Lead (Pb) is a widespread environmental pollutant that induces systemic toxicity in aquatic organisms primarily through oxidative stress, hematological disruption, and immune dysfunction. This study investigated the protective effects of ellagic acid (EA) against Pb-induced toxicity in common carp (Cyprinus carpio) using a multi-biomarker approach and molecular docking. Fish were assigned to six experimental groups: control, EA-treated, Pb-I, Pb-I + EA, Pb-II, and Pb-II + EA. Fish in the Pb-I and Pb-II groups were exposed to 2.5 and 5 mg/L Pb, respectively, while EA was administered via diet at 100 mg/kg for 14 days. At the end of the exposure period, hematological indices, innate immune parameters, and oxidative stress biomarkers were assessed in blood, liver, kidney, and gill tissues. Pb exposure caused marked hematological impairment, suppressed immune responses, increased malondialdehyde levels, and disrupted antioxidant defense by reducing SOD, CAT, GSH-Px, and GSH levels while increasing GST activity. In contrast, dietary EA supplementation significantly mitigated Pb-induced alterations, improved hematological and immunological responses, reduced lipid peroxidation, restored antioxidant capacity, and normalized GST activity to control levels. Molecular docking analyses further showed that EA interacts with hemoglobin and immunoglobulin M, supporting its potential role in preserving oxygen transport and immune functions under Pb-induced stress. Overall, the findings demonstrate that Pb-induced toxicity involves coordinated disruption of redox homeostasis and immune function, whereas EA exerts a multi-target protective effect through biochemical and molecular mechanisms. This study provides mechanistic insight into chemical-biological interactions and supports the potential application of natural bioactive compounds in mitigating heavy metal-induced toxicity.

Ibuprofen, a non-steroidal anti-inflammatory drug, has been detected in the aquatic environment, but little is known about its effects on fish and other aquatic organisms. This study aimed to investigate the effect of ibuprofen on micronucleus levels in erythrocytes and whole-body antioxidant parameters of carp (Cyprinus carpio) fry. Fish were exposed to immersion in two different doses of ibuprofen concentrations (50 and 250 μg/L) for 24, 48, 72 and 96 h and 14 days. No mortalities occurred in any group during the test. Abnormal behaviours such as irregular swimming, loss of balance and hanging vertically in the water were observed in some fish exposed to acute toxicity. In this study, a significant increase in the micronucleus number of red blood cells was observed throughout the study period (p < 0.05). Antioxidant parameters were measured spectrophotometrically. Results obtained showed that ibuprofen significantly increased (p < 0.05) malondialdehyde (MDA) levels, glutathione S-transferase (GST) and catalase (CAT) activities, but decreased (p < 0.05) the glutathione (GSH) levels of whole-body carp fry in a dose-dependent manner. Furthermore, ibuprofen significantly increased MDA, GST and CAT levels, but increased the GSH levels of whole-body carp fry in a time-dependent manner. These results indicate that carp fry is sensible to human pharmaceutical agents in aquatic environments. The manner and the constructed interactions between ibuprofen and two protein targets (immunoglobin and haemoglobin) were investigated using molecular docking and molecular dynamics simulation. The results indicated that ibuprofen could affect the haemoglobin activity. Finally, absorption, distribution, metabolism, excretion and toxicity analysis was performed to examine the drug potential of the studied molecules.

This study examined the adverse effects of sublethal concentrations of cadmium (Cd) and the potential of curcumin (CUR) to mitigate Cd-induced toxicity. We administered 1.5 and 3 mg/L of Cd and 100 mg/kg of CUR to fish (Cyprinus carpio) separately and in combination over 14 days. At the end of the experiments, various haematological and immunological parameters, oxidative stress levels and antioxidant parameters of fish were measured, and the differences between the groups were evaluated. Simultaneous CUR treatment with Cd reversed the changes in haematological parameters in fish, increased immune responses, reduced oxidative stress and normalized the impaired antioxidant enzyme activities. In summary, our study showed that dietary supplementation with CUR may be an effective strategy to reduce the toxic effects of Cd in carp.

In this study, ameliorative effects of ellagic acid (EA) on oxidative stress induced by chlorpyrifos (CPF) in carp, Cyprinus carpio, were investigated. Fish were divided into six groups: C (no treatment), EA (100 mg kg fish−1), CPF-1 (0.040 mg L−1), CPF-2 (0.080 mg L−1), CPF-1 + EA, and CPF-2 + EA. CPF and EA were applied simultaneously for 14 days and, at the end of the study, liver, kidney, and gill samples were collected from fish. On the taken tissue samples, malondialdehyde (MDA) level and some antioxidant enzyme (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and glutathione-S-transferase (GST)) activities were evaluated. The results demonstrated statistically significant increases in the MDA levels of the CPF-1 and CPF-2 groups. On the other hand, the MDA levels were significantly decreased by EA administration. Also, CPF exposure caused statistically significant increases in the SOD and GST activities and statistically significant decreases in the CAT and GPx activities. However, treatment with EA reversed negative alterations in the SOD, CAT, GPx, and GST activities. Therefore, the results of this study results showed that simultaneous treatment with EA alleviates CPF-induced oxidative stress in fish.

Chlorpyrifos (CPF) is an organophosphate pesticide that is frequently and widely used to control both agricultural and domestic pests worldwide. In this study, the protective effect of Fennel (Foeniculum vulgare) essential oil (FEO) was investigated in carp (Cyprinus carpio) exposed to CPF. The fish were divided into six groups that one control group (no treatment) and five experimental groups (FEO (3ml/100g diet) group, CPF1 (0.023 mg/l) group CPF2 (0.046 mg/l) group, CPF1 (0.023 mg/l) plus FEO (3ml/100g diet) group, CPF2 (0.046 mg/l) plus FEO (3ml/100g diet) group). Blood and tissue (liver, kidney, gill, and brain) samples were taken from the fish at the end of 14 days of application. Hemoglobin (Hb) level, nitoblue tetrazolium (NBT) activity, and total immunoglobulin (TI) level were measured in blood samples of fish. Acetylcholinesterase (AChE) activity was determined in brain tissue while malondialdehyde (MDA) level, reduced glutathione (GSH) level, catalase (CAT), and glutathione peroxidase (GPx) activity were determined in liver, kidney, and gill tissues. The results showed that there was a significant decrease in Hb level, NBT activity, and TI levels in CPF-treated fish compared to the control group. In addition, increased in MDA levels and significant decreases in GSH level, AChE, CAT, and GPx activities were observed in CPF-treated groups. However, FEO-treated was showed a significant improvement in all parameters except AChE activity compared to CPF groups. These study findings showed that FEO could improve CPF-induced toxicity in C. carpio, except inhibition of AChE activity.
