Green revolution in nanotechnology: phytogenic CuO nanoparticles for biomedical and pharmaceutical applications

Ceylan, Yusuf; Korkmaz, Nesrin; Kaçan, Fatma N.; Aydin, Ali; Korkmaz, Şengül A.; Yildirim, Ebru Ş.

Copper oxide nanoparticles (CuO NPs) were synthesized using a green approach with Vitis vinifera L. extract, a plant rich in secondary metabolites that act as natural reducing agents. The obtained CuO NPs were characterized by XRD, EDX, and STEM analyses. FT-IR analysis revealed the presence of functional groups originating from plant biomolecules and confirmed the formation of Cu–O bonds, indicating the successful synthesis and stabilization of CuO NPs. XRD confirmed distinctive CuO diffraction peaks at 32.38°, 35.35°, 38.61°, 40.75°, 48.79°, 51.90°, 57.66°, 61.38°, 65.94°, and 75.12°. STEM imaging showed predominantly spherical, well-dispersed nanoparticles with an average size of 23–38 nm. Antibacterial analysis revealed a minimum inhibitory concentration (MIC) of 4 µg mL−1 against Escherichia coli ATCC 25922. In anticancer assays, the CuO NPs demonstrated notable cytotoxicity at both low concentrations and even at levels below the control group in A549, Calu-1, and Saos-2 cancer cell lines, highlighting their strong biomedical potential.

New dinuclear cyanido complexes with amine alcohol ligand: Synthesis, characterization and biotechnological application potential

    Korkmaz, N.; Korkmaz, Ş.A.; Ceylan, Y.; İmamoğlu, R.; Bülbül, A.S.; Karadağ, A.

In this study, the cyanido complexes given by the formula [Ni(Abut)Ni(CN)4 ]·8H2 O (C1), [Cu(Abut)2 Ni(CN)4 ]·7H2 O (C2), [Zn(Abut)Ni(CN)4 ]·8H2 O (C3) and [Cd(Abut)Ni(CN)4 ]·7H2 O (C4) were ob-tained by microwave synthesis method. The powder forms of the complexes were characterized by elemental, FT-IR spectroscopy, and thermal analysis. And also antibacterial, antibiofilm and anticancer activities were investigated. The splitting stretching bands of cyanido groups in the FT-IR spectra of C1-C4 indicated the assets of terminal and end cyanido groups. The antibacterial activities of C1-C4 were tested with nine Gram negative and six Gram positive bacteria. The most efficient antibacterial activity of complexes was observed at 1000 µg/ml-1 concentration. Anticancer activity was tested using HeLa cell line and MTT test. The studied cyanide complexes have been shown to decrease the viability of HeLa cells with IC50 values 14.86, 6.5, 7.2 and 19.2 µg/ml for C1, C2, C3 and C4 complex, respectively.

Evaluation of pharmacological activity of heterobimetallic coordination compounds containing n, n-bis (2-hydroxyethyl)-ethylenediamine on ht29, hela, c6 and vero cells

    Aydin, Ali; Korkmaz, Şengül Aslan

The present study was conducted in order to investigate the pharmacological activities of three heterobimetallic coordination compounds: [Cd(N-bishydeten)2 ][Ni(CN)4 ] (C1), [Cu2 (N-bishydeten)2 Co(CN)6 ].3H2 O (C2), and K[Cd(N-bishydeten)Co(CN)6 ].1.5H2 O (C3) (N-bishydeten = N,N-bis(2-hydroxyethyl)-ethylenediamine). This paper describes the ability of complexes to inhibit cell growth, cell migration and human topoisomerase I and to interact with DNA/BSA; this paper also evaluates the potential mechanisms of action exhibited by these compounds via the use of powerful measurement techniques. Studies on HT29, HeLa, C6 and Vero cells revealed that each compound demonstrated significant antiproliferative activity in conjunction with regressed cell migration velocity and caused apoptotic changes in morphology. There are strong data suggesting that the mechanisms of action exhibited by these compounds are associated with their DNA/BSA binding features. The IC50 and binding constant range for the compounds are 20-180 µM and 1.2-3.2 x 104 M-1, respectively. Moreover, we observed that these compounds alter the P53-Bcl-2 ratio and inhibit the relaxation activity of human topoisomerase I. Furthermore, a correlation between the antiproliferative effects of these compounds and their cytotoxic activity was observed. In conclusion, preliminary information demonstrates that these compounds have been found to exhibit effective antiproliferative activity against cancer cell lines, indicating that they are a potent candidate for further pharmacological study.

