Bioremediation is one of the cheapest and easy method for biological treatment for most kind of industial wastewater. Bioremediation potential of the Phanerochaete chrysosporium for industrial wastewater from Industrial Zone Wastewater plant in Mardin, Turkey, was evaluated. The chemical oxygen demand (COD) and total organic carbon (TOC) reduction efficiencies of P. chrysoporium of on wastewater studied using biomarkers such as catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), Thiobarbituric Acid (TBARS) and reduced glutathione (GSH) levels in Dreissena polymorpha. The level of TBARS, a marker of lipid peroxidation was found to be high because of wastewater exposure after treatment. TBARS levels decreased (from 20.2 to 31.6 and from 20.8 to 37.5). GSH levels (from 52.3 to 15.9), CAT levels (from, 119.7 to 15.3 and from 91.1 to 13.4) SOD levels (from 3.8 to 0.9 and from 3.4 to 1.2) and GPX activity (from 177.2 to 104.4 and 174.2 to 100.5) decreased in the wastewater exposure groups during 24 and 96th hour compared to the control groups. GSH levels, CAT, SOD and GPX activity increased after treatment by P. chrysosporium. Results indicate that industrial wastewater caused oxidative stress in Dreissena polymorpha. The findings revealed that the highest removal efficiency for COD and TOC, at 86.3 % and 80.3 % respectively, occurred in the 1/20 diluted medium. P. chrysosporium has proven to be effective in the bioremediation of wastewater from the Industrial Zone Changes in biochemical parameters before and after bioremediation showed that antioxidant parameters such as CAT, SOD, GPx activities and GSH and TBARS levels can be used as biomarker to evaluate bioremediation efficiency.

Serdar, Osman; Ketenalp, Zozan; Cikcikoglu Yildirim, Nuran
The purpose of this study was to measure the antioxidant parameters to examine the potential ameliorative impacts of the rare earth element (REE) cerium (Ce) in dimethoate (DM) induced toxicity in Dreissena polymorpha. Lethal concentration 50 (LC50) of DM for 96 h was measured. The antioxidant effect of Ce was investigated by exposure of 1/2 of the LC50 value of DM together with 4, 20 and, 100 mg/L of Ce to D. polymorpha. The responses of the biomarkers were determined by measuring the superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione (GSH) and malondialdehyde (MDA) levels. SOD, CAT, GPX and GSH levels were reduced (p < 0.05), while the MDA levels increased significantly (p < 0.05) in mussels exposed to DM. When DM was exposured together with Ce, it was observed that the MDA levels decreased, SOD, CAT, GPX and GSH levels increased during 24 and 96 h. The results of our study showed that Ce exerted a strong antioxidant effect on toxicity induced by DM. It has been shown that Ce may have a compensatory effect with the positive change in the antioxidant response of D. polymorpha to DM. This study is the first report about Ce and insecticide effects in bivalves.

Cikcikoglu Yildirim, Nuran; Serdar, Osman; Basaran, Senay
The aim of this study is to assess the toxicity of ibuprofen (IBU) and propranolol (PRO) drugs usingGammarus pulex as a model organism. Firstly, the 96 h LC50 values of IBU and PRO were determined and then three sublethal concentrations of the drugs were exposed to G. pulex. The activities of superoxide dismutase (SOD), catalase (CAT) and acetylcholinesterase (AChE) were evaluated. SOD activity decreased in G. pulex exposed to IBU and PRO compared to control. In all groups exposed to IBU, CAT activity increased at different concentrations at 24 and 96 h. In the groups exposed to different PRO concentrations, CAT activities increased after 24 h compared to the control group (p < 0.05). AChE activities increased in all application groups exposed to IBU for 96 hours (p < 0.05). In conclusion, exposure to IBU and PRO resulted in increased oxidative damage. PRO has been found to cause neurotoxicity.

