Yazdıç, Ferit Can; Kar, Bülent; Karaman, Altuğ
In this study, phylogenetic relationships and genetic diversity of Salmo spp. populations in different water bodies in Türkiye were investigated, focusing on morphologically similar specimens to Salmo macrostigma. Significant genetic variability and evolutionary differentiation within these populations have been identified using mitochondrial regions sequences (12S rDNA, 16S rDNA, and Cyt-b), morphological observations, and cytogenetic analysis. Samples collected from eight river basins revealed three main phylogenetic clades and local adaptations influenced by ecological pressures. Cytogenetic findings showed that they have chromosome numbers ranging from 78 to 80 (2n), consistent with known Salmo species. For this purpose, three mitochondrial DNA fragments encoding 12S rDNA, 16S rDNA, and Cyt-b regions were amplified by PCR, sequenced, and then used for phylogenetic analysis of the studied species. The study revealed the need for comprehensive integrative taxonomic approaches to clarify the classification of Salmo spp. with critical implications for conservation strategies in Türkiye biodiversity hotspots. In conclusion, this study not only advances our knowledge of Salmo macrostigma or similar Salmo spp., but also suggests innovative approaches for future Salmonid phylogenetics and cytogenetics studies. The findings have important implications for understanding the evolution, taxonomy, and conservation of these economically and ecologically important fish.

Absract: In this study, chloroplast DNA (cpDNA) ndhF sequence analysis was used to determine the sequence analysis and genetic diversity of Chrysophthalmum Schultz Bip. species distributed in Türkiye. Genomic DNA was isolated from green leaves and cpDNA ndhF region was amplified by PCR. The results of the PCR, the forward and reverse sequences were edited using the Sequencher 5.4.6 programme and the contig sequences were obtained. To determine the phylogenetic relationships between species, MEGA 6.0 was used to construct a maximum likelihood (ML) tree. Using the same program, nucleotide composition, phylogenetic trees, genetic distance matrix, and genetic diversity analyses were performed. In addition, the physicochemical properties and 3D structure of ndhF proteins were determined. From the results of the ndhF analyses, the nucleotide length of Chrysophthalmum species was found to vary between 471 and 625. The genetic matrix value was determined to be between 0.000 and 0.009. In the phylogenetic tree constructed using ndhF sequences from Anvillea DC., Pulicaria Gaertn., Limbarda Adans, Inula L., Carpesium L., Pentanema Cass., Dittrichia Greuter, Rhanteriopsis Rauschert and Telekia Baumg species from NCBI, C. montanum was grouped with Inula, Pentanema and Rhanteriopsis species. C. gueneri was grouped with C. dichotomum and Pentanema alanyense. Previous phylogenetic studies were in agreement with our results. In addition, the ndhF results supported that Chrysophthalmum species are not a monophyletic group.

In this study, the phylogenetic relationship of four species of Dianthus genus which are endemic and distributed in Türkiye, including D. zederbaueri Vierch, D. varankii Hamzaoğlu & Koç, D. halisdemirii Hamzaoğlu & Koç, and D. goekayi Kaynak, was determined using nuclear DNA (nrDNA) internal transcribed spacer (ITS) and chloroplast DNA (cpDNA) rbcL regions. After sampling from the plant, DNA was extracted and the ITS (ITS4-ITS5A) and rbcL (rbcLaF-rbcLaR) primers were used to perform PCR amplification. Sequencer 5.4.6. was used to edit the sequences. MEGA 11 software was used to determine the nucleotide ratios, genetic distance matrix, and π = nucleotide diversity. Some species of Dianthus available in the National Center for Biotechnology Information (NCBI) were used to construct maximum likelihood (ML) phylogenetic trees. As a result, based on the ITS analysis π = nucleotide diversity was determined as: 0.001543, and the result of the rbcL analysis π = nucleotide diversity was determined as: 0.000000. In the ITS results, D. halisdemirii, D. superbus, and D. varankii appeared together, while D. zederbaueri and D. aculeatus appeared together. The rbcL results showed that D. zederbaueri and D. varankii were closely related, but the relationship between D. halisdemirii and D. goekayi could not be determined. Furthermore, in the phylogenetic tree constructed by taking the ITS and rbcL sequences of Dianthus, Gypsophila, Silene, Arenaria, Saponaria, Cerastium, Stellaria, Petrorhagia, Minuartia, Agrostemma, and Polycarpon species from NCBI using ITS and rbcL sequences, our endemic Dianthus species appeared in the same group with Petrorhagia, Gypsophila, and Saponaria species. To conclude, our ITS and rbcL results were consistent in determining the phylogenetic relationships of our Dianthus species with other species.

Starch and gelatin offer opportunities for use due to their non-toxicity, biocompatibility, physical and functional properties. In this study, different amounts of nano-zinc borate were added to starch/gelatin composition for biological applications and their properties were studied. The structure of the films was investigated by various techniques. Nanostructured zinc borate was synthesized using a chemical method and particle sizes were determined as 196 nm. The atenolol drug was doped into the polymer composite. The atenolol release was performed at pH 7.4 and in phosphate-buffered saline solution medium. Cumulative drug release was 76% with the inclusion of nano-zinc borate in the polymer mixture. The atenolol release was approximately 50%–70% in films with nano-zinc borate added after 300 min. Furthermore, thermal resistance of the films improved with the addition of nano-zinc borate. The investigated chemical compound exhibited pronounced antibacterial activity against Gram-positive bacteria (Bacillus subtilis and Staphylococcus aureus), with significantly greater potency compared to Gram-negative bacteria (E. coli and Pseudomonas aeruginosa). Notably, the formulation containing 1.5% starch, 0.5% gelatin, and 1% nano-zinc borate demonstrated broad-spectrum efficacy against all tested pathogens, highlighting its potential as a promising antibacterial agent. The observed differential activity against bacterial strains suggests that fine-tuning the concentrations of starch, gelatin, and nano-zinc borate may optimize antibacterial efficacy and broaden the spectrum. The results showed us that non-toxic and biocompatible polymer films can be realized in pharmaceutical industry systems and biomedical applications.
- MakaleYalnız künye2025Scopus SJRQ3Antioxidant Enzyme Responses to Lead Exposure in Daphnia magna
Sahilli, Yeliz Cakir
Abstract: Lead (Pb), a pervasive environmental contaminant, poses significant toxicological risks to freshwater ecosystems. Daphnia magna, a widely used model organism in aquatic toxicology, provides valuable insights into the sublethal effects of heavy metal exposure. This study investigated the effects of Pb exposure on antioxidant defense mechanisms in D. magna. Test organisms were exposed to aqueous Pb solutions at varying concentrations (0, 10, 20, 30 μg/L) for 96 h. The enzyme activities of glutathione peroxidase (GPx), catalase (CAT), and superoxide dismutase (SOD) were measured using enzyme-linked immunosorbent assay (ELISA). SOD activity did not differ significantly between the exposed and control groups. However, CAT activity showed a significant decrease in all Pb-exposed groups compared to controls. GPx activity exhibited a non-significant decrease. In conclusion, Pb exposure may induce oxidative stress by generating reactive oxygen species (ROS) and inhibiting the function of sulfhydryl-containing antioxidant enzymes. Changes in antioxidant enzyme activity may serve as early biomarkers in ecotoxicological assessments of Pb pollution in aquatic environments. It is suggested that future studies incorporate molecular-level endpoints and chronic exposure scenarios to better elucidate the long-term impacts of Pb on aquatic invertebrates.
