Serdar, Osman; Yildirim, Nuran Cikcikoglu; Tatar, Sule; Yildirim, Numan; Ogedey, Aysenur

The discharge of landfill leachate into the environment without effective treatment poses a serious threat for the aquatic ecosystems. This present study was undertaken to evaluate whether electrocoagulation process is efficient for treatment landfill leachate (LL) or not by using antioxidant biomarkers in Gammarus pulex. Glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and catalase (CAT) activities and malondialdehyde (MDA) and glutathione (GSH) levels in G. pulex exposed to untreated, treated, and diluted rates 1/10 and 1/20 in both LL during 24 and 96 h were tested. Physiochemical characteristics of leachate (chemical oxygen demand, electrical conductivity, pH, phosphate, turbidity, NH3, Cl−, and color) were determined pre and post treatment. All physiochemical characteristics of LL decreased after treatment process. GSH-Px and CAT activities and GSH and MDA levels were increased in untreated groups when compared to control (p < 0.05). After treatment by electrocoagulation, MDA and GSH levels and CAT activities were returned to control values. In conclusion, the abilities of LL to stimulate oxidative stress in G. pulex have been proven. The results revealed that antioxidant parameters are useful biomarkers for determining the treatment efficiency of the electrocoagulation process.

Aksu, Onder; Yildirim, Nuran Cikcikoglu; Danabas, Durali; Yildirim, Numan

In this study, we determined the effects of the textile dyes Remazol Brillant Blue R (RBBR) and Congo Red (CR) on the enzymatic activities of Glutathione S-Transferase (GST), cytochrome P450 (CYP1A1) and Lactate dehydrogenase (LDH), in hepatopancreas of Astacus leptodactylus. The crayfishes were exposed to 0.5, 1.0, and 2.0 mg/l of RBBR or CR, for 24 and 48 h. The recorded GST activity clearly increased after 24 or 48 h exposure to CR, compared to a control group (p<0.05), but the changes of GST activity depending on time and dose were not statistically significant in the RRBR group (p<0.05). The activity of CYP1A1 generally decreased, but LDH activity increased in the groups exposed to different doses of CR and RBBR, when compared to their control groups. Detoxification enzymes (as GST, and CYP1A1) and metabolic enzymes (as LDH) can be proven to make useful markers for further evaluating the physiological effects of CR and RBBR on crayfish.

Yonten, Vahap; Alp, Hevidar; Yildirim, Numan; Yildirim, Nuran Cikcikoglu; Ogedey, Aysenur

This study aims to explore the optimum conditions for P. eryngii var. ferulae in adsorption of sulfamethazine (SM) from synthetic solutions via reduction of chemical oxygen demand (COD). The optimum conditions were optimized using the response surface methodology (RSM) developed by the application of the quadratic model associated with the central composite design. For this aim, RSM was employed to determine the effects of some parameters on this adsorption process by fungal biomass as an effective and available adsorbent. The investigated parameters were initial concentration of sulfamethazine (70–235 mg L−1), solution pH (3–7), adsorbent dosage (0.3–1.5 g), and contact time (5–25 min). The significant factors on each experimental design response were identified from the analysis of variance. The results of RSM analyzes revealed that optimum conditions of initial SM concentration, pH, adsorbent dosage, and contact time for maximum COD removal (82%) were achieved as 400 mg L−1, 11, 1.46 g, and 15.4 min, respectively. The results showed that dead biomass of P. eryngii var. ferulae were an appropriate adsorbent for the removal of COD in the SM medium from synthetic solutions.

