The present study focused on changes in the C-reactive protein (CRP) and adrenomedullin (ADM) levels in the liver and gill tissues of Capoeta umbla as biomarkers of environmental pollution and ecosystem health in inland waters of Tunceli (Turkey). The CRP levels in liver and gills were not significantly changed in March and September at every site (p>0.05) except for site 3 in gill tissues (p<0.05). The results of statistical analysis indicate that significant differences in CRP levels were found in liver and gills among sites (p<0.05), but no changes in the gills at March (p>0.05). In March, ADM levels in gills and liver were statistically changed among sites (p<0.05), while no statistical changes were determined in September (p>0.05). The results of this study indicated that CRP levels in the tissues of the C. umbla could be utilized as a significant bioindicator for disease pathogenesis in fish infections.

The poly[2-hydroxy-3-(1-naphthyloxy)propyl] methacrylate [poly(NOPMA)] was used as a novel adsorbent for the removal of Remazol Brilliant Blue R (RBBR) known as a reactive dye. Physico-chemical properties of polymer were analyzed by FT-IR and SEM before and after dye adsorption. The Langmuir and Freundlich isotherms were used to explain the adsorption equilibrium, and when it was compared with each other it was seen that the adsorption process was well explained by the Langmuir isotherm model and showing a monolayer adsorption capacity for RBBR of 238.10 mg/g. Kinetic models known as pseudo-first-order, pseudo-second-order and intra-particle diffusion model were utilized to analyze the mechanism of adsorption, and results indicated that was an adsorption kinetic suitable with the pseudo-second-order. The thermodynamic parameters (∆Ho, ∆So, ∆Go) indicated that the adsorption process was spontaneous and endothermic. To optimize the study conditions, the effects of the concentration of RBBR, adsorbent amount and contact time on the adsorption were investigated by response surface methodology (RSM). According to RSM results, maximum RBBR removal was reached at the optimum initial dye concentration of 60.85 mg/L, adsorbent dosage of 0.04 g/50 mL and contact time of 59.91 min.

- MakaleYalnız künye2017Scopus SJRQ2Optimizing electrocoagulation process using experimental design for COD removal from unsanitary landfill leachate
Ogedey, Aysenur; Tanyol, Mehtap
Leachate is the most difficult wastewater to be treated due to its complex content and high pollution release. For this reason, since it is not possible to be treated with a single process, a pre-treatment is needed. In the present study, a batch electrocoagulation reactor containing aluminum and iron electrodes was used to reduce chemical oxygen demand (COD) from landfill leachate (Tunceli, Turkey). Optimization of COD elimination was carried out with response surface methodology to describe the interaction effect of four main process independent parameters (current density, inter-electrode distance, pH and time of electrolysis). The optimum current density, inter-electrode distance, pH and time of electrolysis for maximum COD removal (43%) were found to be 19.42 mA/m2, 0.96 cm, 7.23 and 67.64 min, respectively. The results shown that the electrocoagulation process can be used as a pre-treatment step for leachate.

- MakaleAçık Erişim2017Scopus SJRQ2Rapid malachite green removal from aqueous solution by natural zeolite: Process optimization by response surface methodology
Tanyol, Mehtap
In this study, malachite green removal from aqueous solutions by natural zeolite was investigated. The interaction between zeolite and malachite green was determined by SEM/EDX and FTIR. The parameters like adsorbent dosage (0.1–1 mg/100 mL), initial malachite green concentration (40–200 mg/L) and the contact time (10–600 s) were optimized using the central composite design of response surface methodology. Maximum removal yield (99.70%) was obtained at an adsorbent dosage of 0.68 mg/100 mL, an initial malachite green concentration of 79.02 mg/L, and at 414 s contact time. The equilibrium process complied better with the Langmuir isotherm model, which exhibits a single layer adsorption, when compared with the Freundlich isotherm model. Adsorption kinetics followed the pseudo-second-order type kinetic and intra-particle diffusion played an important role onto adsorption mechanism of malachite green. The findings of this study demonstrated that zeolite which is found abundant on earth as a cheap and efficient adsorbent could be used for rapid malachite green adsorption from aqueous solutions.

This study investigates preparation of biosorbent Pleurotus eryngii immobilized on Amberlite XAD-4 and the optimal conditions for removal of Remazol Brilliant Blue R (RBBR) reactive dye from synthetic aqueous solutions. The process was optimized using the response surface methodology (RSM) developed by the application of the quadratic model associated with the central composite design. For this purpose, RSM was employed to determine the effects of operational parameters on this material as effective and available adsorbent. The investigated variables were dye initial concentration (10-60 mg L-1), solution pH (2-9), adsorbent dosage (0.1-0.5 g), and temperature (20-45 degrees C). The significant factors on each experimental design response were identified from the analysis of variance (ANOVA). The RSM indicated that optimum conditions of initial dye concentration, pH, adsorbent dosage, and temperature for maximum RBBR removal (98%) were achieved as 36.3 mg L-1, 2.0, 0.304 g, and 38.7 degrees C, respectively. The results showed that this biosorbent was an appropriate adsorbent for the removal of RBBR from aqueous solutions.

