The Effects of Commercial Diet Supplemented with Mannanoligosaccharide (MOS) and Vitamin B12 on the Growth and Body Composition of the Carp (Cyprinus carpio L. 1758)

    Atar, Hasan Huseyin; Ates, Mehmet

Four hundred carp fries (Cyprinus carpio L. 1758) with average initial weight of 1.1 +/- 0.05 g and total average length of 4.04 +/- 0.04 cm, was fed for a period of 90 days with the addition of commercial feed Mannanoligosaccharide (MOS) and vitamin B-12. The carps were divided into four different groups and fed through different feeds (Control; group A with MOS, group B with vitamin B-12, group C with a the combination of MOS + vitamin B-12). The effects of feeding through MOS and vitamin B-12 in different proportions on the parameters such as growth in view of live weight and total length, survival rate, feed conversion, condition factor and protein efficacy for the carp fries was studied. The best growth in view of live weight was maintained through MOS and vitamin B-12 combination in group C (4.51 +/- 0.13 g), while the best growth in terms of length was obtained through MOS in group A (6.68 +/- 0.08 cm). As a consequence, the growth parameters of the carp fries fed through commercial feeds with the addition of MOS and vitamin B-12 in different doses under empirical conditions shown affirmative effects on the survival rate, feed assessment, condition factor and protein efficacy.

The effect of adrenomedullin and cold stress on interleukin-6 levels in some rat tissues

    Yildirim, N. C.; Yurekli, M.

P>Stress known to stimulate sympathetic activity, as well as the hypothalamic-pituitary-adrenal axis (HPA), produces a significant increase in adrenomedullin (AdM) levels, suggesting a regulatory or protective role for AdM in countering HPA activation that follows a variety of stressors. Stressors can modulate the secretion of proinflammatory cytokines. Interleukin (IL)-6 is a potent activator of the HPA and appears to play a pathogenic role in conditions related to stress. In the present study, we investigated the administration of AdM on IL-6 levels in cold exposed rats. Male Wistar rats were divided into four groups as control, adrenomedullin treatment, cold stress and cold stress+adrenomedullin-treated groups. In the adrenomedullin-treated group, animals received intraperitoneal (i.p.) injection of adrenomedullin (2000 ng/kg body weight) once a day for a week. For the cold stress exposure the rats were kept in separate cages at 10 degrees C for a week. Control group rats were kept in laboratory conditions. The concentration of IL-6 was determined using an enzyme-linked immunosorbent assay (ELISA) kit. When compared to control, IL-6 levels increased significantly in the cold stress- and adrenomedullin-treated groups (P < 0 center dot 05). Administration of AdM in addition to cold stress decreased IL-6 levels in lung and liver, but increased in brain and heart when compared to control (P < 0 center dot 05). The results suggest that cold stress may induce increase of rat proinflammatory cytokine IL-6 and adrenomedullin may play a regulatory or protective role for cold stress.

Adrenomedullin administration alters vascular endothelial growth factor levels in rats in cold stress

    Yildirim, Nuran Cikcikoglu; Yurekli, Muhittin

Background: Many endogenous peptides play important regulatory roles in angiogenesis by modulating endothelial cell behavior. Adrenomedullin (AdM) is one of such factors. Angiogenesis and vascular endothelial growth factor (VEGF) are indistinguishable. Exposure to cold environment stimulates capillary angiogenesis. Objectives: Examine the effect of the bioactive peptide AdM on VEGF levels in rat liver, lung, brain, and heart tissues after cold stress treatment. Methods: Male wistar rats were divided into four groups as control, AdM treatment, cold stress and AdM+cold stress treated groups. In AdM-treated group, animals received intraperitoneal injection of AdM (2000 ng/kg body weight) once a day during a week. For the cold stress exposure, the rats were kept in separate cages at 10 degrees C for a week. Results: The administration of AdM increased VEGF levels in all tissues in cold exposed rats. Conclusion: AdM may be a major regulatory factor in angiogenesis by modulating VEGF levels that is closely associated with cold exposure-related metabolic stimulation.