The ability of Phanerochaete chrysosporium (ME446) on chemical oxygen demand remediation in submerged culture medium supplemented with malathion insecticide

Erguven, Gokhan Onder; Yildirim, Numan; Adar, Elanur

This present study aims to evaluating the efficiency of P. chrysosporium (P.c) to degrade the different concentrations of malathion (MLT) (50, 100 and 150 ppm) under agitated (130 rpm) submerged culture conditions via reduction of chemical oxygen demand (COD). We have previously screening the growth of fungus in sabouraud dextrose agar (SDA) medium for 6 days. We have also determined the laccase activity during degradation process for 15 days. According to our results the fungus has achieved 99.6, 98.8 and 98.7 % COD reduction at concentration of 50, 100 and 150 ppm respectively the end of the 15 days. The highest laccase activity (180 ± 5.69 U/L) was measured at concentration of 50 ppm the end of the 15 days and the lowest activity (36 ± 3.26) was determined at concentration of 150 ppm the end of the 15 days. The dried biomass of fungus was weighted at end of the 15 days and we have determined that malathion in culture medium reduced the biomass production compared the control group (SDB + P.c). Our results showed that, P. chrysosporium could be an effective bioremediation tool for treatment of malathion containing wastewater.

Yonten, Vahap; Tanyol, Mehtap; Yildirim, Numan; Yildirim, Nuran Cikcikoglu; Ince, Muharrem

This study investigates preparation of biosorbent Pleurotus eryngii immobilized on Amberlite XAD-4 and the optimal conditions for removal of Remazol Brilliant Blue R (RBBR) reactive dye from synthetic aqueous solutions. The process was optimized using the response surface methodology (RSM) developed by the application of the quadratic model associated with the central composite design. For this purpose, RSM was employed to determine the effects of operational parameters on this material as effective and available adsorbent. The investigated variables were dye initial concentration (10-60 mg L-1), solution pH (2-9), adsorbent dosage (0.1-0.5 g), and temperature (20-45 degrees C). The significant factors on each experimental design response were identified from the analysis of variance (ANOVA). The RSM indicated that optimum conditions of initial dye concentration, pH, adsorbent dosage, and temperature for maximum RBBR removal (98%) were achieved as 36.3 mg L-1, 2.0, 0.304 g, and 38.7 degrees C, respectively. The results showed that this biosorbent was an appropriate adsorbent for the removal of RBBR from aqueous solutions.

Adsorption of bisphenol A from aqueous solutions by Pleurotus eryngii immobilized on Amberlite XAD-4 using as a new adsorbent

Yonten, Vahap; Ince, Muharrem; Tanyol, Mehtap; Yildirim, Numan

The adsorption of bisphenol A (BPA) in a fixed-bed column was evaluated in this work. Fungus Pleurotus eryngii isolated from Tunceli (Turkey) immobilized on XAD-4 resin was used as an adsorbent for decontamination of BPA from aqueous solutions. The extent of BPA removal capacity was evaluated as a function of parameters such as pH, volume of the sample, and column bed height. Adsorption isotherms of BPA on immobilized P. eryngii were determined and correlated with common isotherm equations such as Langmuir and Freundlich models. The mechanism of adsorption of BPA was analyzed preadsorption (XAD-4+fungus) and postadsorption (XAD-4+fungus+BPA) using fourier transform infrared spectroscopy. The results show that adsorption ability of BPA by immobilized P. eryngii is possible and this system yields the best degree of BPA removal (90%) at pH 11 for an initial load of 120mgL(-1). Therefore, this material can be used as a very effective adsorbent for BPA decontamination in aqueous solutions.

Cytokine Responses in Gills of Capoeta umbla as Biomarkers of Environmental Pollution

Danabas, Durali; Yildirim, Nuran Cikcikoglu; Yildirim, Numan; Onal, Ayten Oztufekci; Uslu, Gulsad; Unlu, Erhan; Danabas, Seval; Ergin, Cemil; Tayhan, Nilgun

Immunological biomarkers reflect the effects of exposure to environmental contaminants. In this study, the suitability and sensitivity of cytokine responses, interleukin 1 beta (IL-1 beta), interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-alpha) in gill tissues of Capoeta umbla (Heckel, 1843), collected from different regions, as early warning indices of environmental pollution and ecosystem health was evaluated. Fish and water samples were taken from ten stations in March and September 2011 and 2012. Tumor necrosis factor-alpha, IL-1 beta and IL-6 levels were determined in samples of the gill tissues by using an ELISA kit. Significant variations of TNF-alpha, IL-1 beta and IL-6 levels observed between stations and seasons. The results of this study show that seasonal variations of cytokine responses in gills of Capoeta umbla are sensitive to the contaminants present in Uzuncayir Dam Lake (Tunceli, Turkey) water and are valuable biomarkers for environmental pollution and ecosystem health.

