    Kandemir, F. M.; Sagliyan, A.; Ozkaraca, M.; Gunay, C.; Han, M. C.; Benzer, F.

This study examined the effects of orally given pomegranate seed extract (PSE) on healing of surgically induced full thickness skin wounds in terms of clinical, histopathological and biochemical aspects. A total of 48 healthy, adult male New Zealand rabbits were randomly divided in 2 equal experimental (PSE group) and control groups (C): one week prior the surgery, 100 mg/kg per day via gastric catheter pomegranate seed extract was orally given for 20 days to the assay animals. After one week, all rabitts were anaesthetized by intramuscular injection of xylazine HCl (5 mg/kg) and ketamine HCI (50 mg/kg) and after shaving and disinfection, a circular full thickness skin defect of 2.5 cm diameter was incised on the median line of the back area. Macroscopic and histopathological aspects as well as the tissue oxidant/antioxidant balance were determined on 6 rabbits from each group slaughtered at the 5th, 10th, 15th and 20th days post-surgery.

Beneficial effects of oral administrations of grape seed extract on healing of surgically induced skin wounds in rabbits

    Sagliyan, A.; Benzer, E.; Kandemir, F. M.; Gunay, C.; Han, M. C.; Ozkaraca, M.

This study examined the effects of orally given grape seed extract (GSE) on healing of surgically induced full thickness skin wounds in terms of clinical, histopathological and biochemical aspects. A total of 48 healthy. adult male New Zealand rabbits were randomly divided in 2 equal experimental and control groups: one week prior the surgery, 260 mg/kg grape seed extract was orally given for 20 days to the assay animals. A circular full thickness skin wound (2.5 cm in diameter) was made in the back midline of all rabbits under anaesthesia then, macroscopic and histopathological aspects as well as the tissue oxidant/antioxidant balance were determined on 6 rabbits from each group slaughtered at the 5th, 10th, 15th and 20th days post-surgery. In treated rabbits, it was clinically observed that the wound contraction was significantly amplified and the histological processes of re-epithelisation, granulation tissue development, collagen accumulation and angiogenesis were markedly hastened whereas the inflammatory reaction and the ulcer disappeared more rapidly than in the not-treated controls. Furthermore, the maximal skin MDA formation observed on the 10th day was significantly depressed and the antioxidant catalase (CAT) and glutathione peroxidase (GSH-Px) activities remained significantly elevated until the 15th day compared to the controls. These results demonstrate the beneficial effects of treatment with GSE on skin wound healing in rabbits.

Plasma nitric oxide concentrations and erythrocyte arginase activities in lambs with contagious ecthyma

    Kandemir, F. M.; Issi, M.; Benzer, F.; Gül, Y.; Basbug, O.; Özdemir, N.

The aim of the present study was to investigate the plasma nitric oxide concentrations and the erythrocyte arginase activities in lambs with contagious ecthyma, which frequently occurs in lambs and goat kids. The study was conducted on 10 Akkaraman, 1-2 month old lambs with contagious ecthyma (the clinical diagnosis was based on the presence of typical cauliflower-like proliferative lesions in lips, commissum labials, nostrils and udders) and on 10 apparently healthy lambs (controls) from the same flock. Plasma NO concentrations and erythrocyte arginase activities were determined on all animals using the Griess method and the thiosemicarbazide diacetylmonoxime urea (TDMU) method, respectively. Compared to the control group, the mean plasma NO concentration was significantly increased in the diseased group while the mean arginase activity was markedly depressed. Abnormally high NO concentrations and low enzyme activities were observed in 9 and 10 diseased lambs, respectively. Additionally, a negative correlation was evidenced between the 2 biochemical parameters. These results show that this viral infection is associated with some perturbations in the arginine metabolism, leading preferentially to the synthesis of NO, which is presumed to exhibit some antiviral properties.

Effect of Pomegranate Seed Extract on Free Radical Damage and Antioxidant Activity Under Cisplatin-Induced Oxidative Stress Conditions in Rabbit Testes

    Benzer, F.; Kandemir, F. M.; Yildirim, N. C.; Ozan, S. T.

