Seasonal investigations were conducted on 640 Cyprinus carpio specimens obtained through commercial fishing at four different stations (Gerger, Sitilce, Hosirge, and Samsat) in the Atatürk Dam Lake. MDA (malondialdehyde) levels were measured in the liver and muscle tissues of male and female fish; the results revealed a significant increase in MDA levels at stations exposed to point-source discharge. This indicates that pollution-induced oxidative stress in fish varies significantly across seasonal and regional scales. Among the stations compared, the lowest MDA values were detected in fish tissues from the Gerger station, while the highest values were recorded at the Sitilce and Hosirge stations. Analysis of SOD, GSH-Px, and GSH values across all stations showed that these parameters fluctuated in correlation with MDA levels thereby maintaining the oxidant-antioxidant balance. This study investigated seasonal and spatial variations in oxidative stress biomarkers in common carp (Cyprinus carpio) collected from four stations in Atatürk Dam Lake. A total of 640 specimens were analyzed for malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and reduced glutathione (GSH) levels in liver and muscle tissues. Results showed that MDA levels were significantly higher in fish collected from Sitilce and Hosirge stations, indicating elevated lipid peroxidation associated with point-source pollution. In contrast, the lowest MDA levels were observed at the reference station, Gerger. Antioxidant enzyme activities (SOD, GSH-Px, and GSH) exhibited tissue-, season-, and site-dependent variations, suggesting adaptive responses to oxidative stress. Overall, the findings demonstrate that anthropogenic pressures in Atatürk Dam Lake significantly alter the oxidant–antioxidant balance in fish, highlighting the usefulness of these biomarkers for environmental monitoring.

Lead (Pb) is a heavy metal with high toxicity that can cross the blood-brain barrier and accumulate in brain tissue, causing serious neurological disorders. The aim of this study is to evaluate the potential protective effect of chrysin (CHR) against Pb-induced neurotoxicity. Thirty-five male Sprague Dawley rats were divided into five groups: Control, CHR (50 mg/kg), Pb, Pb+CHR (25 mg/kg), and Pb+CHR (50 mg/kg). Rats were administered CHR (25 or 50 mg/kg) and Pb (30 mg/kg) orally for 7 days. Twenty-four hours after the last treatment, the rats were sacrificed under mild sevoflurane anesthesia by decapitation, and brain tissue samples were collected. Biochemical and histopathological methods were used to measure cholinergic enzyme activity, DNA damage, and oxidative stress parameters in brain tissue. CHR reduced levels of glial fibrillary acidic protein (GFAP), acetylcholinesterase (AChE), and 8-hydroxy-2'-deoxyguanosine (8-OHdG) caused by Pb. Due to its antioxidant properties, CHR decreased Pb-induced lipid peroxidation while increasing glutathione (GSH) levels and the activities of catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx). Based on biochemical and immunohistochemical analyses, CHR reduced cyclooxygenase-2 (COX-2), myeloperoxidase (MPO), nuclear factor kappa-B (NF-κB), neuronal nitric oxide synthase (nNOS), prostaglandin E2 (PGE-2), and p53 levels. These findings suggest that CHR may exert a therapeutic effect by alleviating oxidative stress and inflammation induced by Pb-induced neurotoxicity.

Anti-oxidants are vital for protecting sperm and can mitigate the negative effects associated with cryopreservation. This study was conducted to evaluate the protective effects of adding anti-oxidants (vitamins and amino acids) to extender in sex-reversed rainbow trout (Oncorhynchus mykiss) sperm. The collected sperm was diluted at the ratio of 1: 5 by the extenders supplemented with different anti-oxidants including 1.00 mM of ascorbic acid and L-tryptophan, 2.00 mM of cysteine and α-tocopherol, and their combination. After dilution, the semen was aspirated into 0.50 mL straws, and the straws were placed on the tray, frozen for 10 min, and plunged into liquid nitrogen. Straws were thawed in a 30.00 ˚C water bath for 15 sec. The Sperm Class Analyzer System was used to evaluate sperm kinematics. The activity of anti-oxidant enzymes and lipid peroxidation were determined as oxidative stress indices. Our data indicated that the incorporation of anti-oxidants and amino acids increased sperm motility duration. The elevated activity of glutathione peroxidase, superoxide dismutase, and catalase in post-thaw samples indicates that the anti-oxidant system in sex-reversed rainbow trout sperm likely plays a crucial role in protecting membrane compounds from oxidation. In conclusion, the combination of 1.00 mM L-tryptophan and ascorbic acid to the extender media caused a prolonging effect in sperm motility after thawing and they have the potential to serve as effective agents for improving sperm cryosurvival.

