Drying temperature-dependent changes in grape seed bioactives and oxidative status: integrated HPLC/GC and multivariate analysis

    Karaca Sanyürek, Nesrin

Abstract: Drying temperature (40–80 °C) critically influences the stability of bioactive compounds in grape seeds and their functional responses. In this study, we evaluated the vitamin and phytosterol content of Boğazkere grape seeds using high-performance liquid chromatography (HPLC) and the fatty acid content using gas chromatography (GC). Antioxidant capacity was determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and the oxidative stress marker, malondialdehyde (MDA). Temperature-dependent and compound-specific alterations were observed: while selected vitamins and phytosterols showed distinct thermal susceptibilities, fatty acid profiles exhibited measurable remodeling. Antioxidant activity revealed non-linear dose- and temperature-dependent patterns, whereas MDA consistently decreased with increasing temperature, reflecting the reduced oxidative metabolism in dehydrated tissues. Multivariate analysis, including principal component analysis (PCA) and heatmap visualization, effectively discriminated temperature treatments and revealed correlations between compositional and functional parameters. The combined use of univariate and multivariate analyses revealed that the drying temperature reshaped not only individual bioactive compounds but also the overall biochemical profile of grape seeds. These findings establish a mechanistic link between thermal processing and bioactive stability, providing a chemical basis for optimizing grape seed valorization in food systems.

Biogenic amine profiling of fruit wines by high-performance liquid chromatography with fluorescence detection after OPA derivatisation: effect of fruit type

This study aimed to characterise the biogenic amine (BA) profiles of fruit wines produced from different raw materials and to investigate their association with pH and fruit type. Ten commercially available fruit wines, including pomegranate, black mulberry, sour cherry, strawberry, melon, blackberry, and blueberry, were analysed using HPLC with fluorescence detection (HPLC-FLD) following pre-column derivatisation with o-phthalaldehyde. Putrescine, tyramine (TYR), and histamine were identified as the predominant amines, with total BA concentrations ranging from 11.62 to 53.96 mg/L depending on the fruit type. The pH values of the wines (3.28-3.60) were inversely related to acidity and showed a strong positive correlation with total BA content (r = 0.97, p < .001). Among the detected compounds, TYR exhibited the highest sensitivity to pH variation. Multivariate analysis using principal component analysis clearly differentiated the samples into two distinct groups: low pH-low BA wines (pomegranate and blueberry) and high pH-high BA wines (blackberry and strawberry). The HPLC-FLD method demonstrated excellent analytical performance, with high chromatographic stability (retention time RSD < 1%) and reliable compound identification. Overall, the results indicate that BA formation in fruit wines is closely associated with both pH and fruit type, reflecting the combined effects of raw material composition and fermentation conditions. These findings provide valuable insights into the biochemical characterisation, quality assessment, and safety evaluation of fruit wines in oenological applications.

The pollen morphology of eight endemic species of the genus Inula L. (Asteraceae)–I. helenium subsp. orgyalis (Boiss.) Grierson, I. helenium subsp. vanensis Grierson, I. macrocephala Boiss. & Kotschy ex Boiss., I. discoidea Boiss., I. sarana Boiss., I. fragilis Boiss. & Hausskn., I. anatolica Boiss. and I. sechmenii Hartvig & Strid–distributed in Turkey was studied with light microscopy and scanning electron microscopy. We found that the pollen of these Inula taxa are prolate-spheroidal in shape (except in Inula discoidea), with the polar axis ranging from 19.18 to 33.14 µm and the equatorial diameter ranging from 18.54 to 32.92 µm. The pollen grains are tricolporate and their ornamentation is echinate. The exine thickness ranges from 0.87 to 2.07 µm. As this study demonstrates, results of Principal Component Analysis are revealed that some of the pollen characters, such as spine length and width, are significant morphological characters for taxonomic discrimination of the eight endemic Inula taxa. According to cluster analysis, two main groups of examined taxa are constituted.
