This study investigates the largely untapped therapeutic potential of industrial hemp (Cannabis sativa L.) roots, which are usually discarded as agricultural byproducts. The phytochemical composition and biological activities of extracts obtained using Soxhlet, ultrasound-assisted extraction (UAE), and supercritical CO₂ (SC-CO₂) techniques were comparatively evaluated. Compounds were characterized using FTIR, GC–MS, LC–MS, and ICP-MS. SC-CO₂ extraction yielded the most chemically diverse profile, particularly enriched in fatty acids and triterpenoids. In vitro assays demonstrated that SC-CO₂ and Soxhlet extracts significantly promoted fibroblast migration and reduced inflammatory responses compared to UAE extracts. Gene and protein expression analyses further supported these bioactivities. In conclusion, this study highlights the phytopharmaceutical potential of hemp root extracts obtained using alternative extraction techniques, including supercritical CO₂ extraction. While SC-CO₂ extraction enabled the recovery of a chemically diverse extract, the present study focuses on comparative phytochemical and bioactivity outcomes rather than a quantitative assessment of environmental sustainability. The findings support further investigation of hemp roots, currently treated as an underutilized biomass, for potential applications in natural product research.

A new lectin from Amanita vaginata mushroom with potent anti-inflammatory activity

Seliman, Thoiba Mogahid Hussein; Ozsoy, Nurten; Altiparmak-Ulbegi, Gulsum; Alp-Kavlo, Hevidar; Arcan, Gulsah Gamze; Akev, Nuriye; Can, Ayse; Aksoy-Sagirli, Pinar

This study used 80% ammonium sulphate fractional precipitation, followed by affinity chromatography to purify a new lectin (AVL) from the crude extract of the Amanita vaginata mushroom. The purification procedure achieved a 20% yield, a specific activity of 5120 HU/mg protein, and a 16-fold increase in purity. SDS-PAGE analysis showed a single band of 45 kDa. We assessed AVL's anti-inflammatory effect by measuring levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), induced by lipopolysaccharide (LPS), in RAW 264.7 macrophages. Western blot analysis showed that iNOS and COX-2 protein levels were barely detectable in the control groups, but markedly elevated in the LPS-treated groups. However, AVL treatment significantly inhibited the expression of both iNOS and COX-2 in LPS-stimulated cells. This effect was dose-dependent. These findings suggest that AVL's anti-inflammatory effects are mediated through the suppression of iNOS and COX-2 expression.

Novel formulations ameliorate osteoarthritis in rats by inhibiting inflammation and oxidative stress

Erten, Fusun; Ozdemir, Oguzhan; Tokmak, Muhammed; Durmus, Ali Said; Ozercan, Ibrahim Hanifi; Morde, Abhijeet; Padigaru, Muralidhara; Sahin, Kazim

The study tested new oral plant-based formulations (F) on rats with monosodium iodoacetate (MIA)-induced osteoarthritis, measuring inflammation, antioxidant levels, paw size, stride, and analyzing knee joint images. Fifty-six female Sprague Dawley rats were allocated into 8 groups: (1) Control, (2) MIA (OA induced with MIA), (3) MIA + F1 [curcuminoids+gingerols+acetyl-11-keto-β boswellic acid (AKBA)], (4) MIA + F2 (curcuminoids+Withania glycosides+AKBA), (5) MIA + F3 (curcuminoids+total withanolides+AKBA), (6) MIA + F4 (curcuminoids, AKBA), (7) MIA + UCII (type II collagen), and (8) MIA + GCHON (Glucosamine Chondroitin). Treatments F1 to F4 reduced right joint diameter and improved stride length and paw area in OA rats. Despite improvements with treatments F1 to F4, there was no significant difference between these groups (p >.05). In OA animals, F1 to F4 treatments decreased MDA levels and increased antioxidant enzymes activities (p <.001). This was done by reducing levels of inflammatory markers and enzymes like IL-1β, IL-6, MMP-8, TNF-α, CRP, COMP, and LOX-5, while increasing the anti-inflammatory cytokine IL-10. In conclusion, these plant-based treatments significantly reduced osteoarthritis severity, slowed disease progression by reducing inflammation, and protected joints from damage, showing a protective effect in rats with induced osteoarthritis, likely due to their anti-inflammatory and antioxidant properties.

Screening for Chemical Characterization and Pharmacological Properties of Different Extracts from Nepeta italica

Acquaviva, Alessandra; Di Simone, Simonetta Cristina; Nilofar; Bouyahya, Abdelhakim; Zengin, Gokhan; Recinella, Lucia; Leone, Sheila; Brunetti, Luigi; Uba, Abdullahi Ibrahim; Guler, Osman

Plants from the Nepeta genus have been proved to possess different pharmacological properties, among which are antimicrobial, antioxidant, anti-inflammatory, analgesic, and cytotoxic effects. Nepeta italica is a medicinal plant traditionally used for its analgesic effects, and in the present study, the phytochemical composition and biological effects of hexane, dichloromethane (DCM), ethyl acetate (EA), ethanol, ethanol-water, and water extracts of the aerial parts were investigated for determining phenolic composition, antioxidant effects, and anti-inflammatory effects in isolated mouse colon specimens exposed to lipopolysaccharide (LPS). Polar extracts were the richest in terms of phenolic compounds, especially rosmarinic acid. In parallel, ethanol, ethanol-water, and water extracts were also the most effective as scavenging/reducing and enzyme inhibition agents, especially towards cholinesterases and α-glucosidase, and in inhibiting the LPS-induced cyclooxygenase-2 (COX-2) and tumor necrosis factor α (TNFα) gene expression in mouse colon. This poses the basis for future in vivo investigations for confirming the protective effects of polar extracts of N. italica against inflammatory bowel diseases.

Antioxidant effects of oleuropein on hydrogen peroxide-induced neuronal stress-an in vitro study

Kucukgul, Altug; Isgor, Mehmet M.; Duzguner, Vesile; Atabay, Meryem N.; Kucukgul, Azime

Background: Persistent oxidative stress can lead to chronic inflammation and mediate most chronic diseases including neurological disorders. Oleuropein has been shown to be a potent antioxidant molecule in olive oil leaf having antioxidative properties. Objective: The aim of this study was to investigate the protective effects of oleuropein against oxidative stress in human glioblastoma cells. Methods: Human glioblastoma cells (U87) were pretreated with oleuropein (OP) essential oil 10 µM. After 30 minutes, 100 µM H2 O2 was added to the cells for three hours. Cell survival was quantified by colorimetric MTT assay. Glutathione level, total oxidant capacity, total antioxidant capacity and nitric oxide levels were determined by using specific spectrophotometric methods. The relative gene expression level of iNOS was performed by qRT-PCR method. Results: According to viability results, the effective concentration of H2 O2 (100µM) significantly decreased cell viability and oleuropein pretreatment significantly prevented the cell losses. Oleuropein regenerated total antioxidant capacity and glutathione levels decreased by H2 O2 exposure. In addition, nitric oxide and total oxidant capacity levels were also decreased after administration of oleuropein in treated cells. Conclusion: Oleuropein was found to have potent antioxidative properties in human glioblastoma cells. However, further studies and validations are needed in order to understand the exact neuroprotective mechanism of oleuropein.