- MakaleAçık Erişim2016Scopus SJRQ2Adsorption of bisphenol A from aqueous solutions by Pleurotus eryngii immobilized on Amberlite XAD-4 using as a new adsorbent
Yonten, Vahap; Ince, Muharrem; Tanyol, Mehtap; Yildirim, Numan
The adsorption of bisphenol A (BPA) in a fixed-bed column was evaluated in this work. Fungus Pleurotus eryngii isolated from Tunceli (Turkey) immobilized on XAD-4 resin was used as an adsorbent for decontamination of BPA from aqueous solutions. The extent of BPA removal capacity was evaluated as a function of parameters such as pH, volume of the sample, and column bed height. Adsorption isotherms of BPA on immobilized P. eryngii were determined and correlated with common isotherm equations such as Langmuir and Freundlich models. The mechanism of adsorption of BPA was analyzed preadsorption (XAD-4+fungus) and postadsorption (XAD-4+fungus+BPA) using fourier transform infrared spectroscopy. The results show that adsorption ability of BPA by immobilized P. eryngii is possible and this system yields the best degree of BPA removal (90%) at pH 11 for an initial load of 120mgL(-1). Therefore, this material can be used as a very effective adsorbent for BPA decontamination in aqueous solutions.

Tanyol, Mehtap; Uslu, Gulsad; Yönten, Vahap
The aim of our research was to explore the most cost-efficient and optimal medium composition for the production of lipase from Pseudomonas fluorescens (NRLL B-2641) culture grown on sunflower oil cake (SuOC) by applying response surface methodology (RSM). The oil cake was used instead of carbon sources. Peptone, ammonium sulphate and the carbon source (SuOC) were the most important factors as it is obligatory for microbial growth. Subsequently, the optimum values for the carbon source, peptone and ammonium sulphate were found to be 11.10% (w/v), 1.18% (w/v) and 0.83% (w/v), respectively. Experiments carried out under optimum conditions revealed a maximum lipase activity of 10.8 U mL(-1), which was achieved after 48 h of fermentation. The obtained results were finally verified with batch experiments carried out under the optimum conditions evaluated and it was demonstrated that the SuOC from agro-industrial residue as substrates can be used as an inexpensive base (carbon source) for the production of lipase by P. fluorescens (NRLL B-2641).

- MakaleYalnız künye2015Scopus SJRQ2Removal of Phosphate from Aqueous Solutions by Chemical- and Thermal-Modified Bentonite Clay
Tanyol, Mehtap; Yonten, Vahap; Demir, Veysel
The study investigated phosphate adsorption from aqueous solutions using chemical- and thermal-modified bentonite in batch system. The adsorbent was characterized by SEM, BET, and FTIR spectroscopy. Contact time, beginning phosphate concentration, pH of the solution, and the effects of the temperature on phosphate adsorption capacity were determined by a series of experimental studies. In a wide pH range (3-10), high phosphate removal yields were obtained (between 94.23 and 92.26 %), and with the increase in temperature (from 25 to 45 degrees C), phosphate removal increased. Langmuir and Freundlich isotherms were used to determine the sorption equilibrium, and the results demonstrated that equilibriumdata displayed better adjustment to Langmuir isotherm than the Freundlich isotherm. Phosphate sorption capacity, calculated using Langmuir equation, is 20.37 mg g(-1) at 45 degrees C temperature and pH 3. Mass transfer and kinetic models were applied to empirical findings to determine the mechanism of adsorption and the potential steps that control the reaction rate. Both external mass transfer and intra-particle diffusion played a significant role on the adsorption mechanism of phosphate, and adsorption kinetics followed the pseudo-second-order-type kinetic. Furthermore, thermodynamic parameters (Delta H degrees, Delta G degrees, Delta S degrees) which reveal that phosphate adsorption occur spontaneously and in endothermic nature were determined. The results of this study support that bentonite, which is found abundant in nature and modified as an inexpensive and effective adsorbent, could be used for phosphate removal from aqueous solutions.

Lipases are industrially important enzymes produced by a great variety of microorganisms. In this study, the production of extracellular lipase by Pseudomonas fluorescens (NRLL B-2641) using submerged fermentation was studied: The effects of. initial pH, carbon source, nitrogen source and metal ion addition on lipase activity were investigated using batch experiments. The highest enzyme activity and biomass production obtained were 2.50 U mL(-1) and 2.95 g cell dry wt. L-1, respectively, using a fermentation medium with 1% corn oil as the carbon source, 0.4% peptone as the nitrogen source and an initial pH of 6.0. The addition of calcium increased the enzyme activity to 3.30 U mL(-1). Thus, initial lipase activity, which was 0.92 U mL(-1), increased 356%. Partial characterization of crude lipase was performed to determine the optimum pH and temperature. The crude lipase displayed high activities at pH 6.0-8.5 and 35-65 degrees C, with maximum activity at pH 8.0 and 50 degrees C.

Uslu, Gulsad; Gültekin, Gulden; Tanyol, Mehtap
The bioaccumulation of copper (II) and cadmium (II) ions by viable P. putida was studied as a function of initial pH, temperature, initial glucose and initial metal ion concentration. Optimum pH value for maximum metal ion accumulation was determined to be 3.0 +/- 0.5 and 4.5 +/- 0.5 at 25 and 30 degrees C for each metal ion, respectively. At a constant glucose concentration, although the increase in initial copper (II) and cadmium (II) ion concentration up to approximately 25 mg L-1 increased bioaccumulation capacity of P. putida for both, all metal ion concentrations tested inhibited the growth of P. putida. High microbial growth and copper (II) and cadmium (II) accumulation yields were found at low initial metal ion concentrations. The cell and bioaccumulated metal ion concentrations and uptake yields in-creased with increasing glucose concentration up to 10 g L-1 at a constant metal ion level for each of the metal ions. Although both metal ions caused on inhibition effect on the growth of the microorganism, P. putida was capable of removing copper (II) and cadmium (II) with maximum specific uptake of 58.42 mg g-(1) and 34.65 mg g(-1) at 25 mg L-1 initial copper(II) and cadmium(II) level, respectively. The results also indicated that, P. putida was more efficient in accumulating larger amounts of cadmium (II) than copper (II) ions.