As a potential pollution source, slaughterhouse wastewater (SWW) with development of the meat production and processing sector is a threat in the world. Therefore, the determination of economical, favourable, cost-effective and proper treatment method is crucial for sustainable environmental protection. This study focused on the performance and efficiency of a two-stage process combining biological treatment (BT) with freshwater snails and electrocoagulation (EC) of SWW. Viviparus contectus, Planorbis rubrum, Melanoides tuberculata were used in primary treatment while secondary treatment was realized with EC process. The EC of SWW was performed for reducing both turbidity and chemical oxygen demand (COD) concentration to levels compliant with direct-discharge regulations. In addition, the stirring speed, current density, initial pH and reaction time were assessed for evaluation the optimum EC condition. Oxidative stress biomarkers and some antioxidants were assessed in freshwater snails exposed to SWW after 24 and 96 h. The best removal efficiencies of COD (91.43%) and turbidity (92.42%) were obtained at pH 5 and 20 min of operational time by P. rubrum. Notably, MDA (Malondialdehyde) was highest level in M. tuberculata compared to V. contectus and P. rubrum. Overall, removing organic and inorganic pollutants from SWW could be provided with the hybrid process BT/EC as a combining approach.

The polychlorinated biphenyls (PCBs) in aquatic environment adversely affect non-target organisms, including fish. Especially, the male reproduction and next generation can be damaged through high exposure to these pollutants. Hence, the sperm cells were exposed to sublethal concentrations of Aroclor 1254 (0, 1, 5, 10, or 25 mg/l) for 4 h. The sperm quality parameters were analyzed by SCA (Sperm Class Analyzer). The fertility, eyeing, and hatching rates were determined as gamete markers. Lipid peroxidation (malondialdehyde-MDA), glutathione (GSH), and antioxidant enzymes [superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT)] were measured for determination of oxidative stress. Our results showed that Aroclor 1254 negatively affected the motility rate and duration, fertilization rate, embryogenesis, and hatching and also triggered antioxidant defense mechanisms at the highest concentration (25 mg L−1). Furthermore, linear speed (VSL), linearity index (LIN), and amplitude lateral head (ALH) were significantly changed after exposure to 25 mg L−1, and the lowest concentrations (1 and 10 mg L−1) did not significantly affect the motility and fertilizing capacity. The embryogenesis and hatching were significantly affected by sperm exposure to 1, 10, and 25 mg L−1 of Aroclor 1254. Consequently, Aroclor 1254 causes potential hazards in male germ cells, and the exposure of sperm cells to pollutants can adversely affect next generation of wild populations.

This study was conducted to investigate the effects of chrysin (CH) on lead acetate (PbA) inducing liver injury. In this study, 35 male Sprague Dawley rats were divided into 5 groups as 7 rats in each group. Group 1 (Control): Saline was given orally. Group 2 (CH group): CH (50 mg kg-1 weight day-1 ) were orally given rats for 7 days. Group 3 (PbA group): PbA (30 mg kg-1 ) was orally given to rats at 30 mg kg-1 PbA for 7 days. Group 4 (PbA +CH 25 group): 25 mg/kg CH was orally given to rats for 7 days. In addition, 30 mg kg-1 PbA was given orally to rats for 7 days after 30 minutes of the application of the CH. Group 5 (PbA+CH 50 group): 50 mg kg-1 CH was orally given to rats for 7 days. Furthermore, 30 mg kg-1 PbA was given orally to rats for 7 days after 30 minutes of the application of the CH. The rats were euthanized under mild sevoflurane anesthesia and tissue and blood samples were taken 24 hours after the last application (8th day). AST, ALT and ALP in serum, MDA, GSH and NO levels and SOD, CAT, GPx and arginase enzyme activities in liver tissue were measured. Serum ALT, AST and ALP levels increased in the PbA group when compared with the control, and this increase was partially corrected with CH. Liver MDA and NO levels increased in the PbA administered groups while antioxidant enzyme activities and GSH levels decreased. Although, CH administration decreased the increased MDA and NO levels, it increased the decreased antioxidant enzyme activities and GSH levels. Histologically, it was determined that CH decreased the necrosis, hydropic degenerations and mononuclear cell infiltrations seen in the livers of rats treated with PbA. As a result, CH is effective in the reducing PbA damages in the liver.

