Some nanotoxicity effects of copper (60-80 nm) and copper oxide (40 nm) nanoparticles on Artemia salina

Cimen, Isıl Canan Cicek; Danabas, Durali; Ates, Mehmet

In this study, nanotoxicity tests were made by exposure of Artemia salina to copper (Cu 60-80 nm) and copper oxide (CuO 40 nm) nanoparticles (NPs) at different concentrations (0.2, 1, 5, 10, 25, and 50 mg/L). The LC50 value of Cu (60-80 nm) NPs on the A. salina individuals at the beginning (0), 24th, 48th and 72nd hours and elimination period was 52.37 mg/L while the LC50 value of CuO (40 nm) NPs was 55.39 mg/L. The results of UV-Vis absorbance values showed that all statistical data revealed that maximum effect was observed between 24-30 hours and 25 ppm absorbance concentration was more effective. The multiple R, correlation coefficient (R2) and adjusted R2 values of Cu NP for the suitable Quadratic model were, respectively; 92.96 %, 86.42 % and 76.71 % while they are 98.31 %, 96.64 % and 94.25 % for CuO NP. Also, the data, was indicated effect size significantly changed based on the type and size of NP. Considering the microscope results, it was clearly noticed that A. salina organisms took the NPs in to their body. The accumulation in the gut of A. salina was observed and the images were taken with phase contrast microscope for both of NPs. The highest decrease for survival rates of A. salina individuals exposed to Cu NP was observed in the 10 ppm concentration (43.47 %) and in the 5 ppm concentration (46.20 %) for CuO NP. The results revealed that Cu and CuO NPS showed different toxic effects and that Cu NPs were more toxic than CuO.

Green Synthesis of Gold Nanoparticles Using Aqueous Extract of Asphodelus Aestivus, Coating with Chitosan Biopolymer and Cytotoxicity Studies

Ates, Mehmet; Yilmaz, Ersen; Erbay, Muhammed

The green synthesis of gold nanoparticles (Au-NPs) was carried out by pouring the aqueous extract of East Anatolian origin Asphodelus aestivus plant onto aqueous gold metal ions and reducing them via single-step one- pot method. The absorption peak of the synthesized nanoparticles gave a maximum at 575 nm. All the X-ray diffraction peaks at 2θ = 38.25 , 44.46 , 64.64 and 77.20 that index to (111), (200), (220), and (311) planes verify the successful synthesis of Au-NPs. Mostly spherical shape particles showed a homogeneous distribution with size range 20±5 nm are measured using TEM. From the FTIR spectrum, the peaks are seems to be related to phenolic compounds, flavonoids, benzophenones, terpenoids and anthocyanins which assume that they could act as the reducing agents. The plant extraction, one-pot, single-step method used is environmentally safe without the role of synthetic materials which is highly potential in mild and green synthesis applications. The Au-NPs were coated with chitosan biopolymer in aquatic solution medium and verified by SEM. Then, cytotoxic investigations of the biosynthesized Au-NPs were carried out by HUVEC cells. Au-NPs were showed toxic effects on cell culture, even if in a small amount. However, chitosan biopolymer coating increased cell viability.

Synthesis and Characterization of Silver Nanoparticles and Coating with Chitosan

Ates, Mehmet; Yilmaz, Ersen; Kar, Bulent; Durukan, Ilknur Kars

Recent advancements in nanotechnology have garnered increasing attention due to the improvements in producing and utilizing particles whose sizes are extremely small. There is particular interest in metal nanoparticles due to their distinctive properties. Silver nanoparticles (Ag-NPs) have been of particular interest owing to their unique electrical, optical and physicochemical properties as well as their biomedical applications. Ag-NPs are the fastest growing class of nanoscale materials because of their antibacterial activities. They are now considered to be the next generation antimicrobials. Today, owing to their disinfectant properties, Ag-NPs are extensively used in numerous consumer products, including disinfecting medical devices and home appliances, paints, optical devices, plastics, textiles, soaps and laundry detergents, cosmetics, health-care products, as well as in food packing and food products as preservative and anti-caking agents. Despite their widespread usage, the products containing Ag-NP additives are a serious environmental and human health concern because of their toxicity. Studies on cell and bacterial cultures have shown that Ag-NPs are toxic; and the toxic effects are mainly due to the dissolved Ag ions when Ag-NPs degrade in water. In this study, considering the criteria outlined above, the purpose of the study and Ag-NPs were chemically synthesized in the laboratory environment and modified as chitosan coating. UV-Vis spectroscopy, transmission electron microscopy, scanning electron microscopy, X-ray crystallography and zeta potential analysis were used to characterize the silver nanoparticles.

