Rainbow trout aquaculture is challenging due to the restricted spawning seasons. Sperm cryopreservation technology can help ensure a successful spawning season, but its effectiveness is restricted by oxidative stress during freezing and thawing, which reduces the availability of male gametes. This study aimed to evaluate the effect of several types and concentrations of antioxidants on post-cryopreservation rainbow trout sperm quality. Subsequently, a factorial completely randomized design with three replications was applied, testing four types of antioxidants Ascorbic acid, Myo-inositol, Butylated hydroxytoluene (BHT), and Alpha-tocopherol at concentrations of 0, 20, 40, and 60 mM, respectively. The evaluation parameters included motility rate, sperm lifespan, and sperm motion characteristics using the Computer Assisted Sperm Analysis system. The results showed that the type and concentration of antioxidants significantly affected sperm motility and sperm lifespan (P<0.05). The treatment method carried out using 40 mM Ascorbic acid and 20 mM BHT led to the highest sperm motility of 46.67% and sperm lifespan of 34 seconds. Although both antioxidants showed similar effectiveness, BHT at a concentration of 20 mM is more recommended due to its lower toxicity and technical as well as economic efficiency considerations.

The fertilization and subsequently embryo and larvae development are linked to gamete quality and FA (fatty acid) content is one of the specific factors affecting egg or sperm quality. Hence, FA profile of gametes (egg and sperm) was documented in brook trout (Salvelinus fontinalis) under cultivation. FAs were detected and measured using gas chromatography. These results indicated that a total of 26 and 22 FAs were determined in egg and sperm, respectively. The levels of PUFAs were determined in nearly identical quantities in both egg (47.57%) and sperm (47.53%). DHA (Docosahexaenoic acid), LA (Linoleic acid), EPA (Eicosapentaenoic acid) and ARA (Arachidonic acid) were predominant PUFAs for egg and sperm. Interestingly, vaccenic acid (C18:1 n-11) (24.07%) was major FAs in egg. In conclusion, the analysis of fatty acids in broodstock gametes contributes valuable insights into the dietary requirements necessary for effective aquaculture breeding programs.

The walking catfish Clarias batrachus is popular and affordable in Southeast Asian countries. The development of breeding technology for this species is ongoing, with sperm cryopreservation being a key aspect of study that supports its breeding program. Cryopreservation is an essential method in fish breeding, particularly for sustaining breeding initiatives that supply larvae throughout the years. However, this process can lead to oxidative stress, which may decrease sperm quality due to structural and physiological damage, including DNA fragmentation. Adding an antioxidant to the freezing medium could help mitigate these negative effects and maintain sperm quality. This study aims to evaluate the effectiveness of various types and concentrations of antioxidant on the quality of C. batrachus sperm post-cryopreservation. Four types of antioxidants were tested: ascorbic acid, Myoinositol, alpha-tocopherol, and Butylated Hydroxytoluene, each at concentrations of 0, 20, 40, and 60 mg/L. Ringer solution, 5% DMSO and 5% egg yolk were used as extender and cryoprotectants, respectively. Sperm samples were stored in liquid nitrogen for 2 weeks, after which sperm quality was analyzed. The results showed that both the type and concentration of antioxidant had a significant effect (p<0.05) on all measured sperm quality parameters. Ascorbic acid at a concentration of 40 mg/L produced the best results, achieving 62.33% motility, 66.00% viability, and 61.67% fertility. Electrophoresis analysis showed no DNA fragmentation in any of the treatments, indicating that the genetic structure of sperm remained intact. Ascorbic acid at 40 mg/L is the most effective antioxidant for cryopreservation of sperm C. batrachus.

The commercial aquaculture of seurukan fish (Osteochilus jeruk) is common in Indonesia; however, seed availability from the wild is limited. Therefore, induced breeding technology in seurukan fish using synthetic reproductive hormones is crucial. This study evaluated the efficacy of five different types of synthetic reproductive hormones and their corresponding doses on the reproductive performance of seurukan fish in captivity. The methodology consisted of two experiments; first, four types of synthetic reproductive hormones were tested: ovaprim, ovaspec, human chorionic gonadotropin (HCG), and ovalumon. Second, several doses of ovaprim hormone were stested with doses of 0.25, 0.50, 0.75, and 1.0 ml kg-1 body weight. The results showed that ovaprim, ovaspec, and HCG successfully induced ovulation and spawning in seurukan fish. The hormones significantly affected the latency period, number of eggs released, fertilization, and survival rates (P<0.05), but did not significantly affect the hatching rate and survival of larvae (P>0.05). Ovaprim produced better egg quality compared to ovaspec and HCG. In the second experiment, the results showed a significant difference in the latency period and egg size (P<0.05) of fish treated with varying doses of ovaprim hormone. The hormone doses did not significantly affect the number of eggs, hatching rate, fertilization rate, survival rate, or larval survival (P > 0.05). The dose of 0.5 ml kg-1 BW of ovaprim produced the best results. Therefore, a dose of 0.5 ml kg-1 BW was recommended as an effective dose for induced spawning and seed production of seurukan fish.

