Genotoxic effects of copper nanoparticles on Vicia pannonica Crantz. root meristem cells

    Tasar, Neslihan

With the increasing use of metal nanoparticles, their access to the food chain has become the main concern for scientists. The present study sheds light on the copper nanoparticles (Cu NPs), which are widely used in many industries today. For this purpose, Vicia pannonica Crantz., a common fodder plant, was used and exposed to metal-based Cu NPs. To determine the genotoxic effects on root meristem cells, total of 3 different concentrations (50 ppm, 150 ppm, and 300 ppm) of Cu NPs and the control in deionized water solutions were prepared. The determination of the accumulation and elimination of Cu NPs in Vicia pannonica used in the study was carried out using with X-ray diffraction (XRD), scanning electron microscopy (SEM), SEM element mapping, and energy dispersive X-ray diffraction (EDS) characteristic spectrum analysis. All doses of Cu NPs caused significant decreases in the Mitotic index of V. pannonica meristem cells compared to the control. Multiple analyzes carried out in the study showed that Cu NPs caused abnormalities in cell division such as C-metaphase, polar aberration, deconstructed anaphase, chromosome breakage, loss of genetic material, failure to assemble in the metaphase plate, deconstructed metaphase, asynchronous division remaining chromosome in metaphase, adhesion, deconstructed prophase, advanced chromosomes, and chromosome bridges in anaphase. Results support that Cu NPs have a genotoxic effect on V. pannonica.

Genotoxic effect of iron oxide (Fe2O3) nanoparticles on Triticum aestivum (wheat)

    Tasar, Neslihan

Industrial activities and unconscious consumption of natural resources cause environmental pollution with the rapid increase in the world population. As a result of the widespread use of iron oxide nanoparticles (Fe2O3 NP) with nano-industrial activities, it is predicted that this NP will accumulate in the air, water, and soil. In the present study, the purpose was to find out the genotoxic effects on root meristem cells of the Triticum aestivum L. plant, which is an indicator organism exposed to 20–40 nm Fe2O3 NPs at different concentrations (100, 200, and 400 ppm). The amount of Fe2O3 NP accumulated in T. aestivum used in the study was determined with x-ray diffraction (XRD) spectroscopy, scanning electron microscopy (SEM), SEM element map, and EDS characteristic spectrum. All concentrations of Fe2O3 NP caused significant decreases in the mitotic index. Fe2O3 NPs significantly increased the frequency of mitotic abnormalities in T. aestivum root tip cells at all treatment times and all concentrations when compared to the control. Fe2O3 NPs were formed by various mitotic abnormalities such as loss of genetic material, deconstructed prophase, adhesion, chromosome groupings in metaphase, deconstructed metaphase, C-Metaphase, chromosomal loss, chromosomal fracture, polyploidy, deconstructed anaphase, lagging chromosome, fragment, polar deviation, bridging, propagation, asynchronous division, star anaphase, multipolarity, and deconstructed telophase. All these results show that Fe2O3 NPs are genotoxic and clastogenic and may also cause DNA damage. Briefly, these data show that Fe2O3 NPs taken by organisms may pose a danger to the organism and the upper consumer. These findings also show that the production and use of Fe2O3 NPs, which affect organisms, must be controlled, and ultimately, be safely disposed of to reduce their bioaccumulation. Research Highlights: In the present study, the purpose was to find out the genotoxic effects on root meristem cells of the Triticum aestivum L. (bread wheat) plant, which is an indicator organism exposed to 20–40 nm Fe2O3 NPs at different concentrations (100, 200, and 400 ppm). The amount of Fe2O3 NP accumulated in T. aestivum used in the study was determined with x-ray diffraction (XRD) spectroscopy, scanning electron microscopy (SEM), SEM element map, and EDS characteristic spectrum. The mitotic index was calculated to reveal the genotoxic effect. “Loss of genetic material, deconstructed prophase, adhesion, chromosome groupings in metaphase, deconstructed metaphase, C-Metaphase, chromosomal loss, chromosomal fracture, polyploidy, deconstructed anaphase, lagging chromosome, fragment, polar deviation, bridging, propagation, asynchronous division, star Chromosomal abnormalities such as anaphase, multipolarity, and deconstructed telophase” were visualized. Fe2O3 NPs are genotoxic and clastogenic and may also cause DNA damage.

Mitotic effects of copper oxide nanoparticle on root development and root tip cells of Phaseolus vulgaris L. seeds

    Tasar, Neslihan

Copper oxide nanoparticle (CuO NP) is used widely in many fields in nanotechnology. For this reason, both production, use, and release to the environment are increasing with each passing day. With the increased use of products that contain nanoparticles (NP) (<100 nm), plants and organisms that constitute the food chain are at risk. In the present study, Phaseolus vulgaris L., a very common food plant, was exposed to metal-based CuO NPs. The anomalies that were caused by CuO NP in germination and mitosis of P. vulgaris were investigated. In the trials, a total of 4 groups (Control, 50, 150, and 300 ppm) were formed and examined in three replications. The determination of the accumulation and elimination rate because of NPs in P. vulgaris that was used in the study was made through X-ray diffraction (XRD), scanning electron microscope (SEM), mapping image, and EDX characteristic spectrum analysis. Also, the mitotic effects on germination, root development, and root tip cells of seeds that were grown by treatment with control, 50, 150, and 300 ppm concentrations were investigated. The study was conducted in three replications in a laboratory setting. All concentrations of CuO NPs caused significant decreases in the mitotic index in the root tip cells of P. vulgaris when compared to the control. The mitotic index reached the lowest level, especially at the highest concentration. Multiple analyzes in the study showed that CuO NPs cause abnormalities in cell division such as C-metaphase, distorted metaphase, distorted anaphase and telophase, chromosome breakage, asynchronous division, advanced chromosomes, micronucleus, and loss of genetic material. These findings also support that the Cytogenetic Test of P. vulgaris can be used to evaluate the genotoxicity of new nanomaterials that are used in many consumer products. In this respect, NPs that are taken up by the organisms in the food chain may pose a danger to higher consumer organisms when they accumulate in the tissue. A control mechanism must be established for the use and contamination of these particles and wider studies must be conducted regarding their effects. Highlights: The effects of CuO nanoparticle, which has a very wide usage area, on root development and mitosis of Phaseolus vulgaris L. plant were investigated in the study. The abnormalities of mitotic division on interphase, prophase, metaphase, anaphase, and telophase were visualized. Evaluation was made considering scanning electron microscopy (SEM) and X-ray diffraction (XRD) results as well.