In the present study, the protective effects of chrysin (CHR) against testicular damage caused by lead acetate (PbAc) were examined. In this way, 30 min after rats were given 25 and 50 mg/kg/b.w CHR orally for seven consecutive days, 30 mg/kg/b.w PbAc was administered orally. In biochemical analysis of testicular tissue, it was found that PbAc-reduced antioxidant parameters [glutathione peroxidase (GPx), glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT)], while it increased lipid peroxidation, inflammatory markers [nuclear factor kappa-B (NF-κB), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE-2), and inducible nitric oxide synthase (iNOS)], and 8-hydroxy-2′-deoxyguanosine (8-OHdG). In the immunohistochemical examination, it was determined that PbAc increased the expression of tumor necrosis factor-α (TNF-α) and caspase-3. Accordingly, PbAc was found to cause a decrease in sperm motility and an increase in the percentage of dead sperm. However, it has been observed that CHR relieves oxidative stress due to its antioxidant properties, thus protecting against inflammation and apoptosis. It also allowed the CHR sperm parameters to return to control group levels. The results revealed that CHR could be a natural substance to be used in Pb-induced testicular toxicity. Practical applications: Lead (Pb) is an important environmental contaminant heavy metal. Pb is believed to reduce fertility in men. Oxidative stress plays a significant role in the damage caused by Pb to testicular tissue. CHR is an antioxidant substance that occurs naturally in various plants and has various pharmacological properties. In the present study, it was investigated whether CHR has a protective effect against testicular toxicity induced by PbAc. The results revealed that in rats, CHR protects the testicular tissue from PbAc toxicity by showing antioxidant, anti-inflammatory and anti-apoptotic effects, thus bringing sperm parameters closer to normal.

In this study, the protective effects of chrysin (CR) on lead acetate (PbAc)-induced renal toxicity in Sprague-Dawley rats were investigated with biochemical, histopathological, and immunohistochemical methods. In the study, rats were given orally at 30 mg/kg/body weight (BW) PbAc after CR of 25 and 50 mg/kg/BW was administered to them orally (a total of 7 administrations for 7 days). The results showed that CR reduced urea and creatinine levels by alleviating PbAc-induced kidney damage. It was determined that CR decreases PbAc-induced lipid peroxidation due to its antioxidant properties and increases catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx) activities, and glutathione (GSH) levels. It was also detected that CR protects DNA from the toxic effects of PbAc and reduces 8-hydroxy-2′-deoxyguanosine (8-OHdG) levels. Biochemical and immunohistochemical findings demonstrated that CR had anti-inflammatory and antiapoptotic effects and reduced nuclear factor kappa-B (NF-κB), interleukin-33 (IL-33), prostaglandin-E2 (PGE-2), tumor necrosis factor-α (TNF-α), p53 levels, and the activities of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS), which were increased with PbAc administration. Moreover, CR was found to increase the levels of aquaporin-1 (AQP-1) and nephrine in PbAc-induced kidney tissue. CR decreased the contents of lead (Pb), zinc (Zn), iron (Fe), sodium (Na), and copper (Cu) and increased those of potassium (K) calcium (Ca) in renal tissue. These results indicated that CR considerably alleviates kidney toxicity caused by PbAc.