A comparison of the survival and growth performance in rainbow trout (Oncorhynchus mykiss) and brown trout (Salmo trutta fario) fry

Kizak, Volkan; Guner, Yusuf; Turel, Murat; Can, Erkan; Kayim, Murathan

The rainbow trout is an intensively cultured species because of its more cultivatable character than the brown trout. In this experiment, the brown trout culture did not expand due to low growth performance compared to that of the rainbow trout. This experiment was conducted in a commercial trout farm, Aegean Area (Turkey). Growth performances and survivals of rainbow and brown trouts from fry to fingerling were observed for 155 days. Growth performances, feed conversion rates and survivals were determined. The initial weights of the rainbow and brown trouts were 0.1 +/- 0.01 g. Final weights were 26.5 +/- 5.19 g and 12.97 +/- 2.74 g, respectively at the end of experiment. Survival and FCR of rainbow and brown trouts were 83.9 and 80%; 0.59 and 0.61 respectively. As a result, there is a similarity between these two trout species in point of survivals and FCR's although growth performance of the rainbow trout was significantly better than that of the brown trout.

First record of F2 hybrid striped bass (Morone chrysops ♀ x Morone saxatilis ♂ x Morone chrysops ♀ x Morone saxatilis ♂) in Kemer Dam Lake

    Kizak, Volkan; Güsner, Yusuf

The morphometric and meristic characters of 2 F-2 hybrid striped bass (M. chrysops female x M. saxatilis male x M. chrysops female x M. saxatilis male) from the family Moronidae are described. Two specimens were caught from Kemer Dam Lake. The body of the F-2 hybrid striped bass was elongated, moderately compressed, and scaly. Dorsal surface and sides were silver and black to olive-gray, and the abdomen was white in color. Four or 5 longitudinal broken stripes ran above the lateral line to the caudal fin. The stripes were less visible behind the pectoral fins and below the lateral line. The bodies of the F-2 specimens were deeper than 1/4 the fork length. The 2 dorsal fins were separated entirely. The first dorsal fin had 8-9 spines, and the second dorsal fin had a spine and 13-14 soft rays. The caudal fin was slightly forked. The anal fin had 3 spines with 12-14 soft rays. One tooth patch was present on the anterior of the tongue. According to our results, the fish were F-2 hybrid striped bass offspring of F-1 hybrid striped bass, which can reproduce naturally in Turkey. Morphological criteria showed that the F-2 hybrids tended to resemble white bass (M. chrysops).

    Ince, Muharrem; Yaman, Mehmet

The concentrations and varieties of benzo[a]pyrene were determined in Turkish foodstuffs including grilled meat and chicken samples by high performance liquid chromatography-mass spectrometry (HPLC-MS). The parameters that are thought to affect the response in the HPLC-MS analysis were optimized. The optimized conditions were found to be 0.75 mL min(-1) for flow rate of mobile phase, 45 mu L for injection volume, 55 degrees C for column temperature and 140 V for fragmentor potential. The optimized method was applied for the determination of benzo[a]pyrene concentrations in grilled meat and chicken foods. The obtained concentrations were found in the range of 2.7-8.3 mu g kg(-1) for the normal grilled meat and chicken samples and 72-138 mu g kg(-1) for over-grilled samples. The results showed that the concentrations of benzo[a]pyrene were significantly dependent on the kind of meat and the cooking time and source. The limits of detection and quantitation were found to be 0.7 and 2.33 mu g L-1, respectively.

Storm water retention and actual evapotranspiration performances of experimental green roofs in French oceanic climate

    Yilmaz, D.; Sabre, M.; Lassabatère, L.; Dal, M.; Rodriguez, F.

Green roofs are promising urban management tools from the standpoint of both rainwater management and microclimatology. They are considered as a storm water mitigation technique and may also favour evapotranspiration fluxes, which can be beneficial for urban comfort during summer periods. In France, however, water retention performance of green roofs remains unknown, and published values are often unsuitable. Six experimental roofs, including two thicknesses of growing media, three types of vegetation cover and bare surfaces, were monitored for two years in Nantes and compared to an experimental gravel flat roof. The thickest media combined with the most densely vegetated cover yields the best results in terms of storm water mitigation and actual evapotranspiration. In winter, the rainwater retention performance is clearly dependent on the type of experimental roof vegetation. This kind of experimental set-up is well suited to assisting urban planners design tools for storm water mitigation in buildings.

