Seasonal investigations were conducted on 640 Cyprinus carpio specimens obtained through commercial fishing at four different stations (Gerger, Sitilce, Hosirge, and Samsat) in the Atatürk Dam Lake. MDA (malondialdehyde) levels were measured in the liver and muscle tissues of male and female fish; the results revealed a significant increase in MDA levels at stations exposed to point-source discharge. This indicates that pollution-induced oxidative stress in fish varies significantly across seasonal and regional scales. Among the stations compared, the lowest MDA values were detected in fish tissues from the Gerger station, while the highest values were recorded at the Sitilce and Hosirge stations. Analysis of SOD, GSH-Px, and GSH values across all stations showed that these parameters fluctuated in correlation with MDA levels thereby maintaining the oxidant-antioxidant balance. This study investigated seasonal and spatial variations in oxidative stress biomarkers in common carp (Cyprinus carpio) collected from four stations in Atatürk Dam Lake. A total of 640 specimens were analyzed for malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and reduced glutathione (GSH) levels in liver and muscle tissues. Results showed that MDA levels were significantly higher in fish collected from Sitilce and Hosirge stations, indicating elevated lipid peroxidation associated with point-source pollution. In contrast, the lowest MDA levels were observed at the reference station, Gerger. Antioxidant enzyme activities (SOD, GSH-Px, and GSH) exhibited tissue-, season-, and site-dependent variations, suggesting adaptive responses to oxidative stress. Overall, the findings demonstrate that anthropogenic pressures in Atatürk Dam Lake significantly alter the oxidant–antioxidant balance in fish, highlighting the usefulness of these biomarkers for environmental monitoring.

Background: This study was conducted to determine the effect of various fatty acid ratios added to diets of rainbow trout (Oncorhynchus mykiss, Walbaum) on some sperm parameters and parameters related to arginase activity. Methods: A total of 99 male rainbow trout were used in this study, with the fish averaging 3 years old. Semen was collected by abdominal massage. Four different experimental groups were created in the study: a control group without n-3 series fatty acids added to their diets, and groups with n-3 series fatty acids added at ratios of 1% (D1), 2% (D2), and 3% (D3), respectively. Results: While no significant difference in semen volume was observed between the D1 and D3 groups and the control group, semen volume was significantly higher in the D2 group. Compared with the control group, the time required for sperm motility to decline to both 50% and 0% was significantly prolonged in the D1 and D3 groups. Seminal plasma arginase activity did not differ significantly in the D2 and D3 groups compared with the control group but was significantly higher in the D1 group. Conclusions: Consequently, considering the doses used in the study, fatty acid supplementation was found to lead to changes in sperm quality parameters, specifically volume (only 2%) and arginase activity (only 1%). Given that the rate of decrease in sperm motility was later in groups D1 and D2, it was concluded that the added fatty acids may be beneficial.

