Ahmed, Shakeel; Zengin, Gokhan; Fernández-Ochoa, Álvaro; de la Luz Cádiz-Gurrea, Maria; Leyva-Jiménez, Francisco Javier; Elkiran, Omer; Cakilcioglu, Ugur; Akgul, Bengusu H.; Pereira, Catarina G.; Custódio, Luísa

The current study investigated the chemical composition, antioxidant activity, enzyme inhibition, and cytotoxic activities of extracts from Achillea maritima, a wild medicinal plant used for various therapeutic purposes. The antioxidant activities were assayed through different assays like DPPH, ABTS, CUPRAC, FRAP, and phosphomolybdenum, whereas in enzyme inhibition studies, cholinesterase, tyrosinase, α-amylase, and α-glucosidase were assayed. Cytotoxicity studies are conducted on S17, RAW, and HepG2 to assess its selectivity and effectiveness. Chemical profiling by UHPLC-ESI-QTOF-MS revealed multiple bioactive compounds in the extracts. Polar solvents (ethanol, ethanol/water, and water) resulted in high concentrations of phenolic acids as well as chlorogenic and caffeoylquinic acids, as well as flavonoids like vicenin and apigenin. On the other, the nonpolar (hexane extract) was rich in octadecatrienoic acid hydroperoxy and hydroxyoctadecatrienic acid. Among these, the water extract contained the highest phenolic content of 32.26 mg GAE/g, while the ethyl acetate extract was the richest in flavonoids, with 7.83 mg RE/g. In the antioxidant studies, the water and ethanol/water extracts consistently display the most potent activities, thus indicating their significant free radical scavenging and metal chelation abilities. The studies on enzyme inhibitions showed remarkable BChE inhibitory activities of the ethanol extract in 12.50 mg GALAE/g, thus showing potential in managing disease conditions related to cholinesterase. Tyrosinase inhibition was significant by the ethanol extract, presenting 55.59 mg KAE/g. The ethyl acetate extract exhibited the most potent inhibitory activity against α-amylase with 0.66 mmol ACAE/g, while ethanol extract showed significant inhibition of α-glucosidase with 4.35 ACAE/g. Cytotoxicity results showed that the water extract was most effective against the HepG2 cancer cell line by reducing cell viability to 38.4% at high doses while preserving low toxicity against normal cells, as observed by high viability percentages in S17 and RAW cell lines. These results highlight the usefulness of A. maritima extracts in nutraceutical, pharmaceutical, and cosmeceutical applications.

Phlomis pungens var. hirta Velen has been used by local people for food and to treat diseases but there are limited number of investigations in literature about this plant and cyto/genotoxicity analyses have not been done before. Hence, this study aimed to find out cell-protective activity and DNA damage prevention ability of P. pungens var. hirta by MTS, micronucleus, 8-OHdG and somatic mutation recombination tests (SMART). In addition, liquid chromatography, antioxidant/antibacterial activity analyses and in silico biological activity profiling were performed. The healthy lymphocyte cell growth was supported by the plant extracts and none of the extracts was genotoxic. 8-OHdG test showed that methanolic and ethanolic extracts of the plant were able to decrease the oxidative DNA damage. According to the SMART results, extracts could suppress the genotoxic effects of an important environmental pollutant, H2O2. Rutin hydrate and quercetin were found in highest concentrations and the extracts could be characterized as antioxidant/antibacterial. In silico analysis also showed that the major compounds of this plant can be considered as chemoprotective. This is the first study that assessed the cell protective activity and DNA damage prevention capacity of P. pungens var. hirta, which should be regarded as a potential source for nutritional supplements and therapeutic products.

Tanacetum nitens (Boiss. & Noë) Grierson is an aromatic perennial herb used in Turkish traditional medicine to treat headache, fever, and skin diseases. This study aimed to investigate the chemical composition, antioxidant, enzyme inhibition, and cytotoxic properties of T. nitens aerial parts. Organic solvent extracts were prepared by sequential maceration in hexane, dichloromethane, ethyl acetate, and methanol while aqueous extracts were obtained by maceration or infusion. Nuclear magnetic resonance (NMR) and LC-DAD-MS analysis allowed the identification and quantification of different phytoconstituents including parthenolide, tanacetol B, tatridin B, quinic acid derivatives, β-sitosterol, and glycoside derivatives of quercetin and luteolin. The type and amount of these phytochemicals recovered by each solvent were variable and significant enough to impact the biological activities of the plant. Methanolic and aqueous extracts displayed the highest scavenging and ions-reducing properties while the dichloromethane and ethyl acetate extracts exerted the best total antioxidant activity and metal chelating power. Results of enzyme inhibition activity showed that the hexane, ethyl acetate, and dichloromethane extracts had comparable anti-acetylcholinesterase activity and the latter extract revealed the highest anti-butyrylcholinesterase activity. The best α-amylase and α-glucosidase inhibition activities were obtained from the hexane extract. The dichloromethane and ethyl acetate extracts exhibited the highest cytotoxic effect against the prostate carcinoma DU-145 cells. In conclusion, these findings indicated that T. nitens can be a promising source of biomolecules with potential therapeutic applications.

