Lipid content and wastewater treatment potential of Chlorella vulgaris and Scenedesmus obliquus isolated from Uzuncaylr Dam Lake

Can, Şafak Seyhaneyildiz; Can, Erkan; Yilmaz, Kadir

Production of microalgae for biodiesel, one of the alternative renewable energy sources, is costly due to nutrient media. Industrial and domestic wastewater contains nitrogen, phosphorus and other nutrients that are the primary food source for algae. Releasing this environmentally harmful effluent into receiving bodies of water, such as the sea or a freshwater reservoir, without prior treatment causes serious problems. Combining microalgae cultivation with wastewater treatment is a promising strategy for improving wastewater and reducing the cost of nutrient media required for algae production. In this study, wastewater obtained from a wastewater treatment facility was diluted with clean water to 0.25%, 50%, and 75% concentrations, and Chlorella vulgaris and Scenedesmus obliquus were cultured in these nutrient media for 20 days. As a result, microalgae increased their biomass and lipid content, while consuming nitrite, nitrate, phosphate, and ammonium from the wastewater as nutrients.

The Effects of Probiotic-Prebiotic on the Biomass and Protein Content of Spirulina platensis in Different Temperatures and Illuminations

Seyhaneyıldız Can, Şafak; Seyhaneyıldız, Elif; Dörücü, Mustafa

The blue-green algae Spirulina platensis Geitler 1925 has been used by people since ancient times due to its high protein content. This study was conducted to determine the effect of a Probiotic-Prebiotic (P-P) product, which contains Bifidobacterium infantis, Bifidobacterium thermophilum, Leuconostoc mesenteroides as probiotic bacteria, and beta-glucan and oligosaccharide as a prebiotic mixture on S. platensis. In the study, four groups were one of which was a control. In this study, we investigated whether the P-P product contributed to the biomass development and protein production of S. platensis under different temperatures and lighting conditions. In terms of biomass, the P-P product gave different responses at different temperatures and light intensities, but its results were positive in all groups in terms of protein ratio. The most positive effect on biomass and protein content was in the group where 0.1 ml/L P-P product was added, at 40°C and 3200 lux with 68.71% protein content and an average OD of 0.727.

Potential anesthetic properties of bay leaf (Laurus nobilis) essential oil compared with 2-phenoxyethanol on blue dolphin cichlid, cyrtocara moorii

Kizak, Volkan; Can, Erkan; Seyhaneyildiz Can, Şafak

The efficacy of Bay leaf (Laurus nobilis) essential oil (LnEO) as a herbal anesthetic agent was evaluated with 2-Phenoxyethanol (2-PE) in Blue Dolphin Cichlid, Cyrtocara moorii. Fish were exposed to different anesthetic concentrations and the lowest effective concentrations (LECs) were determined according to deep anesthesia (AD<3 min) and full recovery (RF<5 min) times. LnEO showed anesthetic properties, and all applied concentrations of both agents induced AD in C. moorii. The LECs of LnEO and 2-PE for C. moorii were determined as 800 µl L-1 (AD; RF → 183 ± 6.3 s; 191 ± 2.6 s) and 600 µl L-1 (AD; RF → 171 ± 3.2 s; 239 ± 4.9 s), respectively. Significant differences were found between anesthetic agents in terms of deep anesthesia and full recovery times for the same concentrations (p<0.05). Induction and recovery times decreased with increasing in LnEO concentrations. On the other hand, anesthesia times were decreased with increasing of 2-PE concentrations, while recovery times increased. Induction and recovery times for LnEO were significantly dependent on concentrations and positive relationships were recorded between AD and RF, whereas the relationship was negative in 2-PE. Bay leaf essential oil has not been used as an anesthetic agent in fish until now. Our results showed that LnEO was an effective anesthetic agent with some minor side effects on C. moorii.

Anesthetic Efficiency of Three Medicinal Plant Oils for Aquatic Species: Coriander Coriandrum sativum, Linaloe Tree Bursera delpechiana, and Lavender Lavandula hybrida

Can, Erkan; Kızak, Volkan; Can, Şafak Seyhaneyıldız; Özçiçek, Esin

This study evaluated the potential of three essential oils (EOs) that were obtained from coriander Coriandrum sativum (CEO), linaloe tree Bursera delpechiana (BEO), and lavender Lavandula hybrida (LEO) as anesthetic agents. Convict Cichlids Amatitlania nigrofasciata (Günther 1867) were exposed to eight concentrations of anesthetic (50, 75, 100, 125, 150, 200, 250, and 300 μL/L). After exposure to the anesthetic, the fish were transferred to clean water to recover. All of the EOs produced an anesthetic effect after exposure to the compounds for 30 min at the minimal effective concentration (MEC), which was identified according to deep anesthesia (A5 < 3 min) and full recovery (R3 < 5 min) times. At 50 and 75 μL/L, the total loss of equilibrium was not observed for all tested EOs. The total loss of reflex was induced at a faster rate with higher concentrations of anesthetic in all groups. The recovery time generally increased as the concentration of the anesthetic increased. These findings suggest that CEO, BEO, and LEO are all novel potential anesthetics for aquaculture, and the optimal concentrations were identified as 150 μL/L (A5; 156 ± 1.7 s and R3; 165 ± 2.9 s), 125 μL/L (A5; 176 ± 3.5 s; R3; 125 ± 2.0 s), and 200 μL/L (A5; 20.1 ± 2.4 s and R3; 162 ± 3.4 s), respectively. When considering the active ingredients of EOs, this study also demonstrated that future studies should be focused on the major components such as linalyl acetate, 1.8-cineole, α-pinene, geraniol, and linalool. Their synergistic effects should be examined in herbal anesthetic treatments, since new commercial anesthetics will likely contain them.