Baykara, Murat; Erisir, Mine; Özçelik, Mehtap; Güler, Osman
The antioxidant and radioprotective effects of propolis against ionizing radiation induced oxidativedamage were investigated. Twenty one rats were divided in to 3 groups; 1 st group was the controlgroup; 2nd group was the irradiation group, physiological saline (i.p.)/ 3 days + 9 Gy gammairradiation; 3rd group was irradiation + Propolis group, propolis (i.p. 100 mg / kg BW) / 3 days + 9Gy gamma irradiation. As a result of irradiation, a significant increase was determined inmalondialdehyde (MDA) levels of liver, kidney, and brain tissues of the rats (P<0.001). There was asignificant decrease in glutathione peroxidase (GSH-Px) activity of all the tissues (P˂0.001) andsuperoxide dismutase (SOD) activity and glutathione (GSH) level in the liver (P˂0.01). Also, GSHlevel increased significantly in the kidney (P˂0.001) and ovarium tissues (P˂0.05). The MDA levelsthat increased in the liver as a result of irradiation, decreased significantly due the addition ofpropolis (P˂0.001); whereas, the MDA levels that increased in kidney and brain did not change. Inthe spleen and ovarium tissues, MDA levels decreased significantly as a result of addition ofpropolis compared to the irradiation group (P˂0.05). It was determined that GSH levels, thatdecreased in the liver (P˂0.01), and increased in the kidney due to addition of propolis, and GSHlevels, that decreased in the liver as a result of irradiation (P˂0.01) and increased in the kidney(P˂0.001), approached to the normal level (P˂0.001) and the GSH levels in the spleen and brainincreased compared to both the control and irradiation groups (P˂0.05). The addition of propoliscaused a decrease in the GSH-Px activities of heart, spleen, brain, and ovarium tissues comparedto both the control and irradiation groups (P˂0.001). Also, a decrease in the SOD activity of hearttissue compared to the control group was found (P˂0.05).
- MakaleAçık Erişim2020The Effect of N-Acetylcysteine on Oxidant/Antioxidant Status in Irradiated Rats
Baykara, Murat; Özçelik, Mehtap; Erisir, Mine; Guler, Osman
N-Acetylcysteine (NAC) is both a strong antioxidant and a glutathione precursor. The effect of NAC on the oxidant/antioxidant status of some tissues of the irradiated rats was investigated. Twenty one rats were assigned to 3 groups; the control group, the irradiation group, for which physiological saline solution was administered as i.p. for three days and then, 9 Gy gamma irradiation was applied, and 3rdirradiation + NAC group for which NAC was adminis-tered as i.p. for three days as 300 mg/kg body weight and then the same dose of irradiation was applied. Upon irradia-tion, the increase determined in malondialdehyde (MDA) was significant in the liver, kidney, and brain tissues of the rats (P˂0.05). While glutathione peroxidase (GSH-Px) activity decreased in all of the tissues and superoxide dismutase (SOD) activity only in the liver (P˂0.01), glutathione (GSH) levels significantly increased in the kidney and ovarium tissues (P˂0.001). While NAC administration returned the increased MDA levels in the kidney and brain as a result of irradiation to normal levels (P˂0.05), it was determined that it did not return the increased MDA levels in the liver tissue to the normal level (P˂0.001). While NAC addition led to a significant increase in GSH levels of the liver, heart, spleen, brain and ovarium tissues compared to both control and irradiation groups, it caused a significant decrease in the kid-ney tissue compared to irradiation group (P˂0.001). As a result of NAC addition, a significant decrease was determined in spleen GSH-Px activity, heart and ovarium SOD activity compared to control and irradiation groups (P˂0.05). It can be asserted that GSH increasing by the addition of NAC is the main antioxidant that has a role in decreasing oxidative stress occurring as a result of irradiation. In the examination of MDA values, it was found that the addition of NAC pro-tected the kidney and brain against the oxidative damage induced by irradiation but NAC addition could remain insuffi-cient for the liver.

Ozcelik, Mehtap; Erisir, Mine; Guler, Osman; Baykara, Murat; Bulmus, Funda Gulcu
The antioxidant and protective effect of resveratrol against irradiation induced oxidative damage were researched. Twenty-one rats were seperated into 3 groups; First group was the control group. Second group was the irradiation group; received physiological saline intraperitoneally (i.p.) for three days, then 9 Gray (Gy) irradiation was applied. Third group was irradiation+Resveratrol group; received 10 mg/kg resveratrol i.p. for three days and then the same dose of radiation was applied. As a result of irradiation, a significant increase was found in the level of malondialdehyde (MDA) in the liver, kidney and brain tissues of rats (P<0.001). Glutathione peroxidase (GSH-Px) activity in all the tissues and superoxide dismutase (SOD) activity, Glutathione (GSH) levels in liver decreased (P<0.01), GSH levels in kidney, spleen and ovarium tissues significantly increased (P<0.05). The giving of resveratrol significantly reduced the MDA levels increasing in liver, kidney, and brain as a result of irradiation (P<0.001) and the liver MDA levels reached to the control levels. By giving resveratrol, it was determined that all the antioxidants studied reduced significantly compared to the control group and/or the irradiation group (P<0.05). According to the data obtained as a result; It was concluded that resveratrol shows a corrective effect by changing the oxidant/antioxidant balance against oxidative stress caused by irradiation, therefore resveratrol is a powerful antioxidant.