Benzer, Fulya; Kandemir, Fatih Mehmet; Ceribasi, Songul; Ozkaraca, Mustafa; Yildirim, Nuran Cikcikoglu; Ozan, Sema Temizer

Cisplatin (CP) is one of the most effective cancer chemotherapeutic agent used against various solid tumors. This study was designed to investigate the antioxidant effects of Grape Seed Extract (GSE) on CP induced oxidative damage in rabbit kidney. The material of study was consist of 18 male New Zealand rabbit. Animals were divided into 3 groups. The first group was administered a single intraperitoneal (i.p.) close of 0.9% saline. The second group of rabbits was treated with CP (a single i.p. dose of 5 mg kg(-1) body weight); The third group of rabbits was treated with GSE by gavage at the dose of 250 mg kg(-1) body weight for 6 consecutive days before and 6 consecutive days after a single i.p. CP injection. As a result of CP administration, kidney function tests; urea, BUN (Blood Urea Nitrogen) and creatinine levels were increased (p<0.001). It was observed that CP increase malondialdehide (MDA) levels, the most important indicator of oxidative damage and decreased the activity of catalasc (CAT). Administration of CP did not caused statically a change in glutathione (GSH) levels and glutathione peroxidase (GSH-P-x) activity (p>0.05). GSE treatment has high levels of MDA to compared to normal levels (p<0.001) but not an increase in CAT (p<0.05). In CP treatment group, severe acute tubular necrosis was seen. It was determined that in CP+GSE group mild degenerative changes were received instead of acute tubular necrosis. GSE has been found to be partially protective effect against CP induced biochemical and histopathological changes in rabbit kidney.

Estimation of Confidence Regions and Severity of Undefined Faults in Driving using Synthetic Disturbance Signals

    Yakut, O.; Eren, H.; Kaya, M.; Oksuztepe, E.; Polat, M.; Omac, Z.; Kurum, H.; Celenk, M.

In this study, we aim to determine fault severity in an electric car that may be caused by yawing due to such disturbances as non-uniform road pavements, in-wheel bearing clearance, suspension system, driver under Influence of alcohol (DUI), and tire deformation. The major research contribution herein is to alert drivers about an unforeseen situation on steering wheel whether it refers to a severe fault or contemporary states. In a sense, the undertaken study serves as a state of the art driving assistant system operating under unsteady conditions. We determine the fault severity of the system via classifying it into specified confidence regions by estimating the deviation from a monotonous straight route for any unstable situation. In this way, the proposed system informs driver to gain an insight about the severity level of arising problematic scenario. In order to realize the classification of confidence regions, we initially obtain the overall dynamic model of the system. Then, disturbance functions with different amplitudes and frequencies are characterized and included in the dynamic system specification. Here, the confidence regions have been constructed as to respective fault severity level of the car through the system response. Trajectory of vehicle in dynamic driving conditions considering these perturbations and noises are interrelated through the Kalman filtering to predict deviations from the desired trajectory and the prediction error. In simulation scenarios, Dynamic Time Warping (DTW) is employed to obtain deviation from ground truth under different noise functions, and results are sketched graphically assigning rate of fault severity into specified confidence regions. Initially, we have modeled the proposed system considering an electric car although the idea can readily be generalized for all cars with four tires. Presently, fault severity with classified confidence regions has been investigated under a simple car model.

Stripping voltammetric analysis of dicofol on graphene-modified glassy carbon electrode

Koçak, Berna; Er, Engin; Çelikkan, Huseyin

This study is related to the development of a highly sensitive and selective electrode for the electroanalysis of dicofol (DCF) by the modification of a glassy carbon electrode (GCE) with graphene (GR) nanosheets. GR was characterized by Fourier transform infrared spectroscopy (FTIR), atomic force microscopy (AFM), transmission electron microscopy (TEM), and electrochemical methods. The electrochemical reduction of DCF was observed on both bare GCE and GR-modified GCE (GRE). The electrochemical reaction of DCF on GRE was observed to be an adsorption controlled. Differential pulse stripping voltammetry was chosen in the electrochemical analysis of DCF, and two linear calibration ranges from 5.0 x 10(-8) to 5.0 x 10(-7) M and from 5.0 x 10(-7) to 4.2 x 10(-4) M were obtained with a low detection limit of 1.08 x 10(-7) M (N = 5) under the optimized experimental conditions. The electrode developed was successfully used to monitor dicofol in a series of spiked samples.

