Phylogenetic analysis of the genus conringia heist. Ex fabr. (brassicaceae) in turkey based on nuclear (nrits) and chloroplast ((trnl-f)) dna sequences

Sevindik, Emre; Aydogan, Melike; Paksoy, Mehmet Y.

In this study, phylogenetic analysis of Turkish Conringia (Brassicaceae) species was conducted based on nuclear ribosomal DNA (nrITS) and chloroplast DNA (trnL-F) sequences. In addition, the relationships between the sequences of some Brassicaceae family species retrieved from NCBI, and Conringia species were documented. All of the plant specimens were collected at their flowering and vegetation periods from different regions of Turkey, and brought to the laboratory. Total genomic DNA was extracted using the GeneMark kit. In PCR analyses, ITS4 and ITS5A primers were used for the amplification of the nrITS region, while the trnLe and trnLf primers were used for the cpDNA trnL-F region. The DNA sequences obtained were then edited using BioEdit and FinchTV, and analyzed using MEGA 6.0 software. Neighbor joining (NJ) and bootstrap trees were constructed in order to identify the relationships among Conringia taxa. The nrITS sequences ranged between 573 and 672 nucleotides, while the trnL-F sequences ranged between 346 and 764 nucleotides. The divergence values of nrITS sequences differed between 0.177 and 0.00 and divergence values of trnL-F sequences differed between 0.902 and 0.00. NJ tree generated using nrITS and trnL-F sequences consisted of two clades. In trees generated with both the nrITS and trnL-F sequences, C. orientalis, C. grandiflora and C. austriaca appeared within the same group. In addition, according to the phylogenetic analysis results obtained with other Brassicaceae species, it is revealed that the Conringia genus is polyphyletic.

Potassium chloride recovery from mechanically activated microcline through the chlorination roasting and leaching route

Alyosif, Bahaa; Uysal, Turan; Erdemoğlu, Murat

In this study, effects of mechanical activation in the chlorination roasting and water leaching route known as CaCl2 process and developed for the production of potassium chloride (KCl) from potassium feldspar ores were studied. A microcline containing K-feldspar ore with 10.89% K2O was first intensively dry milled by a planetary ball mill and mixed with calcium chloride (CaCl2) and then roasted at temperatures up to 1000 °C to obtain KCl that will be finally dissolved by the water leaching. Potassium recovery by water leaching increased rapidly up to 800 °C. At higher temperatures, the recovery decreased fast due to the evaporation of KCl. According to the K recovery values per unit energy consumed, the optimum roasting temperature was determined as 750 °C and the milling time was 15 min. It was concluded that intensive milling causes mechanical activation of the microcline to reduce the chlorination roasting temperature, which triggers a rise in the K recovery by the water leaching.

Keller box simulation of magnetic pseudoplastic nano-polymer coating flow over a circular cylinder with entropy optimisation

Al-Mdallal, Qasem; Prasad, V. Ramachandra; Basha, H. Thameem; Sarris, Ioannis; Akkurt, Nevzat

In this study, a Keller box calculation of magnetic nanopolymer coating flow over a circular cylinder in the presence of nonlinear convection is performed. A two-phase nanofluid model (Buongiorno model) is used to model the equations for nanofluid flow and heat and mass transfer. The problem at hand is first formulated in the dimensional form of nonlinear partial differential equations (PDEs) and then transformed into the dimensionless PDEs form by manipulating non-similar variables. The Keller box approach (implicit finite differences) is used for the calculations of the transformed PDEs. The characteristics of critical thermophysical physical parameters on the flow field, viz thermal convection, Brownian motion, Weissenberg number, thermophoresis, magnetic field, heat source/sink, Biot number, buoyancy ration parameter, Eckert number, and mixed convection, are graphically manifested. In addition, the flow control parameters estimate the sheath friction, heat transfer rate, and mass transfer rate in the flow coordinate. A significant increase in the velocity of the Williamson nanofluid is accompanied by an increase in the Biot number, magnetic field, and mixed convection. The isotherms show a higher fluid temperature near the wall in the absence of convection. The Weissenberg number, Biot number, and thermophoresis significantly increase the temperature of the Williamson nanofluid. The density of the streamlines is lowered with a larger buoyancy ratio parameter. The total entropy generation of the Williamson nanofluid is improved by increasing the Brinkman number and the Weissenberg number.