İncele - MakaleAçık Erişim2016Scopus SJRQ1The in vitro effect of cypermethrin on quality and oxidative stress indices of rainbow trout Oncorhynchus mykiss spermatozoa
Kutluyer, Filiz; Benzer, Fulya; Erisir, Mine; Ögretmen, Fatih; Inanan, Burak Evren
There is limited information on the scientific literature about the effect of in vitro exposure of fish sperm to pesticides. In vitro effect of cypermethrin on sperm quality and oxidative stress has not yet been fully investigated. Therefore, the effects of cypermethrin, a type II pyrethroid insecticide, on quality and oxidative stress of spermatozoa were examined in vitro. To explore the potential in vitro toxicity of cypermethrin, fish spermatozoa were incubated with different concentrations of cypermethrin (1.025, 2.05 and 4.1 mu g/l) for 2 h. The motility rate and duration of sperm were determined after exposure to cypermethrin. Reduced glutathione (GSH), glutathione peroxidase (GSH-Px), catalase (CAT) and malondialdehyde (MDA) in spermatozoa were analyzed for determination of oxidant and antioxidant balance. Our results indicated that spermatozoa motility and duration significantly decreased with exposure to cypermethrin. Additionally, activity of GSH-Px (P < 0.05) and MDA and GSH levels increased in a concentration-dependent manner while CAT activity decreased (P < 0.05). Consequently, the oxidant and antioxidant status and sperm quality were affected by quantitative changes and different concentrations of cypermethrin. (C) 2015 Elsevier Inc. All rights reserved.

The purpose of this study was to examine the changes to occur in egg production, egg quality traits, hatchability, and oxidative stress parameters of laying quails fed with high calorie diets and effect of milk thistle (Silybum marianum) seed on these changes. A total of 75 45-day-old quails including 60 females and 15 males were used in the study. The quails were divided into 5 groups with three repetitions. 4 females and 1 male were used in each repetition. The groups of the study were arranged as following; control group (C) consuming corn-soybean based basal diet, oil group (SFO) in which 5% sun flower oil was added into basal diet, oil + milk thistle group (SFO+MT) in which 5% sun flower oil + 1% milk thistle seed was added into basal diet, syrup group (CS) in which 10% corn syrup was added into basal diet, and syrup + milk thistle group (CS+MT) in which 10% corn syrup + 1% milk thistle was added into basal diet. Total egg production was found to be significantly higher in SFO and SFO+MT groups of present study (P<0.05). Feed intake, total egg weight and feed conversion were found similar among the groups (P>0.05). The differences among the groups in albumen rate, yolk rate, dried shell rate and shape index were not statistically significant (P>0.05). The best yolk colour was obtained in SFO+MT and CS groups (P<0.01). There were no statistically differences among the groups in hatchability, hatchability of fertile and embrionic mortality (P>0.05), but the difference in examined fertility were statistically significant (P<0.05). Malonyl dialdehyde (MDA) values of blood, liver and heart tissues were similar among the groups of study (P>0.05). MDA level of kidney significantly increased in CS group, however addition of milk thistle seed into diet was reduced MDA level of kidney in CS+MT group (P<0.01). High calorie diets caused changes in antioxidant system in blood and tissues. Consequently, high-calorie basal diets especially addition of SFO improved only egg production and fertility rate. However feeding quails with corn syrup had significantly higher lipid peroxidation in kidney. Supplementation of milk thistle seed into CS group may protect kidney against free radical damage.

