Spawning and Larval Rearing in Hybrid Striped Bass (Morone chrysops ♀ X Morone saxatilis ♂) in Turkey

Guner, Yusuf; Kizak, Volkan; Altunok, Muhammet; Celik, Ihsan

This study is the first spawning and larval rearing of the F-1 hybrid striped bass (Morone chrysops female X M. saxatilis male) grown in culture conditions in Turkey. The F-1 Hybrid striped bass was fed with commercial pellet feed in floating net cages. They reached the sexual maturity at two years old. Female brood fish were given 30 mu g/kg of LHRH-a (Lutenizing hormone releasing hormone analogue) and male fish given 10 mu g/kg; of CHP(Dried carp pituitary), 4 mg/kg was used for the female and half that amount for the male for induced breeding. The fish injected LHRHa were successfully induced to both strip spawning and tank spawning by a single hormone injection. However, fish injected CPH and saline, untreated control did not spawn. Latency period was found to be 20.41 +/- 0.91 hour. In term of fertilization rate, larval deformity rate no significant difference was found between tank spawning and strip spawning (p>0.05). Fertilization rates ranged from 56.3% to 85.4% and hatch rates were between 0.99% and 46.88%, whereas the level of larval deformities ranged between 81.25% and 100%. Hatched larvae were placed into 400-liter rectangular tanks with water discharge of 4 liter/minute at a stocking density of 30 larvae/liter. After larvae hatching, they were given artemia nauplii for 5-19 days. Trout granule feed (300-500 micron) also started to be given in addition to artemia nauplii afterwards. Granule feed was started eating completely 24 th days. The larvae obtained from a single hatch in the production season reached 2.87 cm length in 32 days with 2.7% survival rate. Of this study was terminated due to white spot disease (Ichtyophthirius multifilis) arising from sudden change in water temperature. The F-2 progenies were not deemed to be available for aquaculture due to poor fertilization, poor hatching, low survival rate and high deformity rate. For the progenies of the hybrid striped bass that grow in natural water, it was suggested that their ecology in waters in Turkey be first studied and brought into aquaculture afterwards.

Korkmaz, Sengul Asian; Karadag, Ahmet; Aydin, Ali; Yerli, Yusuf; Soylu, Mustafa Serkan

Five new complexes of composition [Ni(N-bishydeten)Pt(CN)(4)] (1), [Ni(N-bishydeten)(2)][Pt(CN)(4)] (2), [Cu (N-bishydeten)Pt(CN)(4)] (3), [Cu(N-bishydeten)(2)][Pt(CN)(4)] (4) and [Cd(N-bishydeten)(2)][Pt(CN)(4)] (5) (N-bishydeten = N,N-bis(2-hydroxyethyl)-ethylenediamine) have been obtained and characterized by IR and EPR spectroscopies, elemental and thermal analysis and magnetic measurements. X-ray studies of 2 and 3 exhibited different structures and two different coordination modes of N-bishydeten ancillary ligand with a ratio of M:L. While N-bishydeten acts as a tetradentate with N', N', O and O' atoms in a molar ratio of 1:1 formed 2,2-CT-type zigzag chain for 3, it behaves tridentate ligand by ending up with six coordinated Ni-II center in a complex salt by using 1:2 M ratio for 2. None of the 1 and 3 complexes produced in this study showed sufficient anticancer activity but all did exhibit apoptotic properties and one of them also performed low cytotoxic activity against cell tested. Changes in nitric oxide levels and erythrocyte arginase activities in patient with Down syndrome

