The effects of biosynthesized silver nanoparticle on oxidative stress parameters in model organism Artemia salina (Linnaeus, 1758)

    Ünal, İlkay; Ertürk Gürkan, Selin

Evaluation of the effects of nanomaterials in various habitats and living groups is increasing with in vitro and in vivo studies. This study aims to describe a biological procedure to synthesize silver nanoparticles using an extract of Marrubium astracanicum and to determine the aquatic environment’s oxidative stress on Artemia salina. Silver nanoparticle formation was visually observed with color change. Its structural properties, such as size, shape, morphology, and stability, were characterized by UV-Vis spectrophotometer, TEM, SEM, XRD, and DLS analyses. TEM image showed that the synthesized AgNPs have a size of between 20 and 50 nm. The effects of biosynthesized silver nanoparticles at different concentrations (control,0.2, 1, 5, 10, 25, and 50 mg/mL) and exposure times (24, 48 and 72 h) on total glutathione (GSH) and lipid peroxidation levels (MDA) were investigated on the Artemia salina organism, which is one of the most common saltwater organisms frequently used in ecotoxicity tests.

Effect of type and concentration of antioxidant on sperm motility, viability, and DNA integrity of climbing perch Anabas testudineus Bloch, 1792 (Pisces: Anabantidae) post-cryopreservation

Sperm quality is preserved through the crucial involvement of antioxidants, which play a vital role in minimizing the occurrence of reactive oxygen species (ROS) during the cryopreservation process. The suitability of the type and concentration of antioxidants are species-dependent, and this study is crucial in order to improve the quality of the climbing perch sperm post-cryopreservation. Therefore, this study aimed to determine the best type and concentration of antioxidants for cryopreservation of climbing perch Anabas testudineus sperm. To achieve this, 6 types of antioxidants, namely, ascorbic acid, beta-carotene, glutathione, butylated hydroxytoluene (BHT), myo-inositol, and alpha-tocopherol, with inclusion of a control were tested in 3 replications at three concentration levels of 0 mg/L (control), 20 mg/L, 40 mg/L, and 60 mg/L. Sperm was diluted in a glucose-base extender at a ratio of 1:60 (sperm: glucose base), then 10 % DMSO and 5 % egg yolk was added before cryopreservation for two weeks. The results showed that the type and concentration of antioxidants had a significant effect on the motility and viability of cryopreserved climbing perch sperm (P < 0.05), where the best results for ascorbic acid, beta-carotene, glutathione, myo-inositol, and alpha-tocopherol were obtained at a concentration of 60 mg/L, while BHT was at a concentration of 20 mg/L. The best results for glutathione, myo-inositol, and alpha-tocopherol were significantly different from other treatments, while the best results for ascorbic acid and beta-carotene (60 mg/L) were not significantly different from the 40 mg/L concentration, while the best results for BHT were not significantly different from the control treatments. Therefore, the best concentration of glutathione, myo-inositol, and alpha-tocopherol was 60 mg/L, while for ascorbic acid and beta-carotene it was 40 mg/L, and BHT was not recommended. DNA integrity analysis indicated the absence of fragmentation in all samples, including fresh, control, and treated sperm. Based on practical and economic considerations, myo-inositol at 60 mg/L was recommended for cryopreservation of climbing perch A. testudineus sperm.

THE ROLE OF GLUTATHIONE IN THE DETOXIFICATION OF PERCHLOROETHYLENE

    Sahilli, Yeliz Cakir

Environmental factors have important roles in the development of cancer. Especially, the exposure of chemicals existing in the workplace with people is a very important issue. People at such places are exposed to chemicals more especially via respiratory and dermal route. Thus, their health is under the risk. Dry cleaning centers are intensively subjected to PCE, which is a common chemical used at these work places, and the risk of cancer is very high for them. The main aim of the present study is to investigate, if PCE exposure results in glutathione efflux from human erythrocytes. In the present study, the changes in intracellular and extracellular non-protein sulfhydryl (NPSH) levels were determined in PCE exposed erythrocytes. PCE significantly depleted intracellular NPSH level in a time and concentration-dependent manner. PCE significantly increased the NPSH levels measured in the extracellular media. Along with the change in extracellular nonprotein thiol levels, significant increases in extracellular glutathione levels were detected. Erythrocyte membranes were not damaged over the course of PCE exposure as indicated by the absence of lactate dehydrogenase activity in the extracellular media. Utilization of MK571 an MRP inhibitor decreased the rate of glutathione efflux from erythrocytes suggesting a role for this membrane transporter in the process. The results of the present study indicate that erythrocytes efflux glutathione when exposed to PCE. In addition to glutathione oxidation, glutathione efflux also contributes to intracellular loss of glutathione when exposed PCE.

ANTIMONY INDUCED GLUTATHIONE EFFLUX FROM HUMAN ERYTHROCTES

Sahilli, Yeliz Cakir; Akgun, Mehmet Hanifi; Yildiz, Deniz

Antimony is a trace element which has wide variety of industrial application. Antimony has recently been considered as an environmental pollutant. Among the inorganic forms, SbIII is the more toxic form. SbIII is ten times more toxic compared to the SbV. Consuming plants that are grown in antimony contaminated soil is one of the way by which humans are exposed to antimony. AsIII and SbIII forms strong bonds with free-SH groups. When SbIII or SbV interacts with proteins that contain free-SH groups, protein inactivation usually results in. Antimony may also result in free radical generation and induction of oxidative stress. Even though antimony has some applications in health as a drug, its biological effects have not been investigated in detail. In the present study we investigated the effects of SbIII on glutathione efflux in human erythrocytes. The effects of N-acetyl-L-cysteine on antimony induced glutathione oxidation and glutathione efflux were also investigated. Extracellular glutathione concentration significantly increased and reached to 0.033 +/- 0.0034 micromole/ml erythrocyte with 1 mM of potassium antimony tartrate in four hours. The control value of extracellular glutathione was 0.020 +/- 0.0035 micromole/ml erythrocyte. Our results show that SbIII treatment of erythrocyte results in glutathione efflux.