Effects of subchronic exposure to zinc nanoparticles on tissue accumulation, serum biochemistry, and histopathological changes in tilapia (Oreochromis niloticus)

Kaya, Hasan; Duysak, Muge; Akbulut, Mehmet; Yilmaz, Sevdan; Guerkan, Mert; Arslan, Zikri; Demir, Veysel; Ates, Mehmet

Zinc nanoparticles (ZnNPs) are among the least investigated NPs and thus their toxicological effects are not known. In this study, tilapia (Oreochromis niloticus) were exposed to 1 and 10 mg/L suspensions of small size (SS, 40-60 nm) and large size (LS, 80-100 nm) ZnNPs for 14 days under semi-static conditions. Total Zn levels in the intestine, liver, kidney, gill, muscle tissue, and brain were measured. Blood serum glucose (GLU), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and lactate dehydrogenase (LDH) were examined to elucidate the physiological disturbances induced by ZnNPs. Organ pathologies were examined for the gills, liver, and kidney to identify injuries associated with exposure. Significant accumulation was observed in the order of intestine, liver, kidney, and gills. Zn levels exhibited time- and concentration-dependent increase in the organs. Accumulation in kidney was also dependent on particle size; NPs SS-ZnNPs were trapped more effectively than LS-ZnNPs. No significant accumulation occurred in the brain (p>0.05) while Zn levels in muscle tissue increased only marginally (p0.05). Significant disturbances were noted in serum GOT and LDH (p<0.05). The GPT levels fluctuated and were not statistically different from those of controls (p>0.05). Histopathological tubular deformations and mononuclear cell infiltrations were observed in kidney sections. In addition, an increase in melano-macrophage aggregation intensity was identified on the 7th day in treatments exposed to LS-ZnNPs. Mononuclear cell infiltrations were identified in liver sections for all treatments. Both ZnNPs caused basal hyperplasia in gill sections. Fusions appeared in the gills after the 7th day in fish treated with 10 mg/L suspensions of SS-ZnNPs. In addition, separations in the secondary lamella epithelia were observed. The results indicated that exposure to ZnNPs could lead to disturbances in blood biochemistry and cause histopathological injuries in the tissues of O. niloticus. (c) 2016 Wiley Periodicals, Inc. Acrylamide concentrations in grilled foodstuffs of Turkish kitchen by high performance liquid chromatography-mass spectrometry

Kaplan, Olcay; Kaya, Gokce; Ozcan, Cemile; Ince, Muharrem; Yaman, Mehmet

For over ten years, there has been a considerable interest in determination of acrylamide in foodstuffs. It was known that both protein-rich and carbohydrates-rich foods cooked at high-temperatures can cause acrylamide formation. However, carbohydrates-rich foods such as potato chips and biscuit samples have been the common studied foods compared with protein-rich foods such as meat samples. In this study, determination of acrylamide in these two group foods was examined using HPLC-MS. For this purpose, firstly, the parameters that are thought to affect the response in the HPLC-MS analysis were optimized. The optimized conditions were found to be 0.3 ml min(-1) for flow rate of mobile phase, 40 mu l for injection volume, 5 degrees C for column temperature and 70 V for fragmentor potential. The optimized method was applied for the determination of acrylamide levels in Turkish foodstuffs including grilled meat and chicken samples, potato chips, coffee and biscuit. The obtained concentrations for all studied foods were in the range of 20-250 mu g kg(-1). The results showed that acrylamide concentrations highly varied depending on the kind of food samples. Plasma nitric oxide concentrations and erythrocyte arginase activities in lambs with contagious ecthyma

    Kandemir, F. M.; Issi, M.; Benzer, F.; Gül, Y.; Basbug, O.; Özdemir, N.