Ozbey, Ulku; Seyran, Ayse; Erisir, Mine; Benzer, Fulya; Savucu, Yuksel

Down syndrome is a chromosome abnormality with specific clinical symptoms and mental retardation caused by trisomy of chromosome 21. There is evidence that oxygen free radicals play an important role in the pathophysiology of many neuropsychiatric disorders. Although, it has not been investigated yet, several recent studies proposed that nitric oxide related to oxidative stress and arginase may have a pathophysiological role in mental retardation. The purpose of this study was to determine plasma nitric oxide levels and erythrocyte arginase activities in patients with Down syndrome and in healthy children. Plasma nitric oxide levels and erythrocyte arginase activity were measured in 20 patients with Down syndrome (median age: 15.6 +/- 7.5) and 20 age-matched healthy subjects (median age: 16.2 +/- 7.4). Significant differences were found between erythrocyte arginase activity of the patients with Down syndrome (35.2 +/- 2.7 U/g hemoglobin (Hb)) and those of healthy children (43.5 +/- 2.9 U/g Hb), (P < 0.05). But we have not found any statistical difference between persons with Down syndrome and control group for nitric oxide (P > 0.05). It was concluded that the level of erythrocyte arginase activity could be considered as a useful marker for assessing the level of mental retardation in children with Down syndrome.

ESTIMATION OF THE POPULATION SIZE OF CAPOETA UMBLA (HECKEL, 1843) IN THE LAKE HAZAR (ELAZIG) BY REMOVAL METHOD

    Yüksel, F.; Alp, A.; Gündüz, F.; Çoban, M. Zulfu; Demirol, F.

Fish populations in smaller aquatic environments are affected by the fishing pressure at a much faster pace. Often, compensation of this effect either takes a long time or is not possible at all. For the purposes of establishing fishing regulations in these water environments, the removal method is able to provide results quickly and almost accurately. The Lake Hazar, which is the environment of our study, is 8,600 hectares in size, and is a natural lake where Capoeta umbla species is caught commercially using gill nets. In this study, stock size of Capoeta umbla in the Lake Hazar (Elazig) was estimated using removal method. In addition, the power of fishing and the status of fishing yield in the lake were evaluated. In our research, it was determined that seven commercial fishing boats caught 25,721 kg of fish in the lake in a fishing season. CPUE was calculated as 1.55 kg (kg/day/a gill net). As a result of the regression analysis, the amount of stock in the lake was the estimated to be 37,781 kg (+/- 14,001 kg). When cumulative catch was formulated as recommended by Ricker (1975), the population size was estimated to be 38,649 kg (+/- 15,007 kg). As a result of our study, the use of removal method was found to be appropriate for small reservoirs with reduced population density due to strong fishing pressure.

Effect of Pomegranate Seed Extract on Free Radical Damage and Antioxidant Activity Under Cisplatin-Induced Oxidative Stress Conditions in Rabbit Testes

    Benzer, F.; Kandemir, F. M.; Yildirim, N. C.; Ozan, S. T.

The aim of this study is to evaluate the protective effect of pomegranate seed extract on cisplatin-induced oxidative stress in rabbits testes. Rabbits were divided into three groups; (i) being control group, (ii) cisplatin group and (iii) cisplatin + pomegranate seed extract group. The level of malondialdehyde and glutathione, the activity of catalase, glutathione peroxidase, were measured in testes tissue of rabbits. Malondialdehyde levels were increased depending on cisplatin. In the case of cisplatin + pomegranate seed extract, malondialdehyde levels were found to be lower than cisplatin group (p < 0.05). In cisplatin group, catalase, glutathione peroxidase activity and glutathione levels were decreased when compared to control but in cisplatin + pomegranate seed extract group, catalase, glutathione peroxidase activity and glutathione levels were found higher than cisplatin group (p < 0.05). The pomegranate seed extract supplementation may play a protective role or decrease the side effects of cisplatin-induced testes toxicity from cancer medicines.