Anesthetic potential of geranium (Pelargonium graveolens) oil for two cichlid species, Sciaenochromis fryeri and Labidochromis caeruleus

Can, Erkan; Kizak, Volkan; Seyhaneyildiz Can, Şafak; Özçiçek, Esin

The anesthetic potential of geranium oil (Pelargonium graveolens L.) was evaluated in two freshwater aquarium fish species, the electric blue hap (Sciaenochromis fryeri, 2.00 ± 0.61 g) and blue streak hap (Labidochromis caeruleus, 2.31 ± 0.52 g). Fish were exposed to nine concentrations of anesthetic (25, 50, 75, 100, 125, 150, 200, 250, 300 μl L−1). The induction time generally decreased significantly with increasing concentration of geranium oil for both of the fish species. The recovery time increased with the enhancing geranium oil concentration. Deep anesthesia took between 61.19 ± 7.25 s and 165.43 ± 6.78 s for S. fryeri and between 73.32 ± 8.92 s and 171.12 ± 10.74 s for L. caeruleus. Recovery occurred after 96 ± 6.87 s (L. caeruleus), with the longest being 291.45 ± 8.31 s (S. fryeri). The minimal effective concentration was 75 μl L−1 for deep anesthesia for both two species, and 50 μl L−1 was found as the minimal sedative concentration at the stage of loss of equilibrium. These findings suggested that geranium oil is a new potential anesthetic for two ornamental fish species.

Evaluation of anesthetic potential of rosewood (Aniba rosaeodora) oil as a new anesthetic agent for goldfish (Carassius auratus)

Kizak, Volkan; Can, Erkan; Danabaş, Durali; Can, Şafak Seyhaneyildiz

The aim of this research was to investigate the efficacy of rosewood (Aniba rosaeodora) essential oil (EO) as a new herbal anesthetic compared with camphor (Cinnamomum camphora) EO and commonly used chemical agent 2-phenoxyethanol (2-PE) on goldfish (Carassius auratus). The anesthetic efficacies of these herbal EOs and 2-PE were evaluated in goldfish with mean body weight of 7.36 ± 0.77 g. Fish were exposed to each anesthetic concentrations and the lowest effective concentrations (LECs) were established according to deep anesthesia (AD < 3 min) and full recovery (RF < 5 min) times. RD and CP showed anesthetic characteristics and induced AD. The LECs for each anesthetic were established at 250 μl L−1 for RD (AD; RF → 171 ± 2.28 s; 297 ± 7.69 s), 250 μl L−1 for CP (AD; RF → 129 ± 4.18 s; 291 ± 2.81 s) and 700 μl L−1 for 2-PE (AD; RF → 177 ± 2.35 s; 214 ± 4.93 s). At concentrations of 50, 100, 150, 200 μl L−1 for RD, 50, 100, 150 μl L−1 for CP and 400, 500 μl L−1 for 2-PE failed to induce AD in goldfish. There were significantly differences found among RD, CP and 2-PE concentrations in terms of induction and recovery times (p < 0.05). The fastest AD and RF were obtained by 300 μl L−1 CP (120 ± 1.94 s) and 600 μl L−1 2-PE (190 ± 4.34 s), respectively. Fish exposed to 300 μl L−1 RD and 800 μl L−1 2-PE took slightly longer to achieve AD than 300 μl L−1 CP. Induction and recovery times for EOs and 2-PE were significantly dependent on concentrations. Negative relationships was recorded between AD and RF for all anesthetics. No mortality or adverse effects occurred and fish stayed calm during trials. Results showed that RD was found to be an effective anesthetic as well as CP for goldfish and can be used at least 3-fold lower concentrations than 2-PE. In conclusion, rosewood (Aniba rosaeodora) EO, as a new potential anesthetic for fish, proved to be an effective natural agent resulting in rapid induction and recovery.

Anesthetic efficacy of cymbopogon citratus essential oil as a herbal agent in two ornamental fish species

Kizak, Volkan; Can, Erkan; Can, Şafak Seyhaneyildiz; Özçiçek, Esin

The efficacy of Cymbopogon citratus essential oil (CcEO) as a herbal anesthetic agent was evaluated for two ornamental fish species, Sciaenochromis fryeri and Labidochromis caeruleus. Fish were exposed to various essential oil concentrations and the lowest effective concentrations were determined according to deep anesthesia (AD<3 min) and full recovery (RF<10 min) times. CcEO showed anesthetic traits and induced AD in S. fryeri and L. caeruleus. The lowest effective concentrations was 200 µl/L for S. fryeri (AD; RF → 122 ± 1.8 s; 638 ± 8.9 s) and 200 µl/L for L. caeruleus (AD; RF → 139 ± 6.9 s; 625 ± 11.1 s). At concentration of 25 µl/L C. citratus essential oil did not induce AD in both of ornamental fish species. No significant differences were found between fish species in terms of induction and recovery times for same concentrations excluding AD time of 200 µl/L concentration. Although the fastest AD were obtained by 300 µl/L, recovery times were prolonged and fish did not full recover in 20 min. Induction and recovery times for CcEO were significantly dependent on concentrations and negative relationships were recorded between AD and RF. Neither mortality nor adverse effects were observed on two fish species. As a result, CcEO showed anesthetic properties for S. fryeri and L. caeruleus.