Ecotoxicity Study of Iron Oxide Nanoparticles on Chlorella Sp. and Daphnia Magna

Ertit Tastan, Burcu; Kars Durukan, Ilknur; Ates, Mehmet

Nanoparticles have great impact due to their tremendous industrial applications. However, their applications have produced toxicity effects on the aquatic environments and their detailed analyses are not clearly understood. Iron oxide nanoparticles (Fe2O3 NPs) are being used extensively in many industries but are considered highly toxic to aquatic species residing in surface waters. This paper demonstrates the acute toxicity of alpha-Fe2O3 and gamma-Fe2O3 NPs in two aquatic species. The effects of various concentration (0, 50, 100, 250, 500 and 1000 mg/L) of alpha-Fe2O3 and.-Fe2O3 on the sensitivity response of the Chlorella sp. and D. magna were investigated. The growth of microalgal decreased with increased concentration of the alpha-Fe2O3 and gamma-Fe2O3 NPs concentrations but did not show a significant toxic effect. The EC50 concentration value was 500 mg/L and LD50 concentration value was 1000 mg/L for alpha-Fe2O3 treated daphnids in 72 h, respectively. The findings demonstrate the significant evidence in understanding acute toxicity of Fe2O3 NPs for environmental protection as part of risk assessment strategies.

In this study, the impact of alpha-iron oxide (α-Fe2O3, 20-40 nm) and gamma iron oxide (γ-Fe2O3, 20-40 nm) nanoparticles (NPs) on phytoplankton species Selenastrum capricornutum and Nannochloropsis oculata was investigated Characterizations of the NPs were systematically carried out by TEM, dynamic light scattering, zeta potential, X-ray diffraction, SEM, and Fourier transformation infrared spectroscopy. Acute toxicity was tested between 0.2 and 50 mg/L for each NP for a period of 72 hours exposure. γ-Fe2O3 NP inhibited development of N oculata at the rate of 54% in 0.2 mg/L group with a high mortality rate of up to 82%. α-Fe2O3 NPs were less toxic that induced 97% mortality on N oculata at 10 mg/L suspensions. In contrast, α-Fe2O3 NP inhibited growth of S capricornutum strongly (73%) in 0.2 mg/L group. γ-Fe2O3 NPs showed similar growth inhibition (72%) on S capricornutum in 10 mg/L suspensions. Despite the differential effects, the results indicated acute toxicity of α-Fe2O3 and γ-Fe2O3 NPs on N oculata and S capricornutum.

Comparative effects of Cu (60–80 nm) and CuO (40 nm) nanoparticles in Artemia salina: Accumulation, elimination and oxidative stress

Cimen, Isil Canan Cicek; Danabas, Durali; Ates, Mehmet

In this study, nanotoxicity tests were made by exposure of Artemia salina to copper (Cu 60–80 nm) and copper oxide (CuO 40 nm) nanoparticles (NPs) at different concentrations (0.2, 1, 5, 10, 25, and 50 mg/l) during some exposure duration. Characterization of Cu and CuO NPs were performed using Transmission Electron Microscope (TEM), Dynamic Light Scattering (DLS), Zeta Potential, X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Fourier Transformation Infrared (FT-IR) analyzes. In organisms, the accumulation and elimination rates of NPs was determined by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) analysis and the oxidative stress effects on A. salina were determined by Glutathione (GSH) and Thiobarbituric acid reactive substances (TBARS) analysis methods. Both NPs were found to differ in accumulation and elimination rates at each application time and in parallel with the increase in concentration. In each group, it was determined that ion release increased with application time. The results showed that the accumulation rates in Cu NPs had a tendency to increase at 48 h and to decrease at 72 h in concentrations of 0, 2 and 1 mg/l, respectively. And in the all other concentrations have been seen an increasing trend within the time. In the CuO NPs (40 nm), accumulation rates were a decrease trend at 48th hour and an increase trend at 72nd hour except 10 mg/l concentration. In the 10 mg/l group was an increase trend with the application period. Cu NP, TBARS value increased with increasing concentrations and the highest increase was observed at 24 h of 5 ppm group. The groups showed a tendency to increase-decrease-increase-decrease in TBARS levels in terms of elapsed time (24th, 48th, 72nd hour and elimination) (5 ppm and 10 ppm groups tended to decrease in TBARS level at 72nd hour). TBARS increased with increasing concentration ratios in CuO NPs (40 nm).

