Rare Earth Elements (REE), whose usage areas are increasing day by day, are increasing in the amount of mixing with the environment, causing changes in antioxidant enzyme activities by causing oxidative stress in living organisms. In this study, it was aimed to examine the oxidative stress responses induced by the mixture of 4 different REEs (terbium, gadolinium, Lanthanum, Praseodymium) in Dreissena polymorpha. For this purpose, sublethal concentration values were determined by literature review. Experimental application was carried out within 24 and 96 hours. In the analyzes performed to determine biomarker responses, samples taken from living organisms were weighed and homogenization processes were performed for the analysis of samples taken from the experimental groups, including the control group. After homogenization, samples were centrifuged at 4.000 rpm for 15 minutes. Supernatants were kept at -86 °C until measurements were made. Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities, and glutathione (GSH) and thiobarbituric acid reactive substances (TBARS) levels were determined using ELISA kits. Statistical analyses were performed using SPSS. One-way ANOVA (Duncan’s multiple range test; p < 0.05) was used for comparison of measured parameters among groups. As a result of the application, a decrease in CAT activity and GSH level and an increase in TBARS levels were observed after 96 hours compared to the control group, while no statistically significant difference was detected in SOD and GPx activities.

In parallel with the increase in the need for raw materials used in industry, the area and amount of use of Rare Earth Elements are also increasing. Yttrium (Y), which is used as a raw material source, is thought to form residues in the environment. Gammarus pulex individuals were selected as a model organism to investigate the effects of Yttrium on the environment and aquatic organisms. G. pulexes were exposed to Y at different temperatures (16, 18, and 20 °C) and concentrations (25, 100, and 500 µg/L) for 96 hours and samples were taken at 24 and 96 hours for biochemical analyses. For biomarker analysis, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities, thiobarbituric acid (TBARS) and glutathione (GSH) levels were performed using CAYMAN ELISA kit. Study data revealed that SOD and CAT activities increased significantly with increasing temperature and exposure concentration, while GPx activity showed an irregular response; in contrast, no significant change was observed in TBARS levels at 16 °C, while the increase in TBARS and significant changes in GSH levels at 18 and 20 °C indicate that oxidative stress becomes more pronounced at high temperatures. Nanoparticles (NPs) are increasingly used across various fields of science and technology, including food science from crop harvesting to final food consumption. With their widespread application in daily life, the environmental effects of nanoparticles have become an important area of academic investigation. In this study, Gammarus pulex species was selected as research material/bioindicator species to investigate the effects of zinc oxide (ZnO), copper oxide (CuO), and iron(III) oxide (Fe2O3) on aquatic life. To conduct the experiments, separate setups were established for each of the nanoparticles (ZnO, CuO, and α-Fe2O3). Following ten organisms for each concentration group (10, 20, and 40 ppm) with corresponding control groups, exposures were performed for 24 and 96 hours. Samples were collected at both time points and biochemical analyses were conducted using homogenization procedures. The results were evaluated statistically. According to the findings, SOD activity increased significantly after exposure to ZnO, CuO, and Fe2O3. CAT activity did not show any change under ZnO NP exposure, whereas a decrease was observed under CuO and Fe2O3 NP exposures. However, a significant increase in TBARS levels and a significant increase in GSH levels were observed following nanoparticle exposure. Based on the study data, exposure to ZnO, CuO, and Fe2O3 appears to induce oxidative stress in G. pulex.
- MakaleYalnız künye2026Scopus SJRQ1Determination of microplastic contamination and phthalate ester migration in canned and frozen packaged seafood: A probabilistic human health risk assessment via Monte Carlo simulation
Macit, Arife; Köktürk, Mine; Serdar, Osman; Alak, Gonca
This research was designed to identify the levels of microplastics (MP) and phthalate esters (PAE) in canned and frozen packaged fish and to assess the human health risks associated with these pollutants. Analyses were carried out on a total of 25 sample groups, including 10 different canned tuna samples, 9 different packaged seafood products, and 6 different packaging materials. Microplastics were characterized and polymer identification was performed using μ-Raman and ATR-FTIR spectroscopy, while PAEs were determined by GC–MS/MS. The obtained data were used to calculate estimated daily intake (EDI), hazard index (HI), hazard quotient (HQ), and cancer risk (CR), and uncertainty was assessed using Monte Carlo simulation. The findings revealed that both microplastic and phthalate exposure were higher in the frozen fish group than in canned products. The highest MP concentrations were recorded in packaged Whiting B (0.73 ± 0.25 items/g) and oil-added canned tuna (0.57 ± 0.15 items/g). Regarding PAEs, while DEHP and DBP were generally prominent, significant peak concentrations of BBP and DNOP were also identified, reaching 1.91 ± 0.01 ppm for BBP in canned tuna with sunflower oil and 1.84 ± 0.01 ppm in packaged whiting. Furthermore, higher exposure was observed in males for most PAEs. HQ and HI values were below 1 (maximum HI: 1.57 × 10−2 for males consuming frozen seafood), while CR values remained within an acceptable range (4.74 × 10−7 to 3.49 × 10−6). In conclusion, health risks related to processed fish consumption appear generally low; however, processing and packaging may contribute to contamination and warrant further investigation.
- MakaleAçık Erişim2026Scopus SJRQ2Protective Effect of Propolis Against Oxidative Damage Caused by Cyfluthrin Insecticide on Gammarus pulex
Serdar, Osman
This study aimed to investigate both the protective and therapeutic effects of propolis (PP) against pesticide-induced oxidative stress in aquatic environments. For this purpose, Gammarus pulex was exposed to 0.00 (control), 0.05, 0.1, and 0.2 ppm concentrations of cyfluthrin (CYF) to determine its toxic effects. To evaluate the protective effect of PP, 5 ppm of PP was added to each pesticide concentration. Samples were collected from all experimental groups after 24 and 96 hours under appropriate conditions. SOD activity decreased under CYF exposure but increased significantly after PP supplementation. Although CAT activity did not show significant changes with CYF alone, it increased in the presence of PP. GPx activity remained statistically unchanged under CYF exposure but increased following PP treatment. CYF exposure led to decreased GSH levels; however, PP did not significantly alter GSH levels. TBARS levels increased due to CYF, and this increase was alleviated by PP application.

