Investigation of heavy metal accumulation in Typha latifolia exposed to mine drainage waters

    Tatar, Sule; Okuduci, Ebru

In this study, the accumulation of some heavy metals in water, sediment, and Typha latifolia plant species was investigated in a previously operated mining area located in the Hozat region of Tunceli. In the first phase of the study, water and soil samples were collected from sites where T. latifolia was dominant. The metal uptake capacities from soil to roots, stems, and leaves of the plants were examined for different metals as silver (Ag), barium (Ba), cadmium (Cd), copper (Cu), nickel (Ni), lead (Pb), strontium (Sr), zinc (Zn). Water, sediment, and plant samples collected from the field were analyzed for the above elements at ultra-low detection limits using ICP-MS at ACME Analytical Laboratories in Canada. Bioconcentration factors (BCF-R, BCF-S, BCF-L, respectively) for the root, stem, leaf, and translocation factor (TF) values were calculated. The highest TF value was found to be 0.85 for Sr. The highest bioconcentration factors in the root, stem, and leaves of the plant were also found to be 3.67, 1.55, and 1.79 for Sr, respectively. According to the results obtained, T. latifolia was found to be a suitable plant for the bioaccumulation of Cd, Pb, Cu, and Sr under the existing environmental conditions, but not a suitable bioaccumulator for Ag, Ba, Ni, and Zn. Based on the results, it was determined that some heavy metal pollutants can be remediated using certain plants, and that these plants can function as bioaccumulators for these metals.

Serdar, Osman; Yildirim, Nuran Cikcikoglu; Yildirim, Numan; Tatar, Sule

The assessment of basic blue 41 textile dye decolorisation using the Fenton process: response surface methodology optimisation and toxicity evaluation

Tanyol, Mehtap; Cikcikoglu Yildirim, Nuran; Tatar, Sule; Serdar, Osman; Yildirim, Numan

The biochemical biomarkers (Catalase (CAT), glutathione s-transferase (GST), cytochrome P4501A1 (CYP1A1450) activities and glutathione (GSH), malondialdehyde (MDA) levels) of D. polymorpha exposed to the basic blue 41 (BB41) solutions for 24 and 96 h before and after the Fenton process (FTP) were evaluated to investigate the efficiency of the FTP. The optimum conditions for the removal of BB41 were set using central composite design (CCD), which is a response surface methodology (RSM) module in the Design-Expert® software. The optimal conditions were found to be 27.87 min, 3.49 mg/L, and 0.04 for reaction time, initial BB41 concentration, and Fe2+/H2O2 molar ratio, respectively, resulting in maximum (99%) removal efficiency. After FTP, the MDA and GSH levels decreased significantly during the 24 and 96 h. CAT activities were increased after treatment. No statistically significant changes were observed in the CYP1A1 levels after FTP. GST activities decreased during 24 h but increased during 96 h after FTP. CYP1A1450, catalase, GST activities and MDA, GSH levels of D. polymorpha are useful biomarkers to evaluate the efficiency of FTP. This study suggests that BB41 dye must be further treated after the FTP to minimise the ecotoxicological risk.

Antioxidant Enzyme Responses to Lead Exposure of Gammarus pulex

Serdar, Osman; Yildirim, Numan; Tatar, Sule; Cıkcıkoğlu Yıldırım, Nuran

It is aimed to investigate the activity response of the antioxidant enzymes superoxide dismutase, catalase and glutathione peroxidase of G. pulex exposed to lead. The G. pulex was exposed synthetic solutions containing 10, 30, 50 µg/L lead for 96 h. Superoxide dismutase, catalase, glutathione peroxidase activities were determined by using ELISA kit. The differences in superoxide dismutase activities were statistically insignificant (P>0.05). Catalase decreased in all treatment groups after lead exposure (P<0.05). The glutathione peroxidase activities were decreased depending on lead exposure (P>0.05). Our findings suggest that lead can induce oxidative stress by generating reactive oxygen species and inhibiting sulfhydryl dependent antioxidant enzymes activities. In conclusion, alterations in antioxidant enzymes may potentially be used as sensitive biomarkers for risk assessment of lead in the environment and may contribute to the establishment of discharge regulations.

In order to evaluate the potential ecological risk and the toxic effect of landfill leachate (LL), Phanerochaete chrysosporium was exposed to LL and their biochemical response was observed by using antioxidant parameters. Phanerochaete chrysosporium, ME 446, was kept at 4 °C after being sub-cultured at 28 °C on Sabouraud Dextrose Agar (SDA). Superoxide dismutase (SOD), catalase (CAT) activities, and malaondialdehyde (MDA) and glutathione (GSH) levels of P. chrysosporium exposed to different dilution rates of leachate (1/10 and 1/20) for 24 and 96 h were analyzed by using the ELISA method. The physiochemical parameters such as pH, conductivity, total dissolved solids (TDS), dissolved oxygen (DO), chemical oxygen demand (COD) of leachate, and reference water were analyzed by using the YSI Professional Plus handheld multiparameter meter. In this study, SOD activities were decreased in the application groups compared with the Control Group at the 24th and 96th hours. CAT activities and GSH levels increased in the application groups compared with the Control Group at the 24th hour but decreased at the 96th hours. MDA levels increased in all of the application groups when compared with the Control Group for both 24 and 96 h. Different concentration of LL induces oxidative stress in P. chrysosporium, increased CAT activity and MDA levels, and decreased SOD activity and GSH levels.

Serdar, Osman; Yildirim, Nuran Cikcikoglu; Tatar, Sule; Yildirim, Numan; Ogedey, Aysenur

The discharge of landfill leachate into the environment without effective treatment poses a serious threat for the aquatic ecosystems. This present study was undertaken to evaluate whether electrocoagulation process is efficient for treatment landfill leachate (LL) or not by using antioxidant biomarkers in Gammarus pulex. Glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and catalase (CAT) activities and malondialdehyde (MDA) and glutathione (GSH) levels in G. pulex exposed to untreated, treated, and diluted rates 1/10 and 1/20 in both LL during 24 and 96 h were tested. Physiochemical characteristics of leachate (chemical oxygen demand, electrical conductivity, pH, phosphate, turbidity, NH3, Cl−, and color) were determined pre and post treatment. All physiochemical characteristics of LL decreased after treatment process. GSH-Px and CAT activities and GSH and MDA levels were increased in untreated groups when compared to control (p < 0.05). After treatment by electrocoagulation, MDA and GSH levels and CAT activities were returned to control values. In conclusion, the abilities of LL to stimulate oxidative stress in G. pulex have been proven. The results revealed that antioxidant parameters are useful biomarkers for determining the treatment efficiency of the electrocoagulation process.