The aim of this study is to evaluate the protective effect of pomegranate seed extract on cisplatin-induced oxidative stress in rabbits testes. Rabbits were divided into three groups; (i) being control group, (ii) cisplatin group and (iii) cisplatin + pomegranate seed extract group. The level of malondialdehyde and glutathione, the activity of catalase, glutathione peroxidase, were measured in testes tissue of rabbits. Malondialdehyde levels were increased depending on cisplatin. In the case of cisplatin + pomegranate seed extract, malondialdehyde levels were found to be lower than cisplatin group (p < 0.05). In cisplatin group, catalase, glutathione peroxidase activity and glutathione levels were decreased when compared to control but in cisplatin + pomegranate seed extract group, catalase, glutathione peroxidase activity and glutathione levels were found higher than cisplatin group (p < 0.05). The pomegranate seed extract supplementation may play a protective role or decrease the side effects of cisplatin-induced testes toxicity from cancer medicines.

Compensatory effects of curcumin on cisplatin-induced toxicity in rabbit testis

    Kandemir, F. M.; Benzer, F.; Yildirim, N. C.; Ozdemir, N.

Curcumin, a widely used spice and colouring agent in food, has been shown to possess potent antioxidant, antitumor promoting and anti-inflammatory properties in vitro and in vivo. The present study was designed to investigate the protective effects of curcumin on changes in the levels of lipid peroxidation and endogenous antioxidants induced by cisplatin (cis-diamminedichloroplatinum II, CDDP) in the testiclular tissue of rabbits. 18 healthy male New Zealand white rabbits were equally divided into three groups of six rabbits each, control, cisplatin, and cisplatin+curcumin. The degree of protection produced by cisplatin was evaluated by determining the level of malondialdehyde (MDA) and glutathione (GSH), the activity of catalase (CAT), glutathione peroxidase (GSH-Px), were estimated from testes homogenates. MDA levels were increased with cisplatin compared to control but in cisplatin+curcumin group, MDA levels were found to be lower than cisplatin group (p<0.05). The activity of CAT and GSH-P-X was decreased in cisplatin and cisplatin+curcumin groups compared to control (p<0.05). In the case of cisplatin+curcumin CAT and GSH-Px activity were increased compared to cisplatin group (p<0.05). GSH levels were decreased with cisplatin but administration cisplatin+curcumin increased the levels of GSH comared to cisplatin group (p<0.05). In the present study, co-administration of curcumin with cisplatin prevented the damage to testes induced by this drug and may be considered as a potentially useful candidate in the combination chemotherapy with cisplatin.

MALONDIALDEHYDE LEVELS AND GLUTATHIONE PEROXIDASE ACTIVITIES IN THE TISSUES OF FRESHWATER CRAYFISH (ASTACUS LEPTODACTYLUS ESCH 1823) IN CULTURE CONDITION

    Kandemir, F. M.; Benzer, F.; Erisir, M.; Yildirim, N. C.

The objective of the study was to characterize the effects of sex on lipid peroxidation (as malondialdehyde, MDA), antioxidant defence system (as glutathione peroxidase, GSH-Px) in the hepatopancreas, gonad and muscle tissues of freshwater crayfish, Astacus leptodactylus, in culture condition. The crayfish used in the present study was provided from Keban Dam Lake population of A. leptodactylus. 34 males and 30 females crayfish in different size were hold in stock ponds (6 x 6 x 0.5 m). This study was carried out between June 18, 2004 and December 1, 2004 at the crayfish reproduction unit of Aquaculture Faculty of Firat University, Elazig, Turkey. Plastic pipes (20 cm in length and 7 cm in diameter) were provided as shelters for the crayfish. Crayfish were fed 2 % of their total wet weight daily with prepared ration. Crayfish were exposed to natural photoperiod. During the study, the mean water temperature, dissolved oxygen and mean pH were 4 degrees C, 6.28 +/- 0.32 mg L-1 and 7.82 +/- 0.17, respectively. MDA and GSH-Px levels according to sex and tissue-species were determined. MDA level in the hepatopancreas and gonad of females were higher than males, except muscle tissue (P<0.001). However, the GSH-Px activities in the hepatopancreas and gonad of males were higher than females, except muscle tissue (P<0.001). As a result, MDA and GSH-Px levels in the hepatopancreas, gonad and muscle tissues of freshwater crayfish, Astacus leptodactylus, in culture condition changed both tissues and sex.
