The Effect of Resveratrol on The Oxidative Stress in Rats Treated with Irradiation

Ozcelik, Mehtap; Erisir, Mine; Guler, Osman; Baykara, Murat; Bulmus, Funda Gulcu

The antioxidant and protective effect of resveratrol against irradiation induced oxidative damage were researched. Twenty-one rats were seperated into 3 groups; First group was the control group. Second group was the irradiation group; received physiological saline intraperitoneally (i.p.) for three days, then 9 Gray (Gy) irradiation was applied. Third group was irradiation+Resveratrol group; received 10 mg/kg resveratrol i.p. for three days and then the same dose of radiation was applied. As a result of irradiation, a significant increase was found in the level of malondialdehyde (MDA) in the liver, kidney and brain tissues of rats (P<0.001). Glutathione peroxidase (GSH-Px) activity in all the tissues and superoxide dismutase (SOD) activity, Glutathione (GSH) levels in liver decreased (P<0.01), GSH levels in kidney, spleen and ovarium tissues significantly increased (P<0.05). The giving of resveratrol significantly reduced the MDA levels increasing in liver, kidney, and brain as a result of irradiation (P<0.001) and the liver MDA levels reached to the control levels. By giving resveratrol, it was determined that all the antioxidants studied reduced significantly compared to the control group and/or the irradiation group (P<0.05). According to the data obtained as a result; It was concluded that resveratrol shows a corrective effect by changing the oxidant/antioxidant balance against oxidative stress caused by irradiation, therefore resveratrol is a powerful antioxidant.

The effect of curcumin on lipid peroxidation and selected antioxidants in irradiated rats

Ozcelik, Mehtap; Erişir, Mine; Guler, Osman; Baykara, Murat; Kirman, Esra

The effect of irradiation on oxidants and antioxidants in selected tissues and a possible protective effect of curcumin on these indices were investigated. A total of 28 rats were divided into 3 groups; group 1 was control; group 2 was the irradiation group, saline was administered intraperiotenally (i.p.) for three days and then, 9 Gy gamma irradiation was applied; group 3 was the irradiation + curcumin group: curcumin was given i.p. for three days at 200 mg/ kg body weight and then the same dose of irradiation was applied. A significant increase in malondialdehyde (MDA) was detected in the liver, kidney, and brain tissues of the rats as a result of irradiation (P ˂ 0.01). Glutathione peroxidase (GSH-Px) activity in all the tissues (except for kidneys) decreased (P ˂ 0.01), liver SOD (superoxide dismutase) activity decreased (P ˂ 0.05), and GSH (glutathione) levels in kidney and ovary tissues (P ˂ 0.001) significantly increased. While curcumin administration returned the increased MDA levels in the kidneys and brain in result of irradiation to normal (P ˂ 0.01), it did not return the increased MDA levels in the liver tissue to normal (P ˂ 0.001) despite significantly reducing them. While decreased GSH-Px and SOD activity in the liver in result of irradiation increased with the addition of curcumin (P ˂ 0.05), increased GSH levels in the kidneys and ovaries returned to control levels (P ˂ 0.001). When MDA values were examined, it was found that the addition of curcumin protected the liver, kidneys and brain from the oxidative damage caused by irradiation.

Bioaccumulation of Copper and Zinc in Freshwater Crayfish (Astacus leptodactylus) Fed with Zebra Mussels (Dreissena polymorpha) Collected from Keban Dam Lake, Turkey

Aksu, Onder; Gulec, Azime Kucukgul; Demir, Veysel; Danabas, Durali; Yildirim, Numan; Ozcelik, Mehtap; Can, Erkan; Kocabas, Mehmet

This study is aimed to determine the bioaccumulation of copper and zinc in hemolymph of freshwater crayfish (Astacus leptodactylus) fed with zebra mussel (Dreissena polymorpha) collected from Keban Dam Lake. This is the first study on bioaccumulation of Cu and Zn in hemolymph of males and females of A. leptodactylus. The average concentrations of Cu and Zn in the hemolymph samples were found out as 0.54 and 35.07 ppm respectively for male and 0.42 and 15.59 ppm respectively for female control freshwater crayfish. The levels of Zn in the hemolymph of both male and female ones among control and diet groups were statistically significant (P < 0.05). The Cu levels among control and diet groups in female ones were statistically significant (P < 0.05) but not in the male ones (P > 0.05). The results of current study revealed that feed treatment with boiling was not effective at the levels of metal bioaccumulation in hemolymph. It was observed that this metal bioaccumulation is more related with sex differences.

Some Heavy Metals Accumulation in Tissues in Capoeta umbla (Heckel, 1843) from Uzuncayir Dam Lake (Tunceli, Turkey)

Ural, Mesut; Yildirim, Numan; Danabas, Durali; Kaplan, Olcay; Yildirim, Nuran Cikcikoglu; Ozcelik, Mehtap; Kurekci, Eylem Funda

Concentrations of metals were determined in the gills, liver, kidney, heart and muscle in Capoeta umbla caught from six stations from the Munzur River system. Metal concentrations in the tissues tended to vary significantly among stations (p < 0.05). Liver (Cu, 10.10 +/- A 0.23-23.03 +/- A 9.37 ppm; Zn, 14.67 +/- A 3.01-21.82 +/- A 2.39 ppm; Cd, 18.04 +/- A 4.56-52.69 +/- A 10.65 ppb and Fe, 28.87 +/- A 6.78-115.11 +/- A 34.87 ppm) and kidney (Cu, 1.80 +/- A 0.25-3.70 +/- A 0.62 ppm; Zn, 20.81 +/- A 0.37-29.36 +/- A 0.70 ppm; Cd, 132.06 +/- A 5.29-639.51 +/- A 20.14 ppb and Fe, 24.40 +/- A 1.98-59.39 +/- A 1.97 ppm) tissues showed higher metal concentrations than other tissues. It seems that metal contamination in the river is too high for the health of fish and the people who eat them. The geographical locations of catch, season, nature of diet, and the size of fish used for analyses might lead to different metal concentration in the same fish species.