Sperm quality and oxidative stress in chub Squalius orientalis and Padanian barbel Barbus plebejus (Teleostei: Cyprinidae) after in vitro exposure to low doses of bisphenol A

Kutluyer, Filiz; Çakir Sahilli, Yeliz; Kocabaş, Mehmet; Aksu, Önder

In an aquatic environment, the presence of Bisphenol A (BPA) adversely affects reproduction, biology, behavior, gonads, and early larval development of fish due to being endocrine-disrupting compound. In addition, the detected concentration of BPA in water bodies is reported to be higher than 0.41 μg/L. As an alternative tool, sperm cells are used in toxicological assays for the reliable and practical assessment. For these reasons, we examined the effects of in vitro exposure of BPA on sperm quality of chub Squalius oriantalis and Padanian barbel Barbus plebejus. Spermatozoa were exposed to lower concentrations (0, 0.5, 1.25, 2.5, and 5 µg/L) of BPA for 2 h. The enzymatic activities [glutathione peroxidase (GSH-Px), catalase (CAT), and superoxide dismutase (SOD)] and lipid peroxidation (MDA) were evaluated in spermatozoa. The results demonstrated that BPA exposure significantly decreased activities of SOD and GSH-Px but increased CAT activity and lipid peroxidation (MDA). Compared to control, the percentage and duration of sperm motility significantly decreased. Overall, spermatozoa clearly showed the sensitivity to lower concentrations of BPA.

Chitosan-graft-poly(N‑tert‑butylacrylamide) Copolymer: Synthesis, Characterization and Optimization of Tetracycline Removal Using RSM

    Tanyol, Mehtap; Torğut, Gülben

In the present study, a graft copolymer was synthesized in accordance with the free radical method using N-tertiary butylacrylamide monomer on chitosan, which is an environmentally friendly polymer. The chemical structure and surface morphology of the newly synthesized graft copolymer were determined by Fourier transform infrared spectroscopy, scanning electron microscopy and X-ray diffraction. Moreover, the thermal behavior was analyzed by thermogravimetric analysis. The chitosan-graft-poly(N‑tert‑butylacrylamide) graft copolymer was used as an adsorbent for the rapid removal of tetracycline from aqueous solutions in order to investigate its applicability. Response surface methodology (RSM) was employed for the optimization of the study conditions, namely adsorbent dosage (0.01–0.04 g 100 mL−1), reaction time (10–90 min) and initial tetracycline concentration (10–100 mg 100 mL−1). The effects of these conditions on tetracycline uptake capacity were examined and the results were statistically analyzed. The RSM results showed that the maximum tetracycline uptake capacity of 104.81 mg g−1 was achieved under the following conditions: adsorbent dose of 0.03 mg 100 mL−1, reaction time of 51.12 min and initial tetracycline concentration of 97.99 mg L−1. ANOVA based on central composite design combined with RSM showed a good agreement between the experimental values and quadratic model estimates, thus resulting in a coefficient (R2) of 0.9118 for tetracycline uptake. The kinetics, isotherms, and thermodynamics of adsorption were examined, and results showed that equilibrium data fitted the Freundlich isotherm model, and the adsorption kinetics of tetracycline followed pseudo-second-order model. Thermodynamic parameters like the enthalpy − 10.39 kJ mol−1 (ΔH°), entropy 0.044 kJ mol−1 K−1 (ΔS°) and Gibbs free energy − 23.26 kJ mol−1 (ΔG°) were evaluated and also, ΔG° shows a negative values indicating that the adsorption process was exothermic and spontaneous. The findings demonstrated that the chitosan-graft-poly(N‑tert‑butylacrylamide) copolymer is an effective adsorbent for pharmaceutical wastewater treatment.

Magnesium picolinate improves bone formation by regulation of rank/rankl/opg and bmp-2/runx2 signaling pathways in high-fat fed rats

Sahin, Emre; Orhan, Cemal; Balci, Tansel Ansal; Erten, Fusun; Sahin, Kazim

Magnesium (Mg) deficiency may affect bone metabolism by increasing osteoclasts, de-creasing osteoblasts, promoting inflammation/oxidative stress, and result in subsequent bone loss. The objective of the present study was to identify the molecular mechanism underlying the bone protective effect of different forms of Mg (inorganic magnesium oxide (MgO) versus organic magnesium picolinate (MgPic) compound) in rats fed with a high-fat diet (HFD). Forty-two Wistar albino male rats were divided into six group (n = 7): (i) control, (ii) MgO, (iii) MgPic, (iv) HFD, (v) HFD + MgO, and (vi) HFD + MgPic. Bone mineral density (BMD) increased in the Mg supplemented groups, especially MgPic, as compared with the HFD group (p < 0.001). As compared with the HFD + MgO group, the HFD + MgPic group had higher bone P (p < 0.05) and Mg levels (p < 0.001). In addition, as compared to MgO, MgPic improved bone formation by increasing the levels of osteo-genetic proteins (COL1A1 (p < 0.001), BMP2 (p < 0.001), Runx2 (p < 0.001), OPG (p < 0.05), and OCN (p < 0.001), IGF-1 (p < 0.001)), while prevented bone resorption by reducing the levels of RANK and RANKL (p < 0.001). In conclusion, the present data showed that the MgPic could increase osteogenic protein levels in bone more effectively than MgO, prevent bone loss, and contribute to bone formation in HFD rats.