Centaurea derderiifolia Wagenitz., Centaurea drabifolia Sm. subsp. floccosa (Boiss.) Wagenitz & Greuter and Centaurea kotschyi (Boiss. & Heldr.) Hayek var. floccosa (Boiss.) Wagenitz, which are endemic taxa from Turkey belonging to Centaurea sect. Cheirolepis, were subjected to morphological, karyological and palynological investigations. The somatic chromosome numbers, pollen morphology and detailed morphometric properties were examined. The somatic chromosome numbers were determined as 2n = 2x = 36 in C. derderiifolia, C. kotschyi var. floccosa, and C. drabifolia subsp. floccosa. Karyotype analysis indicated that chromosomes of these Centaurea taxa generally have median region (m), submedian region (sm) and rarely median point (M) karyotypes. The pollen morphology of these endemic taxa of the genus Centaurea was investigated by light and scanning electron microscopy. The pollen grains of the examined species are mainly isopolar and radially symmetric, subprolate, tricolporate. Tectum ornamentation is scabrate and scabrate-perforate, amb triangular. The pollen grains of all taxa studied are 3-zonocolporate. The colpus is usually long, acute at ends and with smooth membrane. Pollen grains belonging to Cheirolepis section reveal homogeneity in their characters.

A new chelating ligand, 5,5-dimethyl-2-(2-(3-nitrophenyl)hydrazono)cyclohexane-1,3-dione (L), and four transition metal complexes were synthesized. The ligand and its complexes were characterized by IR, H-1 and C-13-NMR spectra, elemental analyses, magnetic susceptibility, mass spectra, UV-Vis. and thermogravimetry-differential thermal analysis (TGA-DTA). The metal complexes showed different geometric structures. Antibacterial tests of the ligand and complexes were also done. The Zn(II) complexes showed antibacterial activity against Staphylococcus aureus ATCC-25923 at middle level. The optical properties of the ligand and its complexes were extensively investigated by optical measurements. The average transmittance value of the ligand was the highest value, while the average transmittance value of the [CuL2(NO3)(2)]center dot 6H(2)O complex was the lowest value. The optical band gap of the ligand, [(Zn)(2)L(OH)(4)]center dot 2DMF, [ZnL(NO3)(2)(H2O)(2)]center dot 5H(2)O, [CuL2(OAc)]center dot OAc center dot 3.5H(2)O and [CuL2(NO3)(2)]center dot 6H(2)O complexes were calculated and 3.121 eV, 2.180 eV, 1.551 eV, 2.489 eV and 1.556 eV values were found, respectively. The complexes decreased the optical band gap of the ligand. The optical band gap value of the [ZnL(NO3)(2)(H2O)(2)]center dot 5H(2)O complex is the lowest value. The optical band gap value of the ligand is the highest value. The complexes decrease the angle of incidence of the ligand, while complexes increase the angle of refraction of the ligand. The electric susceptibility of the ligand decreases with its complexes.

    Baskonus, Haci Mehmet; Bulut, Hasan

In this study, we investigate some new analytical solutions to the (1 + 1)-dimensional nonlinear Dispersive Modified Benjamin-Bona-Mahony equation and the (2 + 1)-dimensional cubic Klein-Gordon equation by using the generalized Kudryashov method. After we submitted the general properties of the generalized Kudryashov method in Section 2, we applied this method to these problems to obtain some new analytical solutions, such as rational function solutions, exponential function solutions and hyperbolic function solutions in Section 3. Afterwards, we draw two- and three-dimensional surfaces of analytical solutions by using Wolfram Mathematica 9.

The effects of sebum configuration on Demodex spp. density

Demirdag, Hatice Gamze; Özcan, Hamdi; Gürsoy, Sule; Beker Akbulut, Gulcin

Background/aim: Demodex spp. are ectoparasites living in the pilosebaceous units, which feed on the host's sebum and cellular proteins. The protective barrier of the skin consists of sebum secretion, moisture, and the acid mantle. In this study, we aimed to determine the effects of skin sebum, moisture, pH levels, and sebum configuration on Demodex spp. density Materials and methods: Forty-five patients who had demodicosis were enrolled in the study group, while the control group consisted of 40 subjects without demodicosis. Body fat percentage, serum triglyceride and cholesterol levels, skin sebum, moisture, and pH levels were measured. Demodex spp. density was determined with a standardized skin surface biopsy. Sebum samples were taken from the forehead and a high-performance thin-layer chromatography (HPTLC) method was performed on these samples. Subsequently, densitometric analyses were applied to the HPTLC plates. Results: Demodex spp. were found on the cheeks and lived in an alkali environment. Skin sebum and moisture levels were low in all groups. The skin pH levels and cholesterol ester in the sebum configuration were determined to be significantly higher in the group with demodicosis. Conclusion: We suggest that Demodex spp. may use cholesterol ester in the sebum as nutriment. In other words, cholesterol ester may be a suitable growth medium for the proliferation of Demodex spp.