There is no known artificial breeding method for depik fish Rasbora tawarensis. Sperm extender and the dilution ratio are crucial factors in the artificial breeding of fish. The current study aimed to assess the effect of several common fish sperm extenders and its dilution ratio on sperm characteristics and fertilization of depik fish eggs. Two series of experiments were performed to find the best extender and its dilution ratio. In the first experiment, five types of extenders were tested: Ringer's solution, physiological solution, coconut water, sugarcane water, and roomie water. The second experiment studied four dilution levels of sperm and extender (v/v): 1:20, 1:30, 1:40, and 1:50. Every treatment was conducted in four replicates and the experiments were conducted consequently so that the findings in the first experiment were applied in the second. The first experiment showed that Ringer's is the best extender for depik sperm with sperm motility, fertilization and hatching rates of 71.00%, 69.30%, and 53.66%, respectively; and the second experiment revealed that Ringer's at dilution ratio 1:40 gave higher sperm motility, fertilization, and hatching rates of 74.66, 70.33, and 59.00%, respectively. In conclusion, Ringer's solution was the most favorable extender for sperm which yielded best fertilization results at 1:40 dilution ratio (sperm:extender, v/v) in depik fish.

The study aims to determine the type of extender and the best dilution ratio for short preservation walking catfish Clarias batrachus sperm at a refrigeration temperature of 4°C. The study employed a completely randomized design with two factors, each consisting of 6 treatments and 3 replications. The experiments were divided into 2 stages: the first stage tested several types of extenders, namely tap water, Ringer’s solution, physiological solution, Alsever’s solution, urea solution, and glucose base solution, all at the same dilution level of 1:20 (sperm: extender, v/v). The second stage examined various levels of sperm dilution ratios, namely 1:10, 1:20, 1:30, 1:40, 1:50, and 1:60 (sperm: extender, v/v). The samples were stored at a refrigerated temperature of 4°C. Furthermore, motility and viability were monitored at 24-hour intervals for next 6 days, and on the final day of the experiment. The ANOVA test showed that the type of extender and dilution ratio significantly affected the motility and viability of the spermatozoa and percentage of fertilization (P < 0.05). Generally, the quality of sperm decreased after preservation for 144 hours at 4°C, but diluted with Ringer solution at a ratio of 1:40 (sperm: extender, v/v), still maintains good parameters. Therefore, it is recommended that Ringer’s solution at a dilution ratio of 1:40 is suitable for preserving walking catfish C. batrachus sperm at a refrigerated temperature of 4°C.

Sperm quality is preserved through the crucial involvement of antioxidants, which play a vital role in minimizing the occurrence of reactive oxygen species (ROS) during the cryopreservation process. The suitability of the type and concentration of antioxidants are species-dependent, and this study is crucial in order to improve the quality of the climbing perch sperm post-cryopreservation. Therefore, this study aimed to determine the best type and concentration of antioxidants for cryopreservation of climbing perch Anabas testudineus sperm. To achieve this, 6 types of antioxidants, namely, ascorbic acid, beta-carotene, glutathione, butylated hydroxytoluene (BHT), myo-inositol, and alpha-tocopherol, with inclusion of a control were tested in 3 replications at three concentration levels of 0 mg/L (control), 20 mg/L, 40 mg/L, and 60 mg/L. Sperm was diluted in a glucose-base extender at a ratio of 1:60 (sperm: glucose base), then 10 % DMSO and 5 % egg yolk was added before cryopreservation for two weeks. The results showed that the type and concentration of antioxidants had a significant effect on the motility and viability of cryopreserved climbing perch sperm (P < 0.05), where the best results for ascorbic acid, beta-carotene, glutathione, myo-inositol, and alpha-tocopherol were obtained at a concentration of 60 mg/L, while BHT was at a concentration of 20 mg/L. The best results for glutathione, myo-inositol, and alpha-tocopherol were significantly different from other treatments, while the best results for ascorbic acid and beta-carotene (60 mg/L) were not significantly different from the 40 mg/L concentration, while the best results for BHT were not significantly different from the control treatments. Therefore, the best concentration of glutathione, myo-inositol, and alpha-tocopherol was 60 mg/L, while for ascorbic acid and beta-carotene it was 40 mg/L, and BHT was not recommended. DNA integrity analysis indicated the absence of fragmentation in all samples, including fresh, control, and treated sperm. Based on practical and economic considerations, myo-inositol at 60 mg/L was recommended for cryopreservation of climbing perch A. testudineus sperm.

BACKGROUND: Walking catfish, Clarias batrachus is one of the native and most popular freshwater catfish species in Indonesia. However, cultivation faces challenges, particularly due to the scarcity of larvae resulting from underdeveloped breeding technologies. Cryopreservation is a method of storing sperm to maintain viability for a long period and support the breeding technology of the fish. Cryoprotectant, in this context, plays an important role in determining the success of sperm cryopreservation. OBJECTIVE: To determine the best type and concentration of cryoprotectant for cryopreservation of walking catfish sperm. MATERIALS AND METHODS: A total of five different types of cryoprotectants, namely DMSO, glycerol, ethyl glycol, ethanol, and methanol, were tested at four concentration levels namely 0%, 5%, 10%, 15%, and 20%, each with four replications. RESULTS: The type and concentration of cryoprotectant had a significant effect on sperm motility and viability (P<0.05). The best outcomes were obtained with 5% DMSO and ethyl glycol, 10% glycerol and methanol, as well as 15% ethanol. CONCLUSION: The highest motility and viability values were obtained with 5% DMSO, resulting in its recommendation for cryopreservation of walking catfish sperm.