The authors regret that the total evapotranspiration shown in Fig. 7 (821 mm) was the measured H2O flux for the whole two year measurement period. The correct average value for the two years was 411 mm yr−1, or 33% of the measured annual average precipitation. This left 827 mm yr−1 as the “estimated runoff” (see corrected figure). The sign of the average NEE values in the Abstract and Conclusions should be negative. All other information is correct, including the equations and interpretations. Control measure prioritization in Fine − Kinney-based risk assessment: a Bayesian BWM-Fuzzy VIKOR combined approach in an oil station

Gul, Muhammet; Yucesan, Melih; Ak, Muhammet Fatih

The Fine − Kinney is a risk assessment method widely used in many industries due to its ease of use and quantitative risk evaluation. As in other methods, it is a method that recommends taking a series of control measures for operational safety. However, it is not always possible to implement control measures based on the determined priorities of the risks. It is considered that determining the priorities of these measures depends on many criteria such as applicability, functionality, performance, and integrity. Therefore, this study has studied the prioritization of control measures in Fine − Kinney-based risk assessment. The criteria affecting the prioritization of control measures are hierarchically structured, and the importance weights of the criteria are determined by the Bayesian Best–Worst Method (BBWM). The priorities of control measures were determined with the fuzzy VlseKriterijumska Optimizacija I Kompromisno Resenje (FVIKOR) method. The proposed model has been applied to the risk assessment process in a petrol station’s liquid fuel tank area. According to the results obtained with BBWM, the most important criterion affecting the prioritization of control measures is the applicability criterion. It has an importance weight of about 42%. It is followed by performance with 31%, functionality with 18%, and integrity with 10%, respectively. FVIKOR results show that the “Periodic control of the ventilation device” measure is the top priority for Fine − Kinney risk assessment. “The absence of any ducts or sewer pits that may cause gas accumulation in the tank area and near the dispenser; Yellow line marking of entry and exit and vehicle roads; Placing of speed limit warning signs” has been determined as a secondary priority. On conclusion, this proposed model is expected to bring a new perspective to the work of occupational health and safety analysts, since the priority suggested by Fine − Kinney risk analysis methods is not always in the same order as the one in the stage of taking action, and the source, budget, and cost/benefit ratio of the measure affect this situation in practice.

This study presents the evaluation of biological activities and chemical profiling of Oenanthe aquatica (L.) Poir. and Oenanthe silaifolia M. Bieb. The phytochemical profile, antioxidant, enzyme inhibitory, cytotoxic and antiviral activities of the methanolic and aqueous extracts were investigated. The aqueous extract of O. aquatica possessing the highest content of phenolics (60.85 mg gallic acid equivalent/g extract), also exhibited the strongest radical scavenging potential against 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (79.46 and 148.66 mg Trolox equivalent/g extract, respectively), the highest reducing ability (207.59 and 107.27 mg Trolox equivalent/g extract, for cupric reducing antioxidant capacity and ferric reducing antioxidant activity, respectively), metal chelating potential (33.91 mg ethylenediaminetetraacetic acid equivalent/g extract) and total antioxidant ability (1.60 mmol Trolox equivalent/g extract). Liquid chromatography-electrospray ionization-quadrupole time-of-flight-mass spectrometry (LC-ESI-QTOF-MS/MS) permitted tentative identification of compounds from simple organic acids, phenolic acids, coumarins, flavonoids and their glycosides in O. aquatica and O. silaifolia extracts. The methanolic extract of O. aquatica substantially depressed acetylcholinesterase (3.67 mg galantamine equivalent/g extract), tyrosinase (126.66 mg kojic acid equivalent/g extract), and α-amylase (0.83 mmol acarbose equivalent/g extract) enzymes. The methanolic extract of O. silaifolia showed highest enzymatic inhibitory property against butyrylcholinesterase, and its aqueous extract depressed α-glucosidase activity (0.26 mmol acarbose equivalent/g extract). All tested extracts exerted selective toxicity towards cancer cell lines, and the highest anticancer potential was found for O. aquatica aqueous extract on FaDu and HeLa cells with CC50 of 57.36 and 47.16 µg/mL, respectively. Significant antiviral activity against HSV-1 (HHV-1) was found for both aqueous extracts in concentrations of 1000 µg/mL, which inhibited the HSV-1 cytopathic effect (CPE) in virus infected VERO cells and reduced the virus infective titer by more than 3 log (logCCID50 /mL). This study has produced critical scientific data on O. aquatica and O. silaifolia, which are potential contenders for the development of novel phyto-pharmaceuticals.

