The Effects of Probiotic-Prebiotic on the Biomass and Protein Content of Spirulina platensis in Different Temperatures and Illuminations

Seyhaneyıldız Can, Şafak; Seyhaneyıldız, Elif; Dörücü, Mustafa

The blue-green algae Spirulina platensis Geitler 1925 has been used by people since ancient times due to its high protein content. This study was conducted to determine the effect of a Probiotic-Prebiotic (P-P) product, which contains Bifidobacterium infantis, Bifidobacterium thermophilum, Leuconostoc mesenteroides as probiotic bacteria, and beta-glucan and oligosaccharide as a prebiotic mixture on S. platensis. In the study, four groups were one of which was a control. In this study, we investigated whether the P-P product contributed to the biomass development and protein production of S. platensis under different temperatures and lighting conditions. In terms of biomass, the P-P product gave different responses at different temperatures and light intensities, but its results were positive in all groups in terms of protein ratio. The most positive effect on biomass and protein content was in the group where 0.1 ml/L P-P product was added, at 40°C and 3200 lux with 68.71% protein content and an average OD of 0.727.

Effect of temperature and nitrogen concentration on the growth and lipid content of Spirulina platensis and biodiesel production

Seyhaneyildiz Can, Safak; Koru, Edis; Cirik, Semra

In this study, Spirulina platensis (Cyanophyceae) was cultured at five mediums with different amounts of sodium nitrate and three different temperatures. The study was conducted at 1600 lx light intensity and a pH value of 9. The influences of nitrate amounts and temperature on biomass growth and lipid amount were studied during 20 days. The best biomass growth (1.039 ± 0.668 g L−1 dry weight and 0.780 ± 0.501 optical density) for S. platensis was obtained from the group with the temperature value of 35 °C and 2.5 g L−1 of sodium nitrate. The highest lipid production (4.70 ± 0.067% dry weight) was in the nitrate-free medium at 15 °C. The best results for biomass growth and lipid production were obtained from the group at 35 °C and 1.25 g L−1 of sodium nitrate (P < 0.05). At the end of the study, it was determined that algal biomass increased with increasing the nitrate amount but the produced lipid by the algae decreased. Intensive production was performed in accordance with the culture conditions of the most productive groups and biodiesel was obtained from the algae. The density, viscosity, and flash point of the biodiesel were calculated as 0.78, 4.67, and 107 °C, respectively.

EVALUATING THE DILUTION OF MUNICIPAL WASTEWATER ON BIOMASS INCREASE, LIPID PRODUCTION AND NUTRIENT REMOVAL BY THE BLUE-GREEN ALGAE Spirulina platensis (Geitler)

Can, Safak Seyhaneyildiz; Demir, Veysel; Can, Erkan

Wastewater generated daily in urban localities could provide nourishment to algae for potential production of biofuel and biochemicals. This study presents an integrated approach to the use of Spirulina platensis cultures for the treatment of domestic wastewater and lipid extraction. In this experimental approach, wastewater subjected only to the primary settling process was used as nutrient media at different concentrations of 100%, 75%, 50%, and 25% wastewater diluted with distilled water, and Spirulina platensis was cultured in these mediums for 20 days. The inoculated medium was incubated at 30 degrees C in the presence of light (42 mu mol photon m(-2).s(-1)) by using light/dark cycle of 12 hours. S. platensis consumed nitrite, nitrate, phosphate and ammonium from wastewater, and it increased its biomass and its lipid amount. Although the highest biomass increase was observed at the wastewater medium without dilution (100% wastewater) (the control group was not included), the highest lipid production was found in 25% wastewater. On the other hand, the percent reduction in the level of nitrite, nitrate, phosphate and ammonium in 75% wastewater medium was 91%, 99%, 99% and 99%, respectively. Despite the low lipid content, since Spirulina platensis can live in harsh conditions and increase its biomass rapidly, they can be easily cultivated in wastewater. Therefore, their use in wastewater for biodiesel production was found appropriate. The resulting algae biomass after oil extraction can be also used in different industrial sectors.
