Amino acids have an important biological role for prevention of cell damage during cryopreservation. The objective of this study is to determine the effects of cysteine on postthaw sperm motility, duration of sperm motility, DNA damage, and fertility in the common carp (Cyprinus carpio). Sperm collected from 10 individuals was cryopreserved in extenders containing different cysteine concentrations (2.5, 5, 10, and 20 mM). Semen samples diluted at the ratio of 1:9 by the extenders were subjected to cryopreservation. After dilution, the semen was aspirated into 0.25-mL straws; the straws were placed on the tray, frozen in nitrogen vapor, and plunged into liquid nitrogen. DNA damage was evaluated by comet assay after cryopreservation. Our results indicated that an increase in the concentration of cysteine caused a significant increase in the motility rate and duration of sperm in the common carp (C carpio; P < 0.05). Comparing all concentrations of cysteine, the best concentration of cysteine was 20 mM. Higher postthaw motility (76.00 +/- 1.00%) and fertilization (97.00 +/- 1.73%) rates were obtained with the extender at the concentration of 20 mM. Supplementation of the extender with cysteine was increased the fertilization and hatching rate and decreased DNA damage. Consequently, cysteine affected the motility, fertilization, and DNA damage positively, and extenders could be supplemented with cysteine. (C) 2015 Elsevier Inc. All rights reserved.

In present study, it was examined whether addition of different antioxidants to the cryopreservation extenders had an effect on semen post-thaw fertility and motility in rainbow trout (Oncorhynchus mykiss) and also it was investigated the sperm characteristics post-thaw sperm characteristics and fertility. The collected semen was pooled to minimize individual variation. Each pooled ejaculate was split into 12 equal aliquots and diluted with base extenders supplemented with the antioxidants, and a base extender with no additives (control). The pooled semen samples diluted at the ratio of 1:10 by the extenders were subjected to cryopreservation. Antioxidants were separately added to the extenders (one per experimental group): catalase (250 U/l), superoxide dismutase (250 U/l), peroxidase (250 U/l), oxidized glutathione (1.5 mmol/l), reduced glutathione (1.5 mmol/l), L-methionine (1.5 mmol/l), uric acid (0.25 mmol/l), L-ascorbic acid (0.5 mmol/l), alpha-tocopherol (2.0 mmol/l), beta-carotene (0.5 mmol/l) and camitine (0.5 mmol/l). After dilution the semen was aspirated into 0.25 ml straws, the straws were placed on the tray, frozen for 10 min, and plunged into liquid nitrogen. Our results indicated that the post-thaw motility rate increased in extenders supplemented with uric acid, L-methionine, SOD, L-carnitine, alpha-tocopherol and L-reduced glutathione (p < 0.05). The motility duration of frozen thawed semen increased in extenders supplemented with uric acid, L-methionine, SOD, alpha-tocopherol and L-reduced glutathione (p < 0.05). Fertilization rate and hatching rate of frozen-thawed semen was not affected by the tested antioxidants. Consequently, the tested antioxidants affected the motility parameters and cryopreservation extenders could be supplement with antioxidants. This study suggested usage of antioxidants in the cryopreservation of rainbow trout. (C) 2014 Elsevier Inc. All rights reserved.

- MakaleYalnız künye2014Scopus SJRQ3Use of Clove Oil and Eugenol to Anesthetize Fingerling Shabut Barbus grypus
Ögretmen, Fatih; Gölbasi, Selami; Inanan, Burak E.; Kizak, Volkan; Kayim, Murathan
In this study, the efficacy of four doses of clove oil and eugenol were compared to sedate fingerling Shabut Barbus grypus to various stages of sedation and recovery. The results from the present study indicated that effective concentrations were at 50L/L for eugenol (induction time: 58 8s; recovery time: 199 +/- 15s [mean +/- SD]) and clove oil (induction time: 76 +/- 6s; recovery time: 161 +/- 34s). The induction times decreased significantly with the increasing concentrations of clove oil and eugenol, while recovery times increased with increasing concentrations of anesthetic (P < 0.05). Exposure of fish to 75 or 100L/L clove oil or to 50, 75, or 100L/L eugenol resulted in mean induction times 1min. Fish did not become fully sedated within 3min when treated with 25L/L clove oil. Other concentrations of both anesthetics, fish reached to full induction at 3min and recovered from anesthesia 5min. Our results showed the clove oil and eugenol are effective anesthetics for fingerling Shabut when used at concentrations of 50-100L/L. Received March 25, 2013; accepted July 5, 2013

The probiotics are very important for aquaculture because of their role in enhancing the fish health status, improving disease resistance, growth performance, and body composition, reducing malformations, improving gut morphology and the overall microbial balance. The objective of the presently reported study was to investigate effects of kefir as a probiotic product on the digestive and hepatic enzyme activities, as well as glucose level of Coruh trout, Salmo coruhensis Turan, Kottelat et Engin, 2010. The experiment was performed with four treatments: control group (no kefir), D1, D2, and D3 (kefir-supplemented diet: 10, 20, and 40 g . kg(-1) fish body mass, respectively). The activity of: alanine aminotransferase (ALT), aspartate aminotranferase (AST), lactate dehydrogenase (LDH), lipase, amylase-as well as the glucose levels were determined after 4 months. The results indicated that glucose levels (P < 0.05) and digestive enzymes (lipase and amylase) in the blood serum decreased in fish fed diets containing kefir (P > 0.05). On the contrary, the activity of liver enzymes (ALT, AST, and LDH) increased insignificantly (P > 0.05). In conclusion, digestive and liver enzyme activities were affected by quantitative changes in dietary kefir insignificantly but further research is needed to establish whether the liver and digestive enzyme physiology of Coruh trout can be affected by kefir-supplemented diets in a long-term trial.