Effects of Zn and ZnO Nanoparticles on Artemia salina and Daphnia magna Organisms: Toxicity, Accumulation and Elimination

Danabas, Durali; Ates, Mehmet; Ertit Tastan, Burcu; Cicek Cimen, Isil Canan; Unal, Ilkay; Aksu, Onder; Kutlu, Banu

In the study, Zn in the size of 40–60 nm and 80–100 nm and ZnO in the size of 10–30 nm were applied to A. salina and D. magna individuals in 7 groups with 3 repetitions. Measurements were made at 24th, 48th and 72nd hours and elimination values were examined at +24 h. LC50 values of NPs were determined and chemical analysis (metal accumulation and elimination), ion quantities which were given to the environment and the survival rates of organisms were determined after the exposure. According to the results of phase contrast microscopy, it was found that both experimental organisms absorbed the NPs in the medium level. In the toxicity results of D. magna, it can be said that Zn NP (40–60 nm) has a highly toxic effect only at 50 ppm concentration for 48 h and lethal dose can be accepted as of 5 ppm at the end of 72 h. In A. salina individuals, it is clearly seen that there is an increase in mortality in organisms parallel to the dose increase. Although all NPs were applied to organisms in low doses corresponding to environmental values, it was observed that toxic effect was in parallel with the increase in time. It is clearly known that there is the inverse proportion between the size of NPs and the toxic effect. The smaller the size of NPs is, the higher the toxic effect becomes When the results of Zn accumulation and elimination of A. salina and D. magna individuals exposed to the Zn and ZnO NPs were examined; it was found that accumulation and elimination occurred in parallel with the increase in concentration at each application hour and elimination. Intensive and possible misuse of nanoscale materials is one of the biggest threats to the environment and all living things worldwide.

    CHARACTERIZATION OF ERBIUM OXIDE NANOPARTICLES

Ates, Mehmet; Kar, Bulent; Bogatu, Coneliu

Considering the advantages of nanoscale materials such as being lighter, more robust, programmable materials, less material usage, less energy requirement during production phase, and no longer producing materials, it is important to produce nanomaterials and determine their properties. Therefore, the characterization of nanomaterials has become one of the pillars of material science. An important aspect of nanoscale material science is the method of characterization of materials that we use or study to learn more about them. In recent years, great importance has been given to rare earth materials with nanostructures. One of the rare earth elements is Erbium and it is one of the newly discovered materials that is used in high technology. Erbium oxide (Er2O3) nanoparticles (NPs) are used in electronic devices. It is used in capacitors, metal oxide semiconductor transistors, and as dopants for optical fiber and laser materials. In this study, Erbium based Er2O3 NPs were characterized. The characterization of Er2O3 was realized using X-ray diffraction (XRD), scanning electron microscopy (SEM), the energy dispersive X-ray (EDX), and Fourier transform infrared spectroscopy (FT-IR). The number of specific peaks and their position fit very well with those from standard XRD spectrum and revealed pure Er2O3 nano oxide. SEM analysis of these particles showed their spherical shapes that are in great extent uniform distributed with nano sizes ranged between 40-60 nm.