The objective of this study was to determine the effects of different feeding protocols on compensatory growth, feeding rate, and feed conversion efficiency of Black Sea Trout Salmo trutta labrax. To our knowledge, no previous feeding experiments with Black Sea Trout have incorporated a fasting period to simulate the handling and acclimation conditions experienced by farmed fish. Fifteen fish per tank (10.69 +/- 0.06cm and 13.22 +/- 0.14g, mean +/- SE) were stocked into 40-L fiberglass tanks. Fish were fed with different fasting-feeding regimes for 95 d and were equally allotted to four treatments (T-cont: control; T5-10: 5 d fasting, 10days feeding; T10-10: 10 d fasting, 10 d feeding; and T15-10: 15 d fasting, 10 d feeding) with three replicates per treatment. The results indicated that length (P = 0.0005), weight (P = 0.000), condition factor (P = 0.013), and specific growth rate (P = 0.014) were significantly affected by the interaction between feeding and time. All fasting treatments showed partial compensation during refeeding. There was an increase in daily feeding rate and feed conversion efficiency in fasting treatments compared with the control treatment. In contrast, at the end of the experiment specific growth rate, condition factor, and body weight in fasting treatments were significantly lower compared with the control treatment. We concluded that there was partial compensation of growth with regular refeeding after periods of feed deprivation (e.g., 5, 10, or 15 d) over a long term and a shorter fasting period may be preferred in order to achieve compensatory growth. The objective of the present work is to examine the effects of different doses (0, 10, 20 and 40 g kg(-1) fish diet mass) of kefir on growth performance and oxidant-antioxidant status in the blood and liver tissues of Coruh trout, Salmo coruhensis, in different periods (2 and 3 months). In this study, survival was more than 88.3%, and irrespective of dietary kefir levels at the end of the study. There were no significant differences in SGR (Specific growth rate), FCR (Feed conservation rate), CF (Condition factor) among fish fed diets with 0, 10, 20 and 40 g kg(-1) kefir, however, these growth values were significantly different in terms of durations between 2-month and 3-month treatment (P<0.05). Our results indicated that TAS (Total Antioxidant Status) and TOS (Total Oxidant Status) in blood tissues reduced with kefir treatment at the end of third month (P<0.05). Malondialdehyde (MDA) levels in liver decreased in all groups compared with control group and a considerable extent decrease was observed in 40 g dose of kefir treatment at the end of third month. The data obtained from this experiment indicated that the same dose of kefir was more effective at the end of 3-month treatment than 2-month treatment (P<0.001). Although there was no statistical difference among groups, an increase in the glutathione peroxidase (GSH-Px) enzyme activity was observed in all groups compared to control groups. While catalase (CAT) activity decreased in all groups compared to control group at the end of second month (P<0.01), the decrease was insignificant level at the end of third month. In glutathione (GSH) levels, statistical differences were no observed in all groups compared to the control group with 2-month treatment while there were significant increases with 3-month treatment (P<0.001). It was concluded that kefir could play an antioxidant role and its effectiveness depended on dosage and time of application in Coruh trout, S. coruhensis.

In this study, fecundity and egg size of rainbow trout (Oncorhynchus mykiss), Black Sea trout (Salmo labrax) and brook trout (Salvelinus fontinalis) were examined. Two and six years old female fish reared in commercial fish farm were used and the female size, total and relative fecundity and egg size (mm) were determined for the broodstock fish of three tested species. The average weights of broodfish of rainbow trout, Black Sea trout and brook trout 1.357.27+/-406, 532+/-673.7 and 310.40+/-85.0 g, respectively. The total fecundity (number of eggs fish(-1)) was 2.809+/-791, 1.401+/-1374 and 723+/-320, the relative fecundity (number of eggs kg(-1) fish) was 2.180+/-676, 3.558+/-1307 and 2.571+/-1530 and the egg size (mm); 4.95+/-0.2, 4.51+/-0.67 and 4.49+/-0.21 were determined for rainbow trout, Black Sea trout and brook trout, respectively. A significant linear relationship was found total fecundity and the post-stripped fish weight of Black Sea trout (r = 0.96), but this relationship was low for the rainbow trout (r = 0.28) and brook trout (r = 0.17). A negative relationship was observed between the relative fecundity and brood size in rainbow trout (r = 0.57), Black Sea trout (r = 0.58) and brook trout (r = 0.54). A significant linear relationship was observed in rainbow trout (r = 0.63), Black Sea trout (r = 0.86) and brook trout (r = 0.41) (p<0.01).