The present study was performed to determine the chemical constituents, cytotoxicity, antioxidant and enzyme inhibition activities of the aerial parts of Glaucium acutidentatum Hausskn. and Bornm. (family Papaveraceae). Methanolic and aqueous extracts were prepared by maceration, homogenizer-assisted extraction (HAE) and infusion. Results showed that the highest total phenolic and flavonoids contents were obtained from the methanol extracts obtained by HAE (53.22 ± 0.10 mg GAE/g) and maceration (30.28 ± 0.51 mg RE/g), respectively. The aporphine, beznyltetrahydroisoquinoline, and protopine types of Glaucium alkaloids have been tentatively identified. Among them, glaucine was identified in all extracts. Flavonoids, phenolic acids, coumarins, organic acids and fatty acids were also detected. Methanolic extract obtained using the HAE method displayed the highest anti-DPPH (41.42 ± 0.62 mg TE/g), total antioxidant (1.20 ± 0.17 mmol TE/g), Cu2+ (113.55 ± 6.44 mg TE/g), and Fe3+ (74.52 ± 4.74 mg TE/g) reducing properties. The aqueous extracts obtained by infusion and HAE methods exerted the best anti-ABTS (103.59 ± 1.49 mg TE/g) and chelating (19.81 ± 0.05 mg EDTAE/g) activities, respectively. Methanolic extract from HAE recorded the highest acetylcholinesterase (2.55 ± 0.10 mg GALAE/g) and α-amylase (0.51 ± 0.02 mmol ACAE/g) inhibition activities, while that obtained by maceration showed the best butyrylcholinesterase (3.76 ± 0.31 mg GALAE/g) inhibition activity. Both extracts revealed the best tyrosinase inhibitory activity (25.15 ± 1.00 and 26.79 ± 2.36 mg KAE/g, p ≥ 0.05). G. acutidentatum maceration-derived aqueous extract showed selective anticancer activity against cells originating from human hypopharyngeal carcinoma. In conclusion, these findings indicated that G. acutidentatum is a promising source of alkaloids and phenolic compounds for variable pharmaceutical formulations.

Gürler, Nedim; Paşa, Salih; Erdoğan, Ömer; Cevik, Ozge

The current study focuses on the fabrication, characterization, and cytotoxic effects of polymeric biocomposite films composed of starch, various concentrations of citric acid (3%, 6%, and 9%), and polyvinyl alcohol. The cross-linked corn starch (S)-polyvinyl alcohol (P) biocomposite films are prepared by casting technique using glucose (G) and fructose (F) as plasticizers. The chemical structure of obtained films is characterized by Fourier transform infrared and x-ray diffraction. Physicochemical parameters of the films such as solubility, swelling capacity, and water vapor permeability, optical and mechanical properties are investigated. Furthermore, the cytotoxicity is evaluated to determine the biocompatibility of samples. The physicochemical properties of S-P(Starch-Polyvinyl alcohol) films improve with the addition of citric acid during the esterification reaction. The tensile strength for Starch-Polyvinyl alcohol-Glucose blend composition with 3% citric acid (SPG3) and Starch-Polyvinyl alcohol-Fructose blend composition with 3% citric acid (SFG3) biocomposite films is determined as 10.17 and 8.24 MPa, respectively, compared to 8.16 and 7.94 MPa for without citric acid, respectively. The cell viability values of biocomposite films towards NIH-3T3 and L929 cells are calculated 90% compared to the control group. Thus, it demonstrates that they are also quite biocompatible materials.