There is little information in the scientific literature about effect of in vitro exposure of fish spermatozoa to pesticides. In vitro effect of Lambda-cyhalothrin (La) on sperm quality and oxidative stress has not been fully explored yet. The effects of LCT, which is a synthetic pyrethroid insecticide, on quality and oxidative stress of spermatozoa were investigated in vitro due to extensively use to control a wide range of insect pests in agriculture, public health, and homes and gardens. To explore the potential in vitro toxicity of La, fish spermatozoa were incubated with different concentrations of LCT (0.6, 1.2 and 2.4 mu g/L) for 2 h. Reduced glutathione (GSH), glutathione peroxidase (GSH-Px), catalase (CAT) and malondialdehyde (MDA) in spermatozoa were analyzed for determination of oxidant and antioxidant balance. Our results indicated that the percentage and duration of sperm motility significantly decreased with exposure to LCT. Activity of GSH-Px and MDA (P < 0.05) and GSH levels (P < 0.05) increased in a concentration-dependent manner while CAT activity decreased (P < 0.05). In conclusion, the oxidant and antioxidant status and sperm quality were affected by increasing concentrations of LCT. (C) 2015 Elsevier B.V. All rights reserved.

The objective of the present work is to examine the effects of different doses (0, 10, 20 and 40 g kg(-1) fish diet mass) of kefir on growth performance and oxidant-antioxidant status in the blood and liver tissues of Coruh trout, Salmo coruhensis, in different periods (2 and 3 months). In this study, survival was more than 88.3%, and irrespective of dietary kefir levels at the end of the study. There were no significant differences in SGR (Specific growth rate), FCR (Feed conservation rate), CF (Condition factor) among fish fed diets with 0, 10, 20 and 40 g kg(-1) kefir, however, these growth values were significantly different in terms of durations between 2-month and 3-month treatment (P<0.05). Our results indicated that TAS (Total Antioxidant Status) and TOS (Total Oxidant Status) in blood tissues reduced with kefir treatment at the end of third month (P<0.05). Malondialdehyde (MDA) levels in liver decreased in all groups compared with control group and a considerable extent decrease was observed in 40 g dose of kefir treatment at the end of third month. The data obtained from this experiment indicated that the same dose of kefir was more effective at the end of 3-month treatment than 2-month treatment (P<0.001). Although there was no statistical difference among groups, an increase in the glutathione peroxidase (GSH-Px) enzyme activity was observed in all groups compared to control groups. While catalase (CAT) activity decreased in all groups compared to control group at the end of second month (P<0.01), the decrease was insignificant level at the end of third month. In glutathione (GSH) levels, statistical differences were no observed in all groups compared to the control group with 2-month treatment while there were significant increases with 3-month treatment (P<0.001). It was concluded that kefir could play an antioxidant role and its effectiveness depended on dosage and time of application in Coruh trout, S. coruhensis.

- MakaleAçık Erişim2012Scopus SJRQ2Changes in nitric oxide levels and erythrocyte arginase activities in patient with Down syndrome
Ozbey, Ulku; Seyran, Ayse; Erisir, Mine; Benzer, Fulya; Savucu, Yuksel
Down syndrome is a chromosome abnormality with specific clinical symptoms and mental retardation caused by trisomy of chromosome 21. There is evidence that oxygen free radicals play an important role in the pathophysiology of many neuropsychiatric disorders. Although, it has not been investigated yet, several recent studies proposed that nitric oxide related to oxidative stress and arginase may have a pathophysiological role in mental retardation. The purpose of this study was to determine plasma nitric oxide levels and erythrocyte arginase activities in patients with Down syndrome and in healthy children. Plasma nitric oxide levels and erythrocyte arginase activity were measured in 20 patients with Down syndrome (median age: 15.6 +/- 7.5) and 20 age-matched healthy subjects (median age: 16.2 +/- 7.4). Significant differences were found between erythrocyte arginase activity of the patients with Down syndrome (35.2 +/- 2.7 U/g hemoglobin (Hb)) and those of healthy children (43.5 +/- 2.9 U/g Hb), (P < 0.05). But we have not found any statistical difference between persons with Down syndrome and control group for nitric oxide (P > 0.05). It was concluded that the level of erythrocyte arginase activity could be considered as a useful marker for assessing the level of mental retardation in children with Down syndrome.