Determination of trace element and fatty acid levels in tissues of mirror and scaly carp (Cyprinus carpio L., 1758) living in Keban Dam Reservoir (Elazig)

Aydın, Sevinç; Akkoyun, H. Turan; Bayramoglu Akkoyun, Mahire; Bengu, Aydın Ş.; Ekin, Suat; Harlioğlu, Ayşe G.

This study investigated the concentrations of some accumulated trace metals (iron, manganese, zinc and copper) and fatty acids in liver and muscle tissues of mirror and scaly carp varieties (Cyprinus carpio L., 1758) living in Keban Dam Reservoir (Elazig), which is the second-largest artificial lake in Turkey that hosts commercially valuable and preferred fish varieties for human consumption. The inductively coupled plasma mass spectrometry (ICP-MS) method was used to analyse the concentrations of trace elements. Fatty acid levels were measured in a gas chromatography mass spectrometry (GC–MS) device, using a flame ionization detector (FID). Considering the two fish varieties and tissues that were studied, the mean metal concentrations (μg g−1) were found in the following ranges: Fe 12.32–49.97, Mn 0.12–0.62, Zn 10.45–86.59 and Cu 2.74–11.86. Although the lowest levels were detected in the muscle tissue for all the metals examined, the levels in the liver tissue were significantly higher. According to the results of the analyses on fatty acid concentrations, the mean total saturated fatty acid levels in the muscle tissue of the scaly carp and mirror carp varieties were found to be 31.07 and 31.43 (%), respectively, whereas these values for the liver tissue were consecutively 22.36 and 22.08 (%). Successively, the mean total levels of unsaturated fatty acids were determined as 68.93 and 68.40 (%) in the muscle tissue and 77.64 and 77.92 in the liver tissue. This study, with the finding that the trace (metallic) element accumulation rates were below the upper limits of the tolerable range, and the examined fish varieties were rich in fatty acids, shows that mirror carp and scaly carp living in Keban Dam Reservoir are highly nutritious for human diets.

Toxicological effects of di-(2-ethylhexyl) phthalate in Gammarus pulex: a biochemical and histopathological assessment

Yildirim, Nuran Cikcikoglu; Ak, Tuba Parlak; Samasas, Omer

This study aimed to reveal the toxic characteristics of di-(2-ethylhexyl) phthalate (DEHP) by examining the biochemical and histopathological changes in Gammarus pulex, exposed to different doses of DEHP. For this purpose, the lethal concentration 50 (LC50) value of the DEHP was determined by using a static test and found to be 0.079 ± 0.01 ppm. Three subletal doses of DEHP were applied to the G. pulex for 24 and 96 h. Superoxide dismutase (SOD), catalase (CAT), cytochrome P450 1A1 (CYP1A1), and glutathione S-transferase (GST) activities were measured using commercial ELISA kits. The caspase method, which is an immunohistochemical analysis method, was used to determine the apoptosis that occurred in the G. pulex. The results showed that the CYP1A1 activities decreased in the groups exposed to different doses of DEHP compared to the control group (p > 0.05). CAT activity was found to increase in the application groups at the 24 and 96 h compared to the control group. In addition, it was found that SOD and GST activities increased at the 96 h compared to the control group. In light of the microscope examination of the model organism, hemolymphatic lacunae filled with hemolymph and reduction or absence of hemolymphatic ducts were observed especially in the G. pulex gills. Collapse of the gills and hyperplasia were observed after 96 h. As a result, it is suggested that changes in SOD, CAT, and GST activities can potentially be used as sensitive biomarkers for risk assessment in the environment and increased immunoreactivity in G. pulex caused by DEHP depending on increased application doses and application times.

Enhanced Adhesion and Corrosion Properties of Boron Doped Bioceramic Coated 316L Implants

    Aksoy, M.E.; Aksakal, B.; Aslan, N.; Dikici, B.

