Unal, İlkay
Hydroxyapatite (HAp) is widely used in biomedical applications due to its biocompatibility, osteoconductivity, and bioactivity. However, its low mechanical strength, tendency toward rapid corrosion, and lack of bactericidal properties present significant limitations in applications. This study aimed to improve the properties of HAp by reinforcing it with multi-walled carbon nanotubes (MWCNTs) and doping it with silver nanoparticles (AgNPs) and silver-core selenium-shell nanoparticles (Ag@SeNPs). Ocimum basilicum extract was used as both a reducing and stabilizing agent in the synthesis of nanoparticles using an environmentally friendly and non-toxic method as an alternative to traditional methods. The synthesized HAp, HAp/MWCNT, Ag-HAp/MWCNT, and Ag@Se-HAp/MWCNT nanocomposites were characterized by TEM, SEM, XRD, Raman spectroscopy, and BET analysis. BET analysis showed a reduction in surface area from 109.4 m2/g for pure HAp to 71.4 m2/g, 47.5 m2/g, and 35.3 m2/g for HAp/MWCNTs, Ag- HAp/MWCNTs, and Ag@Se-HAp/MWCNTs, respectively. Antimicrobial activities against P. aeruginosa, E. coli, S. aureus, E. faecalis, and C. albicans were evaluated. HAp and HAp/MWCNT did not show any antimicrobial activity, while Ag-HAp/MWCNTs showed inhibition zones of 14 mm for Escherichia coli and 18 mm for Pseudomonas aeruginosa at 5 mg/mL. Ag@Se-MWCNTs/HAp exhibited superior efficacy with inhibition zones of 18 mm, 12 mm, and 20 mm for S. aureus, E. faecalis, and Candida albicans, respectively. The incorporation of Ag@SeNPs enhanced HAp’s antibacterial and antifungal properties through a synergistic mechanism.

Unal, Ilkay; Egri, Sinan
This study describes an eco-friendly procedure to synthesize silver nanoparticles (AgNPs) using fresh and dried stem extract of Rheum ribes. AgNPs formation was confirmed using UV-vis spectrometry. The effects of parameters such as temperature, reaction time, pH, AgNO3 concentration, and plant extract/AgNO3 ratio on NP formation were evaluated. Morphology and structural features of the particles were characterized using SEM, TEM, XRD, and FTIR. The average size, polydispersity index, and zeta potential were determined using DLS. After characterization, among the two AgNP samples, selected one; AgNP sample synthesized by dried Rheum ribes stem extracts (d-AgNPs), which exhibited narrow size distribution and high surface plasmon resonance in the range of 435-445 nm for toxicity assessment. The cytotoxic effect of d-AgNPs on HUVECs cells was evaluated with MTT analysis. Accumulation measurements were made in three periods as 24, 48, and 72 h and elimination values were examined at +24 h using ICP-MS analysis

- MakaleAçık Erişim2024Evaluation of the Effect of Different Extraction Temperatures on the Synthesis of Silver Nanoparticles from Ocimum basilicum (Basil) Plant
Aydogdu, Burcu; Unal, Ilkay
The eco-friendly green synthesis of silver nanoparticles (AgNPs) using Ocimum basilicum (basil) extract at varying extraction temperatures (40°C, 60°C, 80°C, 100°C) was investigated to determine the optimal conditions for nanoparticle formation. Analysis methods such UV-Vis spectrophotometry, Fourier Transform Infrared Spectroscopy (FTIR), X-ray Diffraction (XRD), and TransmissionElectron Microscopy (TEM) confirmed the crystalline, spherical nature of AgNPs and identified phytochemicals acting as capping and reducing agents. Notably, the extraction temperature was found to influence both the DPPH radical scavenging activity and the structural properties of AgNPs. According to TEM analysis results, it was observed that high extraction temperatures increased the nanoparticle formation efficiency but created a wide size distribution. The crystallite sizes, calculated using the Scherrer equation, for AgNPs synthesized at different extraction temperatures, were determined to be 12.45 nm, 18.77 nm, 17.76 nm, and 16.03 nm, respectively. The hydrodynamic sizes of the AgNPs were found to range between 158.1 and 333.7 nm. The study highlights the critical role of extraction temperature in the synthesis process, suggesting 40°C as the optimal temperature for achieving efficient and environmentally friendly synthesis of AgNPs with enhanced biological activities.

The nanoparticles (NPs) were characterised by various analyses, including TEM, SEM, XRD, FTIR, DSL, and zeta potential. Based on the TEM image, the ZnONPs exhibited a cluster of flower-like structures, whereas the Fe3O4NPs displayed a spherical shape with a varying size distribution. The zeta potential values for ZnO NPs ranged from -5.35 to -16.9, while for Fe3O4NPs ranged from -7.43 to -20.7. All ZnO nanoparticles exhibited antimicrobial activity exclusively against the S. aureus strain, whereas Fe3O4NPs did not demonstrate any antibacterial effect.

İncele - MakaleYalnız künye2024Scopus SJRQ2Plant extract-based biosynthesis of silver nanoparticle and silver–zinc nanocomposites-doped hydroxyapatite and its antimicrobial activity
Unal, Ilkay; Aydoğdu, Burcu; Aytar, Mehmet
Hydroxyapatite (HAp) is an essential material in the biomedical field because of its chemical composition, similar to the apatites found in bones. The lack of bactericidal properties of HAp-based materials cannot prevent the adhesion and growth of bacteria. However, it can develop composite biomaterials using nanoparticles (NPs) such as silver (Ag) and zinc (Zn), which exhibit biocidal behaviours to control resistant microorganisms. This study aimed to prepareHAp–AgNPs and HAp–Ag@ZnNPs nanocomposite and evaluate their antimicrobial properties against Gram-positive and Gram-negative bacteria. HAp powders were produced from sheep bone. An aqueous extract of Marrubium astracanicum was used as a reducing and stabilising agent for the synthesis of NPs. The composites were characterized by XRD, SEM–EDX, SEM and FTIR. In vitro biocompatibility assessment was determined by agar well diffusion tests. The HAp–Ag@ZnNPs nanocomposites showed stronger antimicrobial activity than HAp–AgNPs against bacteria and fungi, while pure HAp did not show any effect.

Aydogdu, Burcu; Kaplan Ince, Olcay; Ince, Muharrem; Unal, Ilkay
Multi-walled carbon nanotubes (fMWCNTs) that functionalized and loaded Ag nanoparticles (AgNPs) were investigated to adsorp doxorubicin hydrochloride (DXR). DXR release behavior efficiency was determined using kinetic studies. DXR agent was treated into fMWCNTs@AgNPs material with different drug loading rates. The material was successfully established and characterized using transmission electron microscopy, and fourier transform–infrared spectroscopy. Thermogravimetric analysis was performed to elucidate the effect of temperature on mass loss and X-ray diffraction analysis was systematically assessed the crystalline state of DXR. Various kinetic models including Higuchi, Baker Hixson-Crowell, and power-law were applied. It was revealed that DXR has more binding affinity to the fMWCNTs@AgNPs material system surface as a nanocarrier at pH 7.4 and showed more release at pH 5.0. Functionalized material confirmed its efficiency as pH-based drug delivery system. Used material can be utilized as promising nanocarriers, and findings will contribute to the rational design of novel delivery tools for drugs.