Anesthesia practices in aquaculture are important for preventing potential stress in fish. For this purpose, interest in natural anesthetic agents as substitute for currently used synthetic anesthetics has increased. In this context, the present study demonstrated the anesthetic efficacy of citronella oil (Cymbopogon nardus) in rainbow trout (Oncorhynchus mykiss) individuals. Based on an artificial neural network (ANN) model derived from preliminary studies, the optimal anesthetic concentration for rainbow trout individuals was estimated as 90 μL L−1. Validation of the concentration predicted by ANN revealed no time-dependent hematological or histological effects on fish to citronella oil. Furthermore, biochemical tests revealed that the partial post-anesthesia increase in ALT and AST values was transient and negligible in light of histological findings. Although oxidative stress was observed during the acute period, the enzymatic activity of SOD and CAT effectively reduced the oxidative damage, demonstrating their role in protecting the fish from oxidative stress. The antioxidant activity observed was temporary, returning to baseline levels without any accompanying tissue damage at the tested concentration. Behavioral monitoring further indicated a swift reduction in motor activity followed by a smooth recovery process, with no evident signs of stress or avoidance behaviour. Overall, these results position citronella oil as a promising natural anesthetic for rainbow trout under the studied parameters. That said, further studies involving various solvents, temperature ranges, and fish species are necessary to comprehensively establish its efficacy and safety.