Biochemical response of crayfish Astacus leptodactylus exposed to textile wastewater treated by indigenous white rot fungus Coriolus versicolor

Aksu, Onder; Yildirim, Nuran Cikcikoglu; Yildirim, Numan; Danabas, Durali; Danabas, Seval

The discharge of textile effluents into the environment without appropriate treatment poses a serious threat for the aquatic organisms. The present study was undertaken to investigate biochemical response of crayfish Astacus leptodactylus exposed to textile wastewater (TW) treated by indigenous white rot fungus Coriolus versicolor. Glutathione S-transferase (GST), cytochrome P450 1A1 (CYP1A1), and acetylcholinesterase (AchE) levels in hepatopancreas and abdomen tissues of crayfish exposed to untreated, treated, and diluted rates (1/10) in both TW during 24 and 96 h were tested. Physiochemical parameters (electrical conductivity (EC), chemical oxygen demand (COD), pH, and total dissolved solid (TDS)) of TW were determined before and after treatment. Physiochemical parameters of TW decreased after treatment. The GST activity and AchE were generally increased, but CYP1A1 activity was decreased in hepatopancreas tissue of crayfish exposed to different kinds of untreated TW. After treatment by indigenous white rot fungus (C. versicolor), GST and CYP1A1 activities were returned to control values, while AchE activities were increasing further. In this study, only GST and CYP1A1 activities of A. leptodactylus confirmed the efficiency of TW treatment with C. versicolor.

Changes in antioxidant defense system in gills of Capoeta umbla caught from Uzuncayir Dam Lake, Turkey

Danabas, Durali; Yildirim, Nuran Cikcikoglu; Yildirim, Numan; Onal, Ayten Oztufekci; Uslu, Gulsad; Unlu, Erhan; Danabas, Seval; Ergin, Cemil; Tayhan, Nilgun

The aim of this work was to determine the seasonal changes in the activities of selected biomarkers in Capoeta umbla (Heckel, 1843) caught from Uzuncayir Dam Lake (Tunceli, Turkey) and to evaluate the effects of environmental factors on these activities. Fish were sampled on seasonal basis, and superoxide dismutase, catalase, glutathione peroxidase activities and levels of glutathione and malondialdehyde in gills were determined. Significant variations of oxidative stress biomarkers were observed between seasons and sites. The results of this study show that seasonal variations of oxidative stress responses and lipid peroxidation in gills of C. umbla are sensitive to the contaminants present in water of Uzuncayir Dam Lake and selected parameters are in valuable biomarkers for monitoring of water systems, since they give an early warning signal of effects of xenobiotics on aquatic organisms at molecular levels which help to prevent their effects at organismal level. Use of acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 in Capoeta umbla as biomarkers for monitoring of pollution in Uzuncayir Dam Lake (Tunceli, Turkey)

Yildirim, Nuran Cikcikogiu; Yildirim, Numan; Danabas, Durali; Danabas, Seual

Uzuncayir Dam Lake is one of the most important water bodies in Tunceli. It is polluted by domestic wastewaters together with natural contamination and pesticides, which have had cumulatively negative effects. This study analyzes the effects of pollution by using the fish species, Capoeta umbla as a sentinel species, with a biomarker approach. The approach comprehends a general biomarker of individual fish health, the condition factor and specific biomarkers of contaminant exposure such as glutathione S-transferase (GST) and cytochrome P450 1A1 (CYP1A1) and acetylcholinesterase (AChE) activity. The general water physico-chemical parameters were measured at each sampling sites during each fish-sampling period. Strong seasonal and locational variations were observed among selected markers. In conclusion, the multibiomarker approach used in the present study clearly revealed differences in the fish health among reference and potentially contaminated sites. The study shows that changes of biomarkers such as acetylcholinesterase, glutathione S-transferase and cytochrome P450 1A1 might be useful for the assessment of environmental contamination in the Uzuncayir Dam Lake (Tunceli, Turkey). Temporal Changes in Concentrations of Some Trace Elements in Muscle Tissue of Crayfish, Astacus leptodactylus (Eschscholtz, 1823), from Keban Dam Lake