Akut toksisite etkileri 24, 48 ve 72 saatlik periyotlar boyunca fitoplankton populasyonunun büyümesi flurometreyle in-vivo chl-a incelenmiştir. Zooplanktonlardaki toksik etkiler, mortalite ve oksidatif stres belirteçlerinden glutatyon (GSH) ve tiyobarbitürat reaktif maddeler (TBARS) testleriyle incelenmiştir. Sonuçlar NP?lerin organizmalar üzerindeki farklı etkilerini göstermekle birlikte, NP?lerin birikim ve eliminasyon oranlarında farklılıklar olduğunu göstermiştir. Organizmadaki birikim NP konsantrasyonuyla artış göstermiştir. NP?lerin metal iyon salınımları, tatlısu ve deniz suyu ortamında farklı olmakla birlikte, herbir NP?ün farklı iyon salınımı gösterdiği gözlenmiştir. NP?lere maruz bırakılan fitoplanktonların yüzeyleri NP?ler ile sarıldığı gözlemlenmiştir. Benzer sonuçlar zooplanktonlarda da gözlemlenmiştir. Oksidatif stres analizlerinde, TBARS değeri D. magna bireylerinde artış gösterirken bazı NP?ler A. salina?da azalmasına neden olmuştur.

Effects of subchronic exposure to zinc nanoparticles on tissue accumulation, serum biochemistry, and histopathological changes in tilapia (Oreochromis niloticus)

Kaya, Hasan; Duysak, Muge; Akbulut, Mehmet; Yilmaz, Sevdan; Guerkan, Mert; Arslan, Zikri; Demir, Veysel; Ates, Mehmet

Zinc nanoparticles (ZnNPs) are among the least investigated NPs and thus their toxicological effects are not known. In this study, tilapia (Oreochromis niloticus) were exposed to 1 and 10 mg/L suspensions of small size (SS, 40-60 nm) and large size (LS, 80-100 nm) ZnNPs for 14 days under semi-static conditions. Total Zn levels in the intestine, liver, kidney, gill, muscle tissue, and brain were measured. Blood serum glucose (GLU), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and lactate dehydrogenase (LDH) were examined to elucidate the physiological disturbances induced by ZnNPs. Organ pathologies were examined for the gills, liver, and kidney to identify injuries associated with exposure. Significant accumulation was observed in the order of intestine, liver, kidney, and gills. Zn levels exhibited time- and concentration-dependent increase in the organs. Accumulation in kidney was also dependent on particle size; NPs SS-ZnNPs were trapped more effectively than LS-ZnNPs. No significant accumulation occurred in the brain (p>0.05) while Zn levels in muscle tissue increased only marginally (p0.05). Significant disturbances were noted in serum GOT and LDH (p<0.05). The GPT levels fluctuated and were not statistically different from those of controls (p>0.05). Histopathological tubular deformations and mononuclear cell infiltrations were observed in kidney sections. In addition, an increase in melano-macrophage aggregation intensity was identified on the 7th day in treatments exposed to LS-ZnNPs. Mononuclear cell infiltrations were identified in liver sections for all treatments. Both ZnNPs caused basal hyperplasia in gill sections. Fusions appeared in the gills after the 7th day in fish treated with 10 mg/L suspensions of SS-ZnNPs. In addition, separations in the secondary lamella epithelia were observed. The results indicated that exposure to ZnNPs could lead to disturbances in blood biochemistry and cause histopathological injuries in the tissues of O. niloticus. (c) 2016 Wiley Periodicals, Inc. Chronic exposure of tilapia (Oreochromis niloticus) to iron oxide nanoparticles: Effects of particle morphology on accumulation, elimination, hematology and immune responses

Ates, Mehmet; Demir, Veysel; Arslan, Zikri; Kaya, Hasan; Yilmaz, Sevdan; Camas, Mustafa

