Unal, Ilkay; Egri, Sinan
This study describes an eco-friendly procedure to synthesize silver nanoparticles (AgNPs) using fresh and dried stem extract of Rheum ribes. AgNPs formation was confirmed using UV-vis spectrometry. The effects of parameters such as temperature, reaction time, pH, AgNO3 concentration, and plant extract/AgNO3 ratio on NP formation were evaluated. Morphology and structural features of the particles were characterized using SEM, TEM, XRD, and FTIR. The average size, polydispersity index, and zeta potential were determined using DLS. After characterization, among the two AgNP samples, selected one; AgNP sample synthesized by dried Rheum ribes stem extracts (d-AgNPs), which exhibited narrow size distribution and high surface plasmon resonance in the range of 435-445 nm for toxicity assessment. The cytotoxic effect of d-AgNPs on HUVECs cells was evaluated with MTT analysis. Accumulation measurements were made in three periods as 24, 48, and 72 h and elimination values were examined at +24 h using ICP-MS analysis

Ünal, İlkay; Ertürk Gürkan, Selin
Evaluation of the effects of nanomaterials in various habitats and living groups is increasing with in vitro and in vivo studies. This study aims to describe a biological procedure to synthesize silver nanoparticles using an extract of Marrubium astracanicum and to determine the aquatic environment’s oxidative stress on Artemia salina. Silver nanoparticle formation was visually observed with color change. Its structural properties, such as size, shape, morphology, and stability, were characterized by UV-Vis spectrophotometer, TEM, SEM, XRD, and DLS analyses. TEM image showed that the synthesized AgNPs have a size of between 20 and 50 nm. The effects of biosynthesized silver nanoparticles at different concentrations (control,0.2, 1, 5, 10, 25, and 50 mg/mL) and exposure times (24, 48 and 72 h) on total glutathione (GSH) and lipid peroxidation levels (MDA) were investigated on the Artemia salina organism, which is one of the most common saltwater organisms frequently used in ecotoxicity tests.

This study aims to describe a simple and environmentally friendly procedure for producing silver nanoparticles (AgNPs) using Paeonia kesrouanensis (P. kesrouanensis) extracts and to determine the toxic effect in the aquatic environment. The morphologies, size, size distributions, and structural properties were analyzed using SEM-EDX, TEM, DLS, zeta potential, FTIR, and XRD. AgNPs were applied to Artemia salina (A.salina), aquatic organism individuals at 7 different concentrations (0.0, 0.2, 1, 5, 10, 25, 50 mg/L) for 24, 48, and 72 h. AgNPs accumulation and elimination, ion release amounts, and the survival rates of organisms were determined at periods of 24, 48, and 72nd hours. The highest accumulation was observed at the 24th hour at the 50 mg/L exposure level. The survival rate decreased as exposure time increased at all concentrations.

- MakaleYalnız künye2020Scopus SJRQ1Comparative effects of Cu (60–80 nm) and CuO (40 nm) nanoparticles in Artemia salina: Accumulation, elimination and oxidative stress
Cimen, Isil Canan Cicek; Danabas, Durali; Ates, Mehmet
In this study, nanotoxicity tests were made by exposure of Artemia salina to copper (Cu 60–80 nm) and copper oxide (CuO 40 nm) nanoparticles (NPs) at different concentrations (0.2, 1, 5, 10, 25, and 50 mg/l) during some exposure duration. Characterization of Cu and CuO NPs were performed using Transmission Electron Microscope (TEM), Dynamic Light Scattering (DLS), Zeta Potential, X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Fourier Transformation Infrared (FT-IR) analyzes. In organisms, the accumulation and elimination rates of NPs was determined by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) analysis and the oxidative stress effects on A. salina were determined by Glutathione (GSH) and Thiobarbituric acid reactive substances (TBARS) analysis methods. Both NPs were found to differ in accumulation and elimination rates at each application time and in parallel with the increase in concentration. In each group, it was determined that ion release increased with application time. The results showed that the accumulation rates in Cu NPs had a tendency to increase at 48 h and to decrease at 72 h in concentrations of 0, 2 and 1 mg/l, respectively. And in the all other concentrations have been seen an increasing trend within the time. In the CuO NPs (40 nm), accumulation rates were a decrease trend at 48th hour and an increase trend at 72nd hour except 10 mg/l concentration. In the 10 mg/l group was an increase trend with the application period. Cu NP, TBARS value increased with increasing concentrations and the highest increase was observed at 24 h of 5 ppm group. The groups showed a tendency to increase-decrease-increase-decrease in TBARS levels in terms of elapsed time (24th, 48th, 72nd hour and elimination) (5 ppm and 10 ppm groups tended to decrease in TBARS level at 72nd hour). TBARS increased with increasing concentration ratios in CuO NPs (40 nm).

