Gürkan, Mert; Gürkan, Selin Ertürk; Yılmaz, Sevdan; Ateş, Mehmet
In this study, alpha and gamma iron oxide nanoparticles, characterizations, toxic effects after being exposed to rainbow trout (Oncorhynchus mykiss) at 0, 1, 10 and 25 mg/L concentrations for 10 days, and then 10-day recovery period without any nanoparticle exposure were examined for histopathological (kidney, liver and gill), hematological, iron accumulation and potential for oxidative stress (TBARS and GSH). Histopathological damages significant at the exposure of increasing concentrations of both nanoparticles (increase in melanomacrophage aggregations, epithelial tissue deformations, cytoplasmic vacuolizations, fatty changes, necrosis, pyknosis, hyperplasia, hypertrophy, lamellar fusions, capillary dilatations). Gamma nanoparticles were determined to accumulate more than the alpha nanoparticles. The most Fe accumulation was detected in the liver. The findings of oxidative stress parameters showed that both nanoparticles have the potential to generate oxidative stress. It was concluded that the exposure of alpha and gamma nanoparticles at specified concentrations and durations had a toxic effect on rainbow trout and the toxicity of these nanoparticles was similar.

Nanoparticles have great impact due to their tremendous industrial applications. However, their applications have produced toxicity effects on the aquatic environments and their detailed analyses are not clearly understood. Iron oxide nanoparticles (Fe2O3 NPs) are being used extensively in many industries but are considered highly toxic to aquatic species residing in surface waters. This paper demonstrates the acute toxicity of alpha-Fe2O3 and gamma-Fe2O3 NPs in two aquatic species. The effects of various concentration (0, 50, 100, 250, 500 and 1000 mg/L) of alpha-Fe2O3 and.-Fe2O3 on the sensitivity response of the Chlorella sp. and D. magna were investigated. The growth of microalgal decreased with increased concentration of the alpha-Fe2O3 and gamma-Fe2O3 NPs concentrations but did not show a significant toxic effect. The EC50 concentration value was 500 mg/L and LD50 concentration value was 1000 mg/L for alpha-Fe2O3 treated daphnids in 72 h, respectively. The findings demonstrate the significant evidence in understanding acute toxicity of Fe2O3 NPs for environmental protection as part of risk assessment strategies.

- MakaleYalnız künye2020Scopus SJRQ3Potential anesthetic properties of bay leaf (Laurus nobilis) essential oil compared with 2-phenoxyethanol on blue dolphin cichlid, cyrtocara moorii
Kizak, Volkan; Can, Erkan; Seyhaneyildiz Can, Şafak
The efficacy of Bay leaf (Laurus nobilis) essential oil (LnEO) as a herbal anesthetic agent was evaluated with 2-Phenoxyethanol (2-PE) in Blue Dolphin Cichlid, Cyrtocara moorii. Fish were exposed to different anesthetic concentrations and the lowest effective concentrations (LECs) were determined according to deep anesthesia (AD<3 min) and full recovery (RF<5 min) times. LnEO showed anesthetic properties, and all applied concentrations of both agents induced AD in C. moorii. The LECs of LnEO and 2-PE for C. moorii were determined as 800 µl L-1 (AD; RF → 183 ± 6.3 s; 191 ± 2.6 s) and 600 µl L-1 (AD; RF → 171 ± 3.2 s; 239 ± 4.9 s), respectively. Significant differences were found between anesthetic agents in terms of deep anesthesia and full recovery times for the same concentrations (p<0.05). Induction and recovery times decreased with increasing in LnEO concentrations. On the other hand, anesthesia times were decreased with increasing of 2-PE concentrations, while recovery times increased. Induction and recovery times for LnEO were significantly dependent on concentrations and positive relationships were recorded between AD and RF, whereas the relationship was negative in 2-PE. Bay leaf essential oil has not been used as an anesthetic agent in fish until now. Our results showed that LnEO was an effective anesthetic agent with some minor side effects on C. moorii.

