Antioxidant Enzyme Responses to Lead Exposure in Daphnia magna

    Sahilli, Yeliz Cakir

Abstract: Lead (Pb), a pervasive environmental contaminant, poses significant toxicological risks to freshwater ecosystems. Daphnia magna, a widely used model organism in aquatic toxicology, provides valuable insights into the sublethal effects of heavy metal exposure. This study investigated the effects of Pb exposure on antioxidant defense mechanisms in D. magna. Test organisms were exposed to aqueous Pb solutions at varying concentrations (0, 10, 20, 30 μg/L) for 96 h. The enzyme activities of glutathione peroxidase (GPx), catalase (CAT), and superoxide dismutase (SOD) were measured using enzyme-linked immunosorbent assay (ELISA). SOD activity did not differ significantly between the exposed and control groups. However, CAT activity showed a significant decrease in all Pb-exposed groups compared to controls. GPx activity exhibited a non-significant decrease. In conclusion, Pb exposure may induce oxidative stress by generating reactive oxygen species (ROS) and inhibiting the function of sulfhydryl-containing antioxidant enzymes. Changes in antioxidant enzyme activity may serve as early biomarkers in ecotoxicological assessments of Pb pollution in aquatic environments. It is suggested that future studies incorporate molecular-level endpoints and chronic exposure scenarios to better elucidate the long-term impacts of Pb on aquatic invertebrates.

Antioxidant Activity and Some Chemical Composition of Polytrichum piliferum Hedw. Extracts

    Alataş, Mevlüt; Sahilli, Yeliz Cakir

In this study, antioxidant, fatty acid, mineral analysis and some chemical properties of Polytrichum piliferum, a species of moss, were investigated. As a result of the research, it was determined that P. piliferum extract had a moderate effect in terms of antioxidant properties and total phenolic content. Regarding fatty acid compositions, oleic and palmitic acids were found to be high. As a result of mineral analysis, it was found that it could accumulate high levels of aluminum (Al), iron (Fe), calcium (Ca) and potassium (K).

THE TOXIC EFFECT OF CADMIUM ON HUMAN ERYTHROCYTES AND NAC'S PROTECTIVE ROLE

    Sahilli, Yeliz Cakir

The toxic effects cadmium (Cd), which is a heavy metal, on human erythrocytes and protective effects of N-acetyl-L-cysteine (NAC) on lipid peroxidation and antioxidant enzymes were investigated. The effects of 1, 50, 150 mu M doses of cadmium and 1mM NAC on the level of malondialdehyde (MDA) in human erythrocytes and superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) enzyme activities were investigated. When the Cd was applied alone, it was found that MDA level increased in erythrocytes and SOD, CAT and GPx activities decreased. No statistically significant difference was observed when 1mM NAC administered erythrocytes were compared with the control cells. However, the beneficial effect was seen only in administration doses of Cd at the levels of I and 50 mu M and NAC was observed to have a protective effect. As a result, NAC partially reduced the toxic effect caused by Cd on erythrocytes but could not completely prevent it.

THE ROLE OF GLUTATHIONE IN THE DETOXIFICATION OF PERCHLOROETHYLENE

    Sahilli, Yeliz Cakir

Environmental factors have important roles in the development of cancer. Especially, the exposure of chemicals existing in the workplace with people is a very important issue. People at such places are exposed to chemicals more especially via respiratory and dermal route. Thus, their health is under the risk. Dry cleaning centers are intensively subjected to PCE, which is a common chemical used at these work places, and the risk of cancer is very high for them. The main aim of the present study is to investigate, if PCE exposure results in glutathione efflux from human erythrocytes. In the present study, the changes in intracellular and extracellular non-protein sulfhydryl (NPSH) levels were determined in PCE exposed erythrocytes. PCE significantly depleted intracellular NPSH level in a time and concentration-dependent manner. PCE significantly increased the NPSH levels measured in the extracellular media. Along with the change in extracellular nonprotein thiol levels, significant increases in extracellular glutathione levels were detected. Erythrocyte membranes were not damaged over the course of PCE exposure as indicated by the absence of lactate dehydrogenase activity in the extracellular media. Utilization of MK571 an MRP inhibitor decreased the rate of glutathione efflux from erythrocytes suggesting a role for this membrane transporter in the process. The results of the present study indicate that erythrocytes efflux glutathione when exposed to PCE. In addition to glutathione oxidation, glutathione efflux also contributes to intracellular loss of glutathione when exposed PCE.

ANTIMONY INDUCED GLUTATHIONE EFFLUX FROM HUMAN ERYTHROCTES

Sahilli, Yeliz Cakir; Akgun, Mehmet Hanifi; Yildiz, Deniz

Antimony is a trace element which has wide variety of industrial application. Antimony has recently been considered as an environmental pollutant. Among the inorganic forms, SbIII is the more toxic form. SbIII is ten times more toxic compared to the SbV. Consuming plants that are grown in antimony contaminated soil is one of the way by which humans are exposed to antimony. AsIII and SbIII forms strong bonds with free-SH groups. When SbIII or SbV interacts with proteins that contain free-SH groups, protein inactivation usually results in. Antimony may also result in free radical generation and induction of oxidative stress. Even though antimony has some applications in health as a drug, its biological effects have not been investigated in detail. In the present study we investigated the effects of SbIII on glutathione efflux in human erythrocytes. The effects of N-acetyl-L-cysteine on antimony induced glutathione oxidation and glutathione efflux were also investigated. Extracellular glutathione concentration significantly increased and reached to 0.033 +/- 0.0034 micromole/ml erythrocyte with 1 mM of potassium antimony tartrate in four hours. The control value of extracellular glutathione was 0.020 +/- 0.0035 micromole/ml erythrocyte. Our results show that SbIII treatment of erythrocyte results in glutathione efflux.
