The effect of Dimethoate on oxidative stress and antioxidant responses of Pontastacus leptodactylus; [El efecto del dimetoato sobre el estrés oxidativo y las respuestas antioxidantes de Pontastacus leptodactylus]

Dimethoate (DMT) pesticide is one of the chemicals used to protect some agricultural areas from harmful organisms. DMT residues released directly or indirectly to the environment cause serious problems in nature. DMT residues mixed with the aquatic environment adversely affect aquatic organisms and this effect is carried to humans through the food chain. In this study, oxidative stress responses induced by DMT pesticide in Pontastacus leptodactylus were investigated. For this purpose, oxidative stress and antioxidant parameters Thiobarbituric acid reactive substances (TBARS), Glutathione (GSH), Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX) caused by dimethoate (DMT) pesticide in P. leptodactylus at 17.5, 35, and 70 mg·L-1 concentrations at 24 and 96 hours were investigated. Results were determined using ELISA kits. No significant difference was observed in GSH levels and SOD activities compared to control. Statistically significant differences were observed between decreases in CAT and GPx activities and increases in TBARS levels. SPSS 24.0 package program one–way ANOVA (Duncan 0.05) was used in the evaluation of biochemical analyzes. As a result, it was determined that DMT caused oxidative stress formation in P. leptodactylus and caused changes in enzyme activities.

The potential ameliorative impacts of rare earth element cerium in dimethoate induced toxicity in freshwater mussel Dreissena polymorpha

Serdar, Osman; Ketenalp, Zozan; Cikcikoglu Yildirim, Nuran

The purpose of this study was to measure the antioxidant parameters to examine the potential ameliorative impacts of the rare earth element (REE) cerium (Ce) in dimethoate (DM) induced toxicity in Dreissena polymorpha. Lethal concentration 50 (LC50) of DM for 96 h was measured. The antioxidant effect of Ce was investigated by exposure of 1/2 of the LC50 value of DM together with 4, 20 and, 100 mg/L of Ce to D. polymorpha. The responses of the biomarkers were determined by measuring the superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione (GSH) and malondialdehyde (MDA) levels. SOD, CAT, GPX and GSH levels were reduced (p < 0.05), while the MDA levels increased significantly (p < 0.05) in mussels exposed to DM. When DM was exposured together with Ce, it was observed that the MDA levels decreased, SOD, CAT, GPX and GSH levels increased during 24 and 96 h. The results of our study showed that Ce exerted a strong antioxidant effect on toxicity induced by DM. It has been shown that Ce may have a compensatory effect with the positive change in the antioxidant response of D. polymorpha to DM. This study is the first report about Ce and insecticide effects in bivalves.

Single and combined effects of dimethoate and malathion on oxidative stress biomarkers in the non-target freshwater mussel Dreissena polymorpha

Serdar, Osman; Yildirim, Numan; Yildirim, Nuran Cikcikoglu

This study evaluates the single and combined effects of dimethoate (DI) and malathion (MA) on oxidative stress biomarkers in the freshwater mussel Dreissena polymorpha, estimating the potential harm of these pesticides on aquatic ecosystems. Superoxide dismutase (SOD) and catalase (CAT) activities, and glutathione (GSH) and lipid peroxidation (TBARS) levels were determined in D. polymorpha exposed to sublethal concentrations of DI, MA and a combination of the two during 24 and 96 h periods. The results showed that the GSH levels were decreased but the TBARS levels were increased in all the exposure groups after 24 and 96 h compared to the control. It was observed that SOD activity decreased but CAT activity increased in all the DI exposure groups after 24 h compared to the control. At the end of 96 hours, it was observed that CAT and SOD activities increased again in some exposure groups compared to the control. Exposure time also had an effect on biomarkers in different levels. According to the results, the cytotoxicity of DI and MA combination depended on their concentrations. DI or/ and MA, in sub-lethal concentrations, induced oxidative damages in D. polymorpha. Combined exposure of the pesticides can alter their toxicity and may be evidence of increased toxicity and oxidative stress.

The effect of dimethoate pesticide on some biochemical biomarkers in Gammarus pulex

    Serdar, Osman

In this presented study, it was aimed to determine the effects of pesticides on non-target organisms on the freshwater amphipod, Gammarus pulex, by biochemical responses. Acute toxicity value (LC50) in G. pulex of the dimethoate pesticide was determined. The superoxide dismutase (SOD), glutathione S-transferaz (GST), glutathione peroxidase (GPx), and catalase (CAT) activities and malondialdehyde (MDA), glutathione (GSH) levels of the G. pulex organism exposed to the subletal concentrations were analyzed by ELISA for 24 and 96 h. In conclusion, the present study demonstrated the abilities of dimethoate pesticide induce to oxidative stress. The results revealed that MDA, GSH levels SOD, CAT, GPx, and GST activities of G. pulex can be used as an effective biomarkers.