Compensatory effects of curcumin on cisplatin-induced toxicity in rabbit testis

    Kandemir, F. M.; Benzer, F.; Yildirim, N. C.; Ozdemir, N.

Curcumin, a widely used spice and colouring agent in food, has been shown to possess potent antioxidant, antitumor promoting and anti-inflammatory properties in vitro and in vivo. The present study was designed to investigate the protective effects of curcumin on changes in the levels of lipid peroxidation and endogenous antioxidants induced by cisplatin (cis-diamminedichloroplatinum II, CDDP) in the testiclular tissue of rabbits. 18 healthy male New Zealand white rabbits were equally divided into three groups of six rabbits each, control, cisplatin, and cisplatin+curcumin. The degree of protection produced by cisplatin was evaluated by determining the level of malondialdehyde (MDA) and glutathione (GSH), the activity of catalase (CAT), glutathione peroxidase (GSH-Px), were estimated from testes homogenates. MDA levels were increased with cisplatin compared to control but in cisplatin+curcumin group, MDA levels were found to be lower than cisplatin group (p<0.05). The activity of CAT and GSH-P-X was decreased in cisplatin and cisplatin+curcumin groups compared to control (p<0.05). In the case of cisplatin+curcumin CAT and GSH-Px activity were increased compared to cisplatin group (p<0.05). GSH levels were decreased with cisplatin but administration cisplatin+curcumin increased the levels of GSH comared to cisplatin group (p<0.05). In the present study, co-administration of curcumin with cisplatin prevented the damage to testes induced by this drug and may be considered as a potentially useful candidate in the combination chemotherapy with cisplatin.

Gurger, Murat; Yilmaz, Erhan; Yilmaz, Seval; Once, Gokhan; Konuk, Mustafa; Kaya, Emre; Say, Yakup; Artas, Gokhan; Artas, Hakan

BackgroundThe positive effects of grape seed proanthocyanidin extract (GSPE) on bone health, which is a potent antioxidant, are known but its effects on fracture healing are not sufficiently covered in the literature. This study aims to investigate the effects of GSPE on fracture healing and biomechanics of healing bone.Materials and methodsSixty-four adult Wistar-Albino male rats were divided into 8 groups of 8 animals in each group. Osteotomy was performed to the right femurs of all groups except the negative control (G1) and positive control (G2) groups, and intramedullary Kirchner wire was used for fixation. GSPE was given to half of the rats (G2-G4-G6-G8) 100mg/kg/day by oral gavage. The rats were sacrificed on the tenth (G3-G4), twentieth (G5-G6), and thirtieth (G1-G2-G7-G8) days, respectively, and histopathological, radiological, and biomechanical examinations were performed.ResultsHistopathological examination of the specimens from the callus tissues revealed that bone healing was more prominent in the groups supplemented with GSPE (G4, G6, G8). There was a statistically significant improvement in radiological recovery scores and callus volumes in groups with GSPE. When biomechanical strengths were evaluated, it was found that GSPE increased bone strength not only in fracture groups but also in the positive control group (G2).ConclusionsAs a result, this study showed that GSPE, a potent anti-oxidant, had a positive effect on bone healing and improved mechanical strength of the healing bone.

