Akbulut, Gulcin Beker; Torgut, Gulben
Salt stress is one of the most important environmental factor limiting plant growth and causing serious agricultural problems in the world. Biodegradable poly(ethylene oxide)-co-(ε-caprolactone) poly(EO-co-ε-CL) copolymer synthesized by ring-opening polymerization (ROP) have been used to reduce this negative effect. For the characterization of this copolymer, the Fourier Transform Infrared (FT-IR), Proton Nuclear Magnetic Resonance (1H-NMR), Differential Scanning Calorimetry (DSC) and Thermogravimetric Analysis (TGA) techniques were used. To investigate the impact of copolymer and salt (200 mM NaCl) singly and in combination, on two different sunflowers (Helianthus annuus L. cv. 'Düet' and Helianthus annuus L. cv. 'Meteor') a series of experiments were established. In the seedlings grown for 21 days, differences in some metabolic parameters such as chlorophyll-a (Chl-a), chlorophyll-b (Chl-b), carotenoids (Car), total chlorophyll (Chl), total carbohydrate content, and malondialdehyde (MDA) content were examined. According to the result of our study, after the application of NaCl and poly(EO-co-ε-CL), significant changes were observed in the studied biochemical parameters. It was observed that the copolymer in both species had a healing effect against salt stress. Application of 200 mM NaCl caused an increase in MDA of both genotype, particularly in 'Meteor'. Higher Chl-a, Chl-b, total Chl, and Car contents were observed in 'Meteor' as compared to that in 'Düet' under saline conditions. Total carbohydrate content decreased with increasing salt stress in both cultivars.

Beker Akbulut, Gulcin; Yigit, Emel; Bayram, Dilek
Glyphosate is an herbicide that is applied after non-selective germination and affects plant growth. In this study, glyphosate was applied to Zea mays L. after germination, at a concentration range from 0.017 to 0.145 M in a growth chamber. The effects of this herbicide on some antioxidant enzymes, lipid peroxidation, total chlorophyll and total carbohydrate content were investigated on days 1, 5 and 10 following the application. Results showed that peroxidase (POD), ascorbate peroxidase (APX), superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST) were increased in the groups treated with glyphosate on days 5 and 10, but the activities of reduced glutathione (GSH) and glutathione reductase (GR) were decreased on day 10. Malondialdehyde (MDA) content indicating lipid peroxidation was increased on days 1, 5 and 10. Changes in total chlorophyll and total carbohydrate contents were found to be time-dependent. These increases and decreases in the antioxidant activities, MDA content, total chlorophyll and total carbohydrate content in Zea mays L. were determined as a symptom of cytotoxicity caused due to glyphosate.

In this study, we evaluated effects of fenoxaprop-p-ethyl herbicide on wheat (Triticum aestivum L.). The wheat plants were grown in pots containing perlite. Fenoxaprop-p-ethyl was applied to adequately grown T. aestivum plants at determined doses (1 mM, 1.5 mM, 2.2 mM, 3.4 mM and 5.1 mM) via spraying. The effects of this herbicide on some antioxidant enzymes, lipid peroxidation, carotenoid, total chlorophyll and total phenolic were investigated on days 1, 5 and 10 following the application. Environmental pollutants cause oxidative stress, production of free radicals and changes in antioxidants. Synthesis of signal molecules such as salicylic acid (SA) is important for the stress response of plants. In addition to this, although selenium is known not to be an obligatory element for plants, low dose selenium has positive effects on the plant growth. We also investigated the effects that are elicited on parameters above mentioned by the treatment of exogenous 0.5 mM SA and 0.004 mM organic selenium (SeI) separately. Our findings reveal that SA and SeI, which are externally applied before the germination of seeds that are exposed to herbicide stress, becomes effective in reduction of the negative effects of fenoxaprop-p-ethyl, with regard to antioxidant system, lipid peroxidation, pigment system and total phenolic. When we evaluate the study results, our findings suggest that especially SA plays a protective role on wheat against herbicide stress.

