Docosahexaenoic acid (DHA) was the major polyunsaturated fatty acid (PUFA) in the sperm plasma membrane, and the levels of PUFAs decreased in treated groups during the cryopreservation process. The supplementation of TT (Tribulus terrestris) to Cyprinus carpio sperm at 200 µg∙L-1 positively affected the maintenance of motility parameters and FAs levels after the freeze-thaw cycle, and TT can be used as an herbal medicine instead of synthetic pharmaceuticals to improve the productive performance in aquaculture.

Kocabaş, Filiz Kutluyer; Kocabaş, Mehmet; Benzer, Fulya; Erişir, Mine

Anti-oxidants are vital for protecting sperm and can mitigate the negative effects associated with cryopreservation. This study was conducted to evaluate the protective effects of adding anti-oxidants (vitamins and amino acids) to extender in sex-reversed rainbow trout (Oncorhynchus mykiss) sperm. The collected sperm was diluted at the ratio of 1: 5 by the extenders supplemented with different anti-oxidants including 1.00 mM of ascorbic acid and L-tryptophan, 2.00 mM of cysteine and α-tocopherol, and their combination. After dilution, the semen was aspirated into 0.50 mL straws, and the straws were placed on the tray, frozen for 10 min, and plunged into liquid nitrogen. Straws were thawed in a 30.00 ˚C water bath for 15 sec. The Sperm Class Analyzer System was used to evaluate sperm kinematics. The activity of anti-oxidant enzymes and lipid peroxidation were determined as oxidative stress indices. Our data indicated that the incorporation of anti-oxidants and amino acids increased sperm motility duration. The elevated activity of glutathione peroxidase, superoxide dismutase, and catalase in post-thaw samples indicates that the anti-oxidant system in sex-reversed rainbow trout sperm likely plays a crucial role in protecting membrane compounds from oxidation. In conclusion, the combination of 1.00 mM L-tryptophan and ascorbic acid to the extender media caused a prolonging effect in sperm motility after thawing and they have the potential to serve as effective agents for improving sperm cryosurvival.

As a potential pollution source, slaughterhouse wastewater (SWW) with development of the meat production and processing sector is a threat in the world. Therefore, the determination of economical, favourable, cost-effective and proper treatment method is crucial for sustainable environmental protection. This study focused on the performance and efficiency of a two-stage process combining biological treatment (BT) with freshwater snails and electrocoagulation (EC) of SWW. Viviparus contectus, Planorbis rubrum, Melanoides tuberculata were used in primary treatment while secondary treatment was realized with EC process. The EC of SWW was performed for reducing both turbidity and chemical oxygen demand (COD) concentration to levels compliant with direct-discharge regulations. In addition, the stirring speed, current density, initial pH and reaction time were assessed for evaluation the optimum EC condition. Oxidative stress biomarkers and some antioxidants were assessed in freshwater snails exposed to SWW after 24 and 96 h. The best removal efficiencies of COD (91.43%) and turbidity (92.42%) were obtained at pH 5 and 20 min of operational time by P. rubrum. Notably, MDA (Malondialdehyde) was highest level in M. tuberculata compared to V. contectus and P. rubrum. Overall, removing organic and inorganic pollutants from SWW could be provided with the hybrid process BT/EC as a combining approach.

First Record of Black Sea Trout (Salmo labrax) and Its Potential Hybrids with Endemic Munzur Trout (Salmo munzuricus) in Uzunçayır Dam Lake, Tunceli

Ölçülü, Abdullatif; Düzgüneş, Zehra Duygu; Kocabaş, Filiz Kutluyer; Kızak, Volkan; Kocabaş, Mehmet

The depletion of unique genetic resources and increased risk of extinction may result from hybridization between native and introduced species. In the present study, morphological and molecular analysis were performed on two putative natural interspecific hybrids of the genus Salmo and their parent species, Salmo labrax and Salmo munzuricus, collected from Uzunçayır Dam Lake, which is fed by Munzur and Pülümür streams. Molecular analyses revealed the presence of S. labrax in Uzunçayır Dam Lake for the first time. Phylogenetic analysis using the cytochrome b (Cytb) gene of mitochondrial DNA to determine the maternal origin revealed that hybrid individuals belonged to the same clades as S. labrax or S. munzuricus. Recombination activating gene 1 (Rag1) sequence of nuclear DNA was analyzed for paternal analysis and it was determined that hybrid individuals carried similar single nucleotide polymorphisms (SNPs) to S. labrax or S. munzuricus. These findings support that individuals derive from different parental species. Therefore, evidence from trout samples assumed to be natural hybrids in this study suggests interspecific hybridization between S. labrax and S. munzuricus, but further studies are required. Kocabaş, Filiz Kutluyer; Kocabaş, Mehmet; Çanakçi, Aykut; Karabacak, A. Hasan

Mussels have great annual production in the world and their shells are discarded as a waste. Shells are calcium and carbon accumulators and, formed as a consequence of calcium carbonate biomineralisation. They can be used as bio‐based composite materials in different fields. Hence, assessment of the chemical compound content and microstructure of shells is important for the prospective utility. For these reasons, structure of Cerastoderma edule, Chamelea gallina, Donax trunculus, Ruditapes decussatus shells were analyzed with X-ray diffraction (XRD), Fourier Transform Infrared Spectroscopy (FT-IR) and Scanning Electron Microscope-Energy Dispersive Spectroscopy (SEM-EDS). In addition, microhardness was measured in shells. Our results indicated that shells have a flake like structure with irregular grains appearance morphologically at micrometric scale. The fractions of aragonite in particular shells were assessed. EDS analysis showed that Ca, C and O were major elements of C. edule, C. gallina, D. trunculus, R. decussatus in accordance with XRD data. The lowest microhardness (2.68±0.16 GPa) was in D. trunculus while the highest microhardness (4.28±0.32 GPa) was in C. edule.