- MakaleYalnız künye2020Scopus SJRQ1Effects of Zn and ZnO Nanoparticles on Artemia salina and Daphnia magna Organisms: Toxicity, Accumulation and Elimination
Danabas, Durali; Ates, Mehmet; Ertit Tastan, Burcu; Cicek Cimen, Isil Canan; Unal, Ilkay; Aksu, Onder; Kutlu, Banu
In the study, Zn in the size of 40–60 nm and 80–100 nm and ZnO in the size of 10–30 nm were applied to A. salina and D. magna individuals in 7 groups with 3 repetitions. Measurements were made at 24th, 48th and 72nd hours and elimination values were examined at +24 h. LC50 values of NPs were determined and chemical analysis (metal accumulation and elimination), ion quantities which were given to the environment and the survival rates of organisms were determined after the exposure. According to the results of phase contrast microscopy, it was found that both experimental organisms absorbed the NPs in the medium level. In the toxicity results of D. magna, it can be said that Zn NP (40–60 nm) has a highly toxic effect only at 50 ppm concentration for 48 h and lethal dose can be accepted as of 5 ppm at the end of 72 h. In A. salina individuals, it is clearly seen that there is an increase in mortality in organisms parallel to the dose increase. Although all NPs were applied to organisms in low doses corresponding to environmental values, it was observed that toxic effect was in parallel with the increase in time. It is clearly known that there is the inverse proportion between the size of NPs and the toxic effect. The smaller the size of NPs is, the higher the toxic effect becomes When the results of Zn accumulation and elimination of A. salina and D. magna individuals exposed to the Zn and ZnO NPs were examined; it was found that accumulation and elimination occurred in parallel with the increase in concentration at each application hour and elimination. Intensive and possible misuse of nanoscale materials is one of the biggest threats to the environment and all living things worldwide.

- MakaleAçık Erişim2016Scopus SJRQ2Toxicity of Engineered Nickel Oxide and Cobalt Oxide Nanoparticles to Artemia salina in Seawater
Ates, Mehmet; Demir, Veysel; Arslan, Zikri; Camas, Mustafa; Celik, Fatih
In this study, the effects of exposure to engineered nickel oxide (NiO 40-60 nm) and cobalt oxide (CoO < 100 nm) nanoparticles (NP) were investigated on Artemia salina. Aggregation and stability of the aqueous NP suspensions were characterized by DLS and TEM. Acute exposure was conducted on nauplii (larvae) in seawater in a concentration range from 0.2 to 50 mg/L NPs for 24 h (short term) and 96 h (long term). The hydrodynamic diameters of NiO and CoO NPs in exposure medium were larger than those estimated by TEM. Accumulation rate of NiO NPs were found to be four times higher than that of CoO NPs under the same experimental conditions. Examinations under phase contrast microscope showed that the nanoparticles accumulated in the intestine of Artemia, which increased with increasing exposure concentration. Differences were observed in the extent of dissolution of the NPs in the seawater. The CoO NPs dissolved significantly while NiO NPs were relatively more stable. Oxidative stress induced by the NP suspensions was measured by malondialdehyde assay. Suspensions of NiO NPs caused higher oxidative stress on nauplii than those of CoO NPs. The results imply that CoO and NiO NPs exhibit toxicity on Artemia (e. g., zooplankton) that is an important source of food in aquatic food chain.
