- MakaleAçık Erişim2023Investigation of the Effects of Ghrelin and Kisspeptin Levels in Liver Tissue of Rats Fed with High Fructose Diet -A Histological Study
Özay, Yusuf; Güler, Osman; Türk, Ahmet; Karadag, Abdullah; Zencirci, Büşra
Purpose: High fructose corn syrup (HFCS) is used commonly in the food industry and has been associated with various diseases including obesity, diabetes and fatty liver. Ghrelin, an orexigenic hormone with widespread receptors in many tissues, exerts various central and peripheral effects. Food intake may reduce its synthesis. Kisspeptin plays a major role in reproductive hormones and its receptors are expressed in tissues with high metabolic activity, such as the liver. This study aims to investigate the effects of HFCS consumption on liver tissue and its relationship with Kisspeptin and Ghrelin levels. Materials and Methods: Fourteen adult male Wistar albino rats, aged eight to ten weeks, were used in this study and divided into two groups; Control and HFCS (n=7). Tumor Necrosis Factor-alpha (TNF-α), Ghrelin, and Kisspeptin levels in the liver tissues of the rats were measured using an immunohistochemical method and subsequently analyzed through histoscoring. Results: Our data revealed a significant increase in Kisspeptin, Caspase 3, and TNF-α levels and a decrease in Ghrelin levels in the HFCS group compared to the Control group. Conclusion: In conclusion, our study demonstrates, for the first time, changes in liver Kisspeptin levels induced by a high fructose diet. Furthermore, the consumption of HFCS in food led to inflammatory activation, tissue damage, and reduced Ghrelin levels.

The objective of this study was to investigate the effects of taurine on PPAR-γ, IRS-1, HSP-27, and HSP-72 levels along with the kidney histopathological changes in streptozotocin (STZ) induced diabetic rats. In the study, 40 Wistar albino male rats were divided into four groups. (i) Control group, (ii) Taurine, rats received taurine with drinking water (2%); (iii) STZ, rats were injected with STZ (60 mg/kg i.p. as single dose) and (iv) STZ+Taurine, rats received taurine and injected with STZ. After 8 weeks of the study, the Taurine group was found to be statistically indifferent to the control group in terms of PPAR-γ, IRS-1, HSP-27, and HSP-72 levels (P>0.05). However, PPAR-γ (P<0.01), IRS-1 and HSP-72 (P<0.001) levels decreased in the STZ group. HSP-27 (P<0.01) levels increased in the STZ group as compared with the control. On the other hand, there were histopathological changes in the STZ group. The intensity of histopathological changes induced by STZ was decreased in the STZ+Taurine group. As a result, the taurine supplementation may prevent and treat the kidney damage in diabetic rats

Objective: The aim of the study is to assess the health management in women with gestational diyabetes mellitus. Method: Designed as descriptive and crosssectional, the study was performed in the pregnancy clinic of a university hospital between 1st January and 31st August 2010. All patients applying to the clinic during the study with gestational diyabetes via oral glucose tolerance testing (n=149) constituted the universe of the study. Not using insulin, pregnant were included in the study that participating in the educational program at least for two months between the weeks 24 and 38 during their pregnancies. Totally 120 individuals were recruited, 60 as insulin-diet-exercise health management program, and 60 in only diet-exercise health management program. Findings: The mean age of the women were 32,8±5,1 and 47,5% were primary school graduates. Of the women, 80.8% were housewives. It was determined that the mean HbA1c (p=0,016) and postprandial blood glucose levels (p=0,034) of women in insulin-diet-exercise health management program were statistically and significantly lower and controlled than those in onlydiet-exercise health management program. The mean fasting blood glucose level was lower in those using insulin, but this score was statistically insignificant (p=0,131). The comparison of the two groups revealed that the rate of compliance with the diet (p=0,003) and exercise (p=0.269) programs prescribed was lower in those using insulin. Conclusion: Forming life style changes by involving women into health management programs effectively plays a key role in decreasing the negative effects of gestational diyabetes mellitus to the lowest levels for both the mothers and the newborns.
