Cisplatin (CP) is one of the most effective cancer chemotherapeutic agent used against various solid tumors. This study was designed to investigate the antioxidant effects of Grape Seed Extract (GSE) on CP induced oxidative damage in rabbit kidney. The material of study was consist of 18 male New Zealand rabbit. Animals were divided into 3 groups. The first group was administered a single intraperitoneal (i.p.) close of 0.9% saline. The second group of rabbits was treated with CP (a single i.p. dose of 5 mg kg(-1) body weight); The third group of rabbits was treated with GSE by gavage at the dose of 250 mg kg(-1) body weight for 6 consecutive days before and 6 consecutive days after a single i.p. CP injection. As a result of CP administration, kidney function tests; urea, BUN (Blood Urea Nitrogen) and creatinine levels were increased (p<0.001). It was observed that CP increase malondialdehide (MDA) levels, the most important indicator of oxidative damage and decreased the activity of catalasc (CAT). Administration of CP did not caused statically a change in glutathione (GSH) levels and glutathione peroxidase (GSH-P-x) activity (p>0.05). GSE treatment has high levels of MDA to compared to normal levels (p<0.001) but not an increase in CAT (p<0.05). In CP treatment group, severe acute tubular necrosis was seen. It was determined that in CP+GSE group mild degenerative changes were received instead of acute tubular necrosis. GSE has been found to be partially protective effect against CP induced biochemical and histopathological changes in rabbit kidney.

This study examined the effects of orally given grape seed extract (GSE) on healing of surgically induced full thickness skin wounds in terms of clinical, histopathological and biochemical aspects. A total of 48 healthy. adult male New Zealand rabbits were randomly divided in 2 equal experimental and control groups: one week prior the surgery, 260 mg/kg grape seed extract was orally given for 20 days to the assay animals. A circular full thickness skin wound (2.5 cm in diameter) was made in the back midline of all rabbits under anaesthesia then, macroscopic and histopathological aspects as well as the tissue oxidant/antioxidant balance were determined on 6 rabbits from each group slaughtered at the 5th, 10th, 15th and 20th days post-surgery. In treated rabbits, it was clinically observed that the wound contraction was significantly amplified and the histological processes of re-epithelisation, granulation tissue development, collagen accumulation and angiogenesis were markedly hastened whereas the inflammatory reaction and the ulcer disappeared more rapidly than in the not-treated controls. Furthermore, the maximal skin MDA formation observed on the 10th day was significantly depressed and the antioxidant catalase (CAT) and glutathione peroxidase (GSH-Px) activities remained significantly elevated until the 15th day compared to the controls. These results demonstrate the beneficial effects of treatment with GSE on skin wound healing in rabbits.

BENEFICIAL EFFECTS OF GRAPE SEED EXTRACT AGAINST CISPLATIN-INDUCED TESTICULER DAMAGE IN RABBITS

Yildirim, Nuran Cikcikoglu; Kandemir, Fatih Mehmet; Benzer, Fulya

Biological compounds with antioxidant properties may contribute to the protection of cells and tissues against deleterious effects of reactive oxygen species (ROS) and other free radicals induced by cisplatin. This study was performed to investigate the possible protective role of antioxidant treatment with grape seed extract (GSE) on cisplatin-induced testes oxidant injury using biochemical approaches. The degree of protection produced by GSE was evaluated by determining the level of malondialdehyde (MDA) and glutathione (GSH), the activity of catalase (CAT), glutathione peroxidase (GSH-Px), were estimated from testes of rabbits. Male New Zealand white rabbits were divided into 3 groups (n=6): 1-Control group(1 ml saline. i.p) 2-Cisplatin group (a single dose of cisplatin (5 mg/kg, i.p) 3-A single dose of cisplatin (5 mg/kg, i.p) + GSE (250 mg/kg/day, i.p) for 6 days. MDA level were significantly higher when compared to Control (p < 0.05). In Cisplatin+grapes groups MDA levels were found significantly lower than cisplatin group (p < 0.05). GSH levels were decreased with cisplatin compared to control but in case of cisplatin+ GSE it increased (p < 0.05). In treated rabbits, the activity of CAT and GSH-Px was decreased in cisplatin and cisplatin+ GSE groups compared to control (p < 0.001). These results indicate that the antioxidant GSE might have a protective effect against cisplatin-induced testiculer damage and oxidative stress in rabbit.