Cytoprotective, antimutagenic/antirecombinogenic and antibacterial properties of Lallemantia iberica extracts

Yilmaz Kardas, Begumhan; Diken, Mehmet Emin; Bayhan, Hamza; Acar, Mikail; Dogan, Serap

BACKGROUND: Plants have important chemical compounds to protect organisms from many mutagens but they may also have dangerous toxic substances that must be tested before use. Lallemantia iberica has been used by local people for treatment of some diseases but it was never tested for its cell protective and antimutagenic/antirecombinogenic properties. In this study, it was aimed to determine the cytoprotective, antimutagenic/antirecombinogenic, antibacterial, antioxidant and phenolic profile of L. iberica by MTS assay, micronucleus test/somatic mutation recombination test (SMART), disc diffusion method, ABTS/DPPH tests and liquid chromatography (LC), respectively. RESULTS: MTS assay results showed that both of the extracts supported the normal growth of healthy human lymphocytes and there was no significant difference between extracts. Although there was no significant increase in micronucleus concentration (‰) of the cultures treated with ethanol or methanol extracts, methanol extract caused slightly lower micronucleus concentration (12.64 ± 3.65‰) than the ethanol extract (24.46 ± 4.50‰). SMART results showed that ethanol and methanol extracts prevented harmful mitotic recombination. Lallemantia iberica also showed antibacterial activity against all of the strains tested, and the largest inhibition zones were observed with Pseudomonas aeruginosa (17.9 mm) and Enterobacter aerogenes (13.8 mm). Methanol extracts showed better antioxidant activities (ABTS IC50: 104.07 μg mL−1, DPPH IC50: 58.8 μg mL−1) than ethanol extracts (ABTS IC50: 344.03 μg mL−1, DPPH IC50: 233.7 μg mL−1). According to the LC results, the most abundant phenolic compounds were rutin hydrate (1796.4 ± 62.9 mg kg−1) and p-coumaric acid (228.1 ± 7.60 mg kg−1). CONCLUSION: Lallemantia iberica extracts had cell-protective, antimutagenic/antirecombinogenic, antibacterial and strong antioxidant characteristics in relation to their rich phenolic content.

Synthesis, Characterization and Biocompatibility of Cross-linked Dicarboxylic Acids from Waste Potato Peel Starch

    Paşa, S.; Gürler, N.; Erdoğan, Ö.; Cevik, O.

Abstract: People have become conscious about recycling or reusing many things they use in the 21st century because diminishing vital resources make it necessary, both personally and socially. The evaluation of waste is also critical in terms of national economies. In this study, starch dicarboxylic compounds were synthesized using waste potato peels (WPP) in the presence of sodium hypophosphite monohydrate as a catalyst for the formation of the ester cross-links. Adipic acid, succinic acid, and glutaric acid were employed with waste potato starch (WPS) to obtain cross-linked samples: WPS-A, WPS-S, and WPS-G, respectively. The obtained compounds were characterized by attenuated total reflectance Fourier transform infrared (FTIR-ATR), scanning electron microscopy with energy-dispersive X-ray analysis (SEM-EDX), X-ray diffraction (XRD), and their cytotoxicity properties were investigated. The biocompatibility of the obtained WPS-dicarboxylic acid compounds against NIH-3T3 and L929 murine fibroblast cells with MTT assay was determined. Studies have shown that these compounds do not harm healthy cells and are found to be bioavailable.

Bunium species have been reported to be used both as food and in traditional medicines. The scientific community has attempted to probe into the pharmacological and chemical profiles of this genus. Nonetheless, many species have not been investigated fully to date. In this study, we determined the phenolic components, antimicrobial, antioxidant, and enzyme inhibitory activities of aerial parts of four Bunium species (B. sayai, B. pinnatifolium, B. brachyactis and B. macrocarpum). Results showed that B. microcarpum and B. pinnatifolium were strong antioxidants as evidenced in the DPPH, ABTS, CUPRAC, and FRAP assays. B. brachyactis was the most effective metal chelator, and displayed high enzyme inhibition against cholinesterase, tyrosinase, amylase, glucosidase, and lipase. The four species showed varied antimicrobial activity against each microorganism. Overall, they showed high activity against P. mirabilis and E. coli (MIC and MBC <1 mg mL−1). B. brachyactis was more effective against Aspergillus versicolor compared to the standard drug ketoconazole. B. brachyactis was also more effective than both ketoconazole and bifonazole against Trichoderma viride. B. sayai was more effective than ketoconazole in inhibiting A. fumigatus. B. sayai was most non-toxic to HEK 293 (cellular viability = 117%) and HepG2 (cellular viability = 104%). The highest level of TPC was observed in B. pinnatifolium (35.94 mg GAE g−1) while B. microcarpum possessed the highest TFC (39.21 mg RE g−1). Seventy four compounds were detected in B. microcarpum, 70 in B. brachyactis, 66 in B. sayai, and 51 in B. pinnatifolium. Quinic acid, chlorogenic acid, pantothenic acid, esculin, isoquercitrin, rutin, apigenin, and scopoletin were present in all the four species. This study showed that the four Bunium species are good sources of biologically active compounds with pharmaceutical and nutraceutical potential.

