (LPS). Effective concentrations for different concentrations of LPS (1, 5, 10, 20 mu M) and IN (1, 5, 10, 20 mu g ml(-1)) were determined. While the 20 mu M concentration of LPS, which was effectively selected from preliminary tests, caused 27% cell loss, the effective 1 mu g ml-1 concentration of IN caused 1.1% proliferation in the cells when compared to the control group. All pro-inflammatory parameters investigated in LPS-induced RTG-2 cells showed up-regulation, with the highest increase in TNF-alpha gene expression level (11.3 fold changes). Down-regulation was determined in the IN together with LPS administered group and IL-1 beta had the highest effect with 96%. IL-6 protein level decreased by LPS at a rate of 4% and IN together with LPS increased by 9%. The IN exhibited significant efficacy against inflammation caused by LPS. However, further studies are needed to determine pharmacological activity of I. viscosa in details.

Küçükgül, Azime; Küçükgül, Altuğ; Gönenci, Ramazan; Özsoy, Şule Yurdagül; Kutlu, Banu; Işgör, Mehmet Mustafa

Yersinia ruckeri is the causal agent of enteric redmouth (ERM) in trout. This study aimed to investigate the effectiveness of nontoxic ozone on Y. ruckeri-infected trout cells in in vitro conditions. Rainbow trout macrophages (RTS-11) were exposed to Y. ruckeri for 24 h after being incubated with the ozonized medium at nontoxic concentration. Effective concentrations of nontoxic ozone (3 gamma) and bacteria (1.5 x 108 cfu/mL) were determined by viability tests. The expression of caspases 1, 3, 8, and 12 was determined by real-time PCR and protein levels investigated by ELISA method. Apoptotic-necrotic cell ratios were determined by acridine orange and ethidium bromide staining. Bacterial stimulations of caspases (1, 3, 8, and 12) were suppressed by ozone mostly at the translational level. While bacterial infection increased the count of apoptotic and necrotic cells at different stages, ozone significantly reduced this condition. Obtained data indicate possible use of nontoxic ozone as a safe and effective treatment alternative for increasing host defenses and eliminating the infection in the case of yersiniosis in fish.

    Küçükgül, Azime

The presence of different phytochemical components of essential oil such as tannins, alkaloids, terpenoids and phenolic compounds has antibacterial, antifungal, and anti-inflammatory effects. The aim of this study is to investigate importance of the major components of three herb essential oils (Thymus vulgaris L., Centauriumerythraea Rafn. And Foeniculumvulgare Mill) on challenging with fish diseases. The components of essential oils provided from a commercial firm were made GC/MS analyzes. The major component of T. vulgarewas carvacrol called as phenol, 2-methyl-5-(1-methylethyl) with 40%.The others were Linalool L (15.11%) and benzene, methyl(1-methylethyl)- (12.12%).The richest oil in C. erythraea was bicyclo[3.1.1]hept-2-ene, 2,6,6-trimethyl- (34.90%) called as alpha-pinene, followed by heptacosane (19.15%) and dotriacontane (17.72%), respectively. The evaluation of the essential oil of F. vulgarispresented benzene, 1-methoxy-4-(1-propenyl)- with 67.99%, followed by dl-Limonene (16.03%) and benzene, 1-methoxy-4-(2-propenyl)- (6.97%). The therapeutic effects of thyme are due to its high content of phenolic compounds, particularly carvacrol. The most important compounds of F. vulgare essential oil is anethole, fenchone, limonene that has antibacterial, antioxidant, antifungal and anticancer effects. The previous studies show that alpha pinene significantly inhibits many pathogenic Gram-negative bacteria. In aquaculture studies, in vitro and in vivo effects of the dominant compounds of essential oils in our study are consistent with the previous findings.

Yersinia ruckeri ile Enfekte Gökkuşağı Alabalığı Makrofajlarında Ozonoterapinin Antiapoptotik Etkinliğinin Araştırılması

Otay, Tuba; Küçükgül, Altuğ; Küçükgül, Azime; Özsoy, Şule Yurdagül; Kutlu, Banu; Gonenci, Ramazan

Yersiniosis is a systemic disease caused by Yersinia ruckeri, which is particularly affecting Enteric red mite (ERM) disease in trout. There are still many issues regarding the mechanism of apoptosis and necrosis that play a role in the pathogen-host relationship in the disease. In the project, it was aimed to maintain the life of the cells and killing the bacteria by providing anti-apoptotic and anti-necrotic effect via application of therapeutic concentration of ozone to the macrophages infected with Y. ruckeri. In this study, the cells of the rainbow trout-originated cell line (RTS-11, mononuclear / macrophage) were exposed to the appropriate number of bacteria (Yersinia ruckeri, ATCC- 29473, serotype 1) for 24 hours after being incubated with ozonized medium (30 min) at the nontoxic concentration. For this purpose, effective concentrations of nontoxic ozone and bacteria were demonstrated by viability tests (3 Gamma, 1.5x108, respectively). After incubation times, the expression levels of caspases 1,3,8 and 12 were determined by realtime polymerase chain reaction and protein levels were investigated by ELISA method from the obtained lysates. In addition, apoptotic-necrotic cell ratios were determined in acridine orange and ethidium bromide staining in experimental groups. According to the findings, bacterial infection caused 47% loss of cells, while nontoxic ozone was found to protect the cells by 27%. Again, while bacterial infection increased apoptotic cells in different processes, ozone was found to significantly reduce this rate. At the same time, ozone suppressed the rate of necrosis caused by bacteria. It was also found that bacterial stimulation of caspases (1, 3, 8 and 12), which were effective in the initiation of apoptosis and necrotic processes and in the formation of cell loss, were suppressed by ozone pre-administration and mostly at the level of protein translation. According to the obtained data, Yersinia ruckeri infection caused cell loss in the macrophages of rainbow trout by stimulating both apoptosis and necrosis. However, nontoxic ozone significantly improved this negative situation. In this context, it has come to the conclusion that nontoxic ozone can be used as a safe and effective treatment alternative for increasing host defenses and eliminating the infection in the case of yersiniosis in fishes. However, in order for this argument to be fully understood, it is inevitable to support it with advanced molecular and in vivo studies.

Activity of Apoptotic and Antiinilammation on Rainbow Trout (Oncorhynchus mykiss Walbaum, 1792) Infection-Induced Experimentally

Küçükgül, Altuğ; Küçükgül, Azime; Kubulay, Banu

In this study investigated thymol effects on gene expressions relative to pro-inflammatory and apoptosis in tissue fish infected four different trials were formed. All fish were infected with E. coli LPS (25 pg ml'l) expect control and only thymol (THY) supplementation (100 pg ml'1 diet) group fish. The last group was the infected fish fed with thymol supplementation diet (+THY). Kidney and liver tissues were removed day after the experiment period. The gene expression levels of interleukin-1B (IL-1B) and interferon gamma (INF-y) from pro-inflammatory cytokines; caspase (Cas 3) and caspase (Cas 8) from apoptotic cytokines were investigated by Real-Time PCR analyses. Decreases recorded on gene levels of IL—lB and IFNy by LPS treatment, however it was observed that returned to the control level. The comparative results ofLPS and +THY treatment showed that all gene levels significantly stimulated by up regulation. The expression levels of Cas and following LPS caused the increases in liver, no in kidney. However, any change was not observed in only diet application (THY). +THY application showed about 3-fold up-regulations on all gene expressions including apoptosis. It was determined to be effective by showing changes on gene levels including pro-inflammatory and apoptosis. However, further studies are required to better understand immune modulatory effect of diet supplementation feeding on the virulence mechanism offish-infected.