Moreover, MDA, GSH levels, SOD, CAT, GPx activities and histopathological responses could be used as an effective biomarker in D. polymorpha.
- MakaleAçık Erişim2025Determination of Oxidative Stress Responses in Dreissena polymorpha Exposed to Rare Earth Elements (Terbium, Lanthanum, Gadolinium and Praseodymium) with Temperature
Serdar, Osman; Aydın, Ayşe Nur
The continuous development of industry pushes people to search for new resources, and for this reason, the usage areas of Rare Earth Elements (REEs) are increasing day by day. Increasing concentrations of REEs, as a result of increased use, create pollution in the environment and harm living organisms. This pollution interacts with increasing temperature and causes more negative synergistic effects of the pollutant in the environment and in the living body. In this study, sublethal concentration values were determined by literature review and the concentration value was determined as 125 mg/L. In the present study tt was aimed to investigate some oxidative stress and antioxidant responses of Terbium, Lanthanum, Gadolinium and Praseodymium REEs in Dreissena polymorpha at 125 mg/L concentration at 3 different temperatures (16, 18, 20 0C) with biomarkers. For this purpose, 24 and 96 hour experimental trial design was created and 7 D. polymorpha were used in each trial group, and the application experiments were carried out with 3 replications. The samples at the end of the experimental phase were stored at -80 degrees Celsius until they were analyzed. In this study, Superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) enzyme activities and glutathione (GSH) and Thiobarbituric acid (TBARS) level biomarker responses were determined by ELISA test microplate reader. CAYMAN brand SOD (Catalog No 706002), CAT Catalog No 707002) and GPx (Catalog No 703102), GSH (Catalog No 703002) and TBARS (Catalog No 10009055) were used in the study. SPSS 24.0 package program one-way ANOVA (Duncan 0.05) was used for the evaluation of biochemical analyzes. According to the study data, statistically significant decreases were observed in SOD and CAT activities in the oxidative stress responses of REEs on D. Polymorpha with increasing temperature, while there was no significant change in GPx activities. It was determined that there were increases in TBARS levels and decreases in GSH levels. It is thought that the temperature factor, application concentration and application time are effective in the formation of these changes. It can be said that temperature change and pollutants cause oxidative stress in organisms and cause cell damage.

Pala, Aysegul; Ince, Muharrem; Serdar, Osman
The aim of this study is to test freshwater gammarids as ecotoxicological biomarkers. Gammarids were collected from fresh water and adapted to the laboratory environment. Then, the influence of various variables was investigated. For monitoring and detecting the bioaccumulation level in Gammarus pulex exposed to controlled copper ions and to evaluate potential human and ecological risks, the use of G. pulex as sentinel indicators of water quality is one rational approach. Copper bioaccumulation rate and fate are currently not sufficiently studied. For optimization, the experimental conditions to ensure bioaccumulation level and to find the effects of various variables on proses, a Box–Behnken experimental design was performed. Used mentioned model with statistical underpinnings for Cu2+ ions bioaccumulation have been analyzed, tested and verified and the viability of G. pulex as a biomonitor. The variables’ optimum levels (temperature; 18.1, pH; 4.3, initial concentration of Cu2+ ions, mg L−1; 11.2 and exposure time, h; 42.2) were achieved by employing the quadratic regression model. Critical role of G. pulex in freshwater biomonitoring and their potential to be evaluated as biomarkers for Cu2+ ions was confirmed. It was also explored to see if G. pulex may serve as a potential tool for estimating aquatic quality.

- MakaleAçık Erişim2025Scopus SJRQ1Unlocking the potential of nutmeg oil: A sustainable alternative for rainbow trout anesthesia in aquaculture
Minaz, Mert; Er, Akif; Ak, Kübra; Serdar, Osman
In aquaculture, interest in natural anesthetic agents as alternatives to synthetic anesthetics has been steadily increasing. This is due to concerns over residue accumulation, environmental impact, and growing demand for sustainable practices. Within this context, the present study focused on the anesthetic effects of nutmeg essential oil at three different concentrations in rainbow trout (Oncorhynchus mykiss, 94.42 ± 23.9 g): low (400 µL/L), medium (600 µL/L), and high (800 µL/L). Induction and recovery times were recorded, and samples were collected at 0, 2, 4, and 8 hours post-anesthesia for antioxidant, cortisol, and hematological analyses. Additionally, gill tissues were sampled immediately after anesthesia for histological examination. While the induction time for the low concentration exceeded 3 minutes, the induction and recovery times for the other concentrations ranged within acceptable limits (3 min for induction and 5 min for recovery). Blood parameters exhibited an increasing trend until the 4th hour post-anesthesia, peaking at the 2nd hour, before returning to control levels by the 4th hour. Histological evaluation revealed healthy gill tissues in the control and low-concentration group (400 µL/L), whereas hyperplasia was observed in the medium (600 µL/L) and high-concentration (800 µL/L) groups. Antioxidant enzyme activity (superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), and glutathione peroxidase (GSH-Px)) indicated potential oxidative stress at the high anesthetic concentration. Similarly, cortisol levels were higher at the high concentration, with plasma cortisol showing a slight initial increase followed by a return to normal levels over time. In conclusion, this study highlights that nutmeg oil at a concentration of 600 µL/L may serve as a promising natural alternative to synthetic anesthetics in aquaculture. However, future studies should include a synthetic anesthetic as a control to allow for direct comparisons and a more comprehensive evaluation of its efficacy and physiological effects.