- MakaleYalnız künye2018Scopus SJRQ2Effect of the novel biodegradable copolymer and soil salinity on the growth of corn plant
Torğut, Gülben; Beker Akbulut, Gülcin
Biodegradable and biocompatible poly (1,2-epoxy-3-phenoxy propane)-co-(ε-caprolactone) poly(EPP-co-ε-CL) copolymers were synthesized by ring-opening polymerization method at four different ε-CL ratios. Copolymers were characterized using Fourier-transform infrared spectroscopy, proton nuclear magnetic resonance (1H-NMR), thermogravimetric analysis, and differential scanning calorimetry. To examine their possible applications in agriculture, germination test and growth analyses of corn plants were carried out. For this purpose, corn (Zea mays L. cv. “72 May 80”) was allowed to germinate under suitable conditions. Copolymers, 200 mM salt (NaCl) solution, and both copolymers and salt solution were applied to the plants and compared with the control groups. On the 21st day, all the plants were harvested and were stored at −80°C until analysis. Differences in growth parameters, lipid peroxidation degrees, pigment analysis, and total carbohydrate analysis were tested in order to put forward copolymer and NaCl sensitivity of the corn. Statistical analysis was carried out using the SPSS 17.0 software program and the results were compared with each other. These results showed that the copolymers both have a positive effect on the carbohydrate and malondialdehyde (MDA), a product of lipid peroxidation content as well as the chlorophyll content. Moreover, poly(EPP-co-ε-CL0.88) was more efficient than was the other copolymers in the improvement of the detrimental effects of salinity in increasing chlorophyll, carbohydrate, and MDA content, and growth of corn plants. As a result of the study, it was demonstrated that all of the investigated parameters were affected by the applications of salt and copolymers.

- MakaleYalnız künye2016Scopus SJRQ3The effects of sebum configuration on Demodex spp. density
Demirdag, Hatice Gamze; Özcan, Hamdi; Gürsoy, Sule; Beker Akbulut, Gulcin
Background/aim: Demodex spp. are ectoparasites living in the pilosebaceous units, which feed on the host's sebum and cellular proteins. The protective barrier of the skin consists of sebum secretion, moisture, and the acid mantle. In this study, we aimed to determine the effects of skin sebum, moisture, pH levels, and sebum configuration on Demodex spp. density Materials and methods: Forty-five patients who had demodicosis were enrolled in the study group, while the control group consisted of 40 subjects without demodicosis. Body fat percentage, serum triglyceride and cholesterol levels, skin sebum, moisture, and pH levels were measured. Demodex spp. density was determined with a standardized skin surface biopsy. Sebum samples were taken from the forehead and a high-performance thin-layer chromatography (HPTLC) method was performed on these samples. Subsequently, densitometric analyses were applied to the HPTLC plates. Results: Demodex spp. were found on the cheeks and lived in an alkali environment. Skin sebum and moisture levels were low in all groups. The skin pH levels and cholesterol ester in the sebum configuration were determined to be significantly higher in the group with demodicosis. Conclusion: We suggest that Demodex spp. may use cholesterol ester in the sebum as nutriment. In other words, cholesterol ester may be a suitable growth medium for the proliferation of Demodex spp.

- MakaleYalnız künye2016Scopus SJRQ3EFFECT OF SALICYLIC ACID AND SELENIUM ON ANTIOXIDANT SYSTEM OF AVENA SATIVA L. UNDER FENOXAPROP-P-ETHYL STRESS
Yigit, Emel; Akbulut, Gulcin Beker; Bayram, Dilek; Kaya, Armagan; Gok, Yetkin
The present study focuses on understanding the role of different antioxidants in plant defense against oxidative stress caused by fenoxaprop-p-ethyl herbicide. Besides ameliorative effect of exogenously applied salicylic acid (SA) and selenium (Se) on fenoxaprop-p-ethyl-induced toxicity was examined in Avena sativa L. It was determined that the MDA (malondialdehyde) content in the groups to which only herbicide was applied was higher than that of the groups to which SA and Se applied together with the herbicide. In plants exposed to fenoxaprop-p-ethyl, the activities of peroxidase (POD), ascorbate peroxidase (APX), superoxide dismutase (SOD), glutathione S-transferase (GST), glutathione reductase (GR) and content of glutathione (GSH) increased in the treatment groups compared to the control groups. The antioxidant activity increased in the control and treatment groups that were pre-treated with SA and Se compared to the groups that were not pre-treated with SA and Se. While catalase activity (CAT) decreased in SA pre-treated plants, it increased in Se pre-treated plants. These results indicate that the fenoxaprop-p-ethyl induces various plant defense responses in target plants and that treatment with exogenous SA and Se can increase stress resistance by altering these responses.

In this study, the effects of Reactive Phthalocyanine Blue (RPB) changes on peroxidase, malondialdehyde (MDA) content and total chlorophyll content of Phaseolus vulgaris L. (bean) were examined. The beans were exposed to different concentrations of RPB between 200-1255 mg/L. In the analyzed plants, different effects were observed on peroxidase activity. When compared to the control group, a significant decrease was identified in the plants which were treated with RPB except for the 1255 mg/L application group. Besides the effects of RPB on MDA content were analyzed. MDA content was found to be lower in all application groups except for 439 mg/L. In addition, total chlorophyll content increased in all application groups except for 338 mg/L when compared to the control group. These findings indicated that RPB caused significant changes in peroxidase activity, MDA content and total chlorophyll. It was shown that the textile dye had a toxic effect on the plants.