Anticancer and Cytotoxic Activities of [Cu(C6H16N2O2)2][Ni(CN)4] and [Cu(C6H16N2O2) Pd(CN)4] Cyanidometallate Compounds on HT29, HeLa, C6 and Vero Cell Lines

Aydin, Ali; Korkmaz, Sengul Aslan; Demir, Veysel; Tekin, Saban

Background: In cancer, apoptosis relevant proteins-such as CaM kinase, Bcl-2 or P53, topoisomerase I, cell migration feature and DNA/BSA-macromolecules represent significant targets for current chemotherapeutics. Objective: We recently reported two coordination compounds-[Cu(C6H16N2O2)(2)][Ni(CN)(4)] (1) and [Cu(C6H16 N2O2) Pd(CN)(4)] (2)-together with their IR spectra, magnetic properties, thermal analyses and crystal structures. Herein, we describe the ability of these complexes to induce apoptosis in relevant proteins and stimulate topoisomerase I activity, cell migration velocity and DNA/BSA binding properties. Method: The in vitro antiproliferative effects and cell toxicity of both compounds were investigated through pharmacological measurement techniques, and interactions between both compounds and CT-DNA/BSA were studied with UV-Vis spectroscopy and fluorescence spectroscopy. Results & Conclusion: Studies on cells revealed that 2 (i) demonstrated a high antiproliferative effect, which was higher toward HeLa and C6 cancer cells than toward healthy Vero cells; (ii) impaired the migration of HeLa cells; (iii) altered the P53-Bcl-2 ratio in favor of apoptosis; (iv) strongly bound to DNA/BSA macromolecules; and (v) inhibited human topoisomerase I and KpnI or BamHI restriction endonucleases. In conclusion, this preliminary information demonstrates that 2 may represent a promising antiproliferative agent and a potential candidate for a therapeutic approach against HeLa.

Binuclear cyanido complexes containing [Pt(CN)4]2- building block: Synthesis, crystal structures, magnetic properties and anticancer activities

Korkmaz, Sengul Asian; Karadag, Ahmet; Aydin, Ali; Yerli, Yusuf; Soylu, Mustafa Serkan

Five new complexes of composition [Ni(N-bishydeten)Pt(CN)(4)] (1), [Ni(N-bishydeten)(2)][Pt(CN)(4)] (2), [Cu (N-bishydeten)Pt(CN)(4)] (3), [Cu(N-bishydeten)(2)][Pt(CN)(4)] (4) and [Cd(N-bishydeten)(2)][Pt(CN)(4)] (5) (N-bishydeten = N,N-bis(2-hydroxyethyl)-ethylenediamine) have been obtained and characterized by IR and EPR spectroscopies, elemental and thermal analysis and magnetic measurements. X-ray studies of 2 and 3 exhibited different structures and two different coordination modes of N-bishydeten ancillary ligand with a ratio of M:L. While N-bishydeten acts as a tetradentate with N', N', O and O' atoms in a molar ratio of 1:1 formed 2,2-CT-type zigzag chain for 3, it behaves tridentate ligand by ending up with six coordinated Ni-II center in a complex salt by using 1:2 M ratio for 2. None of the 1 and 3 complexes produced in this study showed sufficient anticancer activity but all did exhibit apoptotic properties and one of them also performed low cytotoxic activity against cell tested.