The aim of the present study was to investigate the plasma nitric oxide concentrations and the erythrocyte arginase activities in lambs with contagious ecthyma, which frequently occurs in lambs and goat kids. The study was conducted on 10 Akkaraman, 1-2 month old lambs with contagious ecthyma (the clinical diagnosis was based on the presence of typical cauliflower-like proliferative lesions in lips, commissum labials, nostrils and udders) and on 10 apparently healthy lambs (controls) from the same flock. Plasma NO concentrations and erythrocyte arginase activities were determined on all animals using the Griess method and the thiosemicarbazide diacetylmonoxime urea (TDMU) method, respectively. Compared to the control group, the mean plasma NO concentration was significantly increased in the diseased group while the mean arginase activity was markedly depressed. Abnormally high NO concentrations and low enzyme activities were observed in 9 and 10 diseased lambs, respectively. Additionally, a negative correlation was evidenced between the 2 biochemical parameters. These results show that this viral infection is associated with some perturbations in the arginine metabolism, leading preferentially to the synthesis of NO, which is presumed to exhibit some antiviral properties.

Piezoelectric energy harvesters in bridge structures: Evaluation of optimal placement and energy harvesting

Sen, Bugra; Polat, Alper; Kapicioglu, Abdullah; Esen, Hikmet

In the present study, energy is harvested from vibrations in bridge structures caused by vehicle passages using piezoelectric energy harvesters (PEHs). In order to maximize the energy harvested, it was aimed to specify the optimal locations of PEHs. In the study, energy harvesting from vibrations in bridge structures was achieved using three different vehicle loads and three different piezoelectric materials. The project of the Ku & scedil;saray & imath;-2 bridge, located in Ku & scedil;saray & imath; village in Baskil, Elaz & imath;& gbreve;, was modelled in the ANSYS program using real measurements. The bridge was analyzed for modal, harmonic, and piezoelectric energy harvesting, respectively. The type with the highest energy production was determined using three different piezoelectric materials (PZT-4, PZT-5A, and PZT-8) for a total of 9 PEHs, and a total of 13 different analyses were performed under three different vehicle loads (60 kN, 90 kN, and 120 kN) to compare the results. The maximum total power obtained from all nine PEHs was 1.484 & micro;W under the 120 kN load with PZT-5A. The maximum power obtained from a single PEH (PEH-1) was 0.49 & micro;W. This power achieved will be adequate for wireless sensor systems used for monitoring the structural health of bridges, and considering that energy harvesting through vibration is independent of meteorological elements, unlike energy sources such as solar and wind, the environmental damage caused by waste batteries in existing systems will be prevented with the dissemination of these systems. With the development of these systems, the energy needs of services such as lighting and traffic signs can be supplied. The results provide an important foundation for the development of vibration-based energy harvesting systems in bridge structures. Thus, the study stands out as an innovative step toward sustainable energy generation in structural engineering.

Five-step sequential extractions were employed to fractionation of Ni, Cr and Cu in soil polluted by anthropogenic activities and determine the mobility of the metals. Twelve samples were collected on an agricultural area that was located near an airport and intercity roads in Elazig-Turkey. Exchangeable, organically bounded, carbonate bounded, adsorbed species on Fe and Mn oxides and residual species (except silicates) of Ni, Cr and Cu were extracted into solution by using CaCl2, Na4P2O7, Na(2)EDTA, NH2OH-HCl and HNO3-H2O2, respectively. Mobile metal concentrations in fractions and total recoverable in soils were determined by using inductively coupled plasma-optical emission spectrometry (ICP-OES). Total recoverable Ni, Cr and Cu concentrations were in the range of 40-119, 45-126 and 23-72mgkg(-1), respectively. It was observed that total concentrations of metals in some of the samples were higher than the permitted values. The sum of the mobile percentages of metals was found to be lower than 50%. The Ni, Cr and Cu percentages for exchangeable species are in the ranges of 0.18-1.64, 0.03-0.59 and 0.42-2.53%, respectively.