The aim of this work was to determine the seasonal changes in the activities of selected biomarkers in Capoeta umbla (Heckel, 1843) caught from Uzuncayir Dam Lake (Tunceli, Turkey) and to evaluate the effects of environmental factors on these activities. Fish were sampled on seasonal basis, and superoxide dismutase, catalase, glutathione peroxidase activities and levels of glutathione and malondialdehyde in gills were determined. Significant variations of oxidative stress biomarkers were observed between seasons and sites. The results of this study show that seasonal variations of oxidative stress responses and lipid peroxidation in gills of C. umbla are sensitive to the contaminants present in water of Uzuncayir Dam Lake and selected parameters are in valuable biomarkers for monitoring of water systems, since they give an early warning signal of effects of xenobiotics on aquatic organisms at molecular levels which help to prevent their effects at organismal level. Li, Zixiang; Çil, Zeynel Abidin; Mete, Suleyman; Kucukkoc, Ibrahim

In recent years, the interests of disassembly line have increased owing to economic reasons and the increase of environmental awareness. Effective line can provide many advantages in terms of economic aspect and it facilitates competition the companies with others. This study contributes to the relevant literature by a branch, bound and remember algorithm for disassembly line balancing problem with AND/OR precedence. The proposed exact solution method employs the memory-based dominance rule to eliminate the reduplicated sub-problems by storing all the searched sub-problems and to utilise cyclic best-first search strategy to obtain high-quality complete solutions fast. In this paper, minimising the number of stations is taken as the performance measure. The proposed methodology is tested on a set of 260 instances and compared with the mathematical model using CPLEX solver and five well-known metaheuristics. Computational results show that the proposed method is capable of obtaining the optimal solutions for all the tested instances with less than 0.1 seconds on average. Additionally, comparative study demonstrates that the proposed method is the state-of-the-art algorithm and outperforms the CPLEX solver and metaheuristics in terms of both solution quality and search speed aspects.

A Novel Probabilistic Nuclei Segmentation Algorithm for H&E Stained Histopathological Tissue Images

Serin, Faruk; Erturkler, Metin; Gul, Mehmet

In this study, we propose a novel, fast and accurate segmentation algorithm to segment nuclei in H&E stained histopathological tissue images. The proposed algorithm does not require pre-processing, post-processing, and any manual parameter or threshold. The algorithm utilizes probabilistic and statistical properties of the pixels' color value in the images with RGB color space, and determines whether pixels are a part of any nuclei or not by using an automatically calculated threshold value. The algorithm provides time efficiency and reduced overall cost in the segmentation. Two more algorithms are also proposed to distinguish nuclei cluster from the other clusters obtained by K-means, and eliminate false positives in nuclei cluster, which are not nuclei. In order to compare and evaluate the performance of the proposed segmentation algorithm in terms of time and cost efficiency, K -Means is preferred because of its common usage. Expert evaluation is declared as ground truth for determining the accuracy of the results. The experiments are performed on 60 healthy and 60 damaged kidney, and 60 healthy and 60 damaged liver tissue images. The evaluations show that the proposed algorithm can effectively segment nuclei. The comparison results also demonstrate that the deviation between proposed algorithm and the expert is 2%, while the deviation between K -Means and expert is 5%.

ANTIOXIDANT RESPONSES OF RAINBOW TROUT (Oncorhynchus mykiss) GILLS AFTER EXPOSURE TO HYDROGEN PEROXIDE

Seker, Engin; Ispir, Unal; Yonar, Serpil Mise; Yonar, M. Enis; Turk, Cebrahil

We evaluated antioxidant responses of Oncorhynchus mykiss, experimentally exposed to different concentrations of hidrogen peroxide (H2O2). Different concentrations of H2O2 (50 and 100 ppm) were administrated to the fish for 30, 60 and 90 min. Samples of the gill were collected at the end of the experiment, and analysed for their oxidant antioxidant status, including the malondialdehyde (MDA) level, the catalase (CAT) and glutathione peroxidase (GSH-Px) activity as well as the reduced glutathione (GSH) concentration. Results obtained showed that H2O2 significantly increased MDA level. Compared to controls, GSH level and GSH-Px and CAT activities were significantly reduced in the fish that were exposed to different concentrations of H2O2. The current data suggest that H2O2 has an oxidative-stress-inducing potential in fish. The various parameters studied in this investigation can also be used as biomarkers of exposure to H2O2.