Zengin, Gokhan; Mahomoodally, Mohamad Fawzi; Paksoy, Mehmet Yavuz; Picot-Allain, Carene; Glamocilja, Jasmina; Sokovic, Marina; Diuzheva, Alina; Jekő, József; Cziáky, Zoltán; Rodrigues, Maria João

Several species of the Apiaceae family have been employed in traditional cultures for their curative virtues. The present study focused on five Apiaceae species, (Falcaria vulgaris (FV), Smyrniopsis aucheri (SA), Smyrniopsis munzurdagensis (SM), Smyrnium cordifolium (SC), and Actinolema macrolema (AM)). The antioxidant, enzyme inhibitory (α-amylase, α-glucosidase, acetyl- and butyrylcholinesterase, lipase, and tyrosinase), antimicrobial, phytochemical, and cytotoxicity profiles of the methanol extracts of the selected Apiaceae species were determined. SC extract (35.68 mg gallic acid equivalent/g extract) possessed the highest phenolic content while the AM extract (56.79 mg rutin equivalent/g extract) had the highest flavonoid content. HPLC-ESI-MS (High performance liquid chromatography-electrospray tandem mass spectrometry) analyses showed presence of ferulic acid in all the five species. SC extract exhibited high radical scavenging (59.28 and 94.31 mg Trolox equivalent [TE]/g extract, DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS (2,2′-azino-bis(3-ethylbenzothiazoline)-6-sulfonic acid), respectively) and reducing activity (161.44 and 113.62 mg TE/g extract, for CUPRAC (cupric reducing antioxidant capacity) and FRAP (ferric reducing antioxidant power), respectively). SM extract exhibited the highest cholinesterase's inhibitory action (3.82 and 4.76 mg galantamine equivalent/g extract, for acetyl- and butyrylcholinesterase, respectively). The extracts showed higher inhibition against α-glucosidase (7.32-11.99 mmol acarbose equivalent [ACAE]/g extract) compared to α-amylase (0.51-0.55 mmol ACAE/g extract). SC extract was the most active (137.54 mg kojic acid equivalent/g extract) tyrosinase inhibitor and FV extract (113.75 mg Orlistat equivalent/g) the best lipase inhibitor. SM extract showed potent antibacterial effect against B. cereus (MIC (minimum inhibitory concentration) 0.180 mg/mL), P. mirabilis (MIC 0.180 mg/mL), M. flavus (MIC 0.560 mg/mL), P. aeruginosa (MIC 0.275 mg/mL), and S. typhimurium (MIC 1.500 mg/mL). FV extract (MIC 0.140 mg/mL) suppressed A. fumigatus growth. Cytotoxicity was assessed on murine macrophage (RAW 264.7), human embryonic kidney (HEK 293), and human hepatocellular carcinoma (HepG2) cell lines. FV (60.3%) and SM (57.4%) showed the highest reduction on RAW 264.7 cellular viability, whereas SM (74.1%) showed toxicity against HepG2. This study supports that the Apiaceae species could be considered as promising candidates for the development of novel pharmacophores for the management of several human ailments.

Extracellular Synthesis and Characterization of Gold Nanoparticles Using Mycobacterium sp. BRS2A-AR2 Isolated from the Aerial Roots of the Ghanaian Mangrove Plant, Rhizophora racemosa

Camas, Mustafa; Sazak Camas, Anil; Kyeremeh, Kwaku

Through the use of genomes that have undergone millions of years of evolution, marine Actinobacteria are known to have adapted to rapidly changing environmental pressures. The result is a huge chemical and biological diversity among marine Actinobacteria. It is gradually becoming a known fact that, marine Actinobacteria have the capability to produce nanoparticles which have reasonable sizes and structures with possible applications in biotechnology and pharmacology. Mycobacterium sp. BRS2A-AR2 was isolated from the aerial roots of the mangrove plant Rhizophora racemosa. The Mycobacterium was demonstrated for the first time ever to produce AuNPs with sizes that range between 5 and 55 nm. The highest level absorbance of the biosynthesized AuNPs was typical for actinobacterial strains (2.881 at 545 nm). The polydispersity index was measured as 0.207 in DLS and the zeta potential was negatively charged (− 28.3 mV). Significant vibration stretches were seen at 3314, 2358, 1635 and 667 cm−1 in FT-IR spectra. This demonstrated the possible use of small aliphatic compounds containing –COOH, –OH, –Cl and –NH2 functional groups in the stabilization of the AuNPs. The effect of the biosynthesized AuNPs on HUVEC and HeLA cell lines was measured at 48 h. IC50 values were determined at 3500 µg/ml concentration for HUVEC and HeLA cell lines at 45.25 and 53.41% respectively.