Aksu, Onder; Adiguzel, Ragip; Demir, Veysel; Yildirim, Numan; Danabas, Durali; Seker, Sebahat; Can, Safak Seyhaneyildiz; Ates, Mustafa

Crayfish (Astacus leptodactylus Eschscholtz, 1823) is the native crayfish species in Turkey. It was exported regularly to Western Europe. In this study, bioaccumulation and temporal trends of some trace elements (arsenic: As, cadmium: Cd, copper: Cu, mercury: Hg, lead: Pb, and zinc: Zn) in edible abdomenmuscle of crayfish from Keban Dam Lake (Elazig, Turkey) were investigated for the 2006-2012 period. Sequence of metal concentration levels was Zn > Cu > Hg > Pb > Cd > As in muscle tissues. The highest concentration of Zn (21.69mg kg(-1)) was detected in 2006, while the lowest (4.35mg kg(-1)) in 2009. In general, it was found that the concentrations of trace elements investigated were lower than the maximum permissible limits of the food regulations of the Ministry of Food, Agriculture, and Livestock (MFAL), the Turkish Food Codex and Commission Regulation (EC). If the crayfish selected for the study are recognized as bioindicators of environmental pollution, then it is possible to conclude that the changes in studied trace elements concentrations in the Keban Dam Lake are being steady.

This study is aimed to determine the bioaccumulation of copper and zinc in hemolymph of freshwater crayfish (Astacus leptodactylus) fed with zebra mussel (Dreissena polymorpha) collected from Keban Dam Lake. This is the first study on bioaccumulation of Cu and Zn in hemolymph of males and females of A. leptodactylus. The average concentrations of Cu and Zn in the hemolymph samples were found out as 0.54 and 35.07 ppm respectively for male and 0.42 and 15.59 ppm respectively for female control freshwater crayfish. The levels of Zn in the hemolymph of both male and female ones among control and diet groups were statistically significant (P < 0.05). The Cu levels among control and diet groups in female ones were statistically significant (P < 0.05) but not in the male ones (P > 0.05). The results of current study revealed that feed treatment with boiling was not effective at the levels of metal bioaccumulation in hemolymph. It was observed that this metal bioaccumulation is more related with sex differences.

Some Heavy Metals Accumulation in Tissues in Capoeta umbla (Heckel, 1843) from Uzuncayir Dam Lake (Tunceli, Turkey)

Ural, Mesut; Yildirim, Numan; Danabas, Durali; Kaplan, Olcay; Yildirim, Nuran Cikcikoglu; Ozcelik, Mehtap; Kurekci, Eylem Funda

Concentrations of metals were determined in the gills, liver, kidney, heart and muscle in Capoeta umbla caught from six stations from the Munzur River system. Metal concentrations in the tissues tended to vary significantly among stations (p < 0.05). Liver (Cu, 10.10 +/- A 0.23-23.03 +/- A 9.37 ppm; Zn, 14.67 +/- A 3.01-21.82 +/- A 2.39 ppm; Cd, 18.04 +/- A 4.56-52.69 +/- A 10.65 ppb and Fe, 28.87 +/- A 6.78-115.11 +/- A 34.87 ppm) and kidney (Cu, 1.80 +/- A 0.25-3.70 +/- A 0.62 ppm; Zn, 20.81 +/- A 0.37-29.36 +/- A 0.70 ppm; Cd, 132.06 +/- A 5.29-639.51 +/- A 20.14 ppb and Fe, 24.40 +/- A 1.98-59.39 +/- A 1.97 ppm) tissues showed higher metal concentrations than other tissues. It seems that metal contamination in the river is too high for the health of fish and the people who eat them. The geographical locations of catch, season, nature of diet, and the size of fish used for analyses might lead to different metal concentration in the same fish species.