- MakaleYalnız künye2013Scopus SJRQ3Comparison of Growth Performance, Gonadal Structure and Erythrocyte Size in Triploid and Diploid Brown Trout (Salmo trutta fario L, 1758)
Kizak, Volkan; Güner, Yusuf; Türel, Murat; Kayim, Murathan
The aim of this study was to compare growth, gonadal structure and red blood cell sizes of triploid and diploid brown trout (Salmo trutta fario). Triploidy was induced by providing a heat shock treatment to the fertilized eggs. From the 18th month post-fertilization triploid (TBT) and control diploid (CBT) brown trout groups were investigated up to 32nd month. Survival, specific growth rate, feed conversion ratio, condition factor, relative growth rate, absolute growth rate, gonadosomatic index, hepatosomatic index and carcass yield were determined in both groups. It has been determined that the triploid fish showed greater live weight than diploid fish despite the difference was not significant (P>0.05). In triploid group triploidisation success was obtained as 95% and male/female ratio was found as 44.4-55.6%. The red blood cells were significantly larger in triploids than diploid ones (P<0.05). Regarding histological section of gonads, triploidy resulted in reduced gonadal development in female, while triploid males seem to exhibit normal gonadal development. Our results suggest that since triploid females could grow more than males, the growth performance studies on triploid brown trout should be performed with all-female stocks under fish farming practices especially for the production of large size trout.

Seasonal variations of proximate composition and fatty acid profiles of wild brown trout (Salmo trutta macrostigma Dumeril, 1858) caught from the Munzur River, Tunceli-Turkey were investigated in this study. It was determined that wild brown trout was high in mono-unsaturated, poly-unsaturated and saturated fatty acids, respectively. C18:1 omega-9 (oleic acid) was the most abundant fatty acid in all seasons ranging from 20.27 to 29.35 %. C22:6 omega-3 (docosahexaenoic acid-DHA), C18:3 omega-3c (cis-linolenic acid), C18:2 omega-6c (linoleic acid), and C20:5 omega-3 (eicosapentaenoic acid-EPA) were found to be predominant fatty acids in PUFAs. As a conclusion, wild brown trout living in Munzur river could be considered as an important protein and omega-3 fatty acid source according to the nutritional quality results evaluated in terms of proximate and fatty acid composition.

- MakaleYalnız künye2013Scopus SJRQ3The Anesthetic Effects of Clove Oil and 2-Phenoxyethanol on Rainbow Trout (Oncorhynchus mykiss) at Different Concentrations and Temperatures
Yildiz, M.; Kayim, M.; Akin, S.
In this study, anesthetic effects of five different concentrations of 2-phenoxyethanol (0.2, 0.3, 0.4, 0.5 and 0.6 ml/L) and clove oil (0.50, 0.75, 1.00, 1.25 and 1.50 ml/L) on rainbow trout (Oncorhynchus mykiss) were studied at temperatures 7, 13 and 18 degrees C. For this purpose, 900 fish (39.08 +/- 1.13 g and 15.48 +/- 0.21 cm) were used in the experiment. Induction time of 2-phenoxyethanol and clove oil varied between 1.05 and 3.36 min at all concentrations, except for 0.2 ml/L (for 2-phenoxyethanol only) and at every temperature application. Full recovery time occurred between 2.44 and 7.14 min for 2-phenoxyethanol and 3.23 - 6.11 min for clove oil. It was found that full recovery times significantly increased with increase in 2-phenoxyethanol concentrations (r(2)=0.81). The same increasing trend was observed in clove oil, but the increase was not strong compared to 2-phenoxyethanol (r(2)=0.21). On the other hand, full induction times of 2-Phenoxyethanol and clove oil significantly declined with the increase in concentrations (r(2)=0.74; r(2)=0.84 for 2-phenoxyethanol and clove oil, respectively). Based on the ideal induction (less than 3 min) and recovery (less than 5 min) time criteria, it can be suggested that the most appropriate concentrations for rainbow trout were 0.3, 0.4 and 0.5 ml/L for 2-phenoxyethanol and 0.50, 0.75 and 1.00 ml/L for clove oil.

The objective of this study was to determine the effects of different feeding protocols on compensatory growth, feeding rate, and feed conversion efficiency of Black Sea Trout Salmo trutta labrax. To our knowledge, no previous feeding experiments with Black Sea Trout have incorporated a fasting period to simulate the handling and acclimation conditions experienced by farmed fish. Fifteen fish per tank (10.69 +/- 0.06cm and 13.22 +/- 0.14g, mean +/- SE) were stocked into 40-L fiberglass tanks. Fish were fed with different fasting-feeding regimes for 95 d and were equally allotted to four treatments (T-cont: control; T5-10: 5 d fasting, 10days feeding; T10-10: 10 d fasting, 10 d feeding; and T15-10: 15 d fasting, 10 d feeding) with three replicates per treatment. The results indicated that length (P = 0.0005), weight (P = 0.000), condition factor (P = 0.013), and specific growth rate (P = 0.014) were significantly affected by the interaction between feeding and time. All fasting treatments showed partial compensation during refeeding. There was an increase in daily feeding rate and feed conversion efficiency in fasting treatments compared with the control treatment. In contrast, at the end of the experiment specific growth rate, condition factor, and body weight in fasting treatments were significantly lower compared with the control treatment. We concluded that there was partial compensation of growth with regular refeeding after periods of feed deprivation (e.g., 5, 10, or 15 d) over a long term and a shorter fasting period may be preferred in order to achieve compensatory growth.