The biochemical reactions of zebra mussels Dreissena polymorpha (Pallas, 1771) exposed to different concentrations of cadmium (Cd) in the acute period (24 and 96 hours after treatment) were studied. Four variants of experimental treatment were examined: 0 μg/L Cd (control), 10 μg/L Cd, 20 μg/L Cd and 40 μg/L Cd. Changes in the enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione (GSH) and thiobarbituric acid reactive substances (TBARS) levels were determined using ELISA kits. In D. polymorpha exposed to the three experimental concentrations of cadmium, there was a statistically significant decrease (p<0.05) in SOD enzyme activity with increasing concentration or exposure times. No change was observed in CAT enzyme activity with increasing concentrations. The decrease in the activity depending on the exposure times was not statistically significant (p>0.05). As for GPx enzyme activity, a significant increase was detected after Cd application compared to the control (p<0.05). TBARS and GSH levels increased significantly in all Cd application groups compared to the control at 24 and 96 hours. The effects of different concentrations of Cd on D. polymorpha in the acute period were associated with changes in SOD, CAT, GPx enzyme activities. TBARS and GSH levels were effective parameters in determining the oxidative damage caused by Cd.

Materials with dimensions between 0.1 and 100 nm are called nanoparticle (NP) materials. In recent years, the usage areas and quantities of NP materials have increased in parallel with the development of the industry. The need and usage areas of heavy metals such as Cu have also expanded in NP sizes. All these developments have led to problems on the ecosystem that are becoming more difficult to compensate. In this study, Zebra Mussel (Dreissena polymorpha) was chosen as a model to investigate the effect of probiotics on CuNP heavy metal accumulation. The model organism was exposed to three different concentrations of CuNP (5, 10, 50 mg/L) with probiotics and directly for 24 and 96 hours. CuNP accumulation amounts in D. polymorpha tissues treated directly and with probiotics were compared. The amount of accumulation in the test organism directly exposed to CuNP was higher compared to the groups administered with probiotics, but a statistically significant difference (p<0.05) was found only in the treatment group with the highest 24-hour concentration (50 mg/L). As a result, according to the findings obtained from the study, it has been determined that probiotics have positive developmental effects on aquatic organisms, as well as beneficial in the elimination of their accumulation in the organism.

For this reason, in this study, it was aimed to reveal the effect of the temperature variable on the toxic response using the Dreissena polymorpha model organism and some of its biomarkers. For this purpose, acetylcholinesterase (AChE), catalase (CAT), superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA) levels in Dreissena polymorpha exposed to subletal concentrations of λ-cyhalothrin at different temperatures were measured using commercial ELISA kits. According to the results obtained, there was a statistically significant increase in MDA levels in the groups exposed to λ-cyhalothrin, while a decrease in GSH levels was found. AChE levels were inhibited especially in the groups exposed high concentrartion of λcyhalothrin. It was also found that the inhibition levels increased depending on the application times. While SOD enzyme activity decreased, CAT enzyme activity increased depending on the exposure concentration. It has been observed that different temperature have different effects on the toxicity of λ-cyhalothrin. It was observed that λ-cyhalothrin caused oxidative stress and neurotoxicity, and the toxicity of λ-cyhalothrin changed depending on the temperature.
