Lipid content and wastewater treatment potential of Chlorella vulgaris and Scenedesmus obliquus isolated from Uzuncaylr Dam Lake

Can, Şafak Seyhaneyildiz; Can, Erkan; Yilmaz, Kadir

Production of microalgae for biodiesel, one of the alternative renewable energy sources, is costly due to nutrient media. Industrial and domestic wastewater contains nitrogen, phosphorus and other nutrients that are the primary food source for algae. Releasing this environmentally harmful effluent into receiving bodies of water, such as the sea or a freshwater reservoir, without prior treatment causes serious problems. Combining microalgae cultivation with wastewater treatment is a promising strategy for improving wastewater and reducing the cost of nutrient media required for algae production. In this study, wastewater obtained from a wastewater treatment facility was diluted with clean water to 0.25%, 50%, and 75% concentrations, and Chlorella vulgaris and Scenedesmus obliquus were cultured in these nutrient media for 20 days. As a result, microalgae increased their biomass and lipid content, while consuming nitrite, nitrate, phosphate, and ammonium from the wastewater as nutrients.

Efficient biofuel production from algae depends on the microalgal biomass production rate and lipid content. Both biomass production and lipid accumulation are limited by several factors, of which nutrients play a key role. In this study, the marine microalgae Dunaliella bardawil was used as a model organism. D. bardawil was cultured at two different sodium nitrate concentrations as 0 and 0.7 M, at six different amounts of salinity as 0.65, 1.0, 1.5, 2.0, 2.75 and 3.5 M, and at two different light intensity levels as 65 and 800 mu mol foton m(-2) s(-1) The study was done under pH 7. After 15-day growth period, the most efficient culture of D. bardawil for both biomass increase and lipid production was determined for the samples with 39.67 +/- 4.51 lipid rates cultured under 0.7 M nitrate concentration, 2.75 M salinity and 800 mu mol photon m(-2) sn(-1) light intensity. This high level of lipid production is sufficient to use D. bardawil for biodiesel production. Also after the evaluation of fatty acid results of GC-FID analysis, it was determined that D. bardawil contains high rate of medium length (C16-C18) fatty acid chains suitable for the production of biodiesel. These results are satisfactory to use D. bardawil in biodiesel production.

EVALUATING THE DILUTION OF MUNICIPAL WASTEWATER ON BIOMASS INCREASE, LIPID PRODUCTION AND NUTRIENT REMOVAL BY THE BLUE-GREEN ALGAE Spirulina platensis (Geitler)

Can, Safak Seyhaneyildiz; Demir, Veysel; Can, Erkan

Wastewater generated daily in urban localities could provide nourishment to algae for potential production of biofuel and biochemicals. This study presents an integrated approach to the use of Spirulina platensis cultures for the treatment of domestic wastewater and lipid extraction. In this experimental approach, wastewater subjected only to the primary settling process was used as nutrient media at different concentrations of 100%, 75%, 50%, and 25% wastewater diluted with distilled water, and Spirulina platensis was cultured in these mediums for 20 days. The inoculated medium was incubated at 30 degrees C in the presence of light (42 mu mol photon m(-2).s(-1)) by using light/dark cycle of 12 hours. S. platensis consumed nitrite, nitrate, phosphate and ammonium from wastewater, and it increased its biomass and its lipid amount. Although the highest biomass increase was observed at the wastewater medium without dilution (100% wastewater) (the control group was not included), the highest lipid production was found in 25% wastewater. On the other hand, the percent reduction in the level of nitrite, nitrate, phosphate and ammonium in 75% wastewater medium was 91%, 99%, 99% and 99%, respectively. Despite the low lipid content, since Spirulina platensis can live in harsh conditions and increase its biomass rapidly, they can be easily cultivated in wastewater. Therefore, their use in wastewater for biodiesel production was found appropriate. The resulting algae biomass after oil extraction can be also used in different industrial sectors.