Maulida, Siti; Eriani, Kartini; Fadli, Nur; Kocabaş, Filiz Kutluyer; Siti-Azizah, Mohd Nor; Wilkes, Martin; Muchlisin, Zainal Abidin

The climbing perch, Anabas testudineus is a freshwater fish that has economic value in Indonesia. It is cultured in the country, but the breeding technology, specifically sperm storage, is not well developed. Sperm cryopreservation is one of the preservation methods that need to be developed to support fish breeding technology. The type of cryoprotectants and its concentration are species-dependent and determines the success of this approach. Therefore, this study is aimed at determining the optimal type and concentration of cryoprotectant for sperm cryopreservation of A. testudineus. Four separate study series were performed, each of which evaluated one type of cryoprotectant at five concentration levels. The cryoprotectants used were DMSO, methanol, glycerol, and ethanol, and the tested concentrations were 0%, 5%, 10%, 15%, and 20%, which were combined with 5% egg yolks. Each treatment was conducted with three replications. The results showed that the type of cryoprotectant and its concentration significantly affected sperm motility, viability, and fertility of climbing perch (P < 0.05). The best outcome was obtained in DMSO, and methanol at a concentration of 10%, glycerol at 5%, and ethanol at 15%. However, the highest motility, viability, and fertility values were observed at 10% DMSO, indicating it is the best type and concentration for sperm cryopreservation of climbing perch A. testudineus.

New Approaches to Predict the Sperm Quality by the Spermatozoal miRNA Content in Fish: A Review

    Kocabaş, Filiz Kutluyer; Kocabaş, Mehmet

The survival of the progeny and fertilization success depend on high-quality mature spermatozoa containing DNA and coding and non-coding RNAs besides the female gamete quality. Sperm cells and gonads in fish contain different RNA classes. It is possible that the development of fish could be affected by miRNA and/or other non-coding RNAs being abundant in Teleost fish and having important functions. Studies about the presence of miRNA in some fish species (e.g. Danio rerio, Salmo salar, Carassius auratus, Cyprinus carpio) showed that sperm motility is regulated by miRNA. Nevertheless, limited knowledge and several hypotheses have been presented about classes and content of RNAs in fish spermatozoa. More research about the effects of miRNA on fish reproduction and sperm quality are needed for population, aquatic ecosystems, and broodstock management, and the evaluation of molecular sperm quality parameters with miRNA could be used together with traditional techniques.

Ascorbic acid ameliorated the sperm quality of rainbow trout (Oncorhynchus mykiss) against arsenic toxicity: Impact on oxidative stress, fertility ability and embryo development

Kocabaş, Filiz Kutluyer; Kocabaş, Mehmet; Aksu, Önder; Çakir Sahilli, Yeliz

Arsenic (As) is a heavy metal and aquatic pollutant and adversely impacts the reproduction of male fish. As a chain-breaking antioxidant, ascorbic acid (AA) has high water solubility and low toxicity. In this context, the current study was performed to assess the protective role of AA (1 mM) on the sperm cells of the rainbow trout (Oncorhynchus mykiss) exposed to sublethal concentrations of As (8, 16 and 32 mg/L). Sperm quality parameters were analyzed using a sperm class analyzer system. Lipid peroxidation and antioxidant enzyme levels were used as indicators of oxidative stress. The fertilization, eyeing and hatching rates were determined as gamete markers. Reduced sperm quality parameters and fertility capacity resulted from in vitro exposure to As (P < 0.05). The oxidative stress in sperm cells increased after As exposure (P < 0.05). The presence of AA improved sperm movement parameters and fertility potential (P < 0.05). Overall, AA had a positive effect on oxidative stress and fertility ability against As toxicity and AA supplementation ameliorated detrimental effects of As in sperm cells.

Aroclor 1254 impairs sperm quality, fertilization ability, and embryo development of rainbow trout (Oncorhynchus mykiss)

Kocabaş, Mehmet; Kocabaş, Filiz Kutluyer; Aksu, Önder; Benzer, Fulya; Erişir, Mine; Sahilli, Yeliz Çakir

The polychlorinated biphenyls (PCBs) in aquatic environment adversely affect non-target organisms, including fish. Especially, the male reproduction and next generation can be damaged through high exposure to these pollutants. Hence, the sperm cells were exposed to sublethal concentrations of Aroclor 1254 (0, 1, 5, 10, or 25 mg/l) for 4 h. The sperm quality parameters were analyzed by SCA (Sperm Class Analyzer). The fertility, eyeing, and hatching rates were determined as gamete markers. Lipid peroxidation (malondialdehyde-MDA), glutathione (GSH), and antioxidant enzymes [superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT)] were measured for determination of oxidative stress. Our results showed that Aroclor 1254 negatively affected the motility rate and duration, fertilization rate, embryogenesis, and hatching and also triggered antioxidant defense mechanisms at the highest concentration (25 mg L−1). Furthermore, linear speed (VSL), linearity index (LIN), and amplitude lateral head (ALH) were significantly changed after exposure to 25 mg L−1, and the lowest concentrations (1 and 10 mg L−1) did not significantly affect the motility and fertilizing capacity. The embryogenesis and hatching were significantly affected by sperm exposure to 1, 10, and 25 mg L−1 of Aroclor 1254. Consequently, Aroclor 1254 causes potential hazards in male germ cells, and the exposure of sperm cells to pollutants can adversely affect next generation of wild populations.