Abstract: In order to provide an effective prevention to 316L implants, a synthesized bioceramic was coated by using electrophoretic deposition (EPD). The effect of boron addition into hydroxyapatite (HA) on surface morphology, adhesion and corrosion resistance of the implant has been examined. Boron doped HA (B–HA) was synthesized by a simple chemical acid-base method with 5, 10, and 15 wt % boron additions to coat on 316L implants. The synthesized B-HA was coated on the substrates and the coated surfaces with and without B additions. Sintering took place at different temperatures and sintering rates. The coatings were characterized by X-ray diffractometer (XRD), Fourier transform infrared spectroscopy (FT–IR) and scanning electron microscope (SEM). Furthermore, adhesion and in vitro corrosion tests were conducted. The influence of boron additons on adhesion and corrosion resistance of 316L have been revealed for the coated substrates at different B additions. It was shown that boron addition increased the adhesion and corrosion resistance of the bioceramic coatings. Furthermore, the sintering effect on adhesion was also evaluated and the highest adhesion as 25.8 MPa has been reached at 750°C for 90 s and 5°C min–1 sintering conditions. It was found that using iodine as EPD dispersant has increased the charging ability of the B–HA powders in the suspension and hence eased deposition rate. The study revealed that boron-containing hydroxyapatite coatings can provide crack-free surfaces that providing high adhesion and hence increase the corrosion resistance of metallic implants. Through the results, it was also shown that the network between powders was improved and hence adhesion was increased with increasing B addition.

Purpose: The study was aimed at investigatting the changes in body image perception, self-esteem, and self-confidence of female-to-male transsexuals (FtM) after sex reassignment surgery (SRS). Design and Methods: The quasi-experimental study was conducted with the female individuals who undergo FtM SRS between May 2018 and May 2019. The study data were collected using the Personal Information Form, Body Image Scale, Coopersmith Self-Esteem Inventory, and Self-Confidence Scale. Findings: It was observed that the patients' postoperative body image perception, self-esteem, and self-confidence improved. As their postoperative body image perception was higher than was their preoperative body image perception, their postoperative self-esteem and self-confidence levels were also higher. In addition, as their self-esteem increased, so did their self-confidence. Practice Implications: It was found that the patients' postoperative body image perception, self-esteem, and self-confidence improved.

Sequential infiltration analysis of infiltration curves measured with disc infiltrometer in layered soils

    Moret-Fernández, D.; Latorre, B.; Lassabatere, L.; Di Prima, S.; Castellini, M.; Yilmaz, D.; Angulo-Jaramilo, R.

The soil sorptivity, S, and saturated hydraulic conductivity, Ks, can be estimated from the inverse analysis of a cumulative infiltration curve using the quasi-exact implicit (QEI) formulation or its corresponding 4-Terms (4T) approximation. Although these models consider the soil as homogeneous media, there is no information about how heterogeneous profiles can affect the inferred soil properties. This work analyzes the influence of layered soils on Ks and S estimates using QEI and 4T models, and designs a new procedure for treating infiltration curves measured on layered soil profiles. The Sequential Infiltration Analysis (SIA) method considers a sequence of increasing time series from the cumulative infiltration data to estimate Ks and S, and its corresponding RMSE as a function of the number of samples used. A procedure to estimate the thickness of the upper uniform soil layer from the estimated wetting front advance (WFA) is also reported. The SIA method was applied on: (i) synthetic homogeneous profiles of loam soil and six layered profiles involving a 1, 2 and 3 cm thickness loam layer over silty or sandy loam soils, respectively, (ii) stratified laboratory soil columns, and (iii) 20 experimental infiltrations performed in a semiarid region of North-Eastern Spain. Similar results were found between QEI and 4T models for all cases. Erroneous estimates of Ks and S were observed when the total infiltration time series was considered for the analysis, regardless of the presence of soil layering. In opposite, estimates improved when the SIA method was applied to the layered systems. The SIA method exploits the fact that the RMSE increases when the wetting front reaches the interface between the soil layers. Such increase allows: (i) detection of the soil heterogeneity, (ii) determination of the infiltration time, to, required for the wetting front to reach the lower layer, and, (iii) accurate estimates of the upper layer Ks and S along with its thickness. Laboratory experiments on layered soils and field measurements demonstrated that the SIA method could be satisfactorily applied on different curves with contrasting shapes and magnitudes. Although soil layering encountered on most field samplings restricted the treatment of the observed infiltrations to short-medium times, the SIA method allowed robust estimates of Ks and S. These results indicate that the proposed method is a promising tool for characterizing the hydraulic properties of layered and heterogeneous soil profiles.