- MakaleAçık Erişim2020Scopus SJRQ4Investigation of curcumin role on doxorubicin-induced tissue damage in terms of trace metal levels using etaas
Ince, Olcay Kaplan; Kandemir, Fatih Mehmet; Ince, Muharrem; Benzer, Fulya; Onal, Ali; Kucukler, Sefa
Doxorubicin (DXR) is one of the commonly used chemotherapeutic agents, which is highly toxic and causes gastrointestinal and cardiac toxicity. Curcumin is used as a spice and coloring agent in food, but also possesses anti-proliferating, potent antioxidant, antitumor-promoting and anti-carcinogenic properties in vitro and in vivo. In this study, the change in element concentrations in the tissues of healthy animals was examined by administering doxorubicin and curcumin separately, or both at the same time. For this purpose, 35 wistar albino male rats were selected and randomly divided into five groups of seven each. The objective of this study was to assess and compare the levels of copper, iron, zinc and selenium in the liver, kidney, heart, and testis of the control group and in the doxorubicin-induced tissues. According to the results, the Cu, Fe, Zn, Se amounts and the Cu/Fe, Cu/Zn, and Cu/Se ratios were present at different levels in each of the five groups. In terms of element concentration, the most affected tissue by doxorubicin application is the heart, and the least affected is the kidney. The ratios of Cu/Fe and Cu/Se in the liver and the heart were highest in Group II and Group III, respectively.

This study was made to investigate the effects of intramuscular administrations of dexamethasone on seminal plasma nitric oxide levels and arginase activity, and some spermatological parameters in rams. Ten Akkaraman rams weighing 50–60 kg and 2 years old were used as material in this study. The study was performed during the breeding season (September–November) for rams. The semen was collected by artificial vagina at 1st, 4th, 24th, 48th, 72nd and 96th hours for control group before dexamethasone administration. For treatment group, 0.25 mg/kg dexamethasone was administered and semen was collected at the time points described for control group. Spermatological characteristics of semen samples (semen volume, pH, sperm motility, density and abnormal sperm rate), seminal plasma arginase enzyme activities and nitric oxide levels were determined. It was determined that the administration of dexamethasone was detected to decrease seminal plasma arginase activity (p <.05 and.01) and nitric oxide level (p <.05), semen volume (p <.05 and.01), mass activity (p <.05 and.01), sperm density (p <.05) and sperm motility (p <.05 and.01), and to increase abnormal sperm rate (p <.05 and.01). In conclusion, dexamethasone is not recommended to be used during the breeding season as it damages the sperm quality of the rams.

The purpose of this study is to investigate the effects of L-arginine on spermatological parameters, seminal plasma nitric oxide levels and arginase enzyme activities. Fertile rams that are 2–3 years old and weighing 50–60 kg were used as material. The semen was collected by artificial vagina at 1st, 4th, 24th, 48th, 72nd, 96th and 120th hours for the control group before L-arginine administration. For treatment groups, L-arginine was administered intraperitoneally at a dose of 5 mg kg−1 bw−1 and semen was collected at the time point described for the control group. Spermatological characteristics of semen samples (semen volume, pH, sperm motility, concentration and abnormal sperm rate), seminal plasma nitric oxide levels and arginase enzyme activities were determined. Increased seminal plasma nitric oxide level (p <.01), seminal plasma arginase activity (p <.01), semen volume (p <.05), semen mass activity (p <.05), sperm motility (p <.05) and concentration (p <.01) and decreased abnormal sperm rate (p <.05 and p <.01) were observed by L-arginine administration. In conclusion, it may be concluded that L-arginine application in rams during the breeding season may have positive effects on rams' reproductive performance.

Behaviour and physiology in fish are affected from environmental factors. Particularly, habitat and culture environment in aquatic animals are essential for life stages. In this study, biomarkers of oxidative stress and antioxidant were compared in wild and farmed Çoruh trout (Salmo coruhensis) spermatozoa. In spermatozoa of Çoruh trout, lipid peroxidation (malondialdehyde), glutathione and enzymatic antioxidant activities (catalase, superoxide dismutase, glutathione peroxidase) were assessed. Our data demonstrated that superoxide dismutase (0.44±1.25 Umg-1 protein), catalase (15.21±4.69 k.g-1 protein), glutathione peroxidase (42.89±0.61 Ug-1 protein), glutathione (0.18±4.01 μmolg-1 cell) and malondialdehyde (2.49±0.12 nmol.g-1 cell) levels in wild fish were low compared to spermatozoa of farmed fish. Overall, alterations in malondialdehyde levels and the antioxidant status could be differences in habitat.