Tanyol, Mehtap; Cikcikoglu Yildirim, Nuran; Tatar, Sule; Serdar, Osman; Yildirim, Numan
The biochemical biomarkers (Catalase (CAT), glutathione s-transferase (GST), cytochrome P4501A1 (CYP1A1450) activities and glutathione (GSH), malondialdehyde (MDA) levels) of D. polymorpha exposed to the basic blue 41 (BB41) solutions for 24 and 96 h before and after the Fenton process (FTP) were evaluated to investigate the efficiency of the FTP. The optimum conditions for the removal of BB41 were set using central composite design (CCD), which is a response surface methodology (RSM) module in the Design-Expert® software. The optimal conditions were found to be 27.87 min, 3.49 mg/L, and 0.04 for reaction time, initial BB41 concentration, and Fe2+/H2O2 molar ratio, respectively, resulting in maximum (99%) removal efficiency. After FTP, the MDA and GSH levels decreased significantly during the 24 and 96 h. CAT activities were increased after treatment. No statistically significant changes were observed in the CYP1A1 levels after FTP. GST activities decreased during 24 h but increased during 96 h after FTP. CYP1A1450, catalase, GST activities and MDA, GSH levels of D. polymorpha are useful biomarkers to evaluate the efficiency of FTP. This study suggests that BB41 dye must be further treated after the FTP to minimise the ecotoxicological risk.

Cikcikoglu Yildirim, Nuran; Yaman, Metin
The present study was undertaken to determine the toxicity of the methyl orange by using the changes of some antioxidant and detoxification enzyme activities in Gammarus pulex. Lethal Concentration (LC) value of Methyl Orange (MO) was determined. Three sublethal doses of MO (1/4; 1/8 and 1/16 of LC value) were exposed to G. pulex for 24 and 96 h. Superoxide dismutase (SOD), Catalase (CAT), Glutathione peroxidase (GSH-P X ), Cytochrome p450 (CYP1A1), Glutathione S-transferase (GST) activities as well as Glutathione (GSH) and Malondialdehyde (MDA) levels were determined by using The enzyme-linked immunosorbent assay (ELISA) kit. The CAT and CYP1A1 activities were decreased in all the groups exposed to different doses of MO. GST activity and GSH, MDA levels were increased all the groups exposed to different doses of MO. The GSH-P X activities were changed in all the groups. MO affected SOD activity at different levels and in different concentrations. In our study, it has been found that exposure duration didn’t significantly affect the biochemical biomarkers except for GST and GSH. In conclusion, alterations in antioxidant and detoxification enzymes and lipid peroxidation may potentially be used as sensitive biomarkers for risk assessment of dyes in the environment and may contribute to the establishment of discharge regulations.

- MakaleAçık Erişim2017Scopus SJRQ2Antioxidant Response in Duckweed After Exposure to Secondary Effluent from Municipal Wastewater Treatment Plant, Elazığ, Turkey
Tatar, Sule Yuksel; Obek, Erdal; Cikcikoglu Yildirim, Nuran
The aim of this study is to evaluate the effects of the effluent of Elazig Municipality Wastewater Treatment Plant on the oxidative defense capacity of aquatic plants (Lemna minor L. and Lemna gibba L.). For this purpose, malondialdehyde (MDA), glutathione (GSH), oxidized glutathione (GSSG), vitamin A (retinol), vitamin E (α-tocopherol), and vitamin C (Ascorbic acid) levels were determined by the HPLC (high performance liquid chromatography) in the control groups and the groups adapting to reactors fed with discharge water. The depletion of vitamins (A, E, and C), decrease of GSH/GSSG ratio, and increase of MDA that reflect a precarious state of the cell in L. minor L. and L. gibba L. were observed after exposure to wastewater. It can be suggested that the selected biomarkers are useful in understanding the biochemical mechanisms of the secondary effluents from wastewater treatment plant in L. minor L. and L. gibba L. as early warning indicators.
