The aim of this study was to evaluate the effect of chrysin on testicular damage induced by carbon tetrachloride (CCl4) in rats. Twenty-four male Spraque-Dawley rats were divided into four groups as the control group, Chrysin group, CCl4 group, and Chrysin+CCl4 group. Chrysin (100 mg/kg/body weight/day) and CCl4 (0.5 ml/kg/body weight/day in corn oil (1:1)) were administered by gavage for 14 days. Compared to the control group, it was determined that malondialdehyde (MDA) level increased, reduced glutathione (GSH) level, and superoxide dismutase (SOD) and catalase (KAT) activities decreased in the CCl4 group. It was also determined that many testicular lesions were induced, Johnsen’s Mean Testicular Biopsy Score (JMTBS) decreased, and caspase-3 immunoreactivity increased in this group. Compared to the CCl4 group, it was determined that GSH level and SOD and CAT activities increased, MDA level decreased in the Chrysin+CCl4 group. It was also observed that testicular lesions were alleviated, JMTBS increased, and caspase-3 immunoreactivity was decreased in this group. As a result, it was concluded that chrysin might have beneficial effects in the treatment of CCl4-induced testicular toxicity as a result of its antioxidant and anti-apoptotic properties.

Baykalir, Burcu Gul; Arslan, Aslihan Sur; Mutlu, Seda Iflazoglu; Parlak Ak, Tuba; Seven, Ismail; Seven, Pinar Tatli; Yaman, Mine; Gul, Huseyin Fatih

The aim of this study was to investigate the possible protective effects of chrysin on oxidative status and histological alterations against carbon tetrachloride (CCl4)-induced liver and kidney tissue in rats. The animals were randomly divided into four groups; the control, chrysin (100 mg/kg), CCl4 (0.5 ml/kg) and chrysin + CCl4 groups. Liver and kidney injuries were assessed by biochemical and histopathological examinations. The levels of malondialdehyde (MDA), reduced glutathione (GSH), and superoxide dismutase (SOD) activity were measured in tissues. Serum tumor necrosis factor-α (TNF-α), aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, and creatinine levels were also measured in blood samples. MDA, serum TNF-α, AST, ALT, urea, and creatinine levels (p < 0.05) were significantly higher, and SOD activity and GSH level were significantly (p < 0.05) lower in the CCl4 group than in the control group. Treatment with chrysin in the chrysin + CCl4 group decreased MDA, AST, ALT, creatinine, and TNF-α levels (p < 0.05), and increased SOD activity, GSH levels (p < 0.05), and serum TNF-α levels (p < 0.05). In addition, body weight change (BWC) (p < 0.05) and feed intake (FI) were significantly lower (p < 0.001) in the CCl4 group than in the control group. Moreover, treatment with chrysin increased BWC and FI in the chrysin + CCl4 group compared with that in the CCl4 group. These findings also confirmed by histopathological examination. The chrysin treatment ameliorated the CCl4-induced biochemical and pathological alterations. These results demonstrated that chrysin provided amelioration on the rat liver and kidney tissues CCl4-induced injury by increasing the antioxidant activity.

This experiment was conducted to determine the protective effects of flunixin meglumine and propolis on feed intake, antioxidant status and histological parameters in kidney and liver tissues in rats exposed to excessive copper. In this study individually housed thirty-six male Sprague-Dawley rats were used. Animals were randomly divided into six groups; control, copper sulphate (500 mg/kg BW/day, gavage), flunixin meglumine (2.2 mg/kg BW/day, ip), propolis (100 mg/kg BW/day, gavage), copper sulphate+flunixin meglumine (500 mg/kg BW/day of copper sulphate by gavage and 2.2 mg/kg BW/day of flunixin meglumine, ip) and copper sulphate+propolis (500 mg/kg BW/day of copper sulphate and 100 mg/kg BW/day of propolis by gavage). The study demonstrated that body weight change in the copper sulphate+propolis group significantly ameliorated in comparison with copper group (P<0.01). Propolis and flunixin meglumine administration significantly decreased MDA levels in the kidney and liver tissues, and serum TNF-α levels (P<0.001). Propolis supplementation to rats who were also treated with copper significantly increased the superoxide dismutase, catalase and reduced glutathione activities (P<0.001). Flunixin meglumine and propolis treatments decreased the copper-induced degenerative and necrotic changes with the apoptotic cells in the liver and kidney tissues. In conclusion, propolis appeared to ameliorate the adverse effects on feed intake, liver and kidney tissues seeing in the copper treatment and, apparently caused by the copper toxicity, by scavenging the free radicals and increasing activity of antioxidants.

The protective effects of chrysin and flunixin meglumine against excess copper in male rats

Seven, Pinar Tatli; Baykalir, Burcu Gül; Seven, Ismail; Ak, Tuba Parlak; Başak, Neşe; Güllüoglu, Hakan

Thirty-six Sprague Dawley male rats were housed in individual cages and randomly divided into six groups: Control, copper sulfate (Cu; 500 ppm body weight (BW)/day), flunixin meglumine (FM; 2.2 ppm BW/day), chrysin (chrysin; 50 ppm BW/day), copper sulfate + FM (Cu + FM; 500 ppm BW/day of copper sulfate and 2.2 ppm BW/day of FM), and copper sulfate + chrysin (Cu + Chrysin; 500 ppm BW/day of copper sulfate and 50 ppm BW/day of chrysin). Feed intake (FI) in the Cu + Chrysin group significantly increased in comparison with that of the Cu group (P < 0.01). Cu excess significantly increased malondialdehyde, indicating oxidative stress. Chrysin and FM administration significantly decreased malondialdehyde levels and increased the superoxide dismutase and catalase activities in the liver and kidney tissues (P < 0.001). Serum TNF-α levels were significantly lower in the Cu + FM and Cu + Chrysin groups in comparison to the Cu group (P < 0.001). It was seen that FM and chrysin treatments alleviated degeneration, necrosis, and apoptosis in the liver and kidney tissues of the Cu-treated rats. Chrysin appeared to ameliorate the adverse effects on FI and liver and kidney tissues by scavenging where the free radicals are located and increasing the activity of antioxidants.

In this study, the effects of fresh, ensiled or dried alfalfa on retinol, a-tocopherol and cholesterol levels in yearling sheep were investigated. In the study, 40 Akkaraman male yearling sheep, 4 months of age and an average body weight of 21 kg, were used. All diets used in the present study were prepared as isonitrogenous and isoenergetic. The alfalfa forms used in rations composed the experimental groups. With regard to groups were fed with wheat straw as roughage was Control group (C-Group) or fed with fresh alfalfa was AF group, ensiled alfalfa was AS group and dried alfalfa was AD group. Retinol levels in muscular and tail adipose tissue were found to be higher in alfalfa feeding groups compared to control group. The highest a-tocopherol level in deltoideous muscle tissue was detected in AF group. Cholesterol levels in muscular and tail adipose tissue were found to be lower in alfalfa feeding groups compared to control group. As a conclusion it can be said that, decrasing cholesterol levels in yearling sheep by feeding with alfalfa, is extremely important for human beings that consume red meat, as it can reduce the risk of coronary heart diseases.