- MakaleYalnız künye2019Scopus SJRQ4THE TOXIC EFFECT OF CADMIUM ON HUMAN ERYTHROCYTES AND NAC'S PROTECTIVE ROLE
Sahilli, Yeliz Cakir
The toxic effects cadmium (Cd), which is a heavy metal, on human erythrocytes and protective effects of N-acetyl-L-cysteine (NAC) on lipid peroxidation and antioxidant enzymes were investigated. The effects of 1, 50, 150 mu M doses of cadmium and 1mM NAC on the level of malondialdehyde (MDA) in human erythrocytes and superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) enzyme activities were investigated. When the Cd was applied alone, it was found that MDA level increased in erythrocytes and SOD, CAT and GPx activities decreased. No statistically significant difference was observed when 1mM NAC administered erythrocytes were compared with the control cells. However, the beneficial effect was seen only in administration doses of Cd at the levels of I and 50 mu M and NAC was observed to have a protective effect. As a result, NAC partially reduced the toxic effect caused by Cd on erythrocytes but could not completely prevent it.

Sub-Sahara Africa is burdened with a high incidence of parasitic infections, including schistosomiasis, trypanosomiasis, trichomoniasis, and leishmaniasis. Currently, there is a rapid widespread development of resistance to prescription drugs for these neglected diseases. Microbes provide the largest chemical and biological diversity in drug discovery screening programs; therefore, our project seeks to explore microbes in the sub-region for new drugs. The oxylipin (9Z,11E)-13-oxooctadeca-9,11-dienoic acid (1) was isolated from the Ghanaian endophytic fungus, Penicillium herquei strain BRS2A-AR obtained from the leaves of a Laguncularia racemosa tree growing on the banks of the River Butre in the Western Regional wetlands of Ghana. Compound 1 was isolated on reverse phase HPLC at tR of 34.3 minutes. The structure of compound 1 was confirmed by a combination of mass spectrometry, 1D and 2D-NMR techniques. Compound 1 showed minimal antiparasitic activity when tested against Plasmodium falciparum 3d7 (IC50>100µM; aretesunate, IC50 36nm), Trypanosoma brucei brucei (IC50>100µM; Coptis japonica, IC50 8.20µM), Leishmani donovani (IC50>100µM; amphotericin B, IC50 0.32µM) and Leishmania major (IC50>100µM; amphotericin B, IC50 0.31µM). Compound 1 produced IC50 of 44.47µM when tested against Trichomonas mobilensis with metronidazole (IC50 5.20µM) as standard. These results show the potential to further engineer the structure of compound 1 into a potent anti-Trichomonas scaffold.

- MakaleYalnız künye2019Scopus SJRQ3Anesthetic Efficiency of Three Medicinal Plant Oils for Aquatic Species: Coriander Coriandrum sativum, Linaloe Tree Bursera delpechiana, and Lavender Lavandula hybrida
Can, Erkan; Kızak, Volkan; Can, Şafak Seyhaneyıldız; Özçiçek, Esin
This study evaluated the potential of three essential oils (EOs) that were obtained from coriander Coriandrum sativum (CEO), linaloe tree Bursera delpechiana (BEO), and lavender Lavandula hybrida (LEO) as anesthetic agents. Convict Cichlids Amatitlania nigrofasciata (Günther 1867) were exposed to eight concentrations of anesthetic (50, 75, 100, 125, 150, 200, 250, and 300 μL/L). After exposure to the anesthetic, the fish were transferred to clean water to recover. All of the EOs produced an anesthetic effect after exposure to the compounds for 30 min at the minimal effective concentration (MEC), which was identified according to deep anesthesia (A5 < 3 min) and full recovery (R3 < 5 min) times. At 50 and 75 μL/L, the total loss of equilibrium was not observed for all tested EOs. The total loss of reflex was induced at a faster rate with higher concentrations of anesthetic in all groups. The recovery time generally increased as the concentration of the anesthetic increased. These findings suggest that CEO, BEO, and LEO are all novel potential anesthetics for aquaculture, and the optimal concentrations were identified as 150 μL/L (A5; 156 ± 1.7 s and R3; 165 ± 2.9 s), 125 μL/L (A5; 176 ± 3.5 s; R3; 125 ± 2.0 s), and 200 μL/L (A5; 20.1 ± 2.4 s and R3; 162 ± 3.4 s), respectively. When considering the active ingredients of EOs, this study also demonstrated that future studies should be focused on the major components such as linalyl acetate, 1.8-cineole, α-pinene, geraniol, and linalool. Their synergistic effects should be examined in herbal anesthetic treatments, since new commercial anesthetics will likely contain them.