Effects of chronic exposure to alpha and gamma iron oxide nanoparticles (alpha-Fe2O3 and gamma-Fe2O3 NPs) were investigated through exposure of tilapia (Oreochromis niloticus) to 0.1, 0.5 and 1.0 mg/L (9.2 x 10(-4), 4.6 x 10(-3) and 9.2 x 10(-3) mM) aqueous suspensions for 60 days. Fish were then transferred to NP-free freshwater and allowed to eliminate ingested NPs for 30 days. The organs, including gills, liver, kidney, intestine, brain, spleen, and muscle tissue of the fish were analyzed to determine the accumulation, physiological distribution and elimination of the Fe2O3 NPs. Largest accumulation occurred in spleen followed by intestine, kidney, liver, gills, brain and muscle tissue. Fish exposed to gamma-Fe2O3 NPs possessed significantly higher Fe in all organs. Accumulation in spleen was fast and independent of NP concentration reaching to maximum levels by the end of the first sampling period (30th day). Dissolved Fe levels in water were very negligible ranging at 4-6 mu g/L for alpha-Fe2O3 and 17-21 mu g/L for gamma-Fe2O3 NPs (for 1 mg/L suspensions). Despite that, Fe levels in gills and brain reflect more dissolved Fe accumulation from metastable gamma-Fe2O3 polymorph. Ingested NPs cleared from the organs completely within 30-day elimination period, except the liver and spleen. Liver contained about 31% of alpha- and 46% of gamma-Fe2O3, while spleen retained about 62% of alpha- and 35% of the gamma-polymorph. No significant disturbances were observed in hematological parameters, including hemoglobin, hematocrit, red blood cell and white blood cell counts (p > 0.05). Serum glucose (GLU) levels decreased in treatments exposed to 1.0 mg/L of gamma-Fe2O3 NPs at day 30 (p < 0.05). In contrast, GLU levels increased during the elimination period for 1.0 mg/L alpha-Fe2O3 NPs treatments (p < 0.05). Transient increases occurred in glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and lactate dehydrogenase (LDH). Serum Fe levels did not change during exposure (p > 0.05), but increased significantly within elimination period due to mobilization of ingested NPs from liver and spleen to blood. Though respiratory burst activity was not affected (p > 0.05), lysozyme activity (LA) was suppressed suggesting an immunosuppressive effects from both Fe2O3 NPs (p < 0.05). In contrast, myeloperoxidase (MPO) levels increased significantly in treatments exposed to alpha-Fe2O3 NPs (p < 0.05), and the effect from gamma-polymorph was marginal (p > 0.05). The results indicate that morphological differences of Fe2O3 NPs could induce differential uptake, assimilation and immunotoxic effects on O. niloticus under chronic exposure. (C) 2016 Elsevier B.V. Toxicity of Engineered Nickel Oxide and Cobalt Oxide Nanoparticles to Artemia salina in Seawater

Ates, Mehmet; Demir, Veysel; Arslan, Zikri; Camas, Mustafa; Celik, Fatih

In this study, the effects of exposure to engineered nickel oxide (NiO 40-60 nm) and cobalt oxide (CoO < 100 nm) nanoparticles (NP) were investigated on Artemia salina. Aggregation and stability of the aqueous NP suspensions were characterized by DLS and TEM. Acute exposure was conducted on nauplii (larvae) in seawater in a concentration range from 0.2 to 50 mg/L NPs for 24 h (short term) and 96 h (long term). The hydrodynamic diameters of NiO and CoO NPs in exposure medium were larger than those estimated by TEM. Accumulation rate of NiO NPs were found to be four times higher than that of CoO NPs under the same experimental conditions. Examinations under phase contrast microscope showed that the nanoparticles accumulated in the intestine of Artemia, which increased with increasing exposure concentration. Differences were observed in the extent of dissolution of the NPs in the seawater. The CoO NPs dissolved significantly while NiO NPs were relatively more stable. Oxidative stress induced by the NP suspensions was measured by malondialdehyde assay. Suspensions of NiO NPs caused higher oxidative stress on nauplii than those of CoO NPs. The results imply that CoO and NiO NPs exhibit toxicity on Artemia (e. g., zooplankton) that is an important source of food in aquatic food chain.

The effects of alpha-iron oxide (alpha-Fe2O3) and gamma-iron oxide (gamma-Fe2O3) nanoparticles (NPs) on marine microalgae species (Nannochloropsis sp. and Isochrysis sp.) were investigated in this study. Both Fe2O3 NPs covered the surface of algae with the agglomerates of the nanoparticles. This form of physical NP toxicity significantly decreased the sizes of phytoplankton. Both NPs were toxic to the tested algal species, while alpha-Fe2O3 showed less toxicity than gamma-Fe2O3 NPs for both algal species. A comparative analysis of growth data of the two algal species treated with alpha-Fe2O3 or gamma-Fe2O3 NPs revealed that Isochrysis sp. are more sensitive than Nannochloropsis sp. Toxicity of these widely used NPs to primary producers forming the base of the food chain in aquatic environments might result in widespread adverse effects on aquatic environmental health.
